技术领域
本发明涉及工厂化水产养殖技术领域,特别涉及一种稚鲍转饵期增加存活率的方法。
背景技术
鲍在养殖过程中存在许多困难,由于禁药原因,鲍养殖过程中不能使用任何药物辅助养殖,而稚鲍养殖的死亡率很高,尤其是稚鲍转饵期。稚鲍转饵期就是稚鲍从摄食底栖硅藻转换成人工饲料的时期。稚鲍培育后,前期生长主要以底栖硅藻为食,通常底栖硅藻培育在塑料波纹板上,稚鲍附着于波纹板上摄食,当稚鲍长到一定大小时,需要更换饵料,将稚鲍从波纹板上剥离下来,转到脊瓦附着基上,之后投喂稚鲍饲料。但稚鲍在转移到脊瓦附着基上培养半个月至二十天后,稚鲍就会出现大量死亡的现象。推测稚鲍死亡的原因,一方面可能是稚鲍剥离液或毛刷对稚鲍造成不适或机械损伤,导致稚鲍受到致病微生物的侵染;另一方面可能稚鲍对突然变化的环境和饵料的不适应,稚鲍摄食异常或消化不良等问题,最终造成稚鲍死亡。
据报道,蛭弧菌能够快速侵入宿主细菌使其裂解,在防治稚鲍死亡具有见效快的特点(CN 201010270955.0),但蛭弧菌的这种寄生繁殖通常表现出严格的特异性,对特定致病菌的杀灭效果很好,而稚鲍死亡的致病菌很多,采用蛭弧菌进行防治具有偶然性。近年来,随着动物免疫的深入研究发现,提高水产动物的免疫力具有较好的抗病,抗损伤效果。虽然益生菌复合制剂具有较好的抗病及提高免疫力的作用,但益生菌在水产动物体内的定植效果仍不理想,往往需要长期投喂才能见效,从而增加养殖成本。尤其鲍的养殖需要长流水环境,施用益生菌的效果更加缓慢,成本更大。因此,需要寻找有效的防治稚鲍转饵期死亡的措施,特别是能提高其免疫力、存活率的养殖方法,从而降低稚鲍的养殖成本和死亡率。
发明内容
本发明的目的在于针对稚鲍转饵期机械损伤、摄食不佳、病菌侵害、死亡率较高等问题,提供了一种新型、绿色无污染、可显著提高稚鲍存活率的养殖方法。
为实现上述目的,本发明提供如下技术方案为:用于向稚鲍喷洒XS1412C菌株发酵液或投喂添加XS1412C菌株发酵液的饵料,其中棒状菌菌株XS1412C,该菌株已保藏于中国典型培养物保藏中心,其保藏编号为CCTCC NO:M 2015149,保藏时间为2015年3月30日。
一种提高稚鲍转饵期存活率的方法,其特征在于,包括步骤:
(1)稚鲍剥离前一周,每天定时1次,向稚鲍附着的波纹板间播撒XS1412C菌株发酵液;
(2)稚鲍剥离后,将网箱收集的稚鲍,先在干净海水冲洗剥离液,再将稚鲍浸泡于低聚壳聚糖-褐藻寡糖溶液中20-90s;
(3)稚鲍转移至脊瓦附着基后,稚鲍养殖池长流水,保持水质澄清,定期投喂添加XS1412C菌株发酵液的饵料,投喂15-20天。
优选的,所述每天定时1次,是指在每天的19:00-20:00之间进行播撒。
优选的,所述XS1412C菌株发酵液的播撒或添加到饵料中的浓度为1×103-1×108cfu/mL。
进一步的,所述低聚壳聚糖-褐藻寡糖溶液,其制备方法为,低聚壳聚糖先用0.5%醋酸溶液溶解制备壳聚糖溶液,低聚壳聚糖溶液与褐藻寡糖溶液按1:5比例混合,低聚壳聚糖终浓度的质量百分比为0.1-2%,褐藻寡糖终浓度的质量百分比为0.1-3%。
进一步的,所述褐藻寡糖溶液,其制备方法为,海水溶解质量分数为3-7%低粘度褐藻酸钠,加入20-50mLXS1412C菌株发酵液,30-40℃发酵酶解4-10h,沸水浴10min灭活菌株,冷却后既得褐藻寡糖溶液。
进一步的,所述XS1412C菌株发酵液,其制备方法是单菌落接种种子培养液26-30℃培养6-10h,1-5%种子液接种发酵培养液,28-35℃发酵培养12-24h。
进一步的,所述XS1412C菌株种子培养液为2216E,发酵培养液成分质量比为0.5-0.9%褐藻酸钠,0.03-0.1%磷酸二氢钾,0.15-0.35%氯化铵,0.1-0.3%蛋白胨过滤的陈海水溶解,不调pH值。
本发明相较于现有技术,具有如下的优势及效果:
(1)混有XS1412C菌株是从鲍肠道中筛选的一种棒状杆菌,溶血性试验验证其不具有溶血性,是能够产褐藻胶裂解酶、蛋白酶及淀粉酶等多种酶的鲍肠道益生菌,在稚鲍肠道的定植效果优于其他来源益生菌制剂。
(2)稚鲍剥离前后饲喂XS1412C菌株发酵液,其中含有褐藻酸钠降解形成的褐藻寡糖,褐藻寡糖与菌株共同作用,有助于调节稚鲍肠道菌群,饵料消化,抗菌消炎,显著提高其免疫力。
(3)稚鲍剥离后,利用低聚壳聚糖和褐藻寡糖的成膜性及抗菌消炎功能,可有效降低稚鲍的机械性损伤及病菌感染风险。
(4)方法所用菌株及低聚壳聚糖-褐藻寡糖溶液的制备,操作简单,成本低,有利于育苗企业成本控制。
具体实施方式
实施例1
XS1412C菌株发酵液制备:
挑取单菌落接种2216E培养液,26℃振荡培养6-8h,按3%接种量接种发酵培养液,30℃发酵培养12-24h,细菌浓度达到1×103-1×108cfu/mL即可。
低聚壳聚糖-褐藻寡糖溶液制备:10g低聚壳聚糖用100mL 0.5%醋酸溶液溶解;500mL海水溶解35g低粘度褐藻酸钠,加入30mL 1×107-1×108cfu/mL XS1412C菌株发酵液,35℃发酵酶解6h,沸水浴灭活10min,冷却后得褐藻寡糖溶液;低聚壳聚糖溶液与褐藻寡糖溶液按1:5比例混合后,加过滤海水至2L,备用。
一种提高稚鲍转饵期存活率的方法,操作如下:
(1)稚鲍剥离前一周,每天19:00-20:00之间,向稚鲍附着的波纹板间播撒1×103-1×106cfu/mL XS1412C菌株发酵液,由于该时间段稚鲍摄食活动较频繁;;
(2)稚鲍剥离后,将网箱收集的稚鲍,先在干净的过滤海水中冲洗剥离液,再将稚鲍浸泡于低聚壳聚糖-褐藻寡糖溶液中60s;
(3)稚鲍转移至脊瓦附着基后,稚鲍养殖池长流水,保持水质澄清,定期投喂添加浓度为1×106-1×108cfu/mL XS1412C菌株发酵液的饵料,投喂20天。
实施例2
XS1412C菌株发酵液制备:
挑取单菌落接种2216E培养液,26℃振荡培养6-8h,按4%接种量接种发酵培养液,30℃发酵培养12-20h,细菌浓度达到1×103-1×108cfu/mL即可。
低聚壳聚糖-褐藻寡糖溶液制备:16g低聚壳聚糖用100mL 0.5%醋酸溶液溶解;500mL海水溶解21g低粘度褐藻酸钠,加入25mL 1×107-1×108cfu/mL XS1412C菌株发酵液,35℃发酵酶解5h,沸水浴灭活10min,冷却后得褐藻寡糖溶液;低聚壳聚糖溶液与褐藻寡糖溶液按1:5比例混合后,加过滤海水至2L,备用。

本发明设置对照组与处理组。对照组是稚鲍不进行任何处理,正常剥离,清洗,饵料饲喂。对照组和处理组分别设置3个平行,每个平行随机选取2000粒,稚鲍体长在6.47-9.36mm;稚鲍转移至脊瓦附着基30天,统计存活率。
注:初始平均壳长即稚鲍剥离后测得的壳长;30天后平均壳长即稚鲍转移至脊瓦附着基生长30天后的壳长。
通过上述实验可看出,本发明提供的方法能显著提高稚鲍转饵期的存活率,同时也有利于促进稚鲍的生长。
Claims (7)
1.一种提高稚鲍转饵期存活率的方法,其特征在于,包括步骤:
稚鲍剥离前一周,每天定时1次,向稚鲍附着的波纹板间播撒XS1412C菌株发酵液;
稚鲍剥离后,将网箱收集的稚鲍,先在干净海水冲洗剥离液,再将稚鲍浸泡于低聚壳聚糖-褐藻寡糖溶液中20-90s;
稚鲍转移至脊瓦附着基后,稚鲍养殖池长流水,保持水质澄清,定期投喂添加XS1412C菌株发酵液的饵料,投喂15-20天。
2.根据权利要求1所述的一种提高稚鲍转饵期存活率的方法,其特征在于:所述每天定时1次,是指在每天的19:00-20:00之间进行播撒。
3.根据权利要求1所述的一种提高稚鲍转饵期存活率的方法,其特征在于:所述XS1412C菌株发酵液的播撒或添加到饵料中的浓度为1×103-1×108cfu/mL。
4.根据权利要求1所述的一种提高稚鲍转饵期存活率的方法,其特征在于:所述低聚壳聚糖-褐藻寡糖溶液,其制备方法为,低聚壳聚糖先用0.5%醋酸溶液溶解制备壳聚糖溶液,低聚壳聚糖溶液与褐藻寡糖溶液按1:5比例混合,低聚壳聚糖终浓度的质量百分比为0.1-2%,褐藻寡糖终浓度的质量百分比为0.1-3%。
5.根据权利要求1或4所述的一种提高稚鲍转饵期存活率的方法,其特征在于:所述褐藻寡糖溶液,其制备方法为,海水溶解质量分数为3-7%低粘度褐藻酸钠,加入20-50mLXS1412C菌株发酵液,30-40℃发酵酶解4-10h,沸水浴10min灭活菌株,冷却后既得褐藻寡糖溶液。
6.根据权利要求1所述的一种提高稚鲍转饵期存活率的方法,其特征在于:所述XS1412C菌株发酵液,其制备方法是单菌落接种种子培养液26-30℃培养6-10h,1-5%种子液接种发酵培养液,28-35℃发酵培养12-24h。
7.根据权利要求6所述的一种提高稚鲍转饵期存活率的方法,其特征在于:所述种子培养液为2216E,发酵培养液成分质量比为0.5-0.9%褐藻酸钠,0.03-0.1%磷酸二氢钾,0.15-0.35%氯化铵,0.1-0.3%蛋白胨过滤的陈海水溶解,不调pH值。
