Abstract
本发明公开了一种从荷叶中联产多种活性物质的方法,该包括以下步骤:提取、萃取得各活性物质部分,再分别对各部分进行分离纯化,最终得到了8种生物碱单体、3种黄酮单体以及4种多酚单体,且本申请制备得到的荷叶总生物碱、荷叶总黄酮以及荷叶总多酚均可直接应用于不同的领域。本发明提供的方法操作简便,生产周期短,分离效率高,工艺稳定,成本低廉,可实现工业化从荷叶中联产多种活性物质,得到的单体的纯度达95%以上,有效成分回收率大于70%,极大地利用了荷叶资源。
Description
技术领域
本发明属于中药活性成分的分离纯化技术领域,具体涉及一种从荷叶中联产多种活性物质的方法。
背景技术
荷叶为睡莲科莲属植物莲Nelumbo nucifera Gaertn .的干燥叶,世界各国均有分布,在我国大部分地区也均有分布,主产于湖北、湖南、浙江、江苏等地,在湖北种植面积达100万亩左右。本公司地处湖北洪湖,是著名的水产之都,盛产莲藕,其是洪湖的主要特产之一。荷叶苦,平,归肝、脾、胃经,具有解暑清热、生发清阳、散瘀止血等功效。1991 年11月,卫生部的卫监发(1991) 第45 号文件中,荷叶被列入第2 批“既是食品又是药品”的名单之中。荷叶具有减肥降脂作用,《本草纲目》中就有“荷叶服之,令人瘦劣”的记载。荷叶中含有蛋白质、维生素 C、脂肪、碳水化合物、胡萝卜素、硫胺素、尼克酸等物质,同时还含有铁、钙、磷等多种微量元素以及黄酮类化合物和包括荷叶碱、亚美罂粟碱、原荷叶碱、N-去甲基荷叶碱等在内的多种生物碱。荷叶中的黄酮类化合物和生物碱均具有明显的生物活性和生理功能,荷叶提取物具有降脂减肥、抗氧化、抑制高胆固醇血症和动脉硬化和抑菌等功效,因此对于荷叶提取物的研究成为医药、美容、食品界备受关注的焦点。
荷叶中活性物质种类丰富,包括多酚类、黄酮类、生物碱类、皂苷类、甾体类、有机酸、多糖、氨基酸及挥发油等物质,用于化妆品配方中,能够起到显著的抗氧化、抗皱、补水、美白、淡化色斑、亮肤等作用。
由于洪湖莲藕的种植量大,由此衍生的一系列产品也非常多,但其多由藕、莲子等加工制作而成,目前对于荷叶的利用还不充分,造成了资源的极大浪费。目前传统提取荷叶粉有效物质的方法提取得率都不高,而且提取过程中都会使用到大量的有机溶剂,不利于实际生产应用。
本发明为充分利用荷叶资源,拓展荷叶资源的应用领域,减少因为荷叶腐烂而对环境造成的污染问题,对其中的活性成分进行提取、纯化,其工艺简单,成本低廉,同时可联产多种生物活性物质,进而应用于不同的领域。
发明内容
本发明旨在克服现有技术的缺陷,提供一种从荷叶中联产多种生物活性物质的方法。该方法操作简便,生产周期短,分离效率高,工艺稳定,成本低廉,可实现工业化联产荷叶中多种生物活性物质,得到的各活性物质纯度高达95%以上,有效成分回收率大于70%。
为了实现上述发明,本发明采用的技术方案如下:从荷叶中联产多种生物活性物质的方法包括以下步骤:
1)将荷叶清洗烘干后,粉碎过80目筛,以体积分数为65-95%的甲醇或乙醇,超声波辅助加热提取2-3次,提取温度为60-85℃,每次提取时间为0.5-2小时,合并提取液,提取液经真空抽滤去除滤渣,减压浓缩得浸膏;
2)将步骤1)得到的浸膏用水分散后,依次以石油醚、氯仿、乙酸乙酯、正丁醇进行萃取,所得氯仿部分、乙酸乙酯部分和正丁醇部分供进一步分离;
3)将上述步骤2)中得到的氯仿部分经超滤膜设备,所得过滤液再经纳滤膜设备,所得滤液减压浓缩后,经冷冻干燥或喷雾干燥得荷叶总生物碱;
4)将上述荷叶中生物碱用中低压制备型液相色谱进行分离,以0.1-0.2%三乙胺的水溶液和乙腈作为洗脱剂,依次进行梯度洗脱,分瓶收集洗脱液,静置析晶并过滤干燥后,经鉴定,共得到如下具体8种生物碱:荷叶碱、去氢荷叶碱、莲碱、去氢莲碱、亚美罂粟碱、巴婆碱、O-去甲基荷叶碱、N-去甲基荷叶碱;
5)将上述步骤2)中得到的乙酸乙酯部分减压浓缩至干,用蒸馏水溶解,将所得水溶液上大孔吸附树脂,用10倍柱体积的去离子水洗脱,再依次用水、50-75%乙醇洗脱,分别洗至无色,收集乙醇洗脱液,浓缩干燥即荷叶总黄酮;
6)将步骤5)得到的荷叶总黄酮上Sephadex LH-20 柱色谱,以二氯甲烷-甲醇梯度洗脱,经TCL检测合并相同组分,上述各组分再经半制备型HPLC,乙腈-水为洗脱剂,最终分离得到如下3种黄酮化合物:槲皮素、木犀草素、3-甲氧基槲皮素,其中3-甲氧基槲皮素为首次从荷叶中分离得到;
7)将上述步骤2)中得到的正丁醇部分减压浓缩至干,用蒸馏水溶解,将所得水溶液上聚酰胺柱层析,以乙醇-水进行梯度洗脱,每个梯度洗3-5倍柱体积,收集30%-50%乙醇洗脱液,浓缩干燥即荷叶总多酚;
8)上述步骤7)所得的荷叶总多酚上半制备型HPLC,以甲醇-水作为流动相,流速为1ml/min,收集不同时间的流出物,最终分离得到如下4中多酚物质:儿茶素、表儿茶素、Nympholide A2和Nympholide B3。
上述步骤1)中提取溶剂的用量为药材质量:溶液体积为1Kg: 10-40L;超声波功率为350W;
步骤3)中超滤和纳滤的温度均控制在40-60℃、压力均控制在0.6-2.2MPa;
步骤5)中大孔吸附树脂为AB-8大孔树脂,上样浓度为20mg /mL,上样体积20mL,径高比为1:30,乙醇浓度为70%。
本发明还提供了上述步骤3)所得的荷叶总生物碱在制备降脂减肥药品、食品或化妆品中的应用。
本发明还提供了上述步骤5)所得的荷叶总黄酮在制备化妆品中的应用。
本发明还提供了上述步骤7)所得的荷叶总多酚在制备化妆品中的应用。
与现有技术相比,本发明的优点是:
本发明提供了一种从荷叶中联产多种活性物质的方法,该包括以下步骤:提取、萃取得各活性物质部分,再分别对各部分进行分离纯化,最终得到了8种生物碱单体、3种黄酮单体以及4种多酚单体,且本申请制备得到的荷叶总生物碱、荷叶总黄酮以及荷叶总多酚均可直接应用于不同的领域。其中对于荷叶总生物碱的分离提取,本发明采用膜分离技术并联合中低压制备型液相色谱进行分离,快速高效地获得了多种生物碱单体;对于荷叶总黄酮的分离纯化,本发明以大孔吸附树脂、Sephadex LH-20 柱色谱以及半制备型HPLC联合使用,高效分离得到了3种黄酮单体,其中3-甲氧基槲皮素为首次从荷叶中分离得到;对于荷叶总多酚的分离纯化,本申请以聚酰胺柱层析联合半制备型HPLC,同样高效地分离得到了4种多酚单体。上述单体化合物的获得,为进一步开发荷叶可利用价值提供了重要保障。
本发明所提供的一种从荷叶中联产多种活性物质的方法,优化提取分离步骤,降低生产成本,依据该方法所得的活性单体含量大于95%,有效成分回收率大于70%。
具体实施方式
实施例1:
从荷叶中联产多种生物活性物质的方法包括以下步骤:
1)将荷叶25kg清洗烘干后,粉碎过80目筛,以体积分数为75%的乙醇,超声波辅助加热提取3次,提取温度为65℃,每次提取时间为1.5小时,提取溶剂的用量为药材质量:溶液体积为1Kg: 20L;超声波功率为350W,合并提取液,提取液经真空抽滤去除滤渣,减压浓缩得浸膏;
2)将步骤1)得到的浸膏用水分散后,依次以石油醚、氯仿、乙酸乙酯、正丁醇进行萃取,所得氯仿部分、乙酸乙酯部分和正丁醇部分供进一步分离;
3)将上述步骤2)中得到的氯仿部分经超滤膜设备,温度控制在40-60℃、压力控制在0.6-2.2MPa,所得过滤液再经纳滤膜设备,温度控制在40-60℃、压力控制在0.6-2.2MPa,所得滤液减压浓缩后,经冷冻干燥或喷雾干燥得荷叶总生物碱;
4)将上述荷叶中生物碱用中低压制备型液相色谱进行分离,以0.1三乙胺的水溶液和乙腈作为洗脱剂,依次进行梯度洗脱,分瓶收集洗脱液,静置析晶并过滤干燥后,经鉴定,共得到如下具体8种生物碱:荷叶碱、去氢荷叶碱、莲碱、去氢莲碱、亚美罂粟碱、巴婆碱、O-去甲基荷叶碱、N-去甲基荷叶碱,上述化合物纯度均大于98%;
5)将上述步骤2)中得到的乙酸乙酯部分减压浓缩至干,用蒸馏水溶解,将所得水溶液上AB-8大孔吸附树脂,上样浓度为20mg /mL,上样体积20mL,径高比为1:30,用10倍柱体积的去离子水洗脱,再依次用水、70%乙醇洗脱,分别洗至无色,收集乙醇洗脱液,浓缩干燥即荷叶总黄酮;
6)将步骤5)得到的荷叶总黄酮上Sephadex LH-20 柱色谱,以二氯甲烷-甲醇梯度洗脱,经TCL检测合并相同组分,上述各组分再经半制备型HPLC,乙腈-水为洗脱剂,最终分离得到如下3种黄酮化合物:槲皮素、木犀草素、3-甲氧基槲皮素,其中3-甲氧基槲皮素为首次从荷叶中分离得到,上述化合物纯度均大于95%;
7)将上述步骤2)中得到的正丁醇部分减压浓缩至干,用蒸馏水溶解,将所得水溶液上聚酰胺柱层析,以乙醇-水进行梯度洗脱,每个梯度洗3-5倍柱体积,收集30%-50%乙醇洗脱液,浓缩干燥即荷叶总多酚;
8)上述步骤7)所得的荷叶总多酚上半制备型HPLC,以甲醇-水作为流动相,流速为1ml/min,收集不同时间的流出物,最终分离得到如下4中多酚物质:儿茶素、表儿茶素、Nympholide A2和Nympholide B3,上述化合物纯度均大于95%。
实施例2:
从荷叶中联产多种生物活性物质的方法包括以下步骤:
1)将荷叶50kg清洗烘干后,粉碎过80目筛,以体积分数为65%的甲醇,超声波辅助加热提取2次,提取温度为70℃,每次提取时间为2小时,提取溶剂的用量为药材质量:溶液体积为1Kg: 10L;超声波功率为350W,合并提取液,提取液经真空抽滤去除滤渣,减压浓缩得浸膏;
2)将步骤1)得到的浸膏用水分散后,依次以石油醚、氯仿、乙酸乙酯、正丁醇进行萃取,所得氯仿部分、乙酸乙酯部分和正丁醇部分供进一步分离;
3)将上述步骤2)中得到的氯仿部分经超滤膜设备,温度控制在60℃、压力控制在1.5MPa,所得过滤液再经纳滤膜设备,温度控制在60℃、压力控制在2.0MPa,所得滤液减压浓缩后,经冷冻干燥或喷雾干燥得荷叶总生物碱;
4)将上述荷叶中生物碱用中低压制备型液相色谱进行分离,以0.2三乙胺的水溶液和乙腈作为洗脱剂,依次进行梯度洗脱,分瓶收集洗脱液,静置析晶并过滤干燥后,经鉴定,共得到如下具体8种生物碱:荷叶碱、去氢荷叶碱、莲碱、去氢莲碱、亚美罂粟碱、巴婆碱、O-去甲基荷叶碱、N-去甲基荷叶碱,上述化合物纯度均大于98%;
5)将上述步骤2)中得到的乙酸乙酯部分减压浓缩至干,用蒸馏水溶解,将所得水溶液上AB-8大孔吸附树脂,上样浓度为20mg /mL,上样体积20mL,径高比为1:30,用10倍柱体积的去离子水洗脱,再依次用水、70%乙醇洗脱,分别洗至无色,收集乙醇洗脱液,浓缩干燥即荷叶总黄酮;
6)将步骤5)得到的荷叶总黄酮上Sephadex LH-20 柱色谱,以二氯甲烷-甲醇梯度洗脱,经TCL检测合并相同组分,上述各组分再经半制备型HPLC,乙腈-水为洗脱剂,最终分离得到如下3种黄酮化合物:槲皮素、木犀草素、3-甲氧基槲皮素,其中3-甲氧基槲皮素为首次从荷叶中分离得到,上述化合物纯度均大于95%;
7)将上述步骤2)中得到的正丁醇部分减压浓缩至干,用蒸馏水溶解,将所得水溶液上聚酰胺柱层析,以乙醇-水进行梯度洗脱,每个梯度洗3-5倍柱体积,收集30%-50%乙醇洗脱液,浓缩干燥即荷叶总多酚;
8)上述步骤7)所得的荷叶总多酚上半制备型HPLC,以甲醇-水作为流动相,流速为1ml/min,收集不同时间的流出物,最终分离得到如下4中多酚物质:儿茶素、表儿茶素、Nympholide A2和Nympholide B3,上述化合物纯度均大于95%。
实施例3:
从荷叶中联产多种生物活性物质的方法包括以下步骤:
1)将荷叶100kg清洗烘干后,粉碎过80目筛,以体积分数为90%的乙醇,超声波辅助加热提取3次,提取温度为85℃,每次提取时间为1小时,提取溶剂的用量为药材质量:溶液体积为1Kg: 40L;超声波功率为350W,合并提取液,提取液经真空抽滤去除滤渣,减压浓缩得浸膏;
2)将步骤1)得到的浸膏用水分散后,依次以石油醚、氯仿、乙酸乙酯、正丁醇进行萃取,所得氯仿部分、乙酸乙酯部分和正丁醇部分供进一步分离;
3)将上述步骤2)中得到的氯仿部分经超滤膜设备,温度控制在40-60℃、压力控制在0.6-2.2MPa,所得过滤液再经纳滤膜设备,温度控制在40-60℃、压力控制在0.6-2.2MPa,所得滤液减压浓缩后,经冷冻干燥或喷雾干燥得荷叶总生物碱;
4)将上述荷叶中生物碱用中低压制备型液相色谱进行分离,以0.1三乙胺的水溶液和乙腈作为洗脱剂,依次进行梯度洗脱,分瓶收集洗脱液,静置析晶并过滤干燥后,经鉴定,共得到如下具体8种生物碱:荷叶碱、去氢荷叶碱、莲碱、去氢莲碱、亚美罂粟碱、巴婆碱、O-去甲基荷叶碱、N-去甲基荷叶碱,上述化合物纯度均大于98%;

Claims (7)
1.一种从荷叶中联产多种生物活性物质的方法,其特征在于包括以下步骤:
1)将荷叶清洗烘干后,粉碎过80目筛,以体积分数为65-95%的甲醇或乙醇,超声波辅助加热提取2-3次,提取温度为60-85℃,每次提取时间为0.5-2小时,合并提取液,提取液经真空抽滤去除滤渣,减压浓缩得浸膏;
2)将步骤1)得到的浸膏用水分散后,依次以石油醚、氯仿、乙酸乙酯、正丁醇进行萃取,所得氯仿部分、乙酸乙酯部分和正丁醇部分供进一步分离;
3)将上述步骤2)中得到的氯仿部分经超滤膜设备,所得过滤液再经纳滤膜设备,所得滤液减压浓缩后,经冷冻干燥或喷雾干燥得荷叶总生物碱;
4)将上述荷叶中生物碱用中低压制备型液相色谱进行分离,以0.1-0.2%三乙胺的水溶液和乙腈作为洗脱剂,依次进行梯度洗脱,分瓶收集洗脱液,静置析晶并过滤干燥后,经鉴定,共得到如下具体8种生物碱:荷叶碱、去氢荷叶碱、莲碱、去氢莲碱、亚美罂粟碱、巴婆碱、O-去甲基荷叶碱、N-去甲基荷叶碱;
5)将上述步骤2)中得到的乙酸乙酯部分减压浓缩至干,用蒸馏水溶解,将所得水溶液上大孔吸附树脂,用10倍柱体积的去离子水洗脱,再依次用水、50-75%乙醇洗脱,分别洗至无色,收集乙醇洗脱液,浓缩干燥即荷叶总黄酮;
6)将步骤5)得到的荷叶总黄酮上Sephadex LH-20 柱色谱,以二氯甲烷-甲醇梯度洗脱,经TCL检测合并相同组分,上述各组分再经半制备型HPLC,乙腈-水为洗脱剂,最终分离得到如下3种黄酮化合物:槲皮素、木犀草素、3-甲氧基槲皮素,其中3-甲氧基槲皮素为首次从荷叶中分离得到;
7)将上述步骤2)中得到的正丁醇部分减压浓缩至干,用蒸馏水溶解,将所得水溶液上聚酰胺柱层析,以乙醇-水进行梯度洗脱,每个梯度洗3-5倍柱体积,收集30%-50%乙醇洗脱液,浓缩干燥即荷叶总多酚;
8)上述步骤7)所得的荷叶总多酚上半制备型HPLC,以甲醇-水作为流动相,流速为1ml/min,收集不同时间的流出物,最终分离得到如下4中多酚物质:儿茶素、表儿茶素、Nympholide A2和Nympholide B3。

4.根据权利要求1所述的一种从荷叶中联产多种生物活性物质的方法,其特征在于:所述步骤5)中大孔吸附树脂为AB-8大孔树脂,上样浓度为20mg /mL,上样体积20mL,径高比为1:30,乙醇浓度为70%。
5.权利要求1步骤3)所得的荷叶总生物碱在制备降脂减肥药品、食品或化妆品中的应用。
6.权利要求1步骤5)所得的荷叶总黄酮在制备化妆品中的应用。
7.权利要求1步骤7)所得的荷叶总多酚在制备化妆品中的应用。
