Abstract

本发明属于生物医药技术领域,具体涉及一种白及胶低损耗干燥方法,将采挖后的新鲜白及在温度17℃‑35℃,湿度60%‑80%,光照强度13000lux‑20000lux下晒5‑7天,之后在温度55℃,湿度大于95%,光照强度5000lux‑8000lux下阴干1‑2天后,55℃烘干,得到的白及胶含量为68.1‑69.5%,较传统的白及烘干方法,本发明大幅保留了白及中白及胶的有效含量,因此,白及采用晴天晒+阴雨天55℃烘干的干燥方法,可以有效减少白及在干燥过程中所引起的白及胶的耗损。

一种白及胶低损耗干燥方法
技术领域
本发明属于生物医药技术领域,具体涉及一种白及胶低损耗干燥方法。
背景技术
白及(Bletilla striata),为兰科植物白及Bletilla striata(Thunb.)Reichb.f.的干燥块茎,白及胶是白及止血的活性成分,其由4分子甘露糖和1分子葡萄糖组成,在水中溶解时受分子间氢键作用的影响,易形成亲水性凝胶,达到药辅合一的目的。白及胶具有调节身体代谢,调节细胞中生长因子的含量,能增强血小板第三因子活性,显著缩短凝血时间及凝血酶原形成时间,抑制纤维蛋白溶酶活性,对局部出血有很好的治疗效果;同时白及胶还具有抗溃疡作用,并可治疗胃及十二指肠溃疡。由于白及有高度钻性,可在胃内形成胶状膜,从而促使溃疡面愈合;其次白及胶还具有消炎抗菌的功效,白及中的有益成分对葡萄球菌及链球菌有抑菌作用,可在局部形成一保护膜,能控制及防止胃部感染;再次白及胶还可以作为辅料添加到药品中,延长主药的停滞时间,降低药物的毒性。
现有技术通常采用首先直接烘干白及药材随后提取的方式获得白及胶,但该方法在直接烘干过程中存在损耗,导致获得的白及胶及多糖含量均不高。
发明内容
本发明要解决的技术问题是:减少直接烘干白及药材所引起的耗损,提高了白及胶的含量。
为了解决上述技术问题,本发明采用如下的技术方案:
一种白及胶低损耗干燥方法,其包括如下步骤:
1)将新鲜的白及在温度17℃-35℃,湿度60%-80%,光照强度13000lux-20000lux下晒5-7天;
2)将步骤1)中获得白及在温度16℃-23℃,湿度大于95%,光照强度5000lux-8000lux下放置1-2天,再烘干;
3)将步骤2)中获得的白及研磨成白及粗粉;
4)将步骤3)中获得的白及粗粉加水回流提取2-5次,每次提取1-3小时,提取液减压浓缩后,加乙醇使其含醇量为70-90%,放置过夜,离心5-10min,沉淀相继用无水乙醇,丙酮依次洗涤3-5次,同法离心,弃去上清液,置50-70℃烘干得白及胶干粉。
优选地,步骤1)在晴天下进行;
优选地,步骤2)在阴雨天进行;
优选地,步骤2)所述的烘干温度为50-60℃,优选55℃;
优选地,步骤3)中使用普通粉碎机粉碎成粗粉,过80~100目筛,优选90目筛;
优选地,步骤4)中离心转速为9000-12000转/min;
通过该方法可以有效减少白及在干燥过程中所引起的白及胶的耗损。
具体实施方式
实施例1
1、试验材料:贵阳市贵州省农业科学现代中药材研究所试验地生长5年的白及。
2、试验方法:
1)将新鲜白及洗净,除去茎、叶、根;
2)将步骤1)获得的白及在晴天的情况下于温度25℃、湿度70%、光照强度16000lux下晒6天;
3)将步骤2)获得的白及在阴雨天的情况下于外界环境温度16℃、湿度96%、光照强度5000lux下放置1天,50℃烘干;
4)将步骤3)中获得的白及使用普通粉碎机粉碎成粗粉,过90目筛;
5)将步骤4)获得的白及粗粉加水回流提取3次,每次提取2小时,提取液减压浓缩后,加乙醇使其含醇量为75%,放置过夜,离心5min(10000转/min),沉淀相继用无水乙醇,丙酮依次洗涤3次,同法离心,弃去上清,置60℃烘干得白及胶干粉。
白及75%乙醇提取率检测样品处理:取本品粉末约5g,精密称定,置150mL锥形瓶中,加75%乙醇100mL,80℃加热回流提取2次,每次1小时。合并提取液,在水浴上蒸干后,于105℃干燥3小时,置干燥器中冷却30分钟,迅速精密称定重量。
多糖检测方法:采用水提醇沉制备供试品溶液,以0.2%蒽酮-硫酸显色,紫外分光光度法490nm测定多糖含量。
实施例2
将步骤1)中获得的白及在晴天的情况下于温度17℃、湿度80%、光照强度20000lux下晒5天;其它与实施例1相同。
实施例3
将步骤1)获得的白及在晴天的情况下于温度25℃、湿度60%、光照强度13000lux下晒7天;其它与实施例1相同。
实施例4
将步骤2)获得的白及在阴雨天的情况下于温度20℃、湿度98%、光照强度8000lux下放置1.5天,55℃烘干;其它与实施例1相同。
实施例5
将步骤2)获得的白及在阴雨天的情况下于温度23℃、湿度96%、光照强度6500lux下放置2天,60℃烘干;其它与实施例1相同。
对比例1
将步骤2)及步骤3)中的晒及烘干步骤替换为在55℃烘干,其它与实施例1相同。
对比例2
将步骤2)及步骤3)中的晒及烘干步骤替换为在75℃烘干,其它与实施例1相同。
测得的实施例及对比例的白及胶及多糖含量如下表1所示:

试验结果表明:白及55℃烘干,白及胶含量为43.5%;白及75℃烘干,白及胶含量为48.2%;而白及采用晴天晒+阴雨55℃烘干,得到的白及胶含量为68.1-69.5%,差异明显,可见,通过本申请的方法,可以有效减少白及在干燥过程中所引起的白及胶的耗损。

Claims (7)

1.一种白及胶低损耗制备方法,其包括如下步骤:
1)将新鲜的白及在温度 17℃-35℃,湿度60%-80% ,光照强度13000lux-20000lux下晒5-7天;
2)将步骤1)中获得白及在温度16℃-23℃,湿度大于95% ,光照强度5000lux-8000lux下放置1-2天,烘干;
3)将步骤2)中获得的白及研磨成过80~100目筛的白及粉;
4)将步骤3)中获得的过80~100目筛的白及粉加水回流提取2-5次,每次提取1-3小时,提取液减压浓缩后,加乙醇使其含醇量为70-90%,放置过夜,离心5-10min,沉淀相继用无水乙醇,丙酮依次洗涤3-5次,同法离心,弃去上清液,置50-70℃烘干得白及胶干粉。
2.根据权利要求1所述的白及胶低损耗制备方法,其特征在于,步骤1)在晴天下进行。
3.根据权利要求1-2任一项所述的白及胶低损耗制备方法,其特征在于,步骤2)在阴雨天进行。
4.根据权利要求3所述的白及胶低损耗制备方法,其特征在于,步骤2)所述的烘干温度为50-60℃。
5.根据权利要求1-2任一项所述的白及胶低损耗制备方法,其特征在于,步骤3)中使用普通粉碎机粉碎。
6.根据权利要求5所述的白及胶低损耗制备方法,其特征在于,步骤3)中使用普通粉碎机粉碎成过90目筛的白及粉。
7.根据权利要求1-2任一项所述的白及胶低损耗制备方法,其特征在于,步骤4)中离心转速为9000-12000转/min。
