本发明是通过以下技术方案来实现:
一种文冠果风味醒酒保肝肽口服液的制备方法,包括以下步骤:
步骤1,采用碱溶酸沉法提取文冠果粕中的文冠果蛋白,将分离得到的沉淀物洗涤,加水得到文冠果蛋白液;
步骤2,采用菠萝蛋白酶或木瓜蛋白酶对步骤1得到的文冠果蛋白液进行酶解,得到文冠果蛋白酶解液A和沉淀;采用风味蛋白酶对沉淀进行酶解,得到文冠果蛋白酶解液B;
步骤3,对步骤2得到的文冠果蛋白酶解液A和文冠果蛋白酶解液B进行过滤后采用离子交换树脂法对文冠果蛋白酶解液A和文冠果蛋白酶解液B的透过液分别进行脱盐处理,超滤得到3k-10kDa分子量以内的超滤液后对超滤液进行冷冻干燥,得到文冠果肽A和文冠果肽B;
步骤4,向步骤3得到的文冠果肽A和文冠果肽B中加入养肝因子、矫味剂、添加剂和辅料,其余成分为纯净水,充分混合后灭菌,制得文冠果风味醒酒保肝肽口服液。
优选地,步骤1的具体步骤为:
将文冠果粕粉碎后加入文冠果粕质量6-15倍的水混合,调节pH至8.0-10.0,搅拌2-3h后,在4℃条件下以4000-8000r/min的速度离心15-30min,离心后收集上清液,将上清液的pH调节至4.5-4.6,在4℃条件下以4000-8000r/min的速度离心15-30min,将沉淀水洗2-5次后加入沉淀体积10-20倍的纯净水,得到文冠果蛋白液。

步骤3:将文冠果蛋白酶解液A和文冠果蛋白酶解液B分别使用微孔滤膜过滤,将透过液采用离子交换树脂法(001×8,732)进行脱盐处理,脱盐处理后利用超滤技术得到10kDa分子量以内的蛋白多肽液,真空冷冻干燥得文冠果肽A和文冠果肽B。
步骤4:将质量比为5:3:3:4的葛根、枸杞、姜黄、香菇放入萃取罐中,再加入葛根、枸杞、姜黄、香菇总质量8倍的水煮沸40min后过滤得到提取液。将质量比为8:3:4的维生素B1、维生素B6和维生素B12配置成复合维生素。按照以下质量百分比准备原料,文冠果肽A8%,文冠果肽B6%,葛根、枸杞、姜黄、香菇的提取液50%、牛磺酸1%、复合维生素0.5%、蜂蜜9%、葡萄汁和苹果汁的浓缩混合果汁13%,其余成分为纯净水,充分混合,通过UHT灭菌后进行热灌装,冷却后保存,制得文冠果风味醒酒保肝肽口服液。
实施例3
步骤1:将文冠果粕粉碎得文冠果粕粉,加入15倍质量的水混合,调节pH至10.0,搅拌3h,在4℃条件下以8000r/min的速度离心15min,离心后收集上清液,将上清液的pH调节至4.6;在4℃条件下以6000r/min的速度离心30min,将沉淀水洗5次后加入沉淀体积20倍的纯净水,得到文冠果蛋白液。
步骤2:向文冠果蛋白液中加入文冠果粕质量0.8%的木瓜蛋白酶,调节pH至8,在55℃下搅拌3h后煮沸20min灭酶,离心,得到文冠果蛋白酶解液A和沉淀,向沉淀中加入沉淀体积15倍的纯净水,调节pH至8,在55℃下加入文冠果粕质量质量0.6%的风味蛋白酶,搅拌2h,煮沸10min后离心得蛋白酶解液B。
步骤3:将文冠果蛋白酶解液A和文冠果蛋白酶解液B分别使用微孔滤膜过滤,将透过液采用离子交换树脂法(001×8,732)进行脱盐处理,脱盐处理后利用超滤技术得到5kDa分子量以内的蛋白多肽液,真空冷冻干燥得文冠果肽A和文冠果肽B。
步骤4:将质量比为5:3:3:4的葛根、枸杞、姜黄、香菇放入萃取罐中,再加入葛根、枸杞、姜黄、香菇总质量7倍的水煮沸35min后过滤得到提取液。将质量比为8:3:4的维生素B1、维生素B6和维生素B12配置成复合维生素。按照以下质量百分比准备原料,文冠果肽A10%,文冠果肽B8%,葛根、枸杞、姜黄、香菇的提取液55%、牛磺酸1.5%、复合维生素1%、蜂蜜8%、草莓汁、橙汁和蓝莓汁的浓缩混合果汁10%,其余成分为纯净水,充分混合,通过UHT灭菌后进行热灌装,冷却后保存,制得文冠果风味醒酒保肝肽口服液。
本发明制得的文冠果风味醒酒保肝肽口服液解酒功效实验:
将24只健康状态良好体重相近的小白鼠分成A、B、C、D四组,每组8只,四组小白鼠统一灌服52°某市售白酒0.1mL/10g,相当于70kg的成人一次性饮用700mL白酒,5min后A、B、C三组分别灌服实施例1、2、3制得的文冠果风味醒酒保肝肽口服液0.1mL/10g,D组灌服同样剂量的生理盐水。
检测:各组小鼠灌服白酒后1小时、3小时各选4只取血抗凝后用气相色谱法检测小鼠血液中酒精浓度。结果如表1:

以上结果表明本发明的文冠果风味醒酒保肝肽口服液能够促进体内酒精代谢,小鼠灌服口服液后再饮酒,无论是1h还是3h后,其体内的酒精含量均低于生理盐水的对照组。

Claims (9)

1.一种文冠果风味醒酒保肝肽口服液的制备方法,其特征在于,包括以下步骤:
步骤1,采用碱溶酸沉法提取文冠果粕中的文冠果蛋白,将分离得到的沉淀物洗涤,加水得到文冠果蛋白液;
步骤2,采用菠萝蛋白酶或木瓜蛋白酶对步骤1得到的文冠果蛋白液进行酶解,得到文冠果蛋白酶解液A和沉淀;采用风味蛋白酶对沉淀进行酶解,得到文冠果蛋白酶解液B;
步骤3,对步骤2得到的文冠果蛋白酶解液A和文冠果蛋白酶解液B进行过滤后采用离子交换树脂法对文冠果蛋白酶解液A和文冠果蛋白酶解液B的透过液分别进行脱盐处理,超滤得到3k-10kDa分子量以内的超滤液后对超滤液进行冷冻干燥,得到文冠果肽A和文冠果肽B;
步骤4,向步骤3得到的文冠果肽A和文冠果肽B中加入养肝因子、矫味剂、添加剂和辅料,其余成分为纯净水,充分混合后灭菌,制得文冠果风味醒酒保肝肽口服液。
2.根据权利要求1所述的文冠果风味醒酒保肝肽口服液的制备方法,其特征在于,步骤1的具体步骤为:
将文冠果粕粉碎后加入文冠果粕质量6-15倍的水混合,调节pH至8.0-10.0,搅拌2-3h后,在4℃条件下以4000-8000r/min的速度离心15-30min,离心后收集上清液,将上清液的pH调节至4.5-4.6,在4℃条件下以4000-8000r/min的速度离心15-30min,将沉淀水洗2-5次后加入沉淀体积10-20倍的纯净水,得到文冠果蛋白液。
3.根据权利要求1所述的文冠果风味醒酒保肝肽口服液的制备方法,其特征在于,步骤2的具体步骤为:
向步骤1制得的文冠果蛋白液中加入文冠果粕质量0.4%-0.8%的菠萝蛋白酶或木瓜蛋白酶,调节pH至7.5-9,在40-55℃下搅拌3-7h后煮沸15-20min,离心得到文冠果蛋白酶解液A和沉淀;向沉淀中加入沉淀体积10-15倍的纯净水,加入文冠果粕质量0.4%-0.6%的风味蛋白酶,调节pH至7-9,在40-55℃下搅拌2-6h后煮沸10-15min后离心得到文冠果蛋白酶解液B。
4.根据权利要求1所述的文冠果风味醒酒保肝肽口服液的制备方法,其特征在于,步骤3的具体步骤为:
将步骤2得到的文冠果蛋白酶解液A和文冠果蛋白酶解液B分别使用微孔滤膜过滤,将透过液再分别采用离子交换树脂法进行脱盐处理,脱盐处理后利用超滤技术得到1k-10kDa分子量以内的蛋白多肽液,真空冷冻干燥得文冠果肽A和文冠果肽B。
5.根据权利要求1所述的文冠果风味醒酒保肝肽口服液的制备方法,其特征在于,步骤4的具体步骤为:
按照以下质量百分比准备原料,文冠果肽A6%-10%,文冠果肽B4%-8%,养肝因子:葛根、枸杞、姜黄和香菇的水提物40%-60%、牛磺酸0.5%-1.5%、维生素0.05%-1%、糖或蜂蜜7%-10%、浓缩果汁10%-15%,其余成分为纯净水,充分混合,灭菌,制得文冠果风味醒酒保肝肽口服液。
6.根据权利要求5所述的文冠果风味醒酒保肝肽口服液的制备方法,其特征在于,葛根、枸杞、姜黄和香菇的水提物是将质量比为5:3:3:4的葛根、枸杞、姜黄、香菇放入萃取罐中,再加入葛根、枸杞、姜黄和香菇总质量5-8倍的水煮沸30-40min后过滤得到的提取液。
7.根据权利要求5所述的文冠果风味醒酒保肝肽口服液的制备方法,其特征在于,维生素是由质量比为8:3:4的维生素B1、维生素B6和维生素B12复合而成的。
8.根据权利要求5所述的文冠果风味醒酒保肝肽口服液的制备方法,其特征在于,浓缩果汁为猕猴桃汁、梨汁、菠萝汁、葡萄汁、苹果汁、草莓汁、橙汁和蓝莓汁中的一种或几种。
9.采用权利要求1~8中任意一项所述的方法制备得到的文冠果风味醒酒保肝肽口服液。
