背景技术
阿尔茨海默病(AD)是一种起病隐匿的进行性发展的神经系统退行性疾病。临床上以记忆障碍、失语、失用、失认、视空间技能损害、执行功能障碍以及人格和行为改变等全面性痴呆表现为特征,病因迄今未明。65岁以前发病者,称早老性痴呆;65岁以后发病者称老年性痴呆。
该病起病缓慢或隐匿,病人及家人常说不清何时起病。多见于70岁以上(男性平均73岁,女性为75岁)老人,少数病人在躯体疾病、骨折或精神受到刺激后症状迅速明朗化。女性较男性多(女∶男为3∶1)。主要表现为认知功能下降、精神症状和行为障碍、日常生活能力的逐渐下降。根据认知能力和身体机能的恶化程度分成三个时期。
第一阶段(1~3年)为轻度痴呆期。表现为记忆减退,对近事遗忘突出;判断能力下降,病人不能对事件进行分析、思考、判断,难以处理复杂的问题;工作或家务劳动漫不经心,不能独立进行购物、经济事务等,社交困难;尽管仍能做些已熟悉的日常工作,但对新的事物却表现出茫然难解,情感淡漠,偶尔激惹,常有多疑;出现时间定向障碍,对所处的场所和人物能做出定向,对所处地理位置定向困难,复杂结构的视空间能力差;言语词汇少,命名困难。第二阶段(2~10年)为中度痴呆期。表现为远近记忆严重受损,简单结构的视空间能力下降,时间、地点定向障碍;在处理问题、辨别事物的相似点和差异点方面有严重损害;不能独立进行室外活动,在穿衣、个人卫生以及保持个人仪表方面需要帮助;计算不能;出现各种神经症状,可见失语、失用和失认;情感由淡漠变为急躁不安,常走动不停,可见尿失禁。第三阶段(8~12年)为重度痴呆期。患者已经完全依赖照护者,严重记忆力丧失,仅存片段的记忆;日常生活不能自理,大小便失禁,呈现缄默、肢体僵直,查体可见锥体束征阳性,有强握、摸索和吸吮等原始反射。最终昏迷,一般死于感染等并发症。

为解决上述问题,本发明公开了一种防治老年痴呆症状的保健食品,包括如下质量分数的原料组成:牛肉0~1.2份、羊肉0~1.1份、葵花籽0~1.4份、花生0~1.5份、葡萄0~2.7份、香蕉0~2.8份、薏米0~1.1份、小麦0~0.7份、红小豆0~1.2份。

本发明还公开了一种防治老年痴呆症状的保健食品的制作方法,其特征包括如下步骤:
(1)称料;按照比例称牛肉、羊肉、葵花籽、花生、葡萄、香蕉、薏米、小麦、红小豆作为原料备用;
(2)牛肉粉的制备,将牛肉原料进行清洗,熟制,熟制后的牛肉原料进行真空干燥后破碎,即获得牛肉粉;
(3)羊肉粉的制备,将羊肉原料进行清洗,熟制,熟制后的羊肉原料进行真空干燥后破碎,即获得羊肉粉;

(1)称料;按照比例称牛肉、羊肉、葵花籽、花生、葡萄、香蕉、薏米、小麦、红小豆作为原料备用;
(2)牛肉粉的制备,将牛肉原料进行清洗,熟制,熟制后的牛肉原料进行真空干燥后破碎,即获得牛肉粉;
(3)羊肉粉的制备,将羊肉原料进行清洗,熟制,熟制后的羊肉原料进行真空干燥后破碎,即获得羊肉粉;
(4)将葵花籽原料进行清洗,烘干后的葵花籽原料进行真空干燥后破碎,即获得葵花籽粉;
(5)将花生原料进行清洗,烘干后的花生原料进行真空干燥后破碎,即获得花生粉;
(6)将葡萄原料进行清洗,熟制,熟制后的葡萄原料进行真空干燥后破碎,即获得葡萄粉;
(7)将香蕉原料进行清洗,熟制,熟制后的香蕉原料进行真空干燥后破碎,即获得香蕉粉;
(8)将薏米原料进行清洗,烘干,破碎后进行高温膨化,膨化后的薏米原料进行真空干燥后破碎,即获得薏米粉;
(9)将小麦原料进行清洗,烘干,破碎后进行高温膨化,膨化后的小麦原料进行真空干燥后破碎,即获得小麦粉;
(10)将红小豆原料进行清洗,烘干,破碎后进行高温膨化,膨化后的红小豆原料进行真空干燥后破碎,即获得红小豆粉;
(11)混合成品,将步骤(2)~(10)获得的各组合物按照比例进行混合,混合均匀后,即制得成品;
(12)包装,将成品进行分料包装,最终得到产品。

进一步的,其中1)将干净食用牛肉羊肉在温度低于0℃下粉碎成直径≤5.0mm肉粗颗粒;
2)将步骤1)得到的肉粗颗粒置于冷冻粉碎机内,在温度<-150℃下粉碎成直径≤ 50μm微颗粒;3)将步骤2)制备的肉微颗粒放置低温干燥机内,在≤-30℃以下干燥至含水≤2.0%即得肉粉;

1.材料与方法
模型小鼠及分组
60 只 3-4 月龄 SAM-P8 模型小鼠和 20 只 SAM-R1 同源对照小鼠(同源年轻小鼠),分为空白对照组(Control),正常饮食组(SD)和五神组(WS),正常饮食+WS 小鼠粪便灌肠组(FMT),每组 20 只,实验干预 15 周。
生存期检测:60 只 SAMP8 小鼠,SD 组和 WS 组分别 30 只,持续观察小鼠的一般情况及生存情况。
2.记忆功能检测
穿梭箱实验方法
首次训练前先将小鼠放入穿梭箱内 5min 适应环境。实验开始训练时,将小鼠放入箱内任何一侧,背对洞口。一开始先给予蜂鸣音刺激,持续5s,蜂鸣音后给小鼠以电刺激(100V,50Hz,AC)10s.如果在电刺激前小鼠逃至另一侧,则为主动回避反应。如果在电刺激开始后小鼠才完成穿梭,则为被动回避反应。若小鼠在蜂鸣音刺激与电刺激结束后仍留在同侧,则为逃避失败。小鼠每天训练 10 次,每次训练后休息 20s,连续训练5天,计算小鼠主动回避反应率和被动回避反应时间。记忆功能检测:
海马免疫组化检测
海马负责记忆的储存转换和定向等功能,实验终点我们将取下小鼠的海马组织。
(1)比较两组海马的大小
(2)通过 HE 染色观察小鼠海马 CA1 区神经细胞的大体形态学改变;
(3)通过免疫荧光观察小鼠海马 Aβ的沉积情况以及 CA1 区 GFAP、VEGF、TAU 的表达变化。
(4)其他衰老相关特异性分子。
3.实验结果
脑组织 HE染色
Control 组:海马体 CA1 区神经元数量丰富、排列规则,形态结构正常、胞核胞质分界清晰,核仁明显,未见明显炎症。
Aging-SD 组组:海马可见大量神经元皱缩,细胞染色加深,胞核胞质分界不清(黑色箭头),神经元排列较紧密;与 control 组相比,海马体 CA1 区神经元数量及层数明显减少(黄色箭头);未见明显炎症。
Aging-WS 组:海马体 CA1 区未见明显神经元皱缩、细胞深染,神经元层数与control 组相比明显减少,但明显高于 Aging-SD 组;海马体 CA2 区可见少量神经元皱缩,细胞染色加深,胞核胞质分界不清,神经元排列较紧密;未见明显炎症。
Aging-FMT 组:海马 CA1 区均可见较多神经元皱缩,细胞染色加深,胞核胞分界不清,神经元排列较紧密;神经元数量及层数较 Control 组明显减少;未见明显炎症。
胶质纤维酸性蛋白(GFAP )在脑组织中的表达水平
小鼠海马胶质纤维酸性蛋白(GFAP)是星形胶质细胞合成的重要的骨架蛋白,随着年龄的增长表达水平逐渐升高。GFAP 阳性着色可见红色表达,免疫反应产物沉积于星形胶质细胞胞浆、突起。结果表明:衰老小鼠 GFAP 表达水平明显高于 Control 组,WS 组及 FMT 组小鼠脑组织 GFAP 表达明显低于 SD 组。
Claims (8)
1.一种防治老年痴呆症状的保健食品,其特征在于包括如下质量分数的原料组成:牛肉0~1.2份、羊肉0~1.1份、葵花籽0~1.4份、花生0~1.5份、葡萄0~2.7份、香蕉0~2.8份、薏米0~1.1份、小麦0~0.7份、红小豆0~1.2份。
2.根据权利要求1所述的一种防治老年痴呆症状的保健食品,其特征在于:所述各原料均为粉末状态。
3.根据权利要求1所述的一种防治老年痴呆症状的保健食品,其特征在于:所述牛肉和羊肉为冻干粉。
4.根据权利要求1所述的一种防治老年痴呆症状的保健食品的制作方法,其特征包括包括如下步骤:
(1)称料;按照比例称牛肉、羊肉、葵花籽、花生、葡萄、香蕉、薏米、小麦、红小豆作为原料备用;
(2)牛肉粉的制备,将牛肉原料进行清洗,熟制,熟制后的牛肉原料进行真空干燥后破碎,即获得牛肉粉;
(3)羊肉粉的制备,将羊肉原料进行清洗,熟制,熟制后的羊肉原料进行真空干燥后破碎,即获得羊肉粉;
(4)将葵花籽原料进行清洗,烘干后的葵花籽原料进行真空干燥后破碎,即获得葵花籽粉;
(5)将花生原料进行清洗,烘干后的花生原料进行真空干燥后破碎,即获得花生粉;
(6)将葡萄原料进行清洗,熟制,熟制后的葡萄原料进行真空干燥后破碎,即获得葡萄粉;
(7)将香蕉原料进行清洗,熟制,熟制后的香蕉原料进行真空干燥后破碎,即获得香蕉粉;
(8)将薏米原料进行清洗,烘干,破碎后进行高温膨化,膨化后的薏米原料进行真空干燥后破碎,即获得薏米粉;
(9)将小麦原料进行清洗,烘干,破碎后进行高温膨化,膨化后的小麦原料进行真空干燥后破碎,即获得小麦粉;
(10)将红小豆原料进行清洗,烘干,破碎后进行高温膨化,膨化后的红小豆原料进行真空干燥后破碎,即获得红小豆粉;
(11)混合成品,将步骤(2)~(10)获得的各组合物按照比例进行混合,混合均匀后,即制得成品;
(12)包装,将成品进行分料包装,最终得到产品。
5.根据权利要求4所述的一种防治老年痴呆症状的保健食品的制作方法,其特征在于:所述牛肉和羊肉干燥过程破碎中通过冷冻干燥进行。
6.根据权利要求5所述的一种防治老年痴呆症状的保健食品的制作方法,其特征在于:1)将干净食用牛肉羊肉在温度低于0℃下粉碎成直径≤5.0mm肉粗颗粒;
2)将步骤1)得到的肉粗颗粒置于冷冻粉碎机内,在温度<-150℃下粉碎成直径≤ 50μm微颗粒;3)将步骤2)制备的肉微颗粒放置低温干燥机内,在≤-30℃以下干燥至含水≤2.0%即得肉粉。
7.根据权利要求6所述的一种防治老年痴呆症状的保健食品的制作方法,其特征在于:所述牛肉羊肉中还有部分牛骨髓和羊骨髓。
8.根据权利要求7所述的一种防治老年痴呆症状的保健食品的制作方法,其特征在于:所述牛骨髓和羊骨髓的含量不低于2%的重量份数。
