Detailed Description
The technical solutions of the present invention are further described below with reference to specific examples to enable those skilled in the art to better understand the present invention, but the scope of the present invention is not limited to the contents described in the examples.
Example 1: extraction method of sweet potato extract
(1) Cleaning and peeling: taking 5kg of sweet potatoes, and cleaning soil impurities on the surfaces of the sweet potatoes;
(2) adding distilled water, tap water or domestic water, pulverizing, and grinding to obtain mixture containing soluble substances such as starch, fiber, protein, mineral, etc.;
(3) separating the sweet potato residue from the emulsion by using a centrifugal sieve (100 meshes);
(4) sand removal: a sand remover is used to remove some of the sand and particles from the emulsion.
(5) Starch separation: washing starch in a countercurrent mode by using a cyclone, separating the starch from other impurities to generate washing liquid, wherein the obtained washing liquid contains protein and trace elements;
(6) high-efficiency liquid concentration: filtering with a filter membrane (the filtering diameter is 1 μm) to remove macromolecular water and fine particles;
(7) precipitating a small amount of fine starch, centrifuging the supernatant liquid, and collecting pasty mucus protein;
(8) adding a small amount of ethanol with volume fraction of 10% into the pasty mucus, and filtering with filter membrane (filtering diameter of 1 μm) to obtain viscous ointment; because these pastes are insoluble in water, the paste is better separated by adding ethanol with the volume fraction of 10%.
(9) And (4) adding distilled water to dilute the mixed solution of the ethanol and the pasty mucus obtained in the step (8), and standing. After the ethanol is completely volatilized, freezing below zero to obtain transparent solution, freezing and drying below zero to obtain solid white powdery substance, drying, sealing and storing.
The product obtained in this step must be subjected to a cryogenic freezing treatment in order to obtain a solid white powder (fig. 1). In this example, 0.35-0.38 g of sweet potato extract was extracted from 5kg of sweet potato, and the pH was 7.1.
Collecting a little solid powder of sweet potato extract, collecting strong sulphuric acid 10m L, adjusting pH to 4, adding sweet potato
Extracting solid powder 1g, rapidly dissolving without stirring, and detecting pH value to be 7, simultaneously taking sodium hydroxide 10m L, pH value to be 12, adding sweet potato to extract solid powder 1g, rapidly dissolving without stirring, and detecting pH value to be 7.
Therefore, the sweet potato extract prepared by the invention can well adjust the pH value.
Furthermore, at least one hydroxyl group and one carboxyl group in the sweet potato extract have acid-base equilibrium reaction with sulfuric acid or alkali, so that the sulfuric acid and alkali decomposing effect can be achieved. Hydroxyl is a polar group, (OH) is an important active oxygen, is (OH) hydroxyl in molecular view, loses an electron formation, and has extremely strong electron-gaining capacity, namely oxidation capacity, and when meeting a sulfuric acid solution, ions are recombined to generate salt and water, so that the sulfuric acid solution can be decomposed. When it meets alkali, carboxyl and OH are combined to form acid-base reaction to produce water, so that it can decompose alkali to regulate pH value.
Therefore, the obtained sweet potato extract at least comprises one hydroxyl and one carboxyl, the amino acid or the vegetable protein of the sweet potato extract is judged, the amino acid molecules contain two functional groups of amino and carboxyl, similar to hydroxy acid, the amino acid can be α -amino acid respectively according to the amino connected at different positions of a carbon chain, but the amino acid obtained after protein hydrolysis is α -amino acid and twenty amino acids are basic units of the protein, and the sweet potato extract is an organic compound containing basic amino and acidic carboxyl, namely the obtained sweet potato extract is the sweet potato protein.
Further, the obtained product is detected by product quality supervision and inspection in a persistent market, and all indexes of the product obtained in the embodiment all meet the standard Q/YSKJ001-2018 'pesticide residue clearing', and specific indexes are detailed in the following table:
