Abstract
本申请实施例公开了一种健脾除湿功能的组合物、其制备方法及应用。该组合物应用于固体饮料中,由以下重量份数的原料组成:薏苡仁900~1800份;豆蔻300~600份;苦杏仁500~1000份;蓝莓果粉500~1000份。该组合物中的四种药材配伍合用,肺脾同调,通利水道,除一身之湿,协同起到健脾除湿的作用。同时,该组合物制成固体饮料,服用方便,口味较好,接受度高,并且无毒副作用,安全性高。

技术领域
本发明涉及健脾除湿的功能饮品领域,具体涉及一种具有健脾除湿功能的组合物。此外,本发明还涉及该组合物的制备方法及应用。
背景技术
随着时代发展,人们的生活节奏加快,湿热体质的人群越来越多,严重的影响到了健康的生活。湿热体质是常见的九种偏颇体质之一,以面垢油光、口苦、苔黄腻等湿热表现为主要特征。“湿热”涉及中医脏腑、经络7个系统的43种疾病,分属于西医的呼吸、泌尿等1个系统的72种急慢性疾病。脾胃湿热证是临床常见病症,尤其是炎热潮湿的地区,人们更易于感受湿热病邪而发病,症状复杂,病程较久,病势缠绵难愈。在脾胃湿热证发生、发展的过程中,各种致病因素的刺激会使躯体做出应激反应,进而导致神经内分泌免疫功能障碍。
为了延缓或阻止脾胃湿热证的发生或加快其恢复,人们通常会服用一些中草药。其中,单味中药有苍术、白术、茯苓等;中药复方制剂有健脾丸、参苓白术散、补脾益肠丸等。此类药物对症状的产生有一定治疗和缓解作用,但这些制剂多为治疗药物,服用次数多为2~3次/日,且口味较差。
发明内容
为解决上述技术问题,本申请采用特定比例的薏苡仁、豆蔻、苦杏仁和蓝莓果粉四味药材进行配伍,通过四者的协同作用,改善健脾除湿的症状。同时,将该组合物应用于固体饮料中,使得服用更加方便,口味较佳。此外,该组合物无毒副作用,安全性高。
第一方面,本申请提供了一种具有健脾除湿功能的组合物,所述组合物应用于固体饮料中,由以下重量份数的原料组成:
薏苡仁900~1800份;
豆蔻300~600份;
苦杏仁500~1000份;
蓝莓果粉500~1000份。
可选地,所述组合物由以下重量份数的原料组成:
薏苡仁900~1700份;
豆蔻300~600份;
苦杏仁500~1000份;
蓝莓果粉600~900份。
第二方面,本申请提供了一种具有健脾除湿功能的固体饮料的制备方法,包括以下步骤:
将900~1800份薏苡仁、300~600份豆蔻,以及500~1000份苦杏仁放入水中煎煮,得到第一提取液;其中,薏苡仁、豆蔻和苦杏仁的重量总和,与水的重量之比为1:6~10;
将所述第一提取液干燥成粉末;
将所述粉末与500~1000份蓝莓果粉混匀,得到混合物;
将所述混合物制软材,制粒,干燥,得到所述具有健脾除湿功能的固体饮料。
可选地,将900~1800份薏苡仁、300~600份豆蔻,以及500~1000份苦杏仁放入水中煎煮,得到第一提取液的步骤中,可选地,煎煮的次数为1~3次,每次煎煮时间为1~3小时。
第三方面,本申请提供第一方面的任一种组合物在在制备用于治疗脾胃湿热证的固体饮料或保健食品中的应用。
在上述的组合物中,薏苡仁为方中君药,主要起到促进体内血液和水分的新陈代谢的作用。豆蔻和苦杏仁均为方中臣药,蓝莓为方中佐药。该组合物中的四种药材配伍合用,肺脾同调,通利水道,除一身之湿,协同起到健脾除湿的作用。宣上,畅中,渗下,宣畅三焦气机,健脾渗湿,生津润燥,非常适合脾虚湿盛体质的人服用。
本申请采用薏苡仁、豆蔻、苦杏仁和蓝莓果粉四味药材进行配伍制成固体饮料,该组合物由纯天然药物配制而成,处方简单,将其应用于固体饮料中,同样可以起到健脾除湿的功效,并使得服用更加方便,口味也较好,接受度高。需要说明的是,在应用于固体饮料中时,可以仅采用该组合物,也可以再添加其他必要的辅料,本申请对此不作限定。此外,该组合物无毒副作用,具有较高的安全性。
具体实施方式
以下将通过具体实施方式以及实施例对本申请进行详细说明。
本申请提供了一种具有健脾除湿功能的组合物,所述组合物应用于固体饮料中,由以下重量份数的原料组成:薏苡仁900~1800份;豆蔻300~600份;苦杏仁500~1000份;蓝莓果粉500~1000份。
其中,薏苡仁,《本经》云:“味甘微寒。主筋急,拘挛不可屈伸,风湿痹,下气。久服轻身益气。其根下三虫,一名解蠡。生平泽及田野。”薏苡仁入脾肾二经,并可兼入肺,渗湿利水、健脾除痹,最善利水,而不伤真阴之气,为药食两用之品。
现代药理学研究表明,薏苡仁具有抗肿瘤、降血糖、抗炎镇痛、提高免疫力、调节血脂代谢、抑制骨质疏松等作用。薏苡仁中含有的自由脂肪酸、棕榈酸、硬脂酸、油酸等成分为抗癌抗肿瘤的活性成分,通过改变细胞凋亡机制抑制癌症的发展。还有研究表明,薏苡仁中丰富的膳食纤维有预防结肠癌的作用。此外,薏苡仁中的薏苡仁蛋白含有多种对人体有益的有效成分。例如,其所含的植物抗病原真菌的蛋白,具有良好的抑菌活性和热稳定性;薏苡仁油脂能减缓或消除肌肉痉挛、舒张支气管、改善呼吸功能,并可降低血清钙和血糖。薏苡仁中丰富的蛋白质、维生素E、B1、B12可有效抗衰老,保持人体皮肤光泽细腻,在美容减肥方面有客观的疗效。
豆蔻味辛,苦,温,归肺、脾、大肠经。现代药理学研究表明,豆蔻具有抑菌作用,尤其可抑制痢疾杆菌。同时,豆蔻能促进胃液分泌,促进肠管蠕动,驱除肠内积气,抑制肠内异常发酵。此外,豆蔻还具有芳香化湿、行气宽中,开胃消食之功效;辛温而不热,芳香而气清,入肺经宣滞宽胸,入脾胃化湿行气,温中而能止呕。
苦杏仁味苦,微温;降气止咳平喘,润肠通便,降肺气以启利水之功。现代药理学研究,杏仁具有抗炎、镇痛、抑制胃酸及抗癌等作用,同时具有促进肺表面活性物质合成作用。此外,苦杏仁油有驱虫,杀菌作用。对人蛔虫,蚯蚓等均有杀死作用,并能杀死伤寒、副伤寒杆菌,在临床应用中对蛔虫,钩虫及蛲虫均有效,且无副作用。
蓝莓又名蓝浆果、为越橘属植物,被誉为“水果皇后”,“美瞳之果”。《本草纲目》中记载,蓝莓果实成熟后酸甜可食,可入药,有强筋益气、益肾固精之效。蓝莓果实具有防止神经衰老,增强心脏功能,明目,抗癌及抗心血管疾病等独特的医疗保健作用。研究表明,蓝莓可以强化毛细血管,改善血液循环,减弱血小板的黏滞性,防止血凝块的产生,长期服用可增强心脑功能。

优选地,该组合物中由以下重量份数的原料组成:薏苡仁900~1700份;豆蔻300~600份;苦杏仁500~1000份;蓝莓果粉600~900份。从后续的试验结果也可见,本申请的组合物在上述特定配比之下的健脾除湿效果更佳。
本申请还提供前述的具有健脾除湿功能的固体饮料的制备方法,包括以下步骤:
(a)将900~1800份薏苡仁、300~600份豆蔻,以及500~1000份苦杏仁放入水中煎煮,得到第一提取液;
(b)将所述第一提取液干燥成粉末;
(c)将所述粉末与500~1000份蓝莓果粉混匀,得到混合物;
(d)将所述混合物制软材,制粒,干燥,得到所述具有健脾除湿功能的固体饮料。
可选地,薏苡仁、豆蔻和苦杏仁的重量总和,与水的重量之比为1:6~10。煎煮的次数为1~3次,每次煎煮时间为1~3小时。当分多次煎煮时,可以将每次煎煮完毕的液体分别过滤,然后将所有滤液合并,得到前述的第一提取液。在将第一提取液干燥之后,将干燥的粉末与蓝莓果粉混合均匀,然后制软材,制粒,干燥,得到具有健脾除湿功能的固体饮料。可选地,在制备软材时,可以添加湿润剂。可选地,在干燥制粒步骤所制得的颗粒之后,还可以对其进行整粒。上述的具有健脾除湿功能的固体饮料的制备方法工艺简单,便于工业化生产。
以下将通过具体的实施例来进一步说明本申请的技术方案。
需要说明的是,以下实施例中的蓝莓果粉由通化禾韵现代农业股份有限公司提供,薏苡仁、豆蔻、苦杏仁均购自于亳州万草堂中药饮片有限公司。鼠伤寒沙门氏菌浓度:109cfu/ml,由广东省微生物研究所提供。大鼠IFN-γ、IL-4ELISA试剂盒,由上海研吉生物科技有限公司提供。
药效试验所采用的SD大鼠为SPF级SD大鼠,雄性,体重200±20g,由吉林大学实验动物研究中心提供,动物合格证号:SCXK(吉)-2018-0007,基础饲料和高糖高脂饲料(80%基础饲料+12%猪油+8%蜂蜜),均由吉林大学实验动物中心提供。
电子体温计,广东省中山市泛豪电子有限公司;电子天平,梅特勒-托利多仪器有限公司。
此外,实施例中其他未做特别说明的试剂、原料和仪器设备均可通过商业途径直接购得,其他未注明具体条件的实验方法,按照常规方法和条件,或按照商品说明书选择即可。
实施例1
取薏苡仁1200g、豆蔻400g、苦杏仁900g,加入6倍于药材量的水煎煮一次,每次1小时,滤过,合并滤液,浓缩,干燥、粉碎。与蓝莓果粉混合,加润湿剂制软材,制粒,干燥、整粒,即得具有健脾除湿功能的组合物。
实施例2
取薏苡仁900g、豆蔻600g、苦杏仁1000g,加入6倍于药材量的水煎煮二次,每次1小时,滤过,合并滤液,浓缩,干燥、粉碎,与蓝莓果粉混合,加润湿剂制软材,制粒,干燥、整粒,即得具有健脾除湿功能的组合物。

(1)样本制备
(1.1)薏苡仁-豆蔻水提液组:以重量比3:1的比例取薏苡仁、豆蔻,加8~12倍量水煎煮三次,每次煎煮1.5小时,滤过,合并滤液。浓缩,干燥、粉碎,加润湿剂制软材,18目筛制粒,干燥,整粒。取10g,溶于50ml水中,摇匀,即得样本。
(1.2)豆蔻-苦杏仁水提液组:以重量比1:2的比例取豆蔻、苦杏仁,加8~12倍量水煎煮三次,每次煎煮1.5小时,滤过,合并滤液。浓缩,干燥、粉碎,加润湿剂制软材,18目筛制粒,干燥,整粒。取10g,溶于50ml水中,摇匀,即得样本。
(1.3)薏苡仁-苦杏仁水提液组:以重量比2:1的比例取薏苡仁、苦杏仁,加8~12倍量水煎煮三次,每次煎煮1.5小时,滤过,合并滤液。浓缩,干燥、粉碎,加润湿剂制软材,18目筛制粒,干燥,整粒。取10g,溶于50ml水中,摇匀,即得样本。
(1.4)薏苡仁-豆蔻-苦杏仁水提液组:以重量比4:2:1的比例取薏苡仁、豆蔻、苦杏仁,加8~12倍量水煎煮三次,每次煎煮1.5小时,滤过,合并滤液。浓缩,干燥、粉碎,加润湿剂制软材,18目筛制粒,干燥,整粒。取10g,溶于50ml水中,摇匀,即得样本。
(1.5)蓝莓果粉-薏苡仁-豆蔻水提液组:以重量比3:1的比例取薏苡仁、豆蔻,加8~12倍量水煎煮三次,每次1.5小时,滤过,合并滤液。干燥、粉碎,所得粉末按1:2比例与蓝莓果粉混合,加润湿剂制软材,18目筛制粒,干燥、整粒。取10g,溶于50ml水中,摇匀,即得样本。
(1.6)蓝莓果粉-苦杏仁-豆蔻水提液组:以重量比1:2的比例取豆蔻、苦杏仁,加8~12倍量水煎煮三次,每次1.5小时,滤过,合并滤液。干燥、粉碎,所得粉末按1:2比例与蓝莓果粉混合,加润湿剂制软材,18目筛制粒,干燥、整粒。取10g,溶于50ml水中,摇匀,即得样本。
(1.7)固体饮料的制备:取实施例1~9样品各10g,溶于50ml水中,摇匀,即得到相应的实施例样品1~9。
(2)动物分组、造模及给药
将170只SPF级雄性SD大鼠适应性喂养3天后,称重、采用随机数字法随机分为5组:正常对照组、脾胃湿热模型组、薏苡仁-豆蔻水提液组、豆蔻-苦杏仁水提液组、薏苡仁-苦杏仁水提液组、薏苡仁-豆蔻-苦杏仁水提液组、蓝莓果粉-薏苡仁-豆蔻水提液组、蓝莓果粉-苦杏仁-豆蔻水提液组,以及实施例1~9组,每组10只。
其中,脾胃湿热证模型通过下述方法来造模。将脾胃湿热模型组的所有SD大鼠均于造模前禁食12h,自由饮水,测定体重、肛温。在20-28℃、50-70%相对湿度的自然环境下,给予高脂高糖饲料喂养10天,然后置于温度33±0.5℃,相对湿度65±5%人工环境下,继续给予高脂高糖饲料,共7天。第18天移出置于自然环境,按2ml/200g给予鼠伤寒沙门氏菌灌胃1次,24小时后加强感染1次(1ml/200g),完成脾胃湿热证大鼠模型复制。
第20天起,对薏苡仁-豆蔻水提液组、豆蔻-苦杏仁水提液组、薏苡仁-苦杏仁水提液组、薏苡仁-豆蔻-苦杏仁水提液组、蓝莓果粉-薏苡仁-豆蔻水提液组、蓝莓果粉-苦杏仁-豆蔻水提液组及实施例1~9组的大鼠,分别以2ml/200g体重灌胃。对正常对照组和脾胃湿热模型组予等容积生理盐水。1次/d,连续灌胃1周。
(3)指标测定
(3.1)一般情况观察
观察大鼠的精神状态、皮毛色泽、活动度和大便性状等。每日记录大鼠摄食量和饮水量。分别于造模前、造模第10天、第18天和给药后测定大鼠体重。于造模前、造模后、给药后测定肛温,每个时间点连续测2次,取平均值。结果如(4)中表1至表3所示。
(3.2)Th细胞因子检测
于末次给药后禁食12h,腹腔麻醉,腹主动脉采血5ml,3000r/min离心10min后取得血清,于-20℃冰箱保存待测。严格按照ELISA试剂盒说明书操作,分别检测大鼠血清γ-干扰素(IFN-γ)、白细胞介素-4(IL-4)水平,酶标仪450nm处读取吸光度(D值),根据标推曲线计算IFNγ/IL-4的浓度,以ng/L为单位,并计算IFNγ/IL-4比值。结果如(4)中表4所示。
(4)统计学处理
实验数据采用SPSS13.0统计软件进行统计学处理,结果以表示。对饮水量、摄食量、体重、肛温采用重复测量方差分析,组间比较采用单因素方差分析,P<0.05为差异有统计学意义。

正常对照组大鼠精神状态好,皮毛有光泽,反应灵敏,饮水和摄食量均衡。脾胃湿热模型组大鼠逐渐出现精神萎靡,嗜卧懒动,皮毛无光泽,反应迟钝,大便稀或溏泄,肛周污秽,饮水量及摄食量逐渐减少。给药后薏苡仁-豆蔻水提液组、豆蔻-苦杏仁水提液组、薏苡仁-苦杏仁水提液组、薏苡仁-豆蔻-苦杏仁水提液组、蓝莓果粉-薏苡仁-豆蔻提液组、蓝莓果粉-苦杏仁-豆蔻水提液组及实施例1~9组的大鼠,一般状态均逐渐好转,大便湿润或转为正常,饮水量及摄食量有所增加,各组间统计学无显著性差异,具体如表1所示。

(4.2)对脾胃湿热证大鼠体重和肛温的影响
正常对照组大鼠体重呈进行性增长,肛温无明显改变。脾胃湿热模型组大鼠体重增长缓慢,肛温进行性升高。给药后薏苡仁-豆蔻水提液组、豆蔻-苦杏仁水提液组、薏苡仁-苦杏仁水提液组、薏苡仁-豆蔻-苦杏仁水提液组、蓝莓果粉-薏苡仁-豆蔻水提液组、蓝莓果粉-苦杏仁-豆蔻水提液组,以及实施例1~9组的大鼠体重增长较模型组加快(P<0.05),肛温较前明显下降,各组间无显著性差异,具体如表2和表3所示。

(4.3)对脾胃湿热证大鼠血清IFN-γ、IL-4和IFN-γ/IL-4比值的影响
(4.3.1)IFN-γ、IL-4表达水平
与正常对照组相比,脾胃湿热模型组、薏苡仁-豆蔻水提液组、豆蔻-苦杏仁水提液组、薏苡仁-苦杏仁水提液组、薏苡仁-豆蔻-苦杏仁水提液组、蓝莓果粉-薏苡仁-豆蔻水提液组、蓝莓果粉-苦杏仁-豆蔻水提液组,以及实施例1~9组的大鼠的IFN-γ、IL-4水平明显升高,以脾胃湿热模型组为最高,具体如表4所示。
与脾胃湿热模型组相比,薏苡仁-豆蔻水提液组、豆蔻-苦杏仁水提液组、薏苡仁-苦杏仁水提液组、薏苡仁-豆蔻-苦杏仁水提液组、蓝莓果粉-薏苡仁-豆蔻提液组、蓝莓果粉-苦杏仁-豆蔻水提液组以及实施例1~9组,明显降低了脾胃湿热证大鼠血清IFN-γ、IL-4的表达水平。并且,实施例1~9组能更加显著降低湿热证大鼠血清IL-4水平,具体如表4所示。
(4.3.2)IFN-γ/IL-4比值
与正常对照组比较,实施例1~9组IFN-γ/IL-4比值升高(P<0.01),模型组IFN-γ/IL-4比值有升高的趋势,具体如表4所示。
与脾胃湿热模型组比较,实施例1~9组IFN-γ/IL-4比值升高,蓝莓果粉-薏苡仁-豆蔻水提液组、蓝莓果粉-苦杏仁-豆蔻水提液组有降低IFN-γ/IL-4比值的趋势,具体如表4所示。

脾胃湿热模型组大鼠的胃肠道粘膜存在一定程度的炎症损伤,IFN-γ和IL-4表达显著升高,提示脾胃湿热证大鼠的细胞免疫和体液免疫均呈亢进状态,表明脾胃湿热证大鼠以较强的炎症反应为主要表现,处于“邪正剧烈抗争”的病理状态。IFN-γ/IL-4比值稍增高,提示可能存在免疫调节的功能紊乱,即Th1/Th2平衡失调,趋向于以Thl型为主。
在本实验结果中,脾胃湿热模型组大鼠体重增长缓慢,肛温进行性升高。给药后薏苡仁-豆蔻水提液组、豆蔻-苦杏仁水提液组、薏苡仁-苦杏仁水提液组、薏苡仁-豆蔻-苦杏仁水提液组、蓝莓果粉-薏苡仁-豆蔻水提液组、蓝莓-苦杏仁-豆蔻水提液组,以及实施例1~9组均能减轻脾胃湿热证大鼠胃肠道病理损伤,不同程度地降低IFN-γ、IL-4的表达水平,调节IFN-γ/IL-4比值。这表明本申请的组合物对脾胃湿热证大鼠的免疫功能有较好的调控作用,可维持机体的免疫稳态,保护胃肠道粘膜,这可能是其清热祛湿的作用机制之一。由此可见,本申请的组合物的健脾除湿作用显著。
此外,整个实验过程中,小鼠的活动状态无异常,且检测过程中生化指标均无异常。这表明本申请的组合物对受试动物健康无明显影响,具有较高的安全性。
本文中应用了具体个例对本发明的实施方式进行了阐述,以上实施例的说明只是用于帮助理解本发明的方法及其核心思想。应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以对本发明进行若干改进和修饰,这些改进和修饰也落入本发明权利要求的保护范围内。
Claims (5)
1.一种具有健脾除湿功能的组合物,其特征在于,所述组合物应用于固体饮料中,由以下重量份数的原料组成:
薏苡仁900~1800份;
豆蔻300~600份;
苦杏仁500~1000份;
蓝莓果粉500~1000份。
2.根据权利要求1所述的组合物,其特征在于,所述组合物由以下重量份数的原料组成:
薏苡仁900~1700份;
豆蔻300~600份;
苦杏仁500~1000份;
蓝莓果粉600~900份。
3.一种具有健脾除湿功能的固体饮料的制备方法,其特征在于,包括以下步骤:
将900~1800份薏苡仁、300~600份豆蔻,以及500~1000份苦杏仁放入水中煎煮,得到第一提取液;其中薏苡仁、豆蔻和苦杏仁的重量总和,与水的重量之比为1:6~10;
将所述第一提取液干燥成粉末;
将所述粉末与500~1000份蓝莓果粉混匀,得到混合物;
将所述混合物制软材,制粒,干燥,得到所述具有健脾除湿功能的固体饮料。
4.根据权利要求3所述的制备方法,其特征在于,煎煮的次数为1~3次,每次煎煮时间为1~3小时。
5.权利要求1-2任一所述的组合物在制备用于治疗脾胃湿热证的固体饮料或保健食品中的应用。
