Abstract
The invention relates to a hand-washing-free disinfectant and a preparation method thereof, belonging to the technical field of hygienic products. In order to solve the problem that the existing hand-washing-free disinfectant is easy to stimulate the skin and cause the loss of skin moisture, the invention provides a hand-washing-free disinfectant, which comprises the following components in parts by mass: 60-80 parts of absolute ethyl alcohol, 10-30 parts of purified water, 0.1-2.0 parts of thickening agent, 1-10 parts of glycerol, 0.01-1 part of plant essential oil and 0.1-5 parts of ginger extract. Further, the disinfectant also comprises 0.1 to 2 parts of chlorhexidine gluconate or chlorhexidine acetate and 0.1 to 2 parts of sodium hyaluronate. The hand-washing-free disinfectant disclosed by the invention is prepared by mixing and matching various raw materials, has a scientific formula, has complementary advantages of the raw materials, realizes hand disinfection, can prevent hand skin from drying at the same time by utilizing the synergistic effect of the raw materials, has a pH value close to that of the skin, is non-toxic and non-irritant to the skin, is safe and efficient, can be carried about and is convenient to apply.

Technical Field
The invention belongs to the technical field of hygienic products, and particularly relates to a hand-washing-free disinfectant and a preparation method thereof.
Background
Investigation has shown that cross-infection in public places causes staphylococcus aureus and gram-negative bacilli carried by human hands to reach tens of millions of colonies, and if effective sterilization is not carried out, germs are easily spread to the human hands, so that the cleaning and disinfection of the hands are very important.
The hand-washing-free disinfectant is a novel disinfection product, does not need to be washed by water after use, is suitable for cleaning and disinfecting hands under the condition of water shortage, is simple and convenient to use, is deeply favored by consumers, has a large sales volume in recent years, and is widely applied to clinical use in hospitals, public places, geological exploration, field work and daily life.
The main components of the hand-washing-free disinfectant comprise a disinfecting component and a volatile component, and a few of alcohol solvents with low boiling points are usually added, the principle is that antibacterial active factors such as alcohol can effectively destroy cell walls of bacteria so as to kill the bacteria and fall off from the skin, like aged epidermal cells fall off from the skin, and the alcohol is volatile and does not remain, so that no residual substance is left after the hand-washing-free disinfectant is used on the skin.
However, the skin of children and women is delicate and sensitive to environmental factors such as dryness, coldness and humidity, and chemical stimuli such as alkalinity and acidity, and the hand-washing-free disinfectant is easy to cause skin water loss, dryness, dullness and lusterless after long-term use.

The invention provides a hand-washing-free disinfectant and a preparation method thereof, aiming at solving the problem that the existing hand-washing-free disinfectant easily stimulates the skin and causes the loss of skin water.

the hand-washing-free disinfectant comprises the following components in parts by mass: 60-80 parts of absolute ethyl alcohol, 10-30 parts of purified water, 0.1-2.0 parts of thickening agent, 1-10 parts of glycerol, 0.01-1 part of plant essential oil and 0.1-5 parts of ginger extract.

A preparation method of a hand-washing-free disinfectant comprises the following steps:
step one, adding 0.1-2.0 parts of thickening agent into 10-30 parts of purified water at 50-70 ℃, and stirring for 5-10min after the thickening agent is completely soaked by the purified water to form uniform colloidal solution A;
step two, measuring 60-80 parts of absolute ethyl alcohol, adding one third of absolute ethyl alcohol into the colloidal solution A, stirring for 5-30min, and standing for 5-30min after uniform stirring to form a uniform colloidal solution B;
weighing 1-10 parts of glycerol, 0.01-1 part of plant essential oil, 0.1-5 parts of ginger extract and one third of absolute ethyl alcohol, stirring for 5-30min, adding into the colloidal solution B after uniformly stirring, and stirring for 5-30min to form a uniform colloidal solution C;
and step four, adding the remaining one third of the absolute ethyl alcohol into the colloidal solution C, stirring for 5-30min to form uniform colloidal solution, and filling into a container to obtain the hand-washing-free disinfectant.
Further, 0.1-2 parts of chlorhexidine gluconate or chlorhexidine acetate is added in the third step, specifically, 0.1-2 parts of chlorhexidine gluconate or chlorhexidine acetate, 1-10 parts of glycerol, 0.01-1 part of plant essential oil, 0.1-5 parts of ginger extract and one third of absolute ethyl alcohol are weighed, stirred for 5-30min, and after being uniformly stirred, the mixture is added into the colloidal solution B and stirred for 5-30min to form a uniform colloidal solution C.
Further, 0.1-2 parts of sodium hyaluronate are added in the fourth step, and the method specifically comprises the following steps: weighing 0.1-2 parts of sodium hyaluronate and one third of alcohol, stirring for 5-30min, adding into the colloidal solution C after uniformly stirring, stirring for 5-30min to form uniform colloidal solution, and filling into a container to obtain the hand-free disinfectant.
Further, the thickening agent in the first step is hydroxypropyl methylcellulose or hydroxyethyl cellulose; and step three, the plant essential oil is one of ginger oil, peppermint oil or green tea essential oil.
Further, the pH value of the obtained hand-washing-free disinfectant is 6-8.
The invention has the beneficial effects that:
the hand-washing-free disinfectant disclosed by the invention is prepared by mixing and matching various raw materials, has a scientific formula, has complementary advantages of the raw materials, realizes hand disinfection by utilizing the synergistic effect of the raw materials, relieves the stimulation of the disinfectant to the hand skin and prevents the hand skin from drying.
The hand-washing-free disinfectant can completely remove bacteria and viruses on hands, cleans hands at any time and any place, has a disinfecting effect lasting for 4-6 hours after being compounded with the chlorhexidine gluconate or the chlorhexidine acetate, and effectively reduces cross infection in public places. The moisturizing and nourishing skin care ingredients such as glycerin and sodium hyaluronate are added into the formula, so that the stimulation to the skin is reduced, the skin is nourished, the hand is kept moist, the pH value is close to the pH value of the skin, the product is mild in property, and the moisturizing and nourishing skin care agent is non-toxic and non-irritant to normal skin, safe and efficient. The ginger extract in the formula is rich in vitamin C, vitamin E, vitamin B group and gingerol, is a good antioxidant, can effectively eliminate free radicals of a human body, reduces the generation of wrinkles, delays senility and is a good product for maintaining beauty and keeping young. In addition, the gingerol can also promote the heat production of human body, expel the cold of human body and avoid the cold of hands caused by the volatilization of alcohol.
The invention provides the hand-washing-free disinfectant with fixed proportion and good fluidity through a strict preparation process, can be contained by various containers, can be carried about and is convenient to use, and the prepared colloidal solution can effectively prevent ethanol from volatilizing, is convenient to store and avoids fire.
Detailed Description
The technical solutions of the present invention are further described below with reference to the following examples, but the present invention is not limited thereto, and any modifications or equivalent substitutions may be made to the technical solutions of the present invention without departing from the spirit and scope of the technical solutions of the present invention.
Example 1
The embodiment provides a hand-washing-free disinfectant, which comprises the following components in parts by mass: 60-80 parts of absolute ethyl alcohol, 10-30 parts of purified water, 0.1-2.0 parts of thickening agent, 1-10 parts of glycerol, 0.01-1 part of plant essential oil and 0.1-5 parts of ginger extract.
The ethanol solution with the volume concentration of 60-80%, especially the ethanol solution with the volume concentration of 75% is similar to the osmotic pressure of bacteria, and can gradually and continuously permeate into the bacteria before the protein on the surface of the bacteria is not denatured, so that all the protein of the bacteria is dehydrated, denatured and solidified, and finally the bacteria are killed.
The glycerin is colorless, sweet, clear and viscous liquid, and is synthesized by adopting a propylene chlorination method, and comprises the following four steps: high-temperature chlorination of propylene, hypochlorination of chloropropene, saponification of dichloropropanol and hydrolysis of epichlorohydrin. The hydrolysis of epichlorohydrin to prepare glycerol is carried out in 10% sodium hydroxide and 1% sodium carbonate aqueous solution at 150 ℃ and 1.37MPa carbon dioxide pressure to generate sodium chloride-containing glycerol aqueous solution with glycerol content of 5-20%, and the glycerol with purity of more than 98% is obtained by concentration, desalination and distillation.
The thickener used in this example was hydroxypropyl methylcellulose or hydroxyethyl cellulose.
Hydroxypropyl methylcellulose (HPMC), a variety of non-ionic cellulose mixed ethers, is a semi-synthetic, inactive, and viscoelastic polymer commonly used as lubricants in ophthalmology, or as an adjuvant or excipient in oral pharmaceuticals, and is commonly used in a variety of different commercial products. It swells in cold water to a clear or slightly turbid colloidal solution. The aqueous solution has surface activity, high transparency and stable performance. HPMC also has the characteristics of thickening ability, low salt tolerance, low ash content, pH stability, water retention, dimensional stability, excellent film forming property, wide enzyme resistance, dispersibility, cohesiveness and the like.
Hydroxyethyl cellulose (HEC) is a white or light yellow, tasteless and nontoxic fibrous or powdery solid, is prepared by etherification reaction of alkali cellulose and ethylene oxide (or chloroethanol), and belongs to nonionic soluble cellulose ethers. HEC has surface active, thickening, suspending, binding, emulsifying, film forming, dispersing, water retaining and protective effects in solid and liquid pharmaceutical preparations.
The ginger can kill bacteria and remove toxicity, the components contained in the ginger can play a role similar to antibiotics, and the ginger can help to relieve symptoms of acute gastroenteritis caused by eating food infected by bacteria. The ginger extract contains more vitamin C and gingerol, is a good antioxidant, can effectively eliminate free radicals of a human body, reduce the generation of wrinkles and delay aging, and is a good product for maintaining beauty and keeping young. In addition, gingerol can promote heat production of human body, expel cold qi of human body, and treat common cold due to wind-cold.
The plant essential oil used in this embodiment is one of ginger oil, peppermint oil, or green tea essential oil.
Ginger oil is mainly an oily component extracted from ginger. It contains a large amount of volatile oil such as farnesene, linalool, geraniol, etc. After being taken into a human body, the traditional Chinese medicine composition can play a role in inducing sweat and relieving exterior syndrome, and has a specific treatment effect on wind-cold type common cold. The product is rich in vitamin E and vitamin B, and has effects of nourishing nerve, caring skin, and removing wrinkle.
The ginger extract and the plant essential oil used in this example are both commercially available products. Wherein the peppermint oil is obtained from Shanghai Shenbao essence and spice GmbH, the ginger oil is obtained from Baoji City Cheng biological development GmbH, the green tea essential oil is obtained from Shanghai Shenyuan actual industry GmbH, and the ginger extract is obtained from Baoji City Cheng biological development GmbH.
Example 2
The embodiment is a preparation method of the hand-washing-free disinfectant provided in embodiment 1, and the preparation method specifically includes the following steps:
step one, adding 0.1-2.0 parts of thickening agent into 10-30 parts of purified water at 50-70 ℃, and stirring for 5-10min after the thickening agent is completely soaked by the purified water to form uniform colloidal solution A;

Example 3
The embodiment provides a hand-washing-free disinfectant, which comprises the following components in parts by mass: 75 parts of absolute ethyl alcohol, 1.0 part of chlorhexidine gluconate, 16.4 parts of purified water, 1.0 part of hydroxypropyl methyl cellulose, 0.5 part of sodium hyaluronate, 4 parts of glycerol, 0.1 part of ginger oil and 2.0 parts of ginger extract.
Chlorhexidine is a biguanide chlorobenzene, which has a fairly broad antimicrobial activity. Under the condition of pH5.0-8.0, it is effective for gram-positive bacteria and gram-negative bacteria, and can be used for external hand and skin disinfection and wound irrigation. Chlorhexidine, which starts slower than alcohols, has considerable persistence on the skin and therefore has persistent chemical antimicrobial activity on the skin, has little possibility of contact sensitivity or light sensitivity caused by chlorhexidine in long-term clinical use, and is not absorbed through the intact skin even when it is washed with hands many times per day.
Sodium hyaluronate is an acidic mucopolysaccharide, the basic structure of which is a large polysaccharide consisting of two disaccharide units, namely D-glucuronic acid and N-acetylglucosamine, and the hyaluronic molecules of the acid hyaluronate can carry more than 500 times of water, so that the acid hyaluronate can improve the nutrition metabolism of skin, make the skin tender and smooth, remove wrinkles, increase the elasticity and prevent aging, and is a good transdermal absorption enhancer while keeping moisture.

step three, weighing 1.0 part of chlorhexidine gluconate, 4 parts of glycerol, 0.1 part of ginger oil, 2.0 parts of ginger extract and one third of absolute ethyl alcohol, stirring for 20min, adding into the colloidal solution B after uniformly stirring, and stirring for 50min to form a uniform colloidal solution C;
and step four, weighing 0.5 part of sodium hyaluronate and one third of alcohol, stirring for 20min, adding the mixture into the colloidal solution C after uniformly stirring, and stirring for 20min to form uniform colloidal solution, namely the hand-washing-free disinfectant.
Example 5
The embodiment provides a hand-washing-free disinfectant, which comprises the following components in parts by mass: 65 parts of absolute ethyl alcohol, 2.0 parts of chlorhexidine acetate, 20.5 parts of purified water, 1.5 parts of hydroxyethyl cellulose, 0.8 part of sodium hyaluronate, 5 parts of glycerol, 0.2 part of peppermint oil and 5.0 parts of ginger extract.

step three, weighing 2.0 parts of chlorhexidine acetate, 5 parts of glycerol, 0.2 part of peppermint oil, 5.0 parts of ginger extract and one third of absolute ethyl alcohol, stirring for 20min, adding into the colloidal solution B after stirring uniformly, and stirring for 20min to form a uniform colloidal solution C;
and step four, weighing 0.8 part of sodium hyaluronate and one third of alcohol, stirring for 20min, adding the mixture into the colloidal solution C after uniformly stirring, and stirring for 20min to form uniform colloidal solution, namely the hand-washing-free disinfectant.
Example 7
The embodiment provides a hand-washing-free disinfectant, which comprises the following components in parts by mass: 80 parts of absolute ethyl alcohol, 0.5 part of chlorhexidine gluconate, 15.15 parts of purified water, 0.8 part of hydroxypropyl methyl cellulose, 1.0 part of sodium hyaluronate, 2 parts of glycerol, 0.05 part of green tea essential oil and 0.5 part of ginger extract.
Example 8
The embodiment is a preparation method of the hand-washing-free disinfectant provided in embodiment 7, and the preparation method specifically includes the following steps:
step one, adding 0.8 part of hydroxypropyl methyl cellulose into 15.15 parts of purified water at 65 ℃, and stirring for 8min to form uniform colloidal solution A after the thickening agent is completely soaked in the purified water;
step two, measuring 80 parts of absolute ethyl alcohol, adding one third of absolute ethyl alcohol into the colloidal solution A, stirring for 20min, and standing for 20min after uniform stirring to form a uniform colloidal solution B;
weighing 0.5 part of chlorhexidine gluconate, 2 parts of glycerol, 0.05 part of green tea essential oil, 0.5 part of ginger extract and one third of absolute ethyl alcohol, stirring for 20min, adding into the colloidal solution B after uniformly stirring, and stirring for 20min to form a uniform colloidal solution C;
and step four, weighing 1.0 part of sodium hyaluronate and one third of alcohol, stirring for 20min, adding the mixture into the colloidal solution C after uniformly stirring, and stirring for 20min to form uniform colloidal solution, namely the hand-washing-free disinfectant.
Verification test I: sterilization test
The bactericidal test was conducted as follows to determine the bactericidal efficacy.
Test bacteria: staphylococcus aureus [ CMCC (B)26003], Pseudomonas aeruginosa [ CMCC (B)10104], Bacillus subtilis [ CMCC (B)63501], Candida albicans [ CMCC (F)98001], Aspergillus niger [ CMCC (B)98003], Escherichia coli [ CMCC (B)44102 ].
Preparing a culture medium: the culture medium is prepared by autoclaving tryptone soy peptone liquid culture medium, tryptone soy peptone agar culture medium, Sa's glucose agar culture medium and Sa's glucose liquid culture medium at 121 ℃ for 15 minutes for culturing microbial strains.
The preparation of bacterial suspension comprises washing 24h slant culture of test bacteria with 5m L0.03.03 mo 1/L Phosphate Buffer Solution (PBS), diluting with PBS to desired concentration, and making into bacterial suspension (the desired concentration is 100 μ L drops in 5m L sample solution, and recovering bacteria number is 1 × 104~9×104cfu/mL)。
And (2) taking 4 tubes of the hand-washing-free disinfectant liquid samples (5m L), 75% ethanol solution and control sample liquid (the same material as the samples, the same size, but no antibacterial material and subjected to sterilization treatment) of examples 3, 5 and 7, taking the bacterial suspension, respectively dropwise adding 100 mu L into each sample liquid to be tested and each control sample liquid, uniformly mixing by using an electric mixer, starting timing, acting for 2 min, 5 min, 10min and 20min, respectively putting the sample liquid (0.5m L) into a test tube containing a corresponding neutralizer of 5m L, fully and uniformly mixing for proper dilution, then taking 2-3 dilutions, respectively sucking 0.5m L, putting the samples into two flat dishes, pouring agar medium (bacteria) or Sabouraud agar medium (fungi) 15m L at 40-45 ℃, rotating the flat dishes to fully and uniformly stir the plates, turning the flat dishes after agar is solidified, culturing the bacteria at 35 ℃ for 48h or 72h, taking viable bacteria for counting, and repeating the test for 3 times.
And (3) judging the sterilization efficacy: after culturing for 48h or 72h, the bacteriostasis rate is calculated according to the following formula that x is (A-B)/Ax is 100%. Wherein X is the bacteriostasis rate, A is the average colony number of a control sample, and B is the average colony number of a tested sample.
The evaluation standard is that the sterilization rate is more than or equal to 90 percent, and the product has sterilization effect.
The sterilization tests were performed on 75% ethanol in examples 3, 5, and 7 and comparative example, respectively, using the above sterilization test methods, and finally the sterilization efficacy of the germicidal hand sanitizer composition of the present invention was determined based on the percentage of the concentration of the non-viable microorganisms to the initial concentration of the microorganisms. The sterilization results are shown in table 1.

As can be seen from the data in Table 1, the hand-washing-free disinfectant provided by the invention has a remarkable bactericidal effect on common pathogenic bacteria, and the effect is obviously superior to that of a 75% ethanol solution.
And (5) verification test II: adverse hand skin reaction test
Randomly selecting 100 people, dividing into two groups, and checking the two hands of the experimental subject before experiment, wherein the skin is required to be complete, and the experimental subject has no skin disease and is not allergic to ethanol.
The experimental group used 8-10ml of the hand-washing-free disinfectant prepared in example 3 to perform six-step hand washing and disinfection until the hand-washing-free disinfectant was dried.
The control group was sterilized with 75% ethanol in the same manner for one month.
The results of the control experiments are as follows:
in the control group, 75% ethanol was used to give a cooling sensation, but if the skin care product was used repeatedly for about 4 days without any other skin care product, the inner side surfaces of the fingers and the palm were peeled off, and the skin was peeled off by more than 58% of 50 test persons with dry skin. With the continuous use, the peeling phenomenon is more serious for most people who bubble and peel, and the peeling phenomenon is relieved for a few people. After 1 month of continuous use, the subject's skin became dry, rough, and stiff.
The test group shows that the no-clean hand sanitizer prepared in example 3 of the invention is peeled in 50 test persons, the dry person is only 2, and the rest persons do not have the problems of foaming, peeling and keratinization, after the no-clean hand sanitizer is continuously used for one month, the skin of a test person is moist and elastic, the surface of the test person is ruddy and soft, the activity is strong, the moisturizing effect is good, the fingers and the palms do not have the defects of wrinkles, dryness, hardness and the like, and the no hand is too cold after the no-clean hand sanitizer is used, so the using effect is good.
Claims (10)
1. The hand-washing-free disinfectant is characterized by comprising the following components in parts by mass: 60-80 parts of absolute ethyl alcohol, 10-30 parts of purified water, 0.1-2.0 parts of thickening agent, 1-10 parts of glycerol, 0.01-1 part of plant essential oil and 0.1-5 parts of ginger extract.
2. The hand-washing-free disinfectant as claimed in claim 1, further comprising chlorhexidine gluconate or chlorhexidine acetate 0.1-2 parts.
3. The hand sanitizer according to claim 1 or 2, further comprising 0.1 to 2 parts of sodium hyaluronate.
4. The hand sanitizer according to claim 3, wherein the plant essential oil is one of ginger oil, peppermint oil or green tea essential oil; the thickening agent is hydroxypropyl methylcellulose or hydroxyethyl cellulose.
5. The hand sanitizer according to claim 4, wherein the pH is from 6 to 8.
6. A preparation method of a hand-washing-free disinfectant is characterized by comprising the following steps:
step one, adding 0.1-2.0 parts of thickening agent into 10-30 parts of purified water at 50-70 ℃, and stirring for 5-10min after the thickening agent is completely soaked by the purified water to form uniform colloidal solution A;
step two, measuring 60-80 parts of absolute ethyl alcohol, adding one third of absolute ethyl alcohol into the colloidal solution A, stirring for 5-30min, and standing for 5-30min after uniform stirring to form a uniform colloidal solution B;
weighing 1-10 parts of glycerol, 0.01-1 part of plant essential oil, 0.1-5 parts of ginger extract and one third of absolute ethyl alcohol, stirring for 5-30min, adding into the colloidal solution B after uniformly stirring, and stirring for 5-30min to form a uniform colloidal solution C;
and step four, adding the remaining one third of the absolute ethyl alcohol into the colloidal solution C, and stirring for 5-30min to form uniform colloidal solution, namely the hand-washing-free disinfectant.
7. The preparation method of the hand-washing-free disinfectant according to claim 6, wherein 0.1-2 parts of chlorhexidine gluconate or chlorhexidine acetate is further added in the third step, and specifically, 0.1-2 parts of chlorhexidine gluconate or chlorhexidine acetate, 1-10 parts of glycerin, 0.01-1 part of plant essential oil, 0.1-5 parts of ginger extract and one third of absolute ethyl alcohol are weighed, stirred for 5-30min, and added into the colloidal solution B after being uniformly stirred, and stirred for 5-30min to form the uniform colloidal solution C.

The preparation method of the hand-washing-free disinfectant according to claim 6 or 7, wherein 0.1-2 parts of sodium hyaluronate are further added in the fourth step, specifically: weighing 0.1-2 parts of sodium hyaluronate and one third of alcohol, stirring for 5-30min, adding into the colloidal solution C after uniformly stirring, and stirring for 5-30min to form uniform colloidal solution, namely the hand-washing-free disinfectant.
9. The method as claimed in claim 8, wherein the thickening agent is hydroxypropyl methylcellulose or hydroxyethyl cellulose; and step three, the plant essential oil is one of ginger oil, peppermint oil or green tea essential oil.
10. The preparation method of the hand-washing-free disinfectant as claimed in claim 9, wherein the pH value of the obtained hand-washing-free disinfectant is 6-8.
