Abstract
本发明公开了一种用于癌症靶向治疗的短肽及基于其的超声响应纳米载药微泡和应用,属于生物医药技术领域。本发明公开的短肽是一段由9个氨基酸组成的新型短肽,同时,本发明采用超声辐照联合载药靶向微泡原理,建立了基于该新型短肽的针对新生血管的分子靶向超声响应纳米载药微泡,载药纳米粒表面羧基化的PEG末端与短肽末端氨基酸的NH2结合形成酰胺键,末端氨基的连接方法降低了对新型多肽的构象及活性的影响,尽可能地保护了新型多肽与血管内皮细胞表面高表达的Dll4配体的识别和结合能力。相对于之前的hD4R肽段来说,将其中的有效活性成分使用纳米递药系统与超声介导结合将会提供一种新型、高效的抗肿瘤血管生成模式。
Description
技术领域
本发明属于生物医药技术领域,涉及一种用于癌症靶向治疗的短肽及基于其的超声响应纳米载药微泡和应用。
背景技术
近年来,肿瘤发病率的日益增高已经引起人们的高度重视。据统计,每10万人中就有286人患癌,而当中有181人因为癌症死亡,平均每5个患癌者里有3人因为癌症而死亡。新生血管是实体肿瘤生长的必要条件之一,抗新生血管形成治疗因此成为肿瘤分子靶向治疗的有效策略,但目前国内外对于该领域的研究显示治疗靶向较为单一、效果有限。因此,如何寻找一种可针对肿瘤新生血管的分子或靶点是目前亟待解决的问题。
肿瘤新生血管是肿瘤分子靶向治疗的关键之一。目前抑制肿瘤新生血管形成的分子靶向药物主要是响应VEGF-VEGFR信号途径,在临床肿瘤治疗中虽然有可靠的疗效,但是其抑制肿瘤生长的作用往往是暂时性的。这些原因促使人们努力建立新的抗肿瘤新生血管靶向治疗新策略和新药物。文献报道和申请人课题组的前期研究提示Notch信号途径在肿瘤新生血管结构形成中发挥重要作用。虽然阻断Notch信号途径可以破坏肿瘤新生血管形成而抑制肿瘤生长,但载体内阻断Notch信号可引起内脏多发性血管瘤,因此,这一思路难以应用于临床治疗。
肝细胞癌属于多血管实体瘤,其发生发展过程与肿瘤血管生成密切相关。其血管生成是一个涉及多种调节因子的复杂过程,受血管生成促进因子及抑制因子的平衡所控制。因此,通过阻断血管生成促进因子的作用、增强血管生成抑制因子的活性等方式可达到治疗肿瘤的目的。而血管生成通过许多信号通路受到许多血管生成因子的调控。
课题组前期筛选出了hD1R、hD4R多肽,并将该多肽链接了RGD导向肽,这两种配体在VEGF依赖性和VEGF非依赖性的抑制血管生成作用中起到了重要作用。虽然多肽类药物的生物活性较高、特异性强,与受体的亲和性好,且毒性较低,对人体的损伤更小,具有独特的优势。但从另一方面来说,与小分子药物相比,多肽类药物最大的缺点就是稳定性较差,很容易在体内进行降解,在酸、碱、高温、有机溶剂等条件下容易失活,其检测方法也因本身的不足而受到限制。
近年来,随着基因转移技术的日趋成熟,基因治疗己经成为生物科学和临床医学的研究热点之一。纳米粒基因转运体是近年发展起来的一种新型的非病毒基因转运载体。它是将DNA、RNA等基因治疗分子包裹在纳米颗粒之中或吸附在其表面,同时也在颗粒表面偶联特异性的靶向分子,通过靶向分子与细胞表面特异性受体结合,在细胞摄取作用下进入胞内,实现安全有效的靶向性基因治疗。筛选肝癌细胞特异性靶向分子,并将其与纳米基因载体结合,己成为提高纳米基因载体肝癌治疗效果的关键问题之一。
当前,还未有任何报道揭示以Dll4-Notch4为靶点的微泡/微球/微粒/脂质体抗新生血管相关报道。
发明内容
为了克服上述现有技术的缺点,本发明的目的在于提供一种用于癌症靶向治疗的短肽及基于其的超声响应纳米载药微泡和应用。
为了达到上述目的,本发明采用以下技术方案予以实现:
本发明公开了一种用于癌症靶向治疗的短肽,其氨基酸序列如SEQ.ID.NO.1所示。
本发明还公开了一种超声响应纳米载药微泡,该超声响应纳米载药微泡是将上述的短肽修饰于SA-PEG纳米粒表面,得到SA-PEG2000-9肽纳米载药微泡。
优选地,该超声响应纳米载药微泡的粒径为500~10000nm。
进一步优选地,该超声响应纳米载药微泡的平均粒径为679±247nm。
本发明还公开了上述的用于癌症靶向治疗的短肽在制备用于治疗肿瘤的药物中的应用。
本发明还公开了上述的超声响应纳米载药微泡在制备用于治疗肿瘤的药物中的应用。
优选地,所述的药物为在超声辐照作用下显著抑制新生血管生产的药物。
进一步优选地,所述的药物为降低肿瘤细胞增殖的药物。
进一步优选地,所述的药物为促进肿瘤细胞坏死凋亡的药物。
与现有技术相比,本发明具有以下有益效果:
本发明公开了一种能够用于癌症靶向治疗的短肽,证实其是一段由9个氨基酸组成的新型短肽。同时,本发明采用超声辐照联合载药靶向微泡原理,建立了基于该新型短肽的针对新生血管的分子靶向超声响应纳米载药微泡。载药纳米粒表面羧基化的PEG末端与新型多肽(H2N-LCKKRNDHF-CONH2)末端氨基酸的NH2结合形成酰胺键,末端氨基的连接方法降低了对新型多肽的构象及活性的影响,尽可能地保护了新型多肽与血管内皮细胞表面高表达的Dll4配体的识别和结合能力。相对于之前的hD4R肽段来说,将其中的有效活性成分使用纳米递药系统与超声介导结合将会提供一种新型、高效的抗肿瘤血管生成模式。不仅可以联合超声定向爆破微泡来阻断肿瘤新生血管形成,同时还可以消除肿瘤新生血管结构异常造成不良后果,如生长因子和炎性因子增加造成的肿瘤生长和侵袭转移。本研究策略可望建立新型的新生血管分子靶向策略和药物,为探索新生血管形成的细胞和分子机制提供方法,为治疗包括肿瘤在内新生血管形成相关疾病提供新手段,具有重要的理论和实际意义。

下面结合附图对本发明做进一步详细描述:
1、提取新型活性短肽
参见图1,为hDll4结构域示意图,图1中左边部分显示的是本课题组前期研究多肽并成功制备RGD导向肽,右边部分显示的是提取得到的短肽片段(9肽,LCKKRNDHF,氨基酸序列如SEQ.ID.NO.1所示)位点为187-195。
2、超声介导下SA-PEG化学合成载体+hDR4活性片段提取9肽
参见图2,通过DSPE-PEG2000修饰的方法,本实验采用NH2末端PEG化技术(N-terminal PEGylation)将具有靶向肿瘤血管内皮细胞表面Dll4配体的活性多肽hD4R中的活性肽段L→F(LCKKRNDHF,氨基酸序列如SEQ.ID.NO.1所示,提取活性片段部分由上海强耀生物科技有限公司提供),修饰于SA-PEG纳米粒表面,载药纳米粒表面羧基化的PEG末端与上述新型短肽(H2N-LCKKRNDHF-CONH2)末端氨基酸的NH2结合形成酰胺键,末端氨基的连接方法降低了对新型多肽的构象及活性的影响,尽可能地保护了新型多肽与血管内皮细胞表面高表达的Dll4配体的识别和结合能力。可以看出,相对于之前的hD4R肽段来说,将其中的有效活性成分使用纳米递药系统与超声介导结合将会提供一种新型、高效的抗肿瘤血管生成模式。
3、在体外观察新型多肽纳米递药系统(SA-PEG2000-9肽)的理化性质、以及理化性质的检测
1)Maldi-tof结构验证为SA-PEG2000-9肽
SA-PEG2000-L→F化学结构式如下:
参见图3,显示的是SA-PEG2000-L→F段的飞行时间质谱(Maldi-tof)图,从图中可以看出,根据计算直线肽LCKKRNDHF的分子量是加上聚合物,最终的分子量为3360.37,与Maldi-tof图谱表征相符,证实了该结构成功制备。
2)扫描电子显微镜下观察SA-PEG2000-9肽纳米微泡的粒径、大小与分布
参见图4,其中,(a)为SA-PEG2000-9肽的扫描电镜图;(b)为其粒径分析图。可以得出,SA-PEG2000-9肽纳米微泡的粒径分布为500~10000nm,平均粒径为679±247nm。
3)免疫荧光染色分析共聚焦电镜分析图
操作步骤如下:
3.1)标本准备
a.培养细胞系(人内皮细胞系、肿瘤细胞株)
细胞株在96孔板上直接培养、处理、固定和染色→吸干液体,随后在细胞上覆盖一层约2-3mm,用温热PBS稀释的4%甲醛→室温下固定细胞15分钟→吸干固定液,用1×PBS漂洗三次,每次5分钟→继续进行免疫染色操作。
3.2)免疫染色(所有随后的孵育都应当在室温下完成,以防止干燥和荧光物质淬灭)
在封闭缓冲液中封闭标本60分钟→封闭标本时,按照数据表中推荐的稀释比例,在抗体稀释缓冲液中配制一抗→吸去封闭缓冲液,加入稀释后的一抗→4℃孵育过夜→用1×PBS漂洗三次,每次5分钟→用抗体稀释缓冲液将荧光物质标记的二抗稀释后,室温下避光孵育标本1–2小时→用1×PBS漂洗三次,每次5分钟→使用 Gold AntifadeReagent或 Gold Antifade Reagent with DAPI,用盖玻片盖住切片→封片液室温过夜。
激光共聚焦电镜下观察,结果参见图5,为SA-PEG2000-9肽的共聚焦电镜图,从图中可以看出,在人血管内皮细胞中(蓝色标记为细胞,绿色为FITC标记的荧光微泡)。说明该微泡具备靶向标记人血管内皮细胞的作用。
4)超声联合SA-PEG2000-9肽纳米微泡能够抑制人内皮细胞、肿瘤细胞株增殖
结果参见图6,从图中可以看出,超声联合SA-PEG2000-9肽纳米微泡对细胞株新生血管有明显的抑制作用,使肿瘤细胞增殖明显减少,在超声的辐照作用下细胞促进细胞坏死凋亡。说明,超声辐照联合SA-PEG2000-9肽纳米微泡后促使多肽释放,具有定向靶向肿瘤细胞的能力。
