Abstract
本发明公开了一种发酵饲料添加剂的制备方法及其应用,属于动物饲料领域。该添加剂利用液固两步发酵法生产,首先把益生菌枯草芽孢杆菌、嗜酸乳杆菌、丁酸梭菌、酿酒酵母和布拉氏酵母进行液体发酵,发酵结束后与固态原料进行固态混合发酵5‑7天,干燥包装,再复配复合预混料和复合酶,即成成品。原料以玉米皮、玉米浆、玉米淀粉糖糖渣、玉米蛋白粉等为主,营养全面,经微生物发酵后还产生大量活性物质。该发酵饲料添加剂用于蛋鸡养殖,可提高其机体免疫力、增强抗病能力,并且可提高蛋鸡的产蛋率和鸡蛋品质。本产品原料来源广泛、价格低廉,生产过程中无三废产生,环保安全。因此本发明有较好的市场应用价值。

技术领域
本发明涉及动物饲料技术领域,具体是一种发酵饲料添加剂的制备方法及应用。
背景技术
微生态饲料添加剂是一类新型饲料添加剂,对调节畜禽肠道微生物群落,在动物机体内可改善消化道菌群及酶的平衡,迅速提高机体的抗病能力、代谢能力和消化吸收能力,从而达到防治消化道疾病和促进生长的双重作用。微生态制剂是纯生物产品,不含任何化学合成成分,对环境无污染和毒副作用。因此,微生态制剂在饲料添加剂中的推广和应用无疑会对畜禽的健康生长,对保证畜产品的质量及提高人们的生活质量具有非常重要的意义。
近半个世纪以来,饲料抗生素对预防动物疾病,促进动物生长,提高饲料利用率等方面确实起到了积极的作用。但是在饲料中添加抗生素,导致的食品抗生素残留和产生耐药性细菌的概率也大幅增加,加大了人类和动物治疗疾病的难度,对人体健康构成了巨大威胁。抗生素对养殖业和食品安全产生的不利作用也日益凸显,禁止动物饲料中添加抗生素已是大势所趋。
目前,微生态制剂已被世界上公认为有希望代替抗生素的饲料添加剂,包括我国在内,世界上已有许多国家大量使用微生态制剂。使用微生态制剂有望达到饲料抗生素的应用效果,而且动物更健康、食品更安全、无抗药性、不污染环境。益生菌发酵饲料添加剂是将不含抗生素的饲料与益生菌和水等混合后发酵而成的非全价饲料。具有体积小运输使用方便的优点, 是最具市场前景的一类发酵饲料产品。
发明内容
本发明的目的是提供一种发酵饲料添加剂,能促进蛋鸡对饲料的消化和吸收,增强鸡的机体免疫能力,防治腹泻,少发病或不发病,减少抗生素和其它药物的使用,最终降低食品中的药物残留,为人类提供安全的鸡蛋产品。同时,也减少鸡粪便中抗生素含量,降低粪便中抗生素对环境的污染。
本发明的另一个目的是一种发酵饲料添加剂的制备方法,适合大规模推广生产。
本发明的技术方案如下:
一种发酵饲料添加剂,其特征在于,它由微生物混合发酵液10 - 20%、玉米皮10 -20%、玉米浆20 – 40%、玉米淀粉糖糖渣10 - 20%、玉米蛋白粉10 - 20%、水10%等重量比的原料组成。把上述原料搅拌均匀,装入固体发酵容器或者密封袋子中,32 - 37℃发酵7 -10天,烘干。把复合预混料和复合酶加入上述烘干后的固态发酵物中,各物料占重量比为:复合预混料3 - 6%、复合酶3 - 5%、石粉7%、固态发酵物82 - 87%,充分搅拌混合均匀,即为发酵饲料添加剂。
作为优选的,一种发酵饲料添加剂,由微生物混合发酵液20%、玉米皮10%、玉米浆30%、玉米淀粉糖糖渣20%、玉米蛋白粉10%、水10%等重量比的原料组成。把上述原料搅拌均匀,装入固体发酵容器或者密封袋子中,32 - 37℃发酵7 - 10天,烘干。把复合预混料和复合酶加入上述烘干后的固态发酵物中,各物料占重量比为:复合预混料6%、复合酶5%、石粉7%、固态发酵物82%,充分搅拌混合均匀,制成的发酵饲料添加剂。
一种发酵饲料添加剂由益生菌枯草芽孢杆菌、嗜酸乳杆菌、丁酸梭菌、酿酒酵母和布拉氏酵母,混合发酵而成。
一种发酵饲料添加剂,是经三级液体发酵和固态发酵等4次发酵制备而成。一级液体发酵即一级种子发酵,由枯草芽孢杆菌、嗜酸乳杆菌、丁酸梭菌、酿酒酵母和布拉氏酵母等5种微生物分别进行的两级种子扩培纯种发酵,三级发酵是5种种子液共同接种液体发酵培养基后混合发酵而成。最后混合发酵液接种玉米皮、玉米浆、玉米淀粉糖糖渣和玉米蛋白粉,进行固态发酵。固态发酵结束后,烘干,再与复合预混料、复合酶、石粉等原料混合,制成发酵饲料添加剂产品。
所述的枯草芽孢杆菌的种子发酵培养基为:葡萄糖 30 g/L,酵母膏 10 g/L,蛋白胨 10 g/L,CaSO4 2 g/L,MgCl2 2 g/L,pH 7.0。
所述的嗜酸乳杆菌的种子发酵培养基为:牛肉膏5 g/L,蛋白胨10 g/L,葡萄糖20g/L,NaCl 1.0 g/L,pH 6.5。
所述的丁酸梭菌的种子发酵培养基为:胰蛋白胨10 g/L,蛋白胨5 g/L,酵母膏3g/L,葡萄糖10 g/L, 淀粉10 g/L ,NaCl 5 g/L,MgSO4 2 g/L,L-半胱氨酸盐酸盐0.5 g/L,pH 7.0。
所述的酿酒酵母和布拉氏酵母的种子发酵培养基为:葡萄糖30 g/L,酵母膏8.5g/L,(NH4)2SO4 0.5 g/L,MgSO4 0.1 g/L,CaCl2 0.06 g/L,pH 5.5。
所述的混合菌种液体发酵,其培养基组成为葡萄糖 50 g/L,淀粉10 g/L,酵母膏15 g/L,蛋白胨 10 g/L,NaCl 5 g/L,MgSO4 2 g/L,CaCl2 0.1 g/L,pH 6.2。
所述的种子发酵过程,枯草芽孢杆菌和酿酒酵母发酵时,按好氧发酵进行,发酵时间12 h;嗜酸乳杆菌和丁酸梭菌发酵时,按厌氧发酵进行,发酵时间20 h。
种子液接种发酵液时的接种量,枯草芽孢杆菌、酿酒酵母和布拉氏酵母按1%接种,嗜酸乳杆菌和丁酸梭菌按2%接种。
所述的二级发酵过程,按照兼性发酵进行,发酵时间24 - 30 h,活菌数达到2.5×109 CFU/mL以上。活菌数检测,按照平板活菌计数法进行,细菌计数用LB固体培养基,酵母用YPD固体培养基。
所述的LB固体培养基配方为:胰蛋白胨10 g/L,酵母提取物5 g/L,氯化钠10 g/L,琼脂18 g/L,pH 7.0。YPD固体培养基配方为:蛋白胨20 g/L,酵母膏10 g/L,葡萄糖20 g/L,琼脂18 g/L,pH 5.0。
所述的复合预混料,包括维生素和氨基酸,具体是由维生素A 0.4%、维生素D 3%、维生素E 0.4%、维生素K 3.5%、维生素B12 0.01%、维生素B6 0.05%、核黄素0.1%、泛酸钙0.3%、烟酸0.6%、生物素0.01%、叶酸0.3%、L-赖氨酸盐酸盐10%、DL-蛋氨酸1%、L-苏氨酸0.8%、硫酸亚铁5%、硫酸铜0.4%、硫酸锰4%、硫酸锌3.5%、碘化钾1.5%、氯化钴0.8%、喷浆皮34.33%、石粉30%等重量比的原料组成。
所述的复合酶,具体是由植酸酶、葡聚糖酶、木聚糖酶、纤维素酶等各占1/4重量比的原料组成。
所述的一种饲料添加剂,其产品检测项目如下:
(1)粗蛋白(%)≧20,GB/T 6432-1994饲料粗蛋白测定方法;
(2)粗纤维(%)≤8,GB/T 6434-2006饲料粗纤维的测定过滤法;
(3)水分(%)≤15,GB/T 6435 饲料水分含量的测定;
(4)粗灰分(%)≤10,GB/T 6438-2007饲料中粗灰分的测定;
(5)活菌数(×108 CFU/g)≧1.0,按照平板活菌计数法进行,细菌计数用LB固体培养基,酵母用YPD固体培养基;
(6)霉菌总数(×104 CFU/g)≤2.0,GB/T 13092 饲料中霉菌总数的测定;
(7)黄曲霉毒素B1≤50ppb,GB/T 17480 饲料中黄曲霉毒素B1的测定;
(8)总砷≤2.0 mg/kg,GB/T 13079 饲料中总砷的测定;
(9)镉≤0.5 mg/kg,GB/T 13082 饲料中镉的测定方法;
(10)铅≤5.0 mg/kg,GB/T 13080 饲料中铅的测定;
(11)汞≤0.1 mg/kg,GB/T 13081 饲料中汞的测定;
(12)沙门氏菌不得检出,GB/T 13091 饲料中沙门氏菌的检测方法。
所述的一种发酵饲料添加剂,色泽黄亮,有浓郁的发酵醇香味道,各项检测结果合格,数据如下所示:
(1)粗蛋白22.5%;
(2)粗纤维6%;
(3)水分12%;
(4)粗灰分8.5%;
(5)活菌数8.2×108 CFU/g;
(6)霉菌总数0.3×104 CFU/g;
(7)黄曲霉毒素B1 14.31ppb;
(8)总砷0.4 mg/kg;
(9)镉0.1 mg/kg;
(10)铅2.2 mg/kg;
(11)汞0.06 mg/kg;
(12)沙门氏菌未检出。
所述的一种发酵饲料添加剂,按1‰添加量,预先加入蛋鸡基础日粮中,搅拌均匀后饲喂。
本发明创造的有益效果在于:
本发明采用益生菌枯草芽孢杆菌、嗜酸乳杆菌、丁酸梭菌、酿酒酵母和布拉氏酵母等多菌种混合发酵玉米淀粉糖副产品,将植物性原料中的大分子物质分解或转化为微生物菌体蛋白、生物活性小肽类氨基酸、微生物活性益生菌、有机酸和B族维生素等促生长因子、复合酶制剂等为一体的生物发酵饲料,使其更易被养殖动物采食、消化、吸收,营养成分更高且无毒害作用的饲料原料。通过发酵能有效地降低原料中的粗纤维。利用玉米淀粉产业的副产品生产发酵饲料添加剂,这是对饲料原料的补充,而且比豆粕、鱼粉型饲料价格便宜,养殖户易于接受,同时也提高了玉米淀粉糖副产品的附加值,提高了企业的经济效益。饲料经过发酵处理后,营养水平及消化利用率得到显著提高,大大减少了抗生素等药物类添加剂的使用量,改善了动物健康水平,进而不同程度地提高动物的生产性能。另外,发酵饲料使畜禽处于有益微生物的控制之下,减少畜禽发病,提高肉蛋奶品质,达到降低成本、提高养殖效益的目的,可为养殖业的健康、高效、可持续发展提供坚实保障。

实施例1:
微生物混合发酵液10%、玉米皮10%、玉米浆40%、玉米淀粉糖糖渣15%、玉米蛋白粉15%、水10%等重量比的原料组成。把上述原料搅拌均匀,装入固体发酵容器或者密封袋子中,32- 37℃发酵7 - 10天,烘干。把复合预混料和复合酶加入上述烘干后的固态发酵物中,各物料占重量比为:复合预混料6%、复合酶5%、石粉7%、固态发酵物82%,充分搅拌混合均匀,即为发酵饲料添加剂。
实施例2:

实施例1-5所述的复合预混料,是由维生素A 0.4%、维生素D 3%、维生素E 0.4%、维生素K 3.5%、维生素B12 0.01%、维生素B6 0.05%、核黄素0.1%、泛酸钙0.3%、烟酸0.6%、生物素0.01%、叶酸0.3%、L-赖氨酸盐酸盐10%、DL-蛋氨酸1%、L-苏氨酸0.8%、硫酸亚铁5%、硫酸铜0.4%、硫酸锰4%、硫酸锌3.5%、碘化钾1.5%、氯化钴0.8%、喷浆皮34.33%、石粉30%等重量比的原料组成。
实施例1-5所述的复合酶,具体是由植酸酶、葡聚糖酶、木聚糖酶、纤维素酶等各占1/4重量比的原料组成。
一种发酵饲料添加剂,其生产过程如下:
益生菌枯草芽孢杆菌、嗜酸乳杆菌、丁酸梭菌、酿酒酵母和布拉氏酵母进行纯种发酵制备种子液,各种子液按照一定的接种量转接发酵培养基进行液体混合菌种发酵,发酵液接入玉米皮、玉米浆、玉米淀粉糖糖渣和玉米蛋白粉等原料中,混合均匀,进行固态发酵。固态发酵5 - 7天后烘干,再按一定重量比加入复合预混料、复合酶和石粉,搅拌均匀,即为发酵饲料添加剂成品。
选用出生日龄相近、70%产蛋的海兰褐壳蛋鸡240只,随机分为6组,1-5组为试验组,饲喂基础日粮中分别加入实施例1-5发酵饲料添加剂,第6组为对照组只喂基础日粮。其它条件按照养鸡场管理进行。
试验期28天,结束时测蛋鸡产蛋率、破蛋率、蛋均重等数据。实施例1-5组蛋鸡产蛋率分别为75%、76%、78%、74%、76%,而对照组蛋鸡产蛋率为72%,其中实施例3组产蛋率最高,较对照组提高8.3%。实施例1-5组与对照组相比,破蛋率降低81-98%,其中第2组破蛋率降低98%,优于其它4个试验组。5个试验组蛋均重比对照组都有所增加,其中第3组增加最多达到1.2%。
本发明发酵饲料添加剂,明显提高了蛋鸡的生产性能,提高蛋鸡产蛋率和蛋均重,降低了破蛋率和料肉比,并且也降低了饲喂蛋鸡的发病率,提高了鸡对疾病的抵抗能力。
Claims (8)
1.一种发酵饲料添加剂,其特征在于,它由益生菌发酵液、玉米皮、玉米浆、玉米淀粉糖糖渣、玉米蛋白粉、石粉、复合预混料、复合酶等原料组成,按1‰添加量,预先加入蛋鸡基础日粮中,搅拌均匀后饲喂。
2.根据权利要求1所述的一种发酵饲料添加剂,其特征在于,所述的益生菌发酵液由益生菌枯草芽孢杆菌、嗜酸乳杆菌、丁酸梭菌、酿酒酵母和布拉氏酵母,混合发酵而成。
3.根据权利要求2所述的发酵液,其特征在于,种子液由枯草芽孢杆菌、嗜酸乳杆菌、丁酸梭菌、酿酒酵母和布拉氏酵母,分别纯种发酵而成,所使用的种子培养基如下:
枯草芽孢杆菌的种子培养基为:葡萄糖 30 g/L,酵母膏 10 g/L,蛋白胨 10 g/L,CaSO42 g/L,MgCl2 2 g/L,pH 7.0;
嗜酸乳杆菌的种子培养基为:牛肉膏5 g/L,蛋白胨10 g/L,葡萄糖20 g/L,NaCl 1.0g/L,pH 6.5;
丁酸梭菌的种子培养基为:胰蛋白胨10 g/L,蛋白胨5 g/L,酵母膏3 g/L,葡萄糖10 g/L, 淀粉10 g/L ,NaCl 5g/L,MgSO4 2g/L,L-半胱氨酸盐酸盐0.5g/L,pH 7.0;
酿酒酵母和布拉氏酵母的种子培养基为:葡萄糖30 g/L,酵母膏8.5 g/L,(NH4)2SO4 0.5g/L,MgSO4 0.1 g/L,CaCl2 0.06 g/L,pH 5.5。
4.根据权利要求3所述的种子液,其特征在于,种子分两级发酵,由斜面转接摇瓶进行一级种子发酵,再由摇瓶种子转接种子罐进行二级种子发酵;枯草芽孢杆菌于32 - 35℃、180rpm条件下发酵12h;酿酒酵母和布拉氏酵母于28 - 30℃、150rpm条件下发酵12h;嗜酸乳杆菌和丁酸梭菌与于35 - 37℃、厌氧条件下发酵20h;一级种子液转接二级种子培养基时,枯草芽孢杆菌、嗜酸乳杆菌、丁酸梭菌、酿酒酵母和布拉氏酵母等5种一级种子液,分别按2%接种量接入二级种子发酵培养基中;二级种子液的发酵条件为:枯草芽孢杆菌于32 -5℃、1:1 - 1.2的通气比,发酵12 h;酿酒酵母和布拉氏酵母,均于28 - 30℃、1:0.8 - 1.0的通气比,发酵12 h;嗜酸乳杆菌和丁酸梭菌于35 - 37℃,通入氮气,发酵20 h;二级种子液接种发酵液时的接种量,枯草芽孢杆菌、酿酒酵母和布拉氏酵母按1%接种,嗜酸乳杆菌和丁酸梭菌按2%接种。
5.根据权利要求1所述的益生菌发酵液,其特征在于,发酵培养基组成为葡萄糖 50 g/L,淀粉10 g/L,酵母膏 15 g/L,蛋白胨 10 g/L,NaCl 5g/L,MgSO4 2g/L,CaCl2 0.1 g/L,pH6.2;发酵过程按照兼性发酵进行,发酵时间24 - 30 h,活菌数达到2.5×109 CFU/mL以上。
6.根据权利要求1所述的复合预混料,其特征在于,所述的复合预混料包括维生素和氨基酸,是由维生素A 0.4%、维生素D 3%、维生素E 0.4%、维生素K 3.5%、维生素B12 0.01%、维生素B6 0.05%、核黄素0.1%、泛酸钙0.3%、烟酸0.6%、生物素0.01%、叶酸0.3%、L-赖氨酸盐酸盐10%、DL-蛋氨酸1%、L-苏氨酸0.8%、硫酸亚铁5%、硫酸铜0.4%、硫酸锰4%、硫酸锌3.5%、碘化钾1.5%、氯化钴0.8%、喷浆皮34.33%、石粉30%等重量比的原料组成。
7.根据权利要求1所述的复合酶,其特征在于,所述的复合复合酶是由植酸酶、葡聚糖酶、木聚糖酶、纤维素酶等各占1/4重量比的原料组成。
8.根据权利要求1所述的一种发酵饲料添加剂的制备方法,其特征在于:
(1)发酵结束后,固体发酵按照微生物混合发酵液10 - 20%、玉米皮10 - 20%、玉米浆20 – 40%、玉米淀粉糖糖渣10 - 20%、玉米蛋白粉10 - 20%、水10%等重量比的原料组成;把上述原料搅拌均匀,装入固体发酵容器或者密封袋子中,32 - 37℃发酵7 - 10天,烘干;
(2)把复合预混料和复合酶加入上述烘干后的固态发酵物中,各物料占重量比为:复合预混料3 - 6%、复合酶3 - 5%、石粉7%、固态发酵物82 - 87%,充分搅拌混合均匀,即为发酵饲料添加剂。
