Product evaluation
Firstly, evaluating the safety of products
The whitening and freckle-removing creams of examples 1 to 14 and comparative examples 1 to 5 were evaluated for irritation of cosmetics.
The test population: 100 women aged 20-50 years have healthy skin and no allergic history of skin diseases, and accord with the voluntary selection standard of the subjects.
Grouping: the test population was divided into 20 groups of 5 individuals each, with the members in the group differing in age by no more than 2 years.
The spot pasting method comprises the following steps: selecting a qualified spot tester, placing 0.020-0.025g of a tested object in the spot tester in a closed spot patch experimental mode, attaching the spot tester with the tested object to the bent side of the back or the forearm of a tested object by using a non-irritating adhesive tape, slightly pressing the bent side with a palm to uniformly attach the tested object to the skin, removing the tested object after 24 hours, observing skin reactions after 0.5, 6, 12, 24 and 48 hours after the tested object is removed, and recording the results according to skin reaction grading standards in cosmetic hygiene standards and cosmetic safety technical standards, wherein the test results are shown in table 1.
Evaluation criteria: level 0: negative reaction; level 1: suspicious reactions, only faint erythema; and 2, stage: weak positive reaction, erythema, infiltration, edema, and possibly pimple; and 3, level: strong positive reaction, erythema, infiltration, edema, pimple and reaction beyond the tested area; 4, level: very strong positive reaction, obvious erythema, severe infiltration, edema, herpes fusiformis and reaction beyond the tested area.

As can be seen from Table 1, the results of the human skin patch test show that the skin reactions of all groups of test subjects are observed within 0.5 hour, 6 hours, 12 hours, 24 hours and 48 hours after all groups of test subjects pass the patch test of all groups of test substances, and the skin reactions of all the test subjects are negative, so that the whitening and freckle-removing cream prepared by the method is safe to use.
Second, product functionality evaluation
Efficacy tests were performed on the whitening and spot-removing creams of examples 1 to 14 and comparative examples 1 to 5.
(1) Test for Melanin reduction Rate
Selecting 100 women and testing the skin melanin reduction rate between the ages of 20-50, dividing a tested human group into 20 groups, respectively correspondingly coating the whitening and freckle-removing creams of examples 1-14 and comparative examples 1-5 on 5 persons in each group and 20 groups of subjects, taking stratum corneum cells of the skin of the subjects by using a scraping blade before the subjects are coated with the whitening and freckle-removing creams for the first time, storing for later use, coating the whitening and freckle-removing creams for 28 days, taking stratum corneum cells of the same subjects at the same position, respectively dyeing the stratum corneum cells scraped in the two times, respectively, visualizing the melanin content, and performing data analysis on skin images of the subjects to obtain a melanin semi-quantitative analysis result, wherein the test result is shown in a table 2.
(2) Inhibition of tyrosinase assay
Respectively taking 0.025g of the whitening spot-lightening creams of the examples 1 to 14 and the comparative examples 1 to 5, placing the whitening spot-lightening creams into test tubes, wherein the numbers are 1 to 20, respectively adding 120 mu L of phosphate buffer solution and 40 mu L of 2.98 mmol/L of tyrosine solution (2.98 mmol/L of tyrosine solution is used as a substrate) into the test tubes 1 to 20, respectively adding the mixed solution in the test tubes 1 to 20 in equal amount into a 96-well plate, adding 40 mu L of 200U/mL of tyrosinase solution into each well after the constant temperature of water bath at 37 ℃ is kept for 10 min, uniformly mixing the mixture under the condition of keeping out of the sun at room temperature, quickly placing the mixture in a multifunctional microplate detector after the water bath at 37 ℃ is carried out for 15 min, measuring the absorbance value at 475 nm, calculating the tyrosinase inhibition rate, repeating the operation, carrying out parallel measurement for 3 times, and obtaining the test results shown in Table 2.
(3) Determination of DPPH radical scavenging Activity
Antioxidant activity is expressed as the ability to scavenge DPPH free radicals. According to the experimental conditions determined by the preliminary experiment, the activities of the whitening and freckle-removing cream of the examples 1 to 14 and the comparative examples 1 to 5 for removing the DPPH (free fatty radical) are measured. Taking 1.0g of each sample, adding 2.0 ml of 1.52X 10-4mmol/L DPPH absolute ethanol solution, mixing well, standing at room temperature in dark place for 30min, taking a solvent as a blank control, and measuring the absorbance A1 at 517 nm; replacing the sample solution with the same volume of solvent to serve as a negative control, and measuring the absorbance A2; the absorbance A3 was measured using the same volume of absolute ethanol as the blank instead of the DPPH solution. VC was used as a control, 3 replicates of each group were averaged, the test results are shown in table 2, and the clearance was calculated as follows:

From the test results in table 2, it can be seen that the melanin reduction rate of the subjects coated with the whitening and freckle-removing creams of examples 1 to 14 is lower than that of comparative examples 1 and 5 only in example 3, and example 14 is equivalent to comparative example 5 but lower than that of comparative example 1, and the other examples are higher than that of comparative example; the subjects who are smeared with the whitening and freckle-removing cream in the examples 1 to 14 have higher tyrosinase inhibition rate than the subjects who are smeared with the whitening and freckle-removing cream in the comparative examples 1 to 5; the DPPH clearance rate of oxygen free radicals of the subjects coated with the whitening and freckle-removing cream of the examples 1 to 3 is higher than that of the comparative examples 1 to 5, and the DPPH clearance rates of the examples 4 to 14 are equivalent to that of the comparative examples 1 and 2 but are higher than that of the comparative examples 3 to 5; therefore, the whitening and freckle-removing cream provided by the invention can effectively reduce melanin, inhibit the activity of tyrosinase, has strong antioxidant activity and has good whitening and freckle-removing effects. The whitening and freckle-removing cream in example 1 has the best efficacy, and the melanin reduction rate, the tyrosinase inhibition rate and the oxygen radical DPPH clearance rate are the highest, and are 65%, 55% and 35% respectively.
Finally, it should be noted that the above embodiments are only used for illustrating the technical solutions of the present invention and not for limiting the protection scope of the present invention, and although the present invention is described in detail with reference to the preferred embodiments, it should be understood by those skilled in the art that modifications or equivalent substitutions can be made on the technical solutions of the present invention without departing from the spirit and scope of the technical solutions of the present invention.

The whitening and freckle-removing cream is characterized by comprising the following freckle-removing agents in percentage by weight: 0.05-3% of undecylenoyl phenylalanine, 0.01-10% of 3-o-ethyl ascorbic acid, 0.01-5% of a mixture a, 0.01-5% of a mixture b and 0.01-5% of a mixture c; the mixture a comprises phenethyl resorcinol; the mixture b comprises yeast extract; said mixture c comprises hydrolysed plums;
also comprises the following skin conditioning agents in percentage by weight: 0.01-1% of bisabolol, 0.01-5% of phytosterol oleate, 0.01-5% of silanediol salicylate and 0.01-1% of sodium hyaluronate; the preservative also comprises the following preservatives in percentage by weight: 0.01-0.4% of methyl hydroxybenzoate, 0.01-0.1% of propyl hydroxybenzoate, 0.01-1% of phenoxyethanol/ethylhexyl glycerol and 0.01-0.3% of sodium bisulfite; also comprises the following emollients in percentage by weight: 0.1-5% of hydrogenated polyisobutene, 0.1-10% of isononyl isononanoate, 0.01-10% of camellia seed oil, 0.1-5% of polydimethylsiloxane, 0.1-5% of cyclopenta dimethyl siloxane, 0.1-10% of shea butter and 0.1-10% of cocoa seed fat; the emulsifier also comprises the following components in percentage by weight: 0.1-5% of mixture d, 0.1-5% of mixture e and 0.01-3% of potassium cetyl phosphate; the mixture d comprises one or more of arachidyl alcohol, behenyl alcohol, arachidyl alcohol glucoside, water and glucose; the mixture e comprises one or more of glyceryl stearate, cetearyl alcohol, stearic acid and sodium lauroyl glutamate; the thickener also comprises the following components in percentage by weight: 0.01-3% of sodium acrylate copolymer/lecithin, 0.01-1% of carbomer, 0.1-5% of cetearyl alcohol and 0.1-3% of mixture f; the mixture f comprises one or more of sodium acrylate/sodium acryloyldimethyl taurate copolymer, isohexadecane, polysorbate-80, water and sorbitan oleate; the antioxidant also comprises the following antioxidants in percentage by weight: 0.01-0.1% of pentaerythritol tetra (bis-tert-butyl hydroxy hydrocinnamate) and 0.01-3% of tocopherol; the environment-friendly type water-based deodorant also comprises 0.01-1% of a pH regulator, 0.1-10% of a humectant, 0.01-1% of an aromatic and a solvent, wherein the solvent is water, the pH regulator is aminomethyl propanol, the humectant is butanediol, the aromatic is essence.

The cream of claim 1, wherein the undecylenoyl phenylalanine is 1%, the 3-o-ethyl ascorbic acid is 3%, the mixture a is 3%, the mixture b is 2%, the mixture c is 3%, the bisabolol is 0.03%, the phytosterol oleate is 1%, the silanediol salicylate is 1%, and the sodium hyaluronate is 0.03% by weight of the cream.
3. The method for preparing the whitening and freckle-removing cream according to claim 1, which comprises the following steps:
(1) weighing the following raw materials in proportion:
phase A raw materials: water, butanediol, sodium hyaluronate, sodium acrylate copolymer/lecithin, and carbomer;
b phase raw materials: mixture d, mixture e, hydrogenated polyisobutene, isononyl isononanoate, camellia oil, cetostearyl alcohol, polydimethylsiloxane, cyclopentadimethylsiloxane, undecylenoylphenylalanine, methylparaben, propylparaben, pentaerythritol tetrakis (di-tert-butyl hydroxyhydrocinnamate), bisabolol, phytosterol oleate, tocopherol, shea butter, cocoa butter, potassium cetyl phosphate, mixture a;
c phase raw material: a mixture f;
