Abstract
本发明提供了一种补充全营养的组合物及其制备方法,涉及特殊食品领域。本发明提供的补充全营养的组合物包括:蛋白质、脂肪、碳水化合物、膳食纤维、维生素和矿物质,能够补充人体所需的各种营养,满足人体生长发育和代谢,同时还具有增强机体免疫力和降血脂药物的药理活性的作用。另外,本发明通过调整维生素C和肌醇的比例,显著提高了降血脂药物的药理作用。

背景技术:
随着国民经济的快速发展,人民生活水平不断提高,人们在日常饮食中越来越注重营养。食物与营养是人类生存的基本条件,人类生命活动所需营养种类繁多,比如:维生素、矿物质、蛋白质、脂肪和碳水化合物等。但没有一种天然食品或传统食品含有维持人体健康所必需的全部营养素。
α-亚麻酸和亚油酸属于多不饱和脂肪酸,是人体必须脂肪酸,人体不能自身合成,也是无法由其他营养来合成的,必须要依靠膳食来获得。α-亚麻酸是是构成人体脑细胞和组织细胞的重要成分,人体一旦缺乏α-亚麻酸,就会引起人体脂质代谢紊乱,导致免疫力降低、健忘、疲劳、视力减退、动脉粥样硬化等症状的发生。尤其是婴幼儿、青少年,如果缺乏α-亚麻酸,就会严重影响其智力和视力的发育。α-亚麻酸还有益于预防和治疗癌症、心脑血管病、糖尿病、类风湿病、皮炎症、抑郁症、精神分裂症、老年痴呆症等疾病。亚油酸有“血管清道夫”的美誉,具有防治动脉粥样硬化及心血管疾病的功效,能够降低血脂、软化血管、降低血压、促进微循环的作用,可预防或减少心血管病的发病率。另外,大豆磷脂被誉为与蛋白质、维生素并列的“第三营养素”,对改善记忆、保护肝脏、防止衰老,恢复细胞功能有着非常重要的作用。胆碱也具有帮助增进记忆力、促进脂肪和胆固醇代谢的作用。肌醇同样也有促进脂肪和胆固醇代谢的作用,能够降低胆固醇,有助预防动脉硬化,一般与胆碱及其他B族的维生素同时服用。
目前,人们还普遍存在部分维生素营养和某些微量元素不足的问题,影响人民的身体健康,使一些与营养有关的疾病的发病率居高不下。因此,为了平衡天然食品中某些营养素的不足,提高食品的营养价值,补充人体对营养素的需要和防止由于缺乏某种天然营养素所导致的各种特定疾病等,开发一种能够补充全营养的组合物是非常有必要的,尤其是对于比较挑食的人群和与营养有关疾病的患者而言。
发明内容:
本发明的目的在于提供一种补充全营养的组合物及其制备方法。本发明提供的组合物在补充人体所需营养的同时,同时还具有增强机体免疫力药物和降血脂药物的药理活性的作用。
本发明一方面提供了一种补充全营养的组合物,所述的组合物包括:蛋白质10-30g、脂肪10-40g、碳水化合物30-55g、膳食纤维3-10g、维生素0.15-1.8g和矿物质1.2-8g。
优选地,所述的蛋白质是大豆分离蛋白、乳清蛋白、酪蛋白、牛奶蛋白、水解乳清蛋白、大豆肽、胶原蛋白肽和鱼低聚肽中的一种或几种;
所述的脂肪是植物油、鱼油、中链甘油三酯、亚油酸和α-亚麻酸中的一种或几种;
所述的碳水化合物是葡萄糖、结晶果糖、淀粉、预胶化淀粉和麦芽糊精中的一种或几种;
所述的膳食纤维是聚葡萄糖、低聚果糖和菊粉中的一种或几种;
所述的维生素包括维生素A、维生素D、维生素E、维生素K、维生素B1、维生素B2、维生素B6、维生素B12、维生素C、烟酸、叶酸、泛酸、生物素、胆碱和肌醇。

(1)将蛋白质、脂肪、碳水化合物和膳食纤维的原料60℃±2℃干燥1.5-2h,检测水分,干燥湿重1%-3%,粉碎,过80目筛,分别按重量份称量,混合均匀,得到混合物A;
(2)将维生素和矿物质的原料60℃±2℃干燥1.5-2h,检测水分,干燥湿重1%-3%,粉碎,过80目筛,分别按重量份称量,采用梯度递增的方式将粉筛后的维生素和矿物质混合均匀,得到混合物B;
(3)将混合物A和混合物B混合均匀,加入磷脂和助流剂,得到混合物C;
(4)将步骤(3)得到的混合物C制成粉剂、颗粒剂、片剂或胶囊剂中的一种。
具体的,一种上述组合物的制备方法,包括以下步骤:
(1)将蛋白质、脂肪、碳水化合物和膳食纤维的原料均匀摊在洁净的不锈钢料盘中加入干燥机,60℃±2℃干燥1.5-2h,检测水分,干燥湿重1%-3%,万能粉碎机粉碎,并用高效筛粉机过80目筛,分别按重量份称量,多向运动混合机转速30Hz,混合时间30min,混合均匀,得到混合物A;
(2)将维生素和矿物质的原料均匀摊在洁净的不锈钢料盘中加入干燥机,60℃±2℃干燥1.5-2h,检测水分,干燥湿重1%-3%,万能粉碎机粉碎,并用高效筛粉机过80目筛,分别按重量份称量,采用梯度递增的方式将粉筛后的维生素和矿物质加入到多向运动混合机混料筒内,每次扩充量为上次物料量的5倍,多向运动混合机转速30Hz,混合时间30min,混合均匀,得到混合物B;
(3)将混合物A和混合物B在多向运动混合机中转速30Hz,混合时间30min,混合均匀,加入磷脂和助流剂混合均匀,得到混合物C;
(4)将步骤(3)得到的混合物C制成粉剂、颗粒剂、片剂或胶囊剂中的一种。

下述实施例中的实验方法,如无特殊说明,均为常规方法,下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
基础实施例
一种补充全营养的组合物包括:蛋白质10-30g、脂肪10-40g、碳水化合物30-55g、膳食纤维3-10g、维生素0.15-1.8g和矿物质1.2-8g。
上述组合物的各营养组分的组成和原料见表1,每种营养组分的原料可选自表1提供的原料中的一种或几种。

实施例1
一种补充全营养的组合物包括:蛋白质10g、脂肪10g、碳水化合物30g、膳食纤维3g、维生素0.13g和矿物质1.2g。
其中,蛋白质为牛奶蛋白10g;脂肪为鱼油10g;碳水化合物为葡萄糖30g;膳食纤维为菊粉3g;维生素包括维生素A190μg、维生素D 4μg、维生素E 4mg、维生素K 2μg、维生素B10.4mg、维生素B2 0.4mg、维生素B6 0.4mg、维生素B12 0.6μg、维生素C 22.4mg、烟酸1mg、叶酸110μg、泛酸1.4mg、生物素10μg、胆碱55mg和肌醇15mg;矿物质包括钠300mg、钾450mg、铜220μg、镁89.6mg、铁4mg、锌2mg、锰120μg、钙200mg、磷154mg、碘12μg、硒16μg、铬8μg和钼30μg。
上述组合物的制备方法,包括以下步骤:

一种补充全营养的组合物及其制备方法,与实施例7的区别仅在于,肌醇200mg、维生素C 200mg,即肌醇和维生素C重量比为1:1。

一种补充全营养的组合物及其制备方法,与实施例7的区别仅在于,肌醇500mg、维生素C 50mg,即肌醇和维生素C重量比为10:1。

实验例1
降血脂效果的试验:
样品:实施例1-9和对比例1-4所述组合物。
高脂饲料配制重量比为:78.8%基础饲料,1%胆固醇,10%蛋黄粉,10%猪油,0.2%胆盐。
试验动物:选用体重为100-150g的无特定病原体SPF级Wistar大鼠,随机平均分为实验组(实施例1-9和对比例1-4以及血脂康组,共14组)、高脂对照组和普通对照组,每组10只,雌雄各半。
试验方法:实验组和高脂对照组喂养高脂饲料,普通对照组喂养基础饲料,自由饮食、饮水,连续喂养30天。将受试物与1mL生理盐水混合,实验组大鼠灌胃受试物(即:实施例1-9和对比例1-4所述组合物以及血脂康组),小鼠给各试验组剂量为4.0g/kg,同时给予血脂康(北京北大维信生物科技有限公司生产)0.02g/kg,血脂康组只给予血脂康0.02g/kg,高脂对照组和普通对照组给予等量生理盐水。每天观察动物的表现、行为、毒性表现和死亡情况。试验前对大鼠编号,并称量试验前后大鼠体重,喂养30天后,禁食10小时,大鼠腹部主动脉采集血液、分离血清,利用酶法试剂盒检测大鼠的总胆固醇(TCH)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)含量,结果见表2。

表2结果表明,本发明提供的补充全营养的组合物与血脂康组合后能够进一步降低大鼠总胆固醇、甘油三酯和低密度脂蛋白胆固醇的含量,具有促进脂肪代谢,降低血脂的效果,可有效预防高血脂、粥样动脉硬化、心脑血管等相关疾病。
实验例2
增强免疫力效果的试验:
实验动物选用SPF级昆明种雌性小白鼠和雄性小白鼠,体重18-22g。
受试样品为按照本发明实施例1-9、对比例1-4所述组合物、贞芪扶正颗粒组,小鼠给药剂量为4.2g/kg。自由饮水、饮食,动物实验前观察适应5天,复查合格后开始进入正式实验。
(1)实验分组:雌雄小鼠均随机均分为15组,即实施例1-9雄性组、对比例1-4雄性组、贞芪扶正颗粒雄性组、生理盐水对照雄性组;实施例1-9雌性组、对比例1-4雌性组、贞芪扶正颗粒雄性组和生理盐水对照雌性组。每组10只小鼠;在整个实验期内,实施例1-9雄/雌性组、对比例1-4雄/雌性组各组每天分别用1mL生理盐水按照4.0g/kg灌服实施例1-9和对比例1-4的组合物,同时给予贞芪扶正颗粒0.17g/kg,贞芪扶正颗粒组只给予贞芪扶正颗粒0.17g/kg;生理盐水对照雄/雌性组均每天灌服1mL生理盐水。小鼠自由采食、饮水,饲养过程中,每天换水,隔日换笼中的木屑垫料以保持清洁。环境温度20-25℃,至灌胃起到实验结束共15天。
(2)鼠血清的制备:末次灌胃后24h,各组小鼠摘除眼球后眼眶取血1-2mL,置于事先装有抗凝的试管中,3000rpm,离心10min,取上清液作为待测血清备用。
(3)测定指标和方法:溶菌酶是正常机体免疫防御机制的组成部分,它存在于中性粒细胞、单核细胞和巨噬细胞内,也存在于黏膜分泌液中,是体表防御因素之一。采用南京建成生物工程研究所的A050-1溶菌酶(LZM)测试盒(带标准)测定各组小鼠血清溶菌酶含量,计算品均值,测定结果见表3。

由表3可以看出,表3中实施例1-9雄/雌性组的血清溶菌酶含量均高于生理盐水对照组的血清溶菌酶含量,说明本发明实施例1-9所制备的补充全营养的组合物具有进一步增强参芪扶正颗粒免疫力的效果。尤其是实施例7所述的组合物,其增强免疫力效果最为显著。

Claims (8)
1.一种补充全营养的组合物,其特征在于,所述的组合物包括:蛋白质10-30g、脂肪10-40g、碳水化合物30-55g、膳食纤维3-10g、维生素0.15-1.8g和矿物质1.2-8g。
2.根据权利要求1所述的组合物,其特征在于,所述的蛋白质是大豆分离蛋白、乳清蛋白、酪蛋白、牛奶蛋白、水解乳清蛋白、大豆肽、胶原蛋白肽和鱼低聚肽中的一种或几种;
所述的脂肪是植物油、鱼油、中链甘油三酯、亚油酸和α-亚麻酸中的一种或几种;
所述的碳水化合物是葡萄糖、结晶果糖、淀粉、预胶化淀粉和麦芽糊精中的一种或几种;
所述的膳食纤维是聚葡萄糖、低聚果糖和菊粉中的一种或几种;
所述的维生素包括维生素A、维生素D、维生素E、维生素K、维生素B1、维生素B2、维生素B6、维生素B12、维生素C、烟酸、叶酸、泛酸、生物素、胆碱和肌醇;
所述的矿物质包括钠、钾、铜、镁、铁、锌、锰、钙、磷、碘、硒、铬和钼。
3.根据权利要求1所述的组合物,其特征在于,所述的蛋白质包括大豆分离蛋白2-20g和乳清蛋白5-25g;
所述的脂肪包括中链甘油三酯7.5-22g、亚油酸2-10g和α-亚麻酸0.5-2g;
所述的碳水化合物包括结晶果糖10-35g和麦芽糊精15-30g。
4.根据权利要求1所述的组合物,其特征在于,所述的维生素包括维生素A190-1000μg、维生素D 4-14μg、维生素E 4-50mg、维生素K 22-70μg、维生素B1 0.4-2mg、维生素B2 0.4-2mg、维生素B6 0.4-2mg、维生素B12 0.6-4μg、维生素C 30-300mg、烟酸1-6mg、叶酸110-500μg、泛酸1.4-9mg、生物素10-50μg、胆碱105-770mg和肌醇20-600mg;
所述的矿物质包括钠400-2000mg、钾550-2000mg、铜220-2200μg、镁90-800mg、铁4-10mg、锌2-10mg、锰120-2500μg、钙300-1500mg、磷200-1500mg、碘12-50μg、硒16-100μg、铬8-250μg和钼30-200μg。
5.根据权利要求4所述的组合物,其特征在于,所述的肌醇和维生素C重量比为1-10:1。
6.根据权利要求1所述的组合物,其特征在于,所述的组合物还包括磷脂和助流剂;所述的磷脂是大豆磷脂;所述的助流剂是二氧化硅。
7.根据权利要求6所述的组合物,其特征在于,所述的磷脂为大豆磷脂4-6g;所述的助流剂为二氧化硅0.08-0.12g。
8.一种根据权利要求1-7任意一项所述的组合物的制备方法,其特征在于,所述的制备方法包括以下步骤:
(1)将蛋白质、脂肪、碳水化合物和膳食纤维的原料60℃±2℃干燥1.5-2h,检测水分,干燥湿重1%-3%,粉碎,过80目筛,分别按重量份称量,混合均匀,得到混合物A;
(2)将维生素和矿物质的原料60℃±2℃干燥1.5-2h,检测水分,干燥湿重1%-3%,粉碎,过80目筛,分别按重量份称量,采用梯度递增的方式将粉筛后的维生素和矿物质混合均匀,得到混合物B;
(3)将混合物A和混合物B混合均匀,加入磷脂和助流剂,得到混合物C;
(4)将步骤(3)得到的混合物C制成粉剂、颗粒剂、片剂或胶囊剂中的一种。
