Abstract
本发明提供了一种抗肿瘤纳米药物及其制备方法,制备步骤包括:采用Fmoc‑固相合成方法合成短肽HCPT–FFpYG‑PMI;采用酶促多肽自组装的方法,在所述短肽HCPT–FFpYG‑PMI中加入PBS溶液,利用碳酸盐溶液调节pH为7.1~7.9;之后,加入3U/mL~7U/mL的碱性磷酸酶后置于2℃~6℃条件下过夜存放,即得到所述抗肿瘤纳米药物。本发明中的抗肿瘤药物是种基于细胞核靶向肽的药物‑多肽两亲性化合物,该多肽以临床用抗癌药物十羟基喜树碱封端,在碱性磷酸酶的催化作用下可以形成纳米纤维,能够起到协同效应更好地将抗癌药物递送到细胞核,在体外癌实验抑制中显示了很好的癌细胞抑制效果,在小鼠肿瘤模型中也显示出很好的抗肿瘤效应,而其他对照化合物抗肿瘤效应明显降低。

技术领域
本发明涉及一种纳米药物,具体涉及一种抗肿瘤纳米药物及其抗肿瘤药物的制备方法。
背景技术
癌症在全球的发病率依然在迅速增长。各类癌症的发生与发展对人类健康影响重大,也严重阻碍了经济的发展。积极寻找癌症治疗的新策略变得刻不容缓。而大多数临床用抗癌药的作用机制都是抑制或者阻碍与DNA复制相关的酶,而细胞核是细胞生命活动的控制中心。因此大多数抗癌药物必须被递送至细胞核才能更好地发挥其抗癌功效。因此寻找高效的抗癌药物递送手段也引起了科研工作者的广泛关注。
p53是我们熟知的肿瘤抑制基因,在许多癌细胞中会存在此基因表达活性的下降。因此其表达活性的高低在癌症的发生与发展过程中起着很重要的作用。MDM2与MDMX是p53抑癌基因转录活性与稳定性的重要调控因子,抑制其与p53的结合活性可有效激活p53抑癌基因,从而促进肿瘤细胞的凋亡,达到有效的抑制肿瘤的效果。因此,寻找与MDM2核MDMX有效结合的配体,是肿瘤抑制的手段之一。
癌症治疗在传统意义上包括化疗、放疗和手术切除等。但是这几种传统方法存在着很大的弊端,比如副作用强,对正常组织选择性低,而且还存在术后恢复和病人体质差异等各方面的局限性。所以,寻找快速有效的癌症治疗方法变得刻不容缓。近年来,小分子多肽自组装的水凝胶由于其容易制备且生物相容性好等不可比拟的优点,目前已经在癌症诊断与治疗方面显示出了极大的潜力。我们前期一些研究工作主要围绕基于抗癌药物封端的短肽,通过加热冷却,酶催化,离子调控或者pH调控等方法,形成性质优良的的水凝胶并应用于抗癌药物递送,达到了很好的抗肿瘤效果。因此,在之前的研究基础上开发新的临床应用的抗癌药很有必要,以达到更好的抗肿瘤效果。
发明内容
本发明旨在至少解决现有技术中存在的技术问题之一,提供一种抗肿瘤纳米药物及其一种抗肿瘤纳米药物的制备方法。
本发明的第一方面,提供一种抗肿瘤纳米药物的制备方法,包括:采用Fmoc-固相合成方法合成短肽HCPT–FFpYG-PMI;
采用酶促多肽自组装的方法,在所述短肽HCPT–FFpYG-PMI中加入PBS溶液,利用碳酸盐溶液调节pH为7.1~7.9;之后,加入3U/mL~7U/mL的碱性磷酸酶后置于2℃~6℃条件下过夜存放,即得到所述抗肿瘤纳米药物。

将2-Cl-Trt树脂与二氯甲烷混合在固相合成器中,使所述2-Cl-Trt树脂充分溶胀,并将所述二氯甲烷从所述固相合成器中移除;
将第一Fmoc保护的酪氨酸溶解在无水二氯甲烷,加入1mmol~3mmol的N,N-二异丙基乙胺,充分溶解后转移至所述固相合成器中,在室温下反应1h~2h;
除去所述固相合成器中的反应液,再加入体积比为所述无水二氯甲烷∶甲醇∶N,N-二异丙基乙胺=8.2~8.7∶1.5~2.5∶0.5~1.5的封闭液5mL~15mL,在室温下封闭5min~15min;
除去所述固相合成器中的反应液,用N,N-二甲基甲酰胺洗涤,再加入体积百分比为15%~25%哌啶的所述N,N-二甲基甲酰胺溶液,切割所述第一Fmoc保护基,继续用所述N,N-二甲基甲酰胺洗涤切掉的所述第一Fmoc保护基;
将第二Fmoc保护的氨基酸、苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸酯和所述N,N-二异丙基乙胺溶解在所述N,N-二甲基甲酰胺中,充分溶解后加入所述固相反应器中反应;

本发明提供一种抗肿瘤纳米药物及其制备方法,在短肽-FFYG封端引入具有抗肿瘤效果临床用药十羟基喜树碱(HCPT),再与PMI序列连接形成短肽HCPT-FFpYG-PMI,之后在温和的酶促多肽自组装方法下,HCPT-FFpYG-PMI加ALP酶催化组通过自组装得到了更好的组装体以得到本发明的抗癌纳米药物。本发明通过封端引入HCPT以及连接核靶向肽PMI两者的协同作用,使药物更多地聚集在肿瘤部位,实现体内抗癌药物的有效递送,以实现更高的细胞摄取,因此可以达到更好的肿瘤抑制效果,这对于肿瘤的诊断及治疗有着重大的意义。

具体实施方式
为使本领域技术人员更好地理解本发明的技术方案,下面结合附图和具体实施方式对本发明作进一步详细描述。
实施例1
如图1所示,本发明的第一方面为抗肿瘤纳米药物的制备方法,具体包括以下步骤:采用Fmoc-固相合成方法合成短肽HCPT–FFpYG-PMI;
采用酶促多肽自组装的方法,在短肽HCPT–FFpYG-PMI中加入PBS溶液,利用碳酸盐溶液调节pH为7.1~7.9;之后,加入3U/mL~7U/mL的碱性磷酸酶后置于2℃~6℃条件下过夜存放,即得到抗肿瘤纳米药物。
需要说明的是,采用Fmoc-固相合成方法合成短肽HCPT–FFpYG-PMI具有包括:在短肽X-FFpYG封端引入HCPT,以形成短肽HCPT–FFpYG;短肽HCPT–FFpYG再与核靶向肽PMI序列链接,合成短肽HCPT–FFpYG-PMI。
具体地,第一步:采用Fmoc-固相合成方法合成短肽HCPT–FFpYG-PMI的具体步骤如下;
1)溶胀:称取0.5mmol 2-Cl-Trt树脂于固相合成管中,然后加入10mL二氯甲烷(以下均简称为DCM),放置在合成仪上摇晃10-15min,使2-Cl-Trt树脂充分溶胀;
2)用洗耳球把DCM从固相合成器中移除干净;
3)将2eq,即1mmol第一Fmoc保护的酪氨酸溶解在15mL的无水DCM里,加入4eq,即2mmol的N,N-二异丙基乙胺(以下均简称为DIEPA),充分溶解后转移到上述固相合成器中,在室温下反应1.5h;
4)封闭:用洗耳球除去固相合成器中的反应液,无水DCM洗3-5次,每次1min,共加入体积比为无水DCM∶甲醇∶DIEPA=8.5∶2∶1的封闭液10mL,在室温下封闭10min;
5)洗:用洗耳球除去固相合成器中的反应液,先用无水DCM洗涤4次,DCM用量为每次10mL,每次1min,再用N,N-二甲基甲酰胺(以下用DMF表示)洗涤3-5次,洗涤方法同上,加入10mL含体积百分比为20%哌啶的DMF溶液,切割第一Fmoc保护基30-40min,继续用DMF洗涤切掉的Fmoc,每次DMF用量10-15mL,1min/次,共洗3-5次,进行下一步反应;
6)加入的第二个Fmoc保护的氨基酸1mmol、苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸酯(HBTU)1mmol、DIEPA 2mmol,溶解在10ml DMF中,充分溶解后加入固相反应器中,反应2h;
7)DMF洗涤,每次DMF用量10mL,1min/次,共洗3-5次;然后哌啶切割保护基Fmoc35min,然后用DMF洗涤,重复加入氨基酸,直到将封端基团十羟基喜树碱加上;
8)封端基团反应完成后,DMF和DCM分别洗3-5次,每次用量均为10mL,每次1min,洗完后切割,按95%TFA,2.5%TIS,2.5%H2O体积百分比组成的溶液10mL加入到上述固相合成器中反应30min,把产物从2-cl-Trt树脂上切下,旋转蒸发器真空旋干,除去溶剂后用冰乙醚沉降,得到粗品,产率约80%左右。之后再用HPLC分离提纯冻干,得到短肽HCPT–FFpYG-PMI。
需要说明的是,短肽序列为短肽序列为X-FFpYG-PMI,其中为X为十羟基喜树碱(HCPT)。“短肽”为本领域常用术语,指由3~9个氨基酸残基组成的短链肽。另外,本实施例中的核靶向肽PMI序列为TSFAEYWNLLSP。
具体地,X为HCPT时,短肽HCPT–FFpYG-PMI结构式如下:

需要进一步说明的是,本实施例中所涉及的制剂来源如下:
N,N-二异丙基乙胺(DIEPA),西格马奥德里奇公司(Sigma-Aldrich),纯度98%;
苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸酯(以下用HBTU表示),购自吉尔生化(上海)有限公司,纯度98%;
2-Cl-Trt树脂,天津南开大学固相合成公司,活性1.1mmol/mL;
三氟乙酸(以下用TFA表示),西格马奥德里奇公司(Sigma-Aldrich),纯度99%;
碱性磷酸酶(ALP),百灵威科技有限公司;
三异丙基硅烷(TIS),西格马奥德里奇公司(Sigma-Aldrich),纯度99%;
十羟基喜树碱,百灵威科技有限公司,纯度99%;
培养基,DMEM,赛默飞世尔科技(ThermoFisher Scientific),无菌;
胎牛血清,赛默飞世尔科技(ThermoFisher Scientific),无菌;
L构型酪氨酸,吉尔生化(上海)有限公司,纯度98%;
以及本实施例中使用的癌细胞HepG2来源于我们杨志谋课题组,并按以下步骤培养:
1)紫外灭菌:胎牛血清、DMEM培养基、双抗、细胞培养皿、离心管等放入超净台中紫外灯照射半小时以上充分灭菌;
2)复苏与培养:从液氮罐中取出的冻存癌细胞HepG2,迅速置于37℃的水浴锅化开并迅速转移到超净台里(水浴锅提前升温至37℃)。用移液枪把含细胞的冻存液转移到含有2mL已配培养基的离心管中,1000rpm离心3-5分钟,弃去上清并用装有5-6mL已配培养基的培养皿中培养(已配培养基含有10%胎牛血清和1%双抗),然后放入37℃培养箱中培养,二氧化碳浓度为5%;
3)传代:次日观察细胞状态,状态良好且密度达到90%以上以后传代以后进行下面的实验。比如细胞铺板,肿瘤接种等。
第二步:采用酶促多肽自组装的形成抗肿瘤纳米药物,具体包括:取5mg短肽HCPT-FFpYG-PMI置于4mL的小玻璃瓶中,再加入PBS溶液(pH=7.4)体积为1mL,用1M的碳酸钠溶液将其pH值调节至7.4,充分混匀并且室温超声使其完全溶解,加入5U/mL的碱性磷酸酶后置于4℃条件下过夜存放,即可得到多肽自组装的抗肿瘤纳米药物。
需要说明的是,短肽HCPT-FFpYG-PMI在ALP的作用下得到的该抗肿瘤纳米药物的为序列为HCPT-FFYGPMI。
进一步需要说明的是,调控多肽自组装有许多种方法,我们最常用的有pH调控、加热-冷却和酶催化等。酶作为天然产物存在很多不可比拟的优势,比如特异性强,温和且高效等。为了制备药物-多肽两亲化合物,所述短肽首先具备良好的水溶性,磷酸化酪氨酸具有良好的亲水性因此我们将之引入多肽序列当中,为了使多肽更好地组装,我们将成胶因子FF引入多肽序列当中。在千分之五多肽浓度条件下,加入碱性磷酸酶后,4℃条件催化HCPT–FFpYG-PMI过夜后可形成水凝胶抗肿瘤纳米药物,这种酶促成胶(EISA)的方法简单实用、温和可控。
本发明的第二方面为根据上述方法制备得到一种抗肿瘤纳米药物,具体制备方法参考前文相关记载,在此不做赘述。
该抗肿瘤药物的结构式为:
相应的,该化合物的序列为HCPT-FFpYGTSFAEYWNLLSP(TSFAEYWNLLSP=PMI)
具体地,在同样的浓度,同样的制备条件下,我们发现HCPT–FFpYG-PMI经酶催化后得到稳定的水凝胶,并且该水凝胶在室温下放置48h依然很稳定。需要说明的是,实施例中通过本小瓶倒置的方法检验水凝胶的形成与否。另外,如图1所示,通过TEM检测发现HCPT–FFpYG-PMI酶催化的后的组装体形貌是纳米纤维状。
进一步地,如图2所示,将抗肿瘤纳米药物用于肿瘤治疗以评价药物的效果,具体评价过程如下:
(1)细胞的培养和小鼠肿瘤模型的建立
在无菌条件下培养HepG2肿瘤细胞,培养基为DMEM加10%胎牛血清和1%双抗,细胞密度达到后进行消化、离心、PBS清洗等步骤制备成500万/100μL的细胞悬液。取生长周龄为6-8周的Balb/c nude小鼠,在其左侧腋下进行皮下癌细胞移植,数量为500万/只小鼠,体积为100μL/只小鼠。
(2)给药治疗
待肿瘤大小长至100mm3时将小鼠随机分为四组,每组均有5只小鼠,分别记为PBS组(参考实施例4),HCPT组(参考实施例3),HCPT-FFpYG-PMI加ALP酶催化组,HCPT-FFYG-PMI加热冷却组(参考实施例2)。以第一次注射药物时间记为1天,取制备实施例1、实施例2、实施例3和实施例4中制得的不同组别的水凝胶或者溶液,将水凝胶按照抗癌药物给药的安全剂量稀释打散后成溶液后分别以100微升每只老鼠的剂量对小鼠进行尾静脉注射。之后分别在第4天,第7天进行同样剂量的尾静脉注射治疗。
(3)肿瘤大小监测
从第一次给药开始,就对小鼠肿瘤的大小和小鼠的体重进行追踪与监测,每两天对小鼠的肿瘤大小和体重变化进行一次记录,小鼠体重的变化可以反应出药品的安全性。肿瘤的长和宽用游标卡尺测量,体积的计算公式为V=ab2/2(其中a代表肿瘤的长度,b代表肿瘤的宽度)。
(4)结果分析
图2中纵坐标代表了不同组别的肿瘤体积大小,反映出了不同给药治疗组别的肿瘤抑制效果,横坐标代表了整个治疗过程的持续时间,不同曲线代表了不同的给药组别。整个过程一共给药三次,黑色箭头指向对应的不同时间点代表了给药治疗的时间点。
通过图2中分析曲线趋势我们可以看出,经过20天的治疗与追踪过后,PBS对照组(实施例4)的肿瘤平均体积达到1148.73mm3,HCPT-FFpYG-PMI加ALP酶催化组肿瘤相对体积是166.375mm3。相对于PBS组,肿瘤体积百分比为14.48%,相对抑制率为85.52%。由此我们可以看出,HCPT-FFpYG-PMI加ALP酶催化组有着最好的抑制效果,HCPT-FFpYG-PMI加ALP酶催化组通过自组装得到了更好的组装体,加上与核靶向肽的协同作用,使药物更多地聚集在肿瘤部位,实现更高的细胞摄取,因此可以实现更好的肿瘤抑制效果。
本实施例中采用酶促自组装方法以及将核靶向肽的优势结合起来,提高抗癌药物在肿瘤部位的富集,实现协同效应,达到更好的抗肿瘤效果,为肿瘤的检测与治疗提供了有价值的手段。
实施例2
按照实施例1的制备方法合成对照化合物HCPT-FFYG-PMI,具体合成步骤不再赘述,参考上述记载。
第一步,用Fmoc-固相合成方法合成对照化合物HCPT-FFYG-PMI,结构式如下:
相应的,该化合物的序列为:HCPT-FFYGTSFAEYWNLLSP。
对于该结构的表征数据如下:

第二步,取5mg短肽HCPT-FFYG-PMI置于4mL的玻璃瓶中,再加入PBS溶液(pH=7.4)总体积为1mL,用1M的碳酸钠溶液将其pH值调节至7.4,充分混匀并且室温超声使其尽量溶解,酒精灯上稍微加热到60℃~70℃之间,待完全溶解后室温放置几小时充分组装,即可得到多肽自组装的纳米结构药物。
需要说明的是,本实施例中将短肽HCPT-FFYG-PMI未经过碱性磷酸酶催化步骤,而是先经过加热再经降温处理后得到纳米结构药物,用于评价处理肿瘤的效果。
具体地,按照实施例1相同的条件下,即在同样的浓度,同样的制备条件下,我们发现HCPT-FFYG-PMI酶催化后先得到透明溶液,随着时间的推移逐渐变为沉淀。
进一步地,将上述得到的多肽自组装的纳米结构药物用于肿瘤治疗以评价药物的效果。评价过程与实施例1相同,在此不再赘述,参考上述记载。如图2所示,具体结果为HCPT-FFYG-PMI加热冷却的对照组处理的肿瘤相对体积是309.497mm3,相对于实施例4中的PBS组,肿瘤体积百分比分别为26.9%,相对抑制率为73.1%。
实施例3
取1mg抗癌药物十羟基喜树碱HCPT,加20μL二甲基亚砜助溶,剩余用PBS溶液(pH=7.4)补齐为1mL。用1M的碳酸钠溶液将其pH值调节至7.4,充分混匀并且室温超声使其完全溶解,用于小鼠尾静脉注射。
需要说明的是,本实施例中抗癌药十羟基喜树碱HCPT未与短肽及PMI结合,也未经碱性磷酸酶处理,直接将其用于评价抗癌效果。
进一步地,将上述得到的药物用于肿瘤治疗以评价药物的效果。评价过程与实施例1相同,在此不再赘述,参考上述记载。如图2所示,具体结果为HCPT对照组的肿瘤相对体积是962.252mm3,相对于实施例4中的PBS组,肿瘤体积百分比分别为83.8%,相对抑制率为16.2%。
实施例4
无菌1×PBS,作为空白对照组。评价过程与实施例1相同,在此不再赘述,参考上述记载。如图2所示,具体结果为PBS空白对照组的肿瘤平均体积达到1148.73mm3。
由实施例1至实施例4结果可知,HCPT-FFpYG-PMI加ALP酶催化组通过自组装得到了更好的组装体,加上与核靶向肽的协同作用,使药物更多地聚集在肿瘤部位,实现更高的细胞摄取,实施例2与实施例3中的组装体的抑制效果没有那么明显,实施例1中凝胶状态的抗肿瘤纳米药物HCPT-FFYG-PMI具有更好的稳定性,可以实现更好的肿瘤抑制效果。由此可知,将酶促自组装,核靶向肽的优势结合起来,可以显著提高抗癌药物在肿瘤部位的富集,实现协同效应,达到更好的抗肿瘤效果,相比实施例3中单独使用抗肿瘤药物的效果更好,为肿瘤的检测与治疗提供了有价值的手段。
可以理解的是,以上实施方式仅仅是为了说明本发明而采用的示例性实施方式,然而本发明并不局限于此。对于本领域内的普通技术人员而言,在不脱离本发明的精神和实质的情况下,可以做出各种变型和改进,这些变型和改进也视为本发明的保护范围。

2.根据权利要求1所述的抗肿瘤纳米药物的制备方法,其特征在于,所述采用Fmoc-固相合成方法合成短肽HCPT–FFpYG-PMI,包括:
在短肽X-FFpYG封端引入HCPT,以形成所述短肽HCPT–FFpYG;
所述短肽HCPT–FFpYG再与核靶向肽PMI序列链接,合成所述短肽HCPT–FFpYG-PMI。
3.根据权利要求1或2所述的抗肿瘤纳米药物的制备方法,其特征在于,所述采用Fmoc-固相合成方法合成短肽HCPT–FFpYG-PMI,包括:
将2-Cl-Trt树脂与二氯甲烷混合在固相合成器中,使所述2-Cl-Trt树脂充分溶胀,并将所述二氯甲烷从所述固相合成器中移除;
将第一Fmoc保护的酪氨酸溶解在无水二氯甲烷,加入1mmol~3mmol的N,N-二异丙基乙胺,充分溶解后转移至所述固相合成器中,在室温下反应1h~2h;
除去所述固相合成器中的反应液,再加入体积比为所述无水二氯甲烷∶甲醇∶N,N-二异丙基乙胺=8.2~8.7∶1.5~2.5∶0.5~1.5的封闭液5mL~15mL,在室温下封闭5min~15min;
除去所述固相合成器中的反应液,用N,N-二甲基甲酰胺洗涤,再加入体积百分比为15%~25%哌啶的所述N,N-二甲基甲酰胺溶液,切割所述第一Fmoc保护基,继续用所述N,N-二甲基甲酰胺洗涤切掉的所述第一Fmoc保护基;
将第二Fmoc保护的氨基酸、苯并三氮唑-N,N,N',N'-四甲基脲六氟磷酸酯和所述N,N-二异丙基乙胺溶解在所述N,N-二甲基甲酰胺中,充分溶解后加入所述固相合成器中反应;
采用所述N,N-二甲基甲酰胺洗涤,并利用哌啶切割所述第二Fmoc保护基,再用所述N,N-二甲基甲酰胺洗涤,重复加入所述氨基酸,直到将封端基团加上;
采用所述N,N-二甲基甲酰胺和所述二氯甲烷分别洗涤,洗完后切割,体积百分数为90%~98%三氟乙酸,1%~5%三异丙基硅烷,1%~5%H2O组成的溶液加入到所述固相合成器中,将所得产物从所述2-Cl-Trt树脂上切下,制得所述短肽HCPT–FFpYG-PMI。
4.一种抗肿瘤纳米药物,根据权利要求1至3任意一项所述的抗肿瘤纳米药物的制备方法所制得。
6.根据权利要求5所述的抗肿瘤纳米药物,其特征在于,所述抗肿瘤纳米药物为水凝胶。
7.根据权利要求6所述的抗肿瘤纳米药物,其特征在于,所述抗肿瘤纳米药物形貌为纳米纤维。
8.根据权利要求7所述的抗肿瘤纳米药物,其特征在于,所述抗肿瘤纳米药物处理后的肿瘤相对体积是150mm3~180mm3,肿瘤体积百分比为12%~17%,相对抑制率为82%~88%。
