(1)将紫皮石斛碎成粉,得到粉状紫皮石斛;
(2)将西洋参碎成粉,得到粉状西洋参;

(4)将步骤(1)中的粉状紫皮石斛、步骤(2)中的粉状西洋参、步骤(3)中的粉状决明子以及枸杞子饮片、灵芝饮片和黄芪饮片置于容器中,加白酒(即粮食白酒),浸泡,过滤得药酒;
(5)将步骤(4)得到的药酒加入适量的白酒(即粮食白酒)、纯化水调配至所需酒精度,得到活性药酒;
(6)将步骤(5)中得到的活性药酒中添加辅料,精滤,制备紫皮石斛复方酒剂;
或将步骤(4)中得到的药酒浓缩、干燥、添加辅料,制备紫皮石斛复方膏剂或固体制剂。
所述的紫皮石斛复方制剂包括紫皮石斛复方酒剂、紫皮石斛复方膏剂、紫皮石斛复方固体制剂。
步骤(1)中,将紫皮石斛碎成粉,过30目筛,得到粉状紫皮石斛。该尺寸以下的粉状紫皮石斛适合药物有效成分的提取。

全部药材与粮食白酒的重量比为1:10~20,有利于将药物活性成分提取出来。
所述的提取为浸泡30天以上,浸泡期间循环为每周用循环泵循环2~3次,过滤,得活性药酒。
步骤(5)中,所述的酒精度为25°~52°,能有效抑菌,不利于细菌的生长。
步骤(6)中,所述的辅料为蜂蜜、冰糖、乳糖、蔗糖、葡萄糖、淀粉、糊精、木糖醇、甘露糖醇、麦芽糖醇、山梨糖醇等辅料中的一种或两种以上(包括两种),加入适宜的辅料混合均匀,精滤制得。

本发明复方紫皮石斛制剂的原料药中,紫皮石斛系兰科植物紫皮石斛的茎,它的药用始载于《神农本草经》,并被列为上品。祖国医学认为石斛味甘淡、微咸,性寒,入胃、肾经,无毒,具有生津益胃、清热养阴的功效。适用于热病后期阴津耗伤而虚热不退、烦躁口渴者。可治肝肾阴虚,为养阴清热,益胃生津之要药。李时珍在《本草纲目》中称它有“强阴益精,厚肠胃,补内绝不足,平胃气,长肌肉,定志除惊,轻身延年”的神功奇效。
历代医药学著作石斛的奇特功效有明确的记载,认为它是滋阴补虚的极品。现代医药学研究证明石斛中所含的活性成分石斛多糖,具有免疫双向调节作用,对免疫力低下者,可通过调节免疫系统功能抵御疾病的侵袭;对免疫功能亢进者,通过调节免疫系统功能,恢复其正常水平。西洋参味甘微苦,性凉,入心肺肾三经,益肺阴,清虚火,生津止渴,《本草纲目拾遗》谓其“补肺降火,生津液,除烦倦,虚而有火者相宜。”故凡阴虚火旺劳嗽之人,每用西洋参则气平火敛,咳嗽渐平。《神农本草》记载灵芝“可明目、补气、安精魂、强筋骨、好颜色,久食,轻身不老,延年神仙”。《本草纲目》称之为上上之药,其性温味淡,无毒,益心气,补中增智,久食轻身不老,延年益寿。枸杞子是茄科小灌木枸杞子的成熟子实,既可作为坚果食用,又是一味功效卓著的传统中药材,常常被当做滋补调养和抗衰老的良药。古今药理与临床研究均证明,枸杞子确有防病治病、延年益寿之功效。黄芪,性味甘,微温。归经:入肺、脾、肝肾经。有益气固表、敛汗固脱,利水消肿之功效,黄芪具有增强机体免疫功能、保肝、利尿、抗衰老、抗应激、降压和较广泛的抗菌作用。决明子味苦、甘而性凉,具有清热明目,润肠通便的功效。本复方制剂中紫皮石斛滋阴生津,配西洋参补气养阴,配灵芝补气安神,配枸杞子滋补肝肾,配黄芪益气固表,配决明子润肠通便,合而用之,既宜于精气不足之体,又宜于阴津亏损之人,全方补而不滞。

本实施例紫皮石斛复方酒剂,其原料药配比为:
主药:紫皮石斛30g,枸杞子(枸杞子饮片)15g,西洋参10g,灵芝(灵芝饮片)10g,黄芪(黄芪饮片)10g,决明子5g。
按照下述具体方式进行制备:
(1)紫皮石斛、西洋参、决明子粉碎成粗粉,过30目筛,备用,得到粉状紫皮石斛、粉状西洋参和粉状决明子;
(2)将粉状紫皮石斛、粉状西洋参、粉状决明子、枸杞子饮片、灵芝饮片和黄芪饮片置于容器中,加10倍量酒精度数为50°的粮食白酒浸渍提取30天,即粉状紫皮石斛、粉状西洋参、粉状决明子、枸杞子饮片、灵芝饮片和黄芪饮片总重量与粮食白酒的重量比为1:10,浸泡30天以上,浸泡期间每周用循环泵循环2~3次;

按照下述具体方式进行制备:
(1)紫皮石斛、西洋参、决明子粉碎成粗粉,过30目筛,备用,得到粉状紫皮石斛、粉状西洋参和粉状决明子;
(2)将粉状紫皮石斛、粉状西洋参、粉状决明子、枸杞子饮片、灵芝饮片和黄芪饮片置于容器中,加10倍量酒精度数为50°的粮食白酒浸渍提取30天,即粉状紫皮石斛、粉状西洋参、粉状决明子、枸杞子饮片、灵芝饮片和黄芪饮片总重量与粮食白酒的重量比为1:10,浸泡30天以上,浸泡期间每周用循环泵循环2~3次;

本实施例紫皮石斛复方片剂,其原料药配比为:
主药:紫皮石斛30g,枸杞子(枸杞子饮片)15g,西洋参10g,灵芝(灵芝饮片)10g,黄芪(黄芪饮片)10g,决明子5g;辅料:甘露糖醇20g,硬脂酸镁0.5g。
按照下述具体方式进行制备:
(1)紫皮石斛、西洋参、决明子粉碎成粗粉,过30目筛,备用,得到粉状紫皮石斛、粉状西洋参和粉状决明子;
(2)将粉状紫皮石斛、粉状西洋参、粉状决明子、枸杞子饮片、灵芝饮片和黄芪饮片置于容器中,加10倍量酒精度数为50°的粮食白酒浸渍提取30天,即粉状紫皮石斛、粉状西洋参、粉状决明子、枸杞子饮片、灵芝饮片和黄芪饮片总重量与粮食白酒的重量比为1:10,浸泡30天以上,浸泡期间每周用循环泵循环2~3次;
(3)采用板框压滤,安装过滤布(材料成分维纶,100目以上)进行过滤,得活性药酒;
(4)将步骤(3)所制得活性药酒于70℃~80℃减压浓缩,回收酒精,得浸膏,备用;
(5)将步骤(4)所得浸膏经喷雾干燥得提取物细粉,过80目筛,备用;
(6)将步骤(5)所制得提取物细粉与过80目筛的甘露糖醇于沸腾制粒机上沸腾制粒,20目筛网整粒,得颗粒,备用;
(7)将步骤(6)所制得颗粒与过80目筛的硬脂酸镁混合均匀,压片,包装,即得紫皮石斛复方片剂。
本实施例紫皮石斛复方片剂,成人每日服用两次,每次3片。
对比例1
本实施例紫皮石斛普通酒剂,其原料药配比为:
主药:紫皮石斛30g,西洋参15g,枸杞子(枸杞子饮片)10g。
制备方法同实施例1。
药效试验
经动物实验证实本品具有增强免疫力和缓解体力疲劳的功能。
一、增强免疫力功能试验
经口给予小鼠不同剂量的本产品1个月,结果显示,本产品具有升高氢化可的松模型小鼠外周血白细胞数,增加模型鼠胸腺、脾脏指数,增强巨噬细胞的吞噬功能,促进模型鼠细胞免疫功能。因此认为本产品具有增强免疫力的功能。
1材料和方法
1.1材料
1.1.1样品:实施例1制备的紫皮石斛复方酒剂;对比例1制备的紫皮石斛普通酒剂。酒剂浓缩20倍,酒精度调至原来的度数。
1.1.2实验动物:清洁级ICR小鼠180只,雌性,体重18~22g。由浙江省医学科学院实验动物中心提供,动物合格证号:SCXK(浙)2016-0010。
1.1.3主要试剂:氢化可的松(天津生物化学制药有限公司);PBS缓冲液(pH7.2~7.4);1.5%冰醋酸溶液;0.1%Na2CO3溶液;印度墨汁(上海长江日用粘合材料厂);绵羊血红细胞(SRBC,玉环县南方试剂厂)。
1.1.4主要仪器:分析天平(上海精密仪器仪表有限公司);UV-3000分光光度计(日本岛津);游标卡尺(精密度0.02mm);手术器械;计数器;显微镜;冰箱。
1.2方法
1.2.1对氢化可的松模型小鼠白细胞数和胸腺、脾脏指数的影响取健康小鼠60只,雌性,按体重随机分为6组,见表1。正常组,模型对照组小鼠每天灌胃给予37°白酒0.01mL·g-1,给药组按表1剂量分别灌胃给予紫皮石斛复方酒剂,即给药剂量(按浓缩前算)分别为5mL·kg-1体重、15mL·kg-1体重与50mL·kg-1体重,分别为低剂量给药组、中剂量给药组和高剂量给药组,每天1次,连续30天。采用对比例1制备的紫皮石斛普通酒剂以15mL·kg-1体重喂药,为对比例1组。除正常组外,其它各组于实验第18天开始皮下注射氢化可的松(40mg·kg-1)5次,隔天注射1次。末次给药1.5h后,小鼠眼眶采血,测定白细胞数,并用颈椎脱臼法处死动物,剖取胸腺、脾脏并称重量,计算脏器指数,再用t检验进行统计处理。
1.2.2对氢化可的松模型小鼠巨噬细胞吞噬功能的影响小鼠分组、给药及造模同实验1.2.1。末次给药1h后,各组小鼠尾静脉注射印度墨汁(用生理盐水稀释4倍)0.01mL·g-1,于注射后2min、10min时分别眼眶取血20uL,溶于2mL 0.1%(wt)碳酸钠水溶液中,摇匀,于600nm波长处比色,测定吸收度值,计算吞噬指数a。吞噬指数a=(logA1-logA2)÷(t1-t2)。
1.2.3对绵羊红细胞(SRBC)诱导小鼠足跖肿胀度的影响小鼠分组及给药同实验1.2.1。每天1次,连续30天。30天后,除正常组外,腹腔注射2%(wt)SRBC,每只小鼠注射0.2ml(约1×108个SRBC)。免疫4天后,测量左后足跖部厚度或肿胀度,然后在测量部位皮下注射20%(V/V)SRBC,每只鼠20ul(约1×108个SRBC),注射后24h测定左后足跖部厚度或肿胀度,同一部位测定三次,取平均值。
2结果
2.1对小鼠白细胞数及胸腺、脾脏指数的影响
结果如表1所示,模型对照组小鼠的白细胞数、胸腺/脾脏指数与正常对照组相比较,有显著的统计学差异(P<0.01),造模成功。与模型对照组相比较,紫皮石斛复方酒各剂量组均能有效抵制氢化可的松所引起的外周血白细胞数的急剧下降,减轻氢化可的松应用的副作用,并能有效抑制氢化可的松引起的免疫器官分泌淋巴细胞的减少;紫皮石斛复方酒中、高剂量组能够较明显地提升外周血白细胞的效应,促进免疫器官的增殖(P<0.05,P<0.01)。为平均值±标准差。
注:与正常组比较,##:P<0.01;与模型组比较,*:P<0.05。
2.2对氢化可的松模型小鼠碳廓清指数的影响
结果如表2所示,各给药组小鼠的巨噬细胞的吞噬指数a与正常对照组相比,均无统计学差异(P>0.05)。与模型对照组相比,紫皮石斛复方酒中、高剂量组均能有效抑制氢化可的松加入后引起的小鼠碳粒廓清速度减慢,明显提高吞噬速率a,增加小鼠碳粒廓清能力,且增强作用相近(P<0.05)。
注:与正常组比较,##:P<0.01;与模型组比较,*:P<0.05。
2.3对绵羊红细胞(SRBC)诱导小鼠足跖肿胀度的影响
结果如表3所示,模型对照组的足跖肿胀度与正常对照组相比,有显著的统计学差异(P<0.01)。与模型对照组相比,紫皮石斛复方酒中、高剂量组均能显著减少绵羊红细胞致敏引起的足跖肿胀程度,有效促进细胞免疫(P<0.05,P<0.01)。

3、结论
本研究提示紫皮石斛复方酒剂具有升高氢化可的松模型小鼠外周血白细胞数,增加模型鼠胸腺、脾脏指数,增强巨噬细胞的吞噬功能,促进模型鼠细胞免疫功能,表明该制剂具有免疫增强作用,并且其效果要明显优于对比例1组。可见,本发明中,紫皮石斛滋阴生津,配西洋参补气养阴,配灵芝补气安神,配枸杞子滋补肝肾,配黄芪益气固表,配决明子润肠通便,六种成分合而用之能够产生协同作用,具有很好的免疫增强作用,更好地增强机体免疫功能。
Claims (10)
3.根据权利要求1或2所述的紫皮石斛复方制剂的制备方法,其特征在于,包括以下步骤:
(1)将紫皮石斛碎成粉,得到粉状紫皮石斛;
(2)将西洋参碎成粉,得到粉状西洋参;
(3)将决明子碎成粉,得到粉状决明子;
(4)将步骤(1)中的粉状紫皮石斛、步骤(2)中的粉状西洋参、步骤(3)中的粉状决明子以及枸杞子、灵芝和黄芪置于容器中,作为药材,然后加白酒,浸泡,过滤得药酒;
(5)将步骤(4)得到的药酒加入白酒、水调配至所需酒精度,得到活性药酒;
(6)将步骤(5)中得到的活性药酒中选择性添加辅料,之后精滤,制备紫皮石斛复方酒剂;
或将步骤(4)中得到的药酒浓缩、干燥、添加辅料,制备紫皮石斛复方膏剂或紫皮石斛复方固体制剂;
所述的紫皮石斛复方制剂包括紫皮石斛复方酒剂、紫皮石斛复方膏剂、紫皮石斛复方固体制剂。
4.根据权利要求3所述的紫皮石斛复方制剂的制备方法,其特征在于,步骤(1)中,将紫皮石斛碎成粉,过30目筛,得到粉状紫皮石斛;
步骤(2)中,将西洋参碎成粉,过30目筛,得到粉状西洋参;
步骤(3)中,将决明子碎成粉,过30目筛,得到粉状决明子。
5.根据权利要求3所述的紫皮石斛复方制剂的制备方法,其特征在于,步骤(4)中,所述的白酒的浓度为40°~60°。
6.根据权利要求3所述的紫皮石斛复方制剂的制备方法,其特征在于,步骤(4)中,所述的药材与粮食白酒的重量比为1:10~20。
7.根据权利要求3所述的紫皮石斛复方制剂的制备方法,其特征在于,步骤(5)中,所述的所需酒精度25°~52°。
8.根据权利要求3所述的紫皮石斛复方制剂的制备方法,其特征在于,步骤(6)中,所述的辅料为蜂蜜、冰糖、乳糖、蔗糖、葡萄糖、淀粉、糊精、木糖醇、甘露糖醇、麦芽糖醇、山梨糖醇中的一种或两种以上。
9.根据权利要求1或2所述的紫皮石斛复方制剂在制备增强免疫力和缓解体力疲劳的保健食品中的应用。
10.根据权利要求1或2所述的紫皮石斛复方制剂在制备增强免疫力和缓解体力疲劳的药物中的应用。
