本发明属于植物源杀菌剂技术领域,公开了喜树碱用于防治稻瘟病的用途。该化合物可有效的抑制稻瘟病菌的菌丝生长和孢子萌发,该化合物作为活性成分加工制成的农用杀菌剂可用于防治稻瘟病。使用该化合物与其他杀菌剂交替使用防治稻瘟病可延缓抗药性的产生。

Description

喜树碱用于防治稻瘟病的用途
技术领域
本发明属于植物源杀菌剂技术领域,具体涉及喜树碱作为活性成分防治稻瘟病的用途。
背景技术
稻瘟病是水稻的三大病害之一,广泛分布于世界各大水稻产区,一旦流行通常可使水稻减产10%~20%,严重时减产40%~50%,甚至颗粒无收。目前喷施化学农药是预防和控制稻瘟病大面积流行的主要手段,但是长时间使用同一种农药容易引起抗药性问题,因此需要发展新型的农药来进行交替使用。
喜树碱是来源于喜树的一种天然喹啉类生物碱,分子式C20H16N2O4,其化学结构式如下所示。该化合物及其衍生物是一类重要的抗癌药物,同时还被作为一种植物源杀虫剂被应用于农业生产中。喜树碱的抑菌活性已有一些相关报道,其中国家发明专利“含喜树碱或喜树碱衍生物的农用杀菌剂及其应用”(专利号200610051741.8)的报道最为全面,公开了喜树碱及其衍生物在防治各类植物病害中的应用,但是其中并未涉及防治稻瘟病的用途。查阅国内外的相关文献及专利,并未发现喜树碱应用于稻瘟病防治中的报道。

发明内容
鉴于上述情况,本发明提供了喜树碱作为活性成分防治稻瘟病的用途。
本发明是通过以下技术方案实现的。
所述的喜树碱是从喜树侧枝中通过多种植物化学分离纯化手段提取所得。喜树碱作为活性成分和农药助剂加工制成农药允许的任何一种剂型,或作为活性成分与其他农药复配成农药允许的任何一种剂型。
所述的剂型包括粉剂、可湿性粉剂、可溶性粉剂、粒剂、水剂、乳油、水乳剂、微乳剂、水分散粒剂、悬浮剂、烟剂、缓释剂、种衣剂或油剂;所述的其他农药为三环唑、稻瘟酰胺、咪鲜胺、嘧菌酯、百菌清、多菌灵、稻瘟灵、异稻瘟净或甲基托布津。
所述的用途是指喜树碱用于抑制稻瘟病菌菌丝生长、孢子萌发和稻瘟病的防治。
本发明通过研究证明喜树碱对稻瘟病菌具有显著的离体及活体抑菌活性,并且其抑菌活性明显优于对其他植物病原菌的活性,可作为稻瘟病防治药剂进行交替使用,从而缓解抗药性的产生。
具体实施方式
下面对本发明具体实施例作进一步详细描述,但本发明的内容并不仅仅限于下列实施例包含的内容。
实施例1:
所述的喜树碱是将喜树侧枝粉碎后,用1.5%的氢氧化钠浸渍15h后分装于渗漉桶中,然后用0.3%氢氧化钠溶液渗漉,收集18-20倍渗漉液,加盐酸搅拌调pH=2-3,用适量的氯仿萃取,将得到的氯仿相浓缩,回收溶剂至有晶体析出为止,在暗室中冷却结晶,滤取析出的结晶喜树碱粗品,滤液待用;再用氯仿与甲醇等体积的混合液重结晶,得到喜树碱纯品。将结晶喜树碱的母液旋干,用乙酸乙酯溶解固体,加入2倍质量的硅胶,混匀后旋干溶剂。取40倍样品质量的硅胶,用100-200目硅胶拌匀,减压旋干,均匀上样,样品上盖一层石英砂,用二氯甲烷:甲醇=98:2进行洗脱。洗脱各流分用TLC检测,展开剂为二氯甲烷:丙酮=7:3,浓硫酸香草醛显色,喜树碱呈蓝色,Rf值为0.5,把只含有喜树碱部分的流分浓缩结晶,得到喜树碱的纯品。以购买的喜树碱纯品为对照,使用高效液相色谱进行分析,其保留时间一致。
实施例2:
称取按实施例1制备所得的喜树碱,与木质素、黄原胶、乙二醇、有机硅酮类消泡剂、苯甲酸钠等辅料按照常规制备工艺制成有效成分浓度为20%的悬浮剂。
实施例3:
称取按实施例1制备所得的喜树碱,与硅藻土、十二烷基苯磺酸钠、木质素等常规辅料按照常规制备工艺制成有效成分浓度为20%的可湿性粉剂。
实施例4:
称取按实施例1制备所得的喜树碱,与高岭土、木质素磺酸钠等常规辅料按照常规制备工艺制成有效成分浓度为60%的水分散粒剂。
实施例5:
按照重量比1:3的比例称取按实施例1制备所得的喜树碱和三环唑,与木质素、黄原胶、乙二醇、有机硅酮类消泡剂、苯甲酸钠等辅料按照常规制备工艺制成有效成分浓度为40%的悬浮剂。
实施例6:
按照重量比2:1的比例称取按实施例1制备所得的喜树碱和咪鲜胺,与木质素、黄原胶、乙二醇、有机硅酮类消泡剂、苯甲酸钠等辅料按照常规制备工艺制成有效成分浓度为40%的悬浮剂。
实施例7:
按照重量比3:2的比例称取按实施例1制备所得的喜树碱和稻瘟灵,与木质素、黄原胶、乙二醇、有机硅酮类消泡剂、苯甲酸钠等辅料按照常规制备工艺制成有效成分浓度为40%的悬浮剂。
实施例8:
按实施例1制备所得喜树碱的抗菌谱测定:
测定对象包括小麦纹枯病菌、稻瘟病菌、棉花黄萎病菌、水稻纹枯病菌、黄瓜灰霉病菌、油菜菌核病菌、水稻恶苗病菌、小麦赤霉病菌、玉米小斑病菌、葡萄炭疽病菌、番茄早疫病菌、瓜果腐霉病菌、辣椒疫霉病菌、大豆疫霉病菌、水稻白叶枯病菌、十字花科蔬菜黑腐病菌和番茄细菌性斑点病菌等常见农业病原菌。所有供试病原菌均为田间采集分离菌株。
将喜树碱溶于DMSO,配制成浓度为5000μg/mL的母液,并使用DMSO进行梯度稀释后待用。将各浓度喜树碱溶液或等体积DMSO分别加入加热融化并冷却至55℃的PSA(用于真菌测定)或V8培养基(用于卵菌测定)中,混匀后倒入直径为9cm的培养皿中制成含药平板。用直径5mm的打孔器在预培养的真菌或卵菌菌落边缘打取菌碟,并接种至含药平板中间,每个处理3个重复。25℃倒置培养至对照接近长满板,十字交叉法测定菌落直径并计算抑制率。抑制率(%)=(对照菌落直径﹣加药处理菌落直径)/对照菌落直径×100。菌落直径=十字交叉法测定直径平均值-5mm。根据不同浓度的抑制率计算喜树碱的EC50和EC90值。
将各浓度喜树碱溶液或等体积DMSO分别加入NB培养液中制成含药培养液,用分光光度计将预培养至对数生长期的细菌培养物OD600值调至0.02,并按1:200的比例加入含药培养液中,每个处理3个重复。28℃175rpm摇培使对照长至对数生长期,用分光光度计测定OD600并计算抑制率。抑制率(%)=(对照OD600值﹣加药处理OD600值)/对照OD600值×100。根据不同浓度的抑制率计算喜树碱的EC50和EC90值。
试验结果:按实施例1制备所得的喜树碱抗菌谱如表1所示。从表中结果可以看出,喜树碱对农业病原真菌活性较强,尤其是对稻瘟病菌抑菌活性最好,对小麦纹枯病菌和棉花黄萎病菌活性也较高。相比之下,喜树碱对供试病原卵菌抑菌活性较弱,而对供试病原细菌无抑菌效果。

实施例9:
按实施例1制备所得喜树碱对稻瘟病菌孢子萌发活性的测定:
将稻瘟病菌接种在燕麦片番茄汁琼脂培养基上,于25℃条件下培养5-7天后用无菌水清洗下菌丝和分生孢子,将其悬浊液均匀涂布于另一新的平板培养基上,超净工作台吹干。25℃培养箱中培养约35-40h后向菌落上加少量自来水,用棉签轻轻擦去表面的气生菌丝,并用水将培养基表面的菌丝和分生孢子清洗干净,待培养基表面晾干后罩上两层纱布,用皮筋绑好,于25℃光照条件下培养至有大量分生孢子产生。从培养基上洗下分生孢子,离心弃上清,得到的分生孢子用0.025%吐温20水溶液悬浮,血球计数板计数后孢子悬浮液浓度调至2×105 \mL待用。
将各浓度喜树碱溶液或等体积DMSO分别加入加热融化并冷却至55℃的水琼脂培养基,混匀后倒入培养皿中制成含药平板。将孢子悬浮液涂布于含药平板,25℃培养后显微观察孢子萌发情况并计算抑制率。抑制率(%)=(对照孢子萌发率﹣加药处理孢子萌发率)/对照孢子萌发率×100。根据不同浓度的抑制率计算喜树碱对稻瘟病菌孢子萌发的EC50和EC90值。
试验结果:按实施例1制备所得的喜树碱对稻瘟病菌孢子萌发的EC50值为10.16μg/mL,95%置信区间为4.53-22.77μg/mL;EC90值为72.99μg/mL,95%置信区间为17.78-299.70μg/mL。
实施例10:
按实施例2制备所得20%喜树碱悬浮剂对稻瘟病的防治效果:
按1000倍、500倍和250倍比例稀释20%喜树碱悬浮剂,以清水作为空白对照,喷施于温室常规种植的‘苏御糯’水稻。在喷药前一天使用按实施例9制备所得的孢子悬浮液进行人工接种,在喷药一周后进行第二次喷药处理。待清水对照充分发病后,调查发病情况。对病叶进行分级,计算各处理病情指数和防治效果。
试验结果:喷施各浓度喜树碱悬浮液后,对水稻生长无不良影响,250倍稀释液对稻瘟病的防效在65%以上,而1000倍稀释液对稻瘟病的防效在40%以上。

2.按照权利要求1所述的用途,其特征在于所述的喜树碱可用于抑制稻瘟病菌的菌丝生长和孢子萌发;所述的喜树碱作为活性成分加工制成的农用杀菌剂可用于防治稻瘟病。
