Detailed Description
The invention discloses a hair tonic composition which contains a silk protein liquid component, mainly comprises medium and small protein molecules with the molecular weight of 2-20 ten thousand Da (or 10-20 ten thousand Da), is rich in silk protein, peptide, amino acid and the like, and has the functions of nourishing and repairing hair, scalp and hair root.
The invention discloses a hair-fixing and hair-loss-preventing hair-growing liquid composition containing fibroin and a preparation method thereof. The hair restorer composition comprises the following main raw material components:
deionized water, butanediol, sodium hyaluronate, EDTA.2Na, allantoin, D-panthenol, α -bisabolol, hydrogenated castor oil, fibroin liquid, a proper amount of essence and a proper amount of preservative.
The fibroin liquid is used together with other components (α -bisabolol, traditional Chinese medicine extract (optional component) and the like), so that the micro-ecological environment of scalp is improved, the release of inflammatory factors is reduced, the activity of human fibroblast is promoted, and the damaged hair scales are effectively repaired, thereby really achieving the purposes of hair growth and hair fixation.
The specific process of the silk protein liquid is as follows:
the fibroin solution is prepared by fresh cut cocoons by adopting the latest O-discharge double-circulation biochemical hydrolysis technology, controlling the hydrolysis degree of silk through hydrolytic enzyme, disorderly winding protein bundles of the silk, cutting the protein bundles into proteins with different grades and molecular weights, and combining the proteins to obtain the fibroin solution. The method specifically comprises the following process steps:
1) 0.4 to 6 mass percent of Na is used2CO2Or NaHCO3The aqueous solution is used as degumming agent for degumming silkworm cocoons under the conditions that the pH value is 8-11, the hydrolysis temperature is 50-80 ℃ and the time is 2-4 hours; filtering after the degumming is finished to obtain degumming hydrolysate and silk fibroin fibers;
2) removing salt in the degumming hydrolysate through electrodialysis, obtaining sericin hydrolysate and dialysis ion solution from the degumming hydrolysate, retaining the sericin hydrolysate, and transferring the dialysis ion solution to next preparation of the sericin hydrolysate for use;
3) dissolving the silk fibroin fibers obtained by filtering in the step 1) in CaCl with the mass percent of 30-60% at the temperature of 60-100 DEG C2、CaCl2-H2O、CaCl2-2H2Hydrolyzing in neutral salt solution O or L iBr to obtain silk fibroin solution;
4) performing electrodialysis on the hydrolyzed silk fibroin solution obtained in the step 3) to remove ions in the solution, and circulating the solution until the conductivity of the solution reaches 10-500us/cm to obtain a light yellow silk fibroin pure solution, wherein the ion solution after electrodialysis can be circularly used as a neutral salt solution in the step 4);
5) uniformly mixing the sericin hydrolysate obtained in the step 2) and the fibroin pure solution obtained in the step 4) to obtain fibroin liquid; by controlling the hydrolysis end point, the molecular weight of the fibroin of the obtained final product is controlled within the range of 2-20 ten thousand daltons, and the molecular weight is mainly centralized within the range of 10-20 ten thousand daltons.
In the silk hydrolysate disclosed by the invention, small molecular peptides and amino acids can be partially absorbed and utilized by the scalp, and have nourishing and nourishing effects on the scalp and hair. The macromolecular sericin and keratin can form a cell adhesion substance like 'mud' on the surfaces of the scalp and the hair, and the cell adhesion substance can repair epidermal cells to be peeled off and delay the shedding of the scalp. The following are contents of the effect test examples.
Effect experiment example 1
The product of the invention is mainly added with fibroin liquid and other components (traditional Chinese medicine extract) for matching use. In order to achieve the test effect, 100 volunteers with symptoms of hair thinning, hair line backward movement, hair loss and hair loss, dry hair and split ends and the like are selected for testing. After the hair restorer is used for 30 days and a proper amount of the hair restorer of the embodiment 1-4 is used every day for scalp massage and nursing, the scalp massage is carried out for 10-20 minutes until the hair restorer is absorbed by the scalp, the hair restorer can be repeatedly used for 1-3 times a day, and the effective rate reaches 70.23 percent through verification.

Nutritional alopecia: excessive sugar, excessive salt, protein deficiency, vitamin deficiency of B group, and trace elements deficiency all cause alopecia, and at this time, nutrition supplement is required to improve alopecia symptoms.

Explanation of hair growth and hair-securing effect:

In addition, as shown in fig. 1a-1d, 15 volunteers are selected for use by a sample prepared by the verification formula of the invention, and from fig. 1c-1d, it can be seen that the degree of hair scale fragmentation is obviously reduced after the test effect diagram of the blank control combination added with the silk protein liquid is amplified by 1000 times and 4000 times, which indicates that the silk protein liquid has a certain repairing effect on hair scales, so that the invention has a certain effect on nutrition and hair fixation.
Based on the theory of promoting the activity of human fibroblasts and deeply repairing damaged scalp to improve the micro-ecological environment of scalp, the silk protein liquid is used for promoting the activity of skin fibroblasts, balancing immune barrier, improving the micro-ecological environment of scalp consisting of physiological lipid, natural moisturizing factors and scalp microbial population, maintaining the health of scalp and repairing damaged hair scales, thereby achieving the effects of growing hair and strengthening hair.
The results of multiple tests show that the hair growth liquid prepared by the formula of the functional raw materials without any auxiliary hair growth has the hair growth effect; meanwhile, the hair growing and strengthening effect can be more remarkable by matching with other functional components (traditional Chinese medicine extracting solution).
Effect test example 2
We then carried out a relevant test to certify the efficacy of our primary component fibroin solution, and we carried out an immune barrier study on the microecological reconstruction of the scalp and scalp with fibroin solution, with the results shown in tables 2 and 3 below.

The results in FIGS. 2-3 and tables 2-3 above show that the fibroin solutions of the examples of the present invention produced some inhibition of I L-1 a and I L-6 cytokine release from macrophages due to L PS at three concentrations tested, 0.012mg/m L, 0.06mg/m L, and 0.3mg/m L, with a tendency to increase with concentration.
In addition, Table 4 below shows that 0.30mg/m L and 1.2mg/m L promote the viability of human skin fibroblasts, and that 3.5mg/m L may produce some inhibition of the cells by the action of the hydrolysate.

As shown in fig. 4, the fibroin solution and the control group were subjected to 2D cell repair test, which shows that cell migration started at 24h after the test, and that 48h clearly shows that the comparative NC blank group has significant effects on cell migration, proliferation and regeneration.

The hair loss delaying and hair growth promoting agent is characterized in that no traditional Chinese medicine components are added in example 1, the silk protein liquid mainly contains α -bisabolol which has synergistic effects of nutrition supply to scalp hair follicles, activity promotion of fibroblasts and inflammatory factor inhibition, and cell regeneration promotion is achieved, so that hair loss is delayed and hair growth is promoted, the success rate of delaying hair loss and promoting hair growth of the traditional Chinese medicine composition in comparative example 3 is the same as that of example 1, and the synergistic effect of the traditional Chinese medicine composition and the silk protein liquid in comparative example 2 is higher than that of single example 1 and comparative example 2.
The difference between comparative example 4 and comparative example 5 is that one has fibroin solution, and the other has double success rate of the traditional Chinese medicine components without fibroin solution, but the success rate of the two in delaying hair loss or promoting hair is also higher than that of example 1 and comparative examples 1, 2 and 3, which shows that part of the components have the effect of providing nutrition for hair follicle cells, and part of the components have the effect of inhibiting 5 α -reductase, and the proliferation of hair outer root sheath cells is promoted.
Meanwhile, part of the components have the effect of stimulating hair follicle blasts, and other components have the effects of sterilizing and resisting inflammation, so that the hair loss is effectively prevented, and the hair growth is accelerated. In summary, the efficacy of each component in the present invention varies.
Meanwhile, experiments show that the reasonable matching of the components has important influence on the exertion of the efficacy, and the synergistic effect can be generated only by the reasonable matching of the components, so that the anti-hair loss and hair growth effects of the invention are achieved.

2. The fibroin-containing hair loss prevention, hair solidification and hair growth promotion composition of claim 1, wherein: the content range of the traditional Chinese medicine extract is 0.1-10%; a preferred range is 1.0-5%.

The fibroin-containing hair loss prevention, hair solidification and hair growth promotion composition of claim 2, wherein: the traditional Chinese medicine extract is selected from one or more of 0.1-1.0% of Chinese arborvitae extract, 0.1-1.0% of polygonum multiflorum extract, 0.1-1.0% of south dodder seed extract, 0.1-1.0% of wormwood extract, 0.1-1.0% of privet extract, 0.1-1.0% of angelica sinensis extract, 0.1-1.0% of ginseng extract, 0.1-1.0% of ganoderma atrum extract, 0.1-1.0% of liquorice extract, 0.1-1.0% of citrus peel extract and 0.1-1.0% of zanthoxylum fruit extract.

The fibroin-containing hair loss prevention, hair solidification and hair growth promotion composition of claim 1, wherein: the fibroin solution is prepared by fresh cut cocoons by adopting the latest O-discharge double-circulation biochemical hydrolysis technology, controlling the hydrolysis degree of silk through hydrolytic enzyme, disorderly winding protein bundles of the silk, cutting the protein bundles into proteins with different grades and molecular weights, and combining the proteins to obtain the fibroin solution.

3.0% of silk protein liquid; 0.1% of arborvitae extract; 0.1% of polygonum multiflorum extract; 0.1% of south dodder seed extract; 0.1% of Artemisia princeps Pampanini extract; 0.1% of privet extract; 0.1% of angelica extract; 0.1% of ginseng extract; 0.1% of ganoderma atrum extract; 0.1% of licorice extract; citrus peel extract 0.1%; 0.1% of zanthoxylum bungeanum fruit extract; 0.03 percent of citric acid; a proper amount of essence; proper amount of preservative.
7. The fibroin-containing hair loss prevention, solidification and growth promotion liquid composition as set forth in claim 2 is characterized by comprising the following raw material components in parts by weight:
80.2% of deionized water; 3.0% of butanediol; 0.1% of EDTA.2Na; 0.05% of sodium hyaluronate; 0.05% of allantoin; 0.5 percent of D-panthenol;
α -bisabolol 0.5%, hydrogenated castor oil 0.5%;
4.0% of silk protein liquid; 1.0% of arborvitae extract; 1.0% of polygonum multiflorum extract; south dodder seed extract 1.0%; 1.0% of artemisia scoparia extract; 1.0% of privet extract; 1.0% of angelica sinensis extract; 1.0% of ginseng extract; 1.0% of ganoderma atrum extract; 1.0% of liquorice extract; 1.0% of citrus peel extract; 1.0% of zanthoxylum bungeanum fruit extract; 0.03 percent of citric acid; a proper amount of essence; proper amount of preservative.
8. The method for preparing the fibroin-containing anti-hair loss, hair-fixing and hair-growing liquid composition as claimed in any one of claims 1 to 7, which comprises the following steps:
1) firstly, stirring, mixing and dissolving deionized water, butanediol and sodium hyaluronate completely, adding EDTA-2Na, allantoin and D-panthenol after the materials are dissolved completely, stirring and dissolving completely at the temperature of 80-85 ℃, and mixing a material body A;
