经查询专利文献已知发明1:公开日:2015年1月28日申请日期:2014年10月21日优先权日:2014年10月21日,对黄精进行特殊处理并与木糖醇、柠檬酸、花生衣提取物、瓜尔豆胶、CMC、乳酸链球菌素等原辅料进行合理调配生产黄精保健饮料。
已知发明2:中国专利申请号CN201610741361.0涉及一种黄精植物饮料及其制备方法,该饮料由黄精、玉竹和铁皮石斛构成,经提取、酶解后加入活性炭过滤、浓缩,然后加入低聚糖混合液,灌装、灭菌、检验、包装即得。
已知发明3:一种黄精饮料,该黄精饮料主要由以下重量比组份组成:黄精1.6~2%、麦芽糖醇5~6%、罗汉果甜苷0.1~0.15%、柠檬酸0.02~0.03%、其余为水。后加香精调配制得产品。

第三、大多数饮料中加入了低聚糖,糖精,甘蔗等辅料,不适宜糖尿病及肥胖人群饮用。
第四、普遍加入防腐剂和香精,长期饮用不利于饮用者的身体健康。
从已知的技术文献中尚未发现利用原产地泰山黄精制备的黄精饮料。
发明内容
为克服上述不足,本发明提供一种黄精饮料及其制备方法,旨在实现原生态,零添加和利用传统沸煮萃取方法,结合现代灭菌工艺,实现规模化流水线生产制备得到。
本发明的技术方案是:一种黄精饮料,其特征在于由黄精、大枣、山楂、甘草、灰树花和水组成,其所述的组份重量百分比为:黄精1.5-2.0%、大枣0.5-1.0%,山楂0.35-0.5%,甘草0.15-0.20%,灰树花0.2-0.3%,水96.00-97.30%。

步骤4:将灌装后的黄精饮料液体送入灭菌釜加温加压灭菌,灭菌温度120℃,灭菌时间20分钟,即制得产品。
本发明所述的步骤2还包括二次萃取,再次向微波萃取罐加入60℃温水,然后加热至95℃,微波萃取5分钟,冷却至常温,将两次萃取冷却后的液体经过滤机过滤后放入定容罐。
本发明将微波萃取罐加入60℃温水,然后将上述泰山酒制黄精、大枣、山楂三味中药分别装入过滤药袋中,放入微波萃取罐中浸泡1个小时,继续加温至90℃,微波萃取5分钟,然后停止加温,冷却至70-80℃后再加入甘草保温浸泡30分钟,微波萃取5分钟,再次降温至60-70℃加入灰树花保温浸泡30分钟,再次微波萃取5分钟。
本发明黄精可采用泰山黄精,泰山黄精为泰山四大名药之一,由于泰山地区特有的地理环境和气候条件,泰山黄精被历代名人誉为黄精中的上品,据传,杜甫喜欢种药,在登泰山时有幸寻觅到泰山四大名药之首的黄精(泰山四大草药包括:黄精、何首乌、紫草、四叶参),杜甫挖取黄精后,带着继续爬山,在山道上遇到一位仙风鹤骨的道长,道长见杜甫身怀黄精,遂与杜甫攀谈,经过一番交流后,了解到原来泰山黄精是道家辟谷、修炼、修道的“仙人饭”,泰山黄精为辟谷上药,峻补黄庭,调和五脏,坚强骨髓,同时泰山黄精能宁心安神、引火下行归于丹田和肾脏,辅助修炼。诗圣杜甫用“扫除白发黄精在,君看他年冰雪容”的佳句来赞美黄精。
黄精能作为泰山四大名药之首是有原因的,李时珍在《本草纲目》中记载,黄精有补肾虚、填精髓的作用,称黄精为“服食安药”。
《神仙芝草经》这样记述:“黄精宽中益气,使五脏调良,肌肉充盛,骨髓坚强,其力增倍,多年不老,颜色鲜明、发白更黑,齿落更生。”
历代医药验方和现代医学的研究都表明黄精营养价值极高,具有诸多对人体有益的功用。近代研究发现,泰山野生黄精为百合科植物,其地下根状茎入药,根茎结节状圆锥形,一端粗,另一端渐细,其关节处有茎痕,又有侧芽,形似鸡头,故名“鸡头黄精”,是药典规定中质量最上品的黄精品种,可谓是黄精中的“泰山”。
黄精为百合科黄精属多年生草本植物干燥根茎,既可以作为药,又可以做食物;黄精性平、味甘,具备补脾润肺、滋阴生津、补中益气;黄精根茎中含有黄精低聚糖、黄精多糖、黄酮类、醌类、黏液质、氨基酸,等多种人体必需的元素。黄精富含低聚糖、黄酮类等具有生物活性的化学成分;低聚糖有利于改善人体肠道生物环境,使有益菌群增殖,减少和抑制有害物质对人体的侵害;此外,黄精还含有较多的黄酮类化合物,黄酮类化合物对高血压引起的头痛、头晕等症状有明显疗效,黄酮类化合物具有较强的抗癌防癌作用,黄酮类化合物具有明显的消炎、抗溃疡作用,黄酮类化合物还具有降血糖、抗氧化、抗突变性、抗衰老、保护神经系统,改善记忆力等作用;同时黄精还具备降血压和防治动脉粥样硬化与肝脂肪浸润等药理功用;还具有显著的抗菌和抗病毒功用,可增强人体免疫力,增强体内超氧化物歧化酶的活性,可减速机体衰老;黄精还可以抑制杆菌及致病性皮肤真菌,以黄精为主治疗皮肤真菌手足癣和甲癣,白癣等多种癣症,疗效明显。
本发明采用的大枣为“泰山宁阳不老枣”,1.枣能提高人体免疫力,并可抑制癌细胞:药理研究发现,红枣能促进白细胞的生成,降低血清胆固醇,提高血清白蛋白,保护肝脏,红枣中还含有抑制癌细胞,甚至可使癌细胞向正常细胞转化的物质;
2.经常食用鲜枣的人很少患胆结石,这是因为鲜枣中丰富的维生素C,使体内多余的胆固醇转变为胆汁酸,胆固醇少了,结石形成的概率也就随之减少;
3.枣中富含钙和铁,它们对防治骨质疏松、贫血有重要作用,中老年人更年期经常会骨质疏松,正在生长发育高峰的青少年和女性容易发生贫血,大枣对他们会有十分理想的食疗作用,其效果通常是药物不能比拟的;
4.对病后体虚的人也有良好的滋补作用;
5.枣所含的芦丁,是一种使血管软化,从而使血压降低的物质,对高血压病有防治功效;
6.枣还可以抗过敏、除腥臭怪味、宁心安神、益智健脑、增强食欲。
本发明所述的山楂为蔷薇科植物山里红的干燥成熟果实。秋季果实成熟时采收,切片,干燥。即《中国药典》(2015年版,一部)收载的山楂。山楂的主要成分为黄酮类及有机酸类化合物。黄酮类化合物主要有牡荆素、槲皮素、槲皮苷、金丝桃苷、四羟基黄酮、葡萄糖苷和芦丁;有机酸主要有山楂酸、柠檬酸、熊果酸等,另外尚含有磷脂、维生素C、维生素B2等。
本发明所述的甘草功效:和中缓急,润肺,解毒,调和诸药。炙用,治脾胃虚弱,食少,腹痛便溏,劳倦发热,肺痿咳嗽,心悸,惊痫;生用,治咽喉肿痛,消化性溃疡,痈疽疮疡,解药毒及食物中毒。
甘草药理作用分析:
抗溃疡作用:甘草的主成分甘草甜素对由组胺及幽门结扎所形成的大鼠实验性溃疡亦有明显的保护作用。对心血管系统的影响:抗心律失常作用,降脂作用和抗动脉粥样硬化作用:甘草浸膏和甘草合剂口服后能覆盖发炎的咽部粘膜,缓和炎症对它的刺激,从而发挥镇咳作用。抗炎作用:其抗炎成分为甘草甜素和甘草次酸。
本发明所述的灰树花:又名栗蘑、贝叶多孔菌、莲花菌、云蕈属担子菌亚门。含有丰富的氨基酸、蛋白质等多种微量元素。具有松蕈样芳香,肉质柔嫩,味如鸡丝,脆似玉兰。栗蘑菇具有极高的医疗保健功能。据文献报导,它有利于大脑发育,抑制常见的高血压、糖尿病,抗衰老;
1、较高的锌含量有利大脑发育、保持视觉敏锐,促进伤口愈合。2、富含铁、铜和维他命C,它能预防贫血、坏血病、白癜风,防止动脉硬化和脑血栓的发生。
3、它的硒和铬含量较高,有保护肝脏、胰脏,预防肝硬化和糖尿病的作用;硒含量高使其还具有防治克山病、大骨节病和某些心脏病的功能。
4、它兼含钙和维生素D,两者配合,能有效地防治佝偻病。
5、高含量的维生素E和硒配合,使之能抗衰老、增强记忆力和灵敏度。
6、灰树花(栗蘑)还是引人注目的抗癌药源,一方面,较高的硒含量有抗御癌肿的作用,尤其是所含栗蘑多糖(Grifolan),以β-葡聚糖(glucan)为主,其中抗癌活性最强,带6条支链的β-(1,3)-葡聚糖占相当大的比重,据说比已经面市的香菇多糖(Lentinan,日本的PSK,)、云芝多糖(上海的PSP)等有更强的抗癌能力;同时它又是极好的免疫调节剂。
作为中药,灰树花(栗蘑)能益气健康,清热,渗湿。用于小便不利、水肿脚气、肝硬化、肝腹水、糖尿病、高血压,并对癌症有防治作用。(《食用菌栽培新技术》1997)
灰树花(栗蘑)周身是宝,素有“真菌之王”和“华北人参”的美誉。其所含的Fe、Zn、Ca、P及多种微量元素食之可避免多种营养缺乏症。VB1和Ve含量较其它菇类高10-20倍,此两种营养素能促进人体生长,增进食欲以及防止细胞膜的脂质过氧化而达到延缓衰老和增寿的功效;Vc含量是其同类的3-5倍,加之所富含的Fe、Cu可预防贫血、坏血病、白癜风,防止动脉硬化和脑血栓的发生,具有抑制高血压和肥胖病的功效。精氨酸、赖氨酸的含量比有“增智菇”之称的金针菇还要高,可促进儿童身体健康成长和智力发育;含量较高的谷氨酸是鲜味较高的氨基酸,天冬氨酸常做营养添加剂,它具有抗疲劳,保护人脑神经的作用;支链氨基酸(缬氨酸、亮氨酸、异亮氨酸)具有促氨保留和蛋白质合成、抑制蛋白质分解的作用。富含的膳食纤维是一般脱水蔬菜的3-5倍,食物中的纤维素具有预防肠道癌发生的作用。硒和铬含量较高,具有保护肝脏、胰脏、预防肝硬化和糖尿病的作用,特别富含的生物有机硒,在人体中吸收利用率高达97.5%,中国营养委员会将硒列为15种营养素之一,它可抑制高血压、肥胖病,预防动脉硬化、克山病和某些心脏病,尤其是能降低癌症的发病率。富含的真菌多糖(主要是β-D-葡聚糖)达8%以上,食之能提高身体免疫力,具有防癌、抗癌、防艾滋病之功效。
本发明的配伍机理,黄精作为君药主理降血压、降血糖、降血脂,提高身体免疫力,适应广大人群饮用。大枣和山楂作为臣药滋阴补阳,健胃消食,促进新陈代谢。灰树花作为佐药具有通经活络,益气安神,强筋键骨之功效。甘草作为使药调和诸药,具有补脾止咳润肺之功效。
采用本发明方法制得的黄精饮料,经消费者饮用证明:具有通经活络,益气安神,滋阴补阳,健胃消食,促进新陈代谢,增强身体免疫力,抗疲劳,强身健体的功效,特别适宜中老年人饮用,对于降血压、降血糖、降血脂有明显效果。
具体实施方式:
所述泰山酒制黄精的制备:将鲜泰山黄精清洗干净,经三蒸三晒晾干,然后用黄酒浸泡晾干切成薄片待用,以备所述制备黄精饮料。本发明采用微波萃取工艺,既降低萃取温度,又提高了萃取效率。微波萃取的机理分两方面,一方面微波辐射过程是高频电磁波穿透萃取介质,到达物料的内部维管束和腺胞系统。由于吸收微波能,细胞内部温度迅速上升,使其细胞内部压力超过细胞壁膨胀承受能力,细胞破裂。细胞内有效成分自由流出,在较低的温度条件下萃取介质捕获并溶解。通过进一步过滤和分离,便获得萃取物料。另一方面,微波所产生的电磁场加速被萃取部分成分向萃取溶剂界面扩散速率,用水作溶剂时,在微波场下,水分子高速转动成为激发态,这是一种高能量不稳定状态,或者水分子汽化,加强萃取组分的驱动力;或者水分子本身释放能量回到基态,所释放的能量传递给其他物质分子,加速其热运动,缩短萃取组分的分子由物料内部扩散到萃取溶剂界面的时间,从而使萃取速率提高数倍,同时还降低了萃取温度,最大限度保证萃取的质量。
实施例1
本实施例提供了本发明的黄精饮料的制备,具体为:
步骤1:按重量份称取各原料,所述原料为泰山酒制黄精20.00Kg、大枣10.00Kg、山楂5.00Kg、甘草2.00Kg、灰树花3Kg、水960Kg。
步骤2:将微波萃取罐加入60℃960Kg温水,然后将上述五味中药分别装入过滤药袋中,放入萃取罐中浸泡1个小时,继续加温至95℃,微波萃取5分钟,冷却待用。
步骤3:将冷却后的液体经过滤机过滤后放入定容罐混合。
步骤4:将定容罐中的药液加温至87℃,然后采用巴氏灭菌法灭菌进入灌装工艺装罐。
步骤5:将灌装后的黄精饮料液体送入灭菌釜加温加压灭菌,灭菌温度120℃,灭菌时间20分钟。然后装箱包装出厂。
实施例2
本实施例提供了本发明的黄精饮料的制备,具体为:
步骤1:将泰山酒制黄精、大枣、山楂、甘草、灰树花分别破碎粉磨至100目左右备用。
步骤2:按重量份称取各原料,所述原料为泰山酒制黄精15.000Kg、大枣5.00Kg、山楂3.50Kg、甘草1.50Kg、灰树花2.00Kg、水500Kg。
步骤3:将微波萃取罐加入60℃500Kg温水,然后将上述五味中药分别装入过滤药袋中,放入萃取罐中浸泡1个小时,加温至95℃,微波萃取7分钟,冷却。
步骤4:将冷却后的液体经过滤机过滤后放入定容罐。
步骤5:二次萃取,再次向微波萃取罐加入60℃473Kg温水,然后加热至95℃,微波萃取5分钟,冷却至常温。将冷却后的液体经过滤机过滤后再次放入定容罐。
步骤6:将两次放入定容罐中的液体搅拌混合,加温至87℃,然后采用巴氏灭菌法灭菌进入灌装工艺灌装。
步骤7:将灌装后的黄精饮料液体送入灭菌釜加温加压灭菌,灭菌温度120℃,灭菌时间20分钟。制得产品,然后装箱包装出厂。
最佳实施例
所述泰山酒制黄精的制备:将鲜泰山黄精清洗干净,经三蒸三晒晾干,然后用黄酒浸泡晾干切成薄片待用,以备所述制备黄精饮料。将大枣、山楂、甘草、灰树花分别切片备用。
步骤1:按重量份称取各原料,所述原料为泰山酒制黄精60.00Kg、大枣30.00Kg、山楂15.00Kg、甘草6.00Kg、灰树花9.00Kg、水2880Kg。
步骤2:将微波萃取罐加入2880Kg60℃温水,然后将上述泰山酒制黄精、大枣、山楂三味中药分别装入过滤药袋中,放入微波萃取罐中浸泡1个小时,继续加温至90℃,微波萃取5分钟,然后停止加温,冷却至70-80℃后再加入甘草保温浸泡30分钟,微波萃取5分钟,再次降温至60-70℃加入灰树花保温浸泡30分钟,再次微波萃取5分钟。
步骤3:将冷却后的液体经过滤机过滤后放入定容罐混合。
步骤4:将定容罐中的药液加温至87℃,然后采用巴氏灭菌法灭菌进入灌装工艺灌装。
步骤5:将灌装后的泰山黄精露送入灭菌釜加温加压灭菌,灭菌温度120℃,灭菌时间20分钟。然后制得产品装箱包装出厂。
本发明具体实验数据如下(以下实验所用到的实验原理数据均采用最佳实施例数据):
目的优选黄精多糖的微波提取工艺,为产业化生产提供依据。
方法以黄精多糖得率为指标,以提取时间、微波功率、料液比为提取因素,利用正交试验设计对微波法提取黄精多糖的工艺进行优化研究。并与热水浸提法进行黄精多糖得率作比较。
结果微波法适宜的提取工艺条件是300W的功率下,以1∶30的固液比提取5min,此条件下粗多糖得率为28.84%。
结论微波法提取黄精多糖,具有提取效率高、用时少、耗能低的特点,优于传统热水浸提法,是一种值得大力推广的提取方式。
多糖提取实验及结果
1.微波法提取黄精多糖
1.1将干燥的黄精放入粉碎机中粉碎,并过16目筛
1.2精确称取粉碎的黄精粉末10g,9份,分别倒入9个烧杯中,用记号笔将其标号1~9.分别加入100ml、200ml、300ml、200ml、300ml、100ml、300ml、100ml、200ml蒸馏水浸泡12h。
1.3正交试验设计
按照下表组合进行处理,基于已有文献确定微波提取法中的主要影响因素,优化工艺操作条件以微波功率(A)、提取时间(B)、料液比(C)为因素,作如下3因素、3水平的正交试验。选择正交表L9(33),因素-水平设计如表1。

1.4将烧杯依次按照正交试验顺序进行,处理完毕后,倒入量筒中记录原提取液。
1.5将原提取液倒入实验锅中浓缩,浓缩到原体积的1/4
1.6按照浓缩液的体积,依次加入浓缩液体积两倍的无水乙醇,加入时用玻璃棒迅速搅拌,用保鲜膜封口,静置12h
1.7将上清液倒去,将沉淀物滤纸过滤,放入烘箱中烘焙12h,取出称重,计算多糖含量

本实验初步优选得到从黄精提取粗多糖的最佳工艺条件为:微波功率300w,处理时间5min,料液比1∶20,该条件下黄精粗多糖得率可达到28.84%。
2.热水浸提法提取黄精多糖
2.1精确称取粉碎的黄精粉末10g,按照1:20料液比加入200ml蒸馏水浸取,在水浴锅浸提2h后,用纱布过滤除去残渣,取上清液
2.2将原提取液倒入实验锅中浓缩,浓缩到原体积的1/4
3.2.3按照浓缩液的体积,依次加入浓缩液体积两倍的无水乙醇,加入时用玻璃棒迅速搅拌,用保鲜膜封口,静置12h
2.4将上清液倒去,将沉淀物滤纸过滤,放入烘箱中烘焙12h,取出称重,计算粗多糖含量
热水浸提多糖重量2.3987g,黄精粗多糖得率为24%。
1、采用本发明最佳实施例方法制得的泰山黄精饮料,经100人品尝,结果95人表示口感细腻,酸甜适宜,非常喜欢;3人表示口感一般;2人表示口感不好,即参与试验的人员中有95.00%的认为本发明口感非常好。
2、采用本发明配方及工艺制得的泰山黄精饮料功效大为提高。
动物实验数据如下:
黄精饮料对小鼠免疫功能的影响试验
1黄精饮料给小鼠的灌胃试验
取健康小鼠30只,随机分为3组,每组10只,分别作为阴性对照组、低剂量组、高剂量组,称重并标记,按照0.27ml/10g的剂量对小鼠给予黄精饮料灌胃。阴性对照组用生理盐水进行灌胃,高剂量组用浓缩1倍后的黄精露进行灌胃,低剂量组直接用黄精饮料进行灌胃。依上述剂量灌胃30天。
2.黄精饮料对小鼠的免疫影响的试验结果
2.1巨噬细胞的吞噬试验
实验结束前1d各组小鼠均给予腹腔注射1%淀粉50ml·kg-1,以诱导巨噬细胞游出。末次给药24h后腹腔注射2%鸡红细胞悬液50ml·kg-1,轻揉腹部使鸡血球分散。0.5h后处死小鼠并称重,打开腹腔,用洁净小吸管吸取腹腔液约0.2ml涂在玻片上,待玻片自然干燥后,瑞氏染色,油镜观察。顺序计数100个巨噬细胞,分别记录有吞噬作用和没有吞噬作用的细胞数,对有吞噬作用的巨噬细胞同时记录吞噬的鸡红细胞数,每只小鼠同时制作3张玻片,取平均值。以吞噬率和吞噬指数作为评价巨噬细胞吞噬活性的指标[10]

2水提液对小鼠免疫功能影响
泰山黄精露对小鼠免疫器官及吞噬细胞吞噬能力的影响
由表得出用黄精饮料灌胃后小鼠的胸腺指数、脾脏指数及吞噬指数与对照组相比均有显著提高,并且随浓度的增大其提高作用也增加,说明黄精饮料可以增加小鼠免疫器官的重量,提高小鼠对鸡血红细胞的吞噬作用,提高小鼠免疫功能,为黄精饮料的产业化开发奠定了基础。
Claims (4)
1.一种黄精饮料,其特征在于由黄精、大枣、山楂、甘草、灰树花和水组成,其所述的组份重量百分比为:黄精1.5-2.0%、大枣0.5-1.0%,山楂0.35-0.5%,甘草0.15-0.20%,灰树花0.2-0.3%,水96.00-97.30%。
2.一种如根据权利要求1所述一种黄精饮料的制备方法,其特征在于:步骤1:黄精的制备,将鲜黄精清洗干净,经三蒸三晒晾干,然后用黄酒浸泡晾干切成薄片待用;
步骤2:将微波萃取罐加入60℃温水,然后将上述黄精、大枣、山楂、甘草、灰树花五味中药分别装入过滤药袋中,放入微波萃取罐中浸泡1个小时,加温至60-95℃,微波萃取5分钟,然后停止加温,冷却至70-80℃后再加入甘草保温浸泡30分钟,微波萃取5分钟,再次降温至60-70℃加入灰树花保温浸泡30分钟,微波萃取5分钟,冷却待用;
步骤3:将冷却后的液体经过滤机过滤后放入定容罐混合,将定容罐中的药液加温至80-87℃,然后采用巴氏灭菌法灭菌进入灌装工艺灌装;
步骤4:将灌装后的黄精饮料液体送入灭菌釜加温加压灭菌,灭菌温度120℃,灭菌时间20分钟,即制得产品。
3.根据权利要求2所述一种黄精饮料的制备方法,其特征在于:所述的步骤2还包括二次萃取,再次向微波萃取罐加入60℃温水,然后加热至95℃,微波萃取5分钟,冷却至常温,将冷却后的液体经过滤机过滤后再次放入定容罐。
