空气污染、生活饮食不规律、工作压力大等原因导致现代人的口腔及咽喉疾病的发生率很高,相关口腔及咽喉疾病的治疗产品也非常多,涉及的中草药原料包含罗汉果、菊花、桔梗、薄荷、胖大海、冬凌草等,这些原料均为药食同源原料,功效温和,无不良滋味或气味,对一些急症或严重的病情,这些产品的效果略显不足。百药煎是一味通过发酵而成的中药饮片,《本草述钩元》记载“五倍子为粗末,每斤以真茶一两,煎浓汁,入酵四两,擂烂拌和,器盛,置糠缸署之,待发起如麹即成已。经过临床验证,百药煎在治疗牙周炎、牙龈炎、牙龈出血、复发性口腔溃疡、口舌生疮、口臭、牙痛、小儿原发性疱疹性口炎方面表现出色。百药煎对急慢性咽炎,尤其对于吸烟人士咽炎具有十分显著功效。对于感冒引起的扁桃体炎、咳嗽、失声、声音嘶哑均有良好效果。
关于百药煎应用也有相关的专利,专利CN 102742698 B公开了一种百药煎保健茶,由百药煎、地血香果、空桶参、建兰花按一定重量配比而制成;该保健茶具有润肺化痰,生津止咳,止咳祛痰,消炎等功效。CN 103463376 B公开了一种由矮地茶、瓜子金、百药煎、竹根七、肿节风等组成的中药配方,其水煎制成汤剂用于治疗小儿咳嗽具有良好的疗效。但由于现有的百药煎产品的酸涩味非常明显,口感较差,虽然效果明显,但服用依从性较差且服用不方便,大大限制了该产品的应用。
发明内容
为了克服上述现有技术的不足,本发明的首要目的在于提供一种口感好且方便使用的喷雾剂,该喷雾剂喷入口腔,药液即与口腔、咽喉充分接触,能有效缓解口腔咽喉不适感,具有较好的消炎止痛利咽功效,且口感好,提高了产品的使用依从性。
本发明的另一目的在于提供上述缓解口腔及咽喉不适的喷雾剂的制备方法。
本发明是通过以下技术方案实现:
一种缓解口腔及咽喉不适的喷雾剂,由下述重量份的原料制成:百药煎3-5份,绿茶3-5份,薄荷3-5份,聚乙烯吡咯烷酮0.05-0.1份,甜味剂0.12-0.15份,薄荷脑0.03-0.05份,乙醇5-10份,水90-95份。
作为本发明进一步优选的技术方案,所述一种缓解口腔及咽喉不适的喷雾剂,由下述重量份的原料制成:百药煎5份,绿茶3份,薄荷2份,甜味剂0.15份,聚乙烯吡咯烷酮0.10份薄荷脑0.05份,乙醇10份,水90份。
本发明中各原料的药理作用如下所述:
百药煎:性平,味酸、甘;归肺、胃经。清热化痰,生津止渴。用于肺热咳嗽,风火牙痛,口舌糜烂,风湿诸疮等症。百药煎由五倍子、茶叶经过发酵而得,产品中含有丰富的没食子酸,没食子酸具有抗炎、抗突变、抗氧化、抗自由基等多种生物学活性,同时具有抗菌抗病毒作用,体外对金黄色葡萄球菌、绿脓杆菌、弗氏痢疾杆菌等有抑制作用,对流感病毒亦有一定抑制作用。
薄荷:性凉,味辛;归肺、肝经。疏散风热,清利头目,利咽,透疹,疏肝行气。可用于风热头痛,目赤,喉痹,口疮,胸胁胀闷等症。薄荷含有挥发性油、蛋白质、脂肪、碳水化合物、矿物质、维生素等成分。薄荷挥发油中的薄荷脑成分具有杀菌止痒功效,涂于局部可刺激神经而引起凉感,并抑制痛觉神经,还可舒缓呼吸道,具有清咽止咳功能。
绿茶:绿茶中含有丰富的茶多酚,茶多酚除具有抗氧化作用外,还具有抑菌作用,如对葡萄球菌、大肠杆菌、枯草杆菌等有抑制作用。
本发明百药煎具有较强的药效,配以香味浓郁宜人,风味独特的绿茶和薄荷,三者协同合用,一方面起到增加产品抗菌消炎效果,另一方面可以降低产品酸涩味,口感好,提高了产品的依从性,可以快速缓解因口腔溃疡、咳嗽痰多或咽喉肿痛引起的不适。
本发明还公开了上述一种缓解口腔及咽喉不适的喷雾剂的制备方法,包括如下步骤:
a)取百药煎粗碎成10-30目;取薄荷洗净,于50℃以下低温干燥,粉碎成14-30目粗颗粒;取绿茶粉碎成14-30目粗颗粒;
b)取百药煎、薄荷、绿茶10-15倍量水,煮沸,加入步骤a)项下粗颗粒,保温浸提20-30分钟,过滤,滤液高速离心,得离心液;
c)取薄荷脑,用乙醇溶解,加入聚乙烯吡咯烷酮,混匀,得薄荷脑液;
d)取b)的离心液,搅拌下加入甜味剂及薄荷脑液,混匀;
e)紫外灭菌40-60分钟,灌装,即得。
优选的,步骤a)中,所述百药煎、薄荷、绿茶粉碎后30目以下的细粉低于10%。
优选的,步骤b)中,所述滤液用高速管式离心机离心,离心的转速为11000-14000r/min。
为了更好地实现本发明的技术效果,本发明分别对配方筛选、甜味剂、辅料量进行了优化研究,具体过程如下:
试验1:配方筛选
按表中比例按相同工艺分别制备不同喷雾剂产品,对口感、风味进行比较,确定较佳的处方比例。
工艺:取原料10倍量水,煮沸,加入原料,保温浸提0.5小时,滤过,滤液加入甜菊糖苷,加纯化水定容到100份,即得,结果见下表。
由表中结果可知,百药煎的重量份数为6份以上时,溶液不稳定,容易产生沉淀,且酸涩味明显。为了保证疗效,本发明添加了同样对口腔和咽喉疾病有治疗作用的绿茶和薄荷,通过筛选三者的比例,可以达到口感、效果、稳定性兼顾的目的。由上述结果,确定本发明中各药物的重量份数为:百药煎3-5份,绿茶3-5份,薄荷3-5份。同时由表中可知,按该比例制取的提取液,除了酸涩味,均有明显苦味,需要对甜味剂进一步筛选。
试验2:甜味剂的筛选
实验方法:按百药煎5份,绿茶3份,薄荷2份比例称取原料,加水提取定容,以不同的甜味剂配比,经同一制备工艺制备得到喷雾剂产品,分别对其口感进行评分,包括喷雾剂甜度、品尝后味;
实验结果:评分结果如下表所示;
由实验数据可得,本发明配方加了甜味剂后,产品苦味得到很好改善,优选的,甜味剂的配比为甜菊糖苷与三氯蔗糖的质量比1-3:1,更优选为2:1。
试验3:辅料的筛选:
实验方法:按百药煎5份,绿茶3份,薄荷2份比例称取原料,加水提取定容,甜味剂添加量为甜菊糖苷为0.1份,三氯蔗糖为0.05份,分成9份。取薄荷脑加乙醇溶解后,加入聚乙烯吡咯烷酮(PVPK30)溶解,搅拌下到水液中,分别制成喷雾剂,分别对其口感进行评分。
实验结果如下表所示;
由实验数据可得,本发明在配方中通过添加适量的薄荷脑及聚乙烯吡咯烷酮可以有效改善其酸涩味,优选的,乙醇用量为5份-10份,薄荷脑用量为0.03份-0.05份,PVPK30用量为0.05份-0.10份。更优的比例为乙醇用量为10份,薄荷脑为0.05份,PVPK30用量为0.10份。
本发明与现有技术相比,具有如下有益效果:
(1)本发明通过配方及添加剂的筛选,很好地解决了百药煎口感酸涩明显,依从性较差的问题,制得的喷雾剂喷入口腔,药液即与口腔、咽喉充分接触,能有效缓解口腔及咽喉不适感,具有较好的消炎止痛利咽功效,使用方便,且具有绿茶薄荷特殊风味,使用后口气清新,是一种居家必备良药。
(2)本发明的提取工艺具有以下优点:提取原料粗碎成粗粉,利于提取有效成分,同时避免粉碎过细,溶液糊化难以过滤;沸水投料保温提取可以减少百药煎冷水煮的糊化现象,利于有效成分溶出,同时利于保留薄荷及绿茶香气;提取液经过筛网粗滤后再经过高速管式离心机离心,可以有效去除药材细粉、淀粉等物质,避免溶液产生沉淀,影响产品品质和质量,保证产品的稳定性。
具体实施方式
下面通过具体实施方式来进一步说明本发明,以下实施例为本发明具体的实施方式,但本发明的实施方式并不受下述实施例的限制。
本发明的原料均来源于市购。
实施例1:
原料配比:百药煎5份,绿茶3份,薄荷2份,甜味剂(甜菊糖苷0.10份,三氯蔗糖0.05份),聚乙烯吡咯烷酮0.10份,薄荷脑0.05份,乙醇10份,水90份。
其制备工艺包括如下步骤:
a)取百药煎粗碎成10-30目(30目以下细粉低于10%);取薄荷洗净,于50℃以下低温干燥,粉碎成14-30目粗颗粒(30目以下细粉低于10%),绿茶粉碎成14-30目粗颗粒(30目以下细粉低于10%);
b)取百药煎、薄荷、绿茶12倍量水,煮沸,加入步骤a)项下粗颗粒,保温浸提30分钟,过滤,滤液用高速管式离心机(转速12000r/min)离心,得离心液;
c)取薄荷脑,用乙醇溶解,加入聚乙烯吡咯烷酮(PVPK30),混匀,得薄荷脑液;
d)取b)的离心液,搅拌下加入甜味剂及薄荷脑液,混匀;
e)紫外灭菌40-60分钟,灌装,即得。
实施例2:
原料配比:百药煎4份,绿茶3份,薄荷3份,甜味剂(甜菊糖苷0.08份,三氯蔗糖0.04份),聚乙烯吡咯烷酮0.06份,薄荷脑0.05份,乙醇10份,水90份。
其制备工艺包括如下步骤:
a)取百药煎粗碎成10-30目(30目以下细粉低于10%);取薄荷洗净,于50℃以下低温干燥,粉碎成14-30目粗颗粒(30目以下细粉低于10%),绿茶粉碎成14-30目粗颗粒(30目以下细粉低于10%);
b)取百药煎、薄荷、绿茶15倍量水,煮沸,加入步骤a)项下粗颗粒,保温浸提20分钟,过滤,滤液用高速管式离心机(转速13000r/min)离心,得离心液;
c)取薄荷脑,用乙醇溶解,加入聚乙烯吡咯烷酮(PVPK30),混匀,得薄荷脑液;
d)取b)的离心液,搅拌下加入甜味剂及薄荷脑液,混匀;
e)紫外灭菌40-60分钟,灌装,即得。
实施例3:
原料配比:百药煎3份,绿茶3份,薄荷5份,甜味剂(甜菊糖苷0.10份,三氯蔗糖0.05份),聚乙烯吡咯烷酮0.05份,薄荷脑0.03份,乙醇10份,水90份。
其制备工艺包括如下步骤:
a)取百药煎粗碎成10-30目(30目以下细粉低于10%);取薄荷洗净,于50℃以下低温干燥,粉碎成14-30目粗颗粒(30目以下细粉低于10%),绿茶粉碎成14-30目粗颗粒(30目以下细粉低于10%);
b)取百药煎、薄荷、绿茶10倍量水,煮沸,加入步骤a)项下粗颗粒,保温浸提25分钟,过滤,滤液用高速管式离心机(转速11000r/min)离心,得离心液;
c)取薄荷脑,用乙醇溶解,加入聚乙烯吡咯烷酮(PVPK30),混匀,得薄荷脑液;
d)取b)的离心液,搅拌下加入甜味剂及薄荷脑液,混匀;
e)紫外灭菌40-60分钟,灌装,即得。
对比例1:
原料配比同实施例1;
制备工艺:取薄荷,洗净,切段2-3cm,取百药煎粗碎成0.5-1cm小块状;取绿茶,一起加15倍水,加热煮沸提取1小时,滤过,滤液不离心;另外薄荷脑加入乙醇溶解,再加入聚乙烯吡咯烷酮溶解,边搅拌边加入上述滤液中,加入甜菊糖苷和三氯蔗糖,搅拌溶解,混匀,即得。
对比例2:
百药煎4份,绿茶3份,薄荷3份,甜味剂(甜菊糖苷0.08份,三氯蔗糖0.04份),加水至100份。
制备工艺:取薄荷,洗净,切段2-3cm,取百药煎粗碎成0.5-1cm小块状;取绿茶,一起加15倍水,加热煮沸提取2小时,滤过,滤液不离心,加入甜菊糖苷和三氯蔗糖,搅拌溶解,混匀,即得。
分别取实施例和对比例的喷雾剂,进行感官评测,其结果如下表所示。
人群试食试验:
选取40位年龄35-50岁的有慢性咽炎或感冒后咳嗽超过5天的志愿者,随机分成两组,第一组喷实施例2样品,每天喷2次,每次喷雾5次,第二组使用对比例2喷雾剂,两组均使用5天后观察结果,收集反馈信息,结果见下表。
由上述结果可以看出,本发明将百药煎与绿茶、薄荷组合制备得到的喷雾剂产品,具有很好消炎止痛及止咳化痰效果,可有效缓解口腔及咽喉适感,使用方便,口感风味良好,容易被患者接受,是一种居家必备良药。
Claims (8)
1.一种缓解口腔及咽喉不适的喷雾剂,其特征在于,由下述重量份的原料制成:百药煎3-5份,绿茶3-5份,薄荷3-5份,聚乙烯吡咯烷酮0.05-0.1份,甜味剂0.12-0.15份,薄荷脑0.03-0.05份,乙醇5-10份,水90-95份。
2.根据权利要求1所述的一种缓解口腔及咽喉不适的喷雾剂,其特征在于,由下述重量份的原料制成:百药煎5份,绿茶3份,薄荷2份,甜味剂0.15份,聚乙烯吡咯烷酮0.10份薄荷脑0.05份,乙醇10份,水90份。
3.根据权利要求1或2所述的一种缓解口腔及咽喉不适的喷雾剂,其特征在于,所述甜味剂为甜菊糖苷或三氯蔗糖中一种或两种混合物。
4.根据权利要求3所述的一种缓解口腔及咽喉不适的喷雾剂,其特征在于,所述甜味剂为甜菊糖苷与三氯蔗糖的质量比为1-3:1。
5.根据权利要求3所述的一种缓解口腔及咽喉不适的喷雾剂,其特征在于,所述甜味剂为甜菊糖苷与三氯蔗糖的质量比为2:1。
6.根据权利要求1-5任一项所述的一种缓解口腔及咽喉不适的喷雾剂的制备方法,其特征在于,包括如下步骤:
a)取百药煎粗碎成10-30目粗颗粒;取薄荷洗净,于50℃以下低温干燥,粉碎成14-30目粗颗粒; 取绿茶粉碎成14-30目粗颗粒;
b) 取百药煎、薄荷、绿茶10-15倍量的水,煮沸,加入步骤a)项下粗颗粒,保温浸提20-30分钟,过滤,滤液高速离心,得离心液;
c)取薄荷脑,用乙醇溶解,加入聚乙烯吡咯烷酮,混匀,得薄荷脑液;
d)取b)的离心液,搅拌下加入甜味剂及薄荷脑液,混匀;
e)紫外灭菌40-60分钟,灌装,即得。
7.根据权利要求6所述的一种缓解口腔及咽喉不适的喷雾剂的制备方法,其特征在于,步骤a)中,所述百药煎、薄荷、绿茶粉碎后30目以下的细粉低于10%。
8.根据权利要求6所述的一种缓解口腔及咽喉不适的喷雾剂的制备方法,其特征在于,步骤b)中,所述滤液用高速管式离心机离心,离心的转速为11000-14000r/min。
