一种黄鳝弹状病毒CrERV及RT-PCR检测引物及应用
技术领域
本发明属于病毒分子检测技术领域,具体涉及一种黄鳝弹状病毒CrERV及RT-PCR检测引物及应用。
技术背景
弹状病毒科(Rhabdoviridae)病毒是一类具有广泛宿主的负链RNA病毒,其颗粒呈现子弹状或棒状。其中狂犬病病毒(Rabies virus)最为人熟知,能引起人或其他温血动物的狂犬病,病死率几乎为100%,引起了世界范围的广泛关注,造成社会对养殖宠物安全度的恐慌。因此,对弹状病毒进行深入研究具有重要意义。此外,弹状病毒还能引起家畜多种急性传染病(如牛流行热,ephemeral fever),以及鱼类和多种农作物的感染性疾病。国际病毒分类委员会(International Committee on Taxonomy of Viruses,ICTV)于2017年发表了最新的病毒分类第十次报告,将弹状病毒科(Rhabdoviridae)划分为18个属,其中能够感染鱼类的弹状病毒有Perhabdovirus,Sprivivirus和Novirhabdovirus三个属。目前以上三个属能够感染多种养殖鱼类,具有强烈的致病性,死亡率可以达到100%,给全球的海淡水养殖业造成重大的经济损失,尤其根据《进出口活水生动物检验检疫协议》,进出境的养殖鱼类,例如鲈鱼、鲤鱼、虹鳟、鲫鱼、牙鲆等养殖和种苗用鱼种,均要进行弹状病毒(IHNV、SVCV、HIRRV)疫病检测,口岸安全意义重大,影响进出口经济贸易。
目前对感染鱼类弹状病毒科的研究主要集中在Sprivivirus和Novirhabdovirus两个属,而Perhabdovirus属作为2017年弹状病毒科新命名的一个属,对其研究甚少。目前该属中包括鳜鱼弹状病毒,该病毒能够引起鳜鱼大量死亡。目前,黄鳝弹状病毒(Chineserice-field eel rhabdovirus,CrERV)全长序列尚无公开报道,更也无有效的检测方法。
本申请首次从染病的黄鳝中分离得到黄鳝弹状病毒(Chinese rice-field eelrhabdovirus,CrERV),针对鳜鱼弹状病毒和黄鳝弹状病毒在P基因结构的区别设计特异性引物,建立RT-PCR检测方法不仅用于检测和监测养殖黄鳝的健康状况,而且通过序列比对能够区别黄鳝弹状病毒和鳜鱼弹状病毒。为黄鳝和鳜鱼养殖及早病原检测和监测提供保障。
发明内容
本发明的目的在于提供了一种黄鳝弹状病毒CrERV,所述的病毒分离自黄鳝病变细胞,该病毒已于2018年4月24日送至中国典型培养物保藏中心进行保藏,分类命名:黄鳝弹状病毒(Chinese rice-field eel rhabdovirus)CrERV,保藏编号:CCTCC NO:V201819,地址:中国武汉武汉大学。
本发明的另一个目的在于提供了黄鳝弹状病毒CrERV的基因,所述基因为SEQ IDNO.1所示。
本发明还有一个目的在于提供一种黄鳝弹状病毒CrERVRT-PCR检测引物,所述的引物为F1:5'-GAGATGAAGGAAGGACTGAGGAGAA-3',R1:5'-CCCGACACCACTAAAGTATAAAA-3';F2:5'–GAGGAGCATTAGTTACTTAAGGGTC-3',R2:5'-TCACAATTTGAATCGTCATGACATA-3'。
本发明还有一个目的在于提供了黄鳝弹状病毒CrERV的应用,所述的应用包括利用该病毒制备成黄鳝弹状病毒病疫苗,或是制备成黄鳝弹状病毒的实验室对照品。
本发明的最后一个目的在于提供一种黄鳝弹状病毒CrERVRT-PCR检测引物的应用,包括利用所述的引物制备成黄鳝弹状病毒检测试剂盒。
为了达到上述目的,本发明采取以下技术措施:
一种黄鳝弹状病毒CrERV,所述的病毒分离自黄鳝病变细胞,该病毒已于2018年4月24日送至中国典型培养物保藏中心进行保藏,分类命名:黄鳝弹状病毒(Chinese rice-field eel rhabdovirus)CrERV,保藏编号:CCTCC NO:V201819,地址:中国武汉武汉大学。
将黄鳝弹状病毒CrERV接种到鲫脑细胞系(GibelCarpBrain,GiCB)单层,25℃培养7天,产生细胞病变效应(CPE),可见GiCB细胞收缩变圆、折光度增加,部分细胞开始脱落,细胞单层开始破裂。
培养基:M199,10%胎牛血清,pH 7.0~7.2。
黄鳝弹状病毒CrERV的形态特征:病毒呈子弹状或梭形,病毒具有双层脂质囊膜,与典型的弹状病毒形态相符。
黄鳝弹状病毒CrERV的应用,所述的应用包括利用该病毒制备成黄鳝弹状病毒病疫苗,或是制备成黄鳝弹状病毒的实验室对照品。
本发明的保护内容还包括基于黄鳝弹状病毒CrERV的全序列(SEQ ID NO.1所示)与其他病毒的全序列的差异设计的引物,所述的引物可用于检测黄鳝弹状病毒CrERV。

与现有技术相比,本发明具有以下优点:
1.本发明首次自黄鳝中分离并克隆了弹状病毒的全长序列,对于黄鳝弹状病毒的研究具备开拓性的意义。
2.本发明的方法快速简便:本发明所建的RT-PCR方法只需数小时,相比较传统病毒分离方法数天时间,极大的提高了检测效率。
3.本发明的方法特异性强:两对引物对靶序列的特异序列区的识别,保证了PCR方法扩增的高度特异性。

本发明所述技术方案,如未特别说明,均为本领域的常规方案,所用试剂或原料,如未特别说明,均购自商业渠道或是已公开。
实施例1:
一种黄鳝弹状病毒CrERV,其分离过程如下:
1)将携带病毒的黄鳝脾、肾等组织取出并碾磨后,培养基稀释100倍,2000rpm离心后取上清过滤,接种到鲫脑细胞系(GibelCarpBrain,GiCB)单层,25℃培养7天,产生细胞病变效应(CPE),可见GiCB细胞收缩变圆、折光度增加,部分细胞开始脱落,细胞单层开始破裂。
培养基:M199,10%胎牛血清,pH 7.0~7.2
2)收集病变细胞,利用透射电镜进行形态学鉴定。
3)收集病毒,对其进行全基因组测序,序列为SEQ ID NO.1所示。
黄鳝弹状病毒的形态特征:病毒子弹状或梭形,病毒具有双层脂质囊膜,与典型的弹状病毒形态相符,该病毒已于2018年4月24日送至中国典型培养物保藏中心进行保藏,分类命名:黄鳝弹状病毒(Chinese rice-field eel rhabdovirus)CrERV,保藏编号:CCTCCNO:V201819,地址:中国武汉武汉大学。
有研究表明鳜鱼弹状病毒可以感染黄鳝,但是通过临床症状特征对比结果显示,感染鳜鱼弹状病毒的黄鳝主要出现出血现象,但是本申请中从患病黄鳝中分离的弹状病毒临床症状主要为头部肿大,两者形成鲜明对比(图1),全序列分析结果显示,鳜鱼弹状病毒与本申请中分离鉴定的黄鳝弹状病毒同源性最高为92%,但是通过基因结构分析发现本申请中分离鉴定的黄鳝弹状病毒磷酸化蛋白基因(phosphoprotein gene,P)由一个大的开放阅读框(ORF)组成(864bp),与鳜鱼弹状病毒P基因包含的两个ORF形成明显区别(大的ORF大小为849bp,小的ORF大小为219bp),而且两株弹状病毒的开放阅读框的大小也不同。两株病毒在宿主感染的倾向性不同,而且感染后表现的临床症状也有明显区别,因此,两株病毒无法归为一株病毒。其中,本申请中涉及到的黄鳝弹状病毒的分类地位属于Perhabdovirus属。
实施例2:
基于黄鳝弹状病毒CrERV的全序列(SEQ ID NO.1所示)与其他病毒的全序列的差异设计的引物:
一种黄鳝弹状病毒CrERV RT-PCR检测引物,包括F1:5'-GAGATGAAGGAAGGACTGAGGAGAA-3',R1:5'-CCCGACACCACTAAAGTATAAAA-3';F2:5'–GAGGAGCATTAGTTACTTAAGGGTC-3',R2:5'-TCACAATTTGAATCGTCATGACATA-3'。
实施例3:
利用黄鳝弹状病毒CrERV RT-PCR检测引物对黄鳝弹状病毒的检测,方法包括:
1.黄鳝组织或感染细胞总RNA的提取及cDNA的合成:采用提取待测样品的总RNA。配置逆转录反应体系:3μL总RNA,Oligo(dT)18primer 1μL,灭菌双蒸水8μL,混匀65℃5min后立刻冰浴;将5×Reaction Buffer 4μl,RNase抑制剂1μl,5mM dNTPs 2μL,M-MLV逆转录酶1μL加入冰浴的反应液中,混匀后,42℃1h,95℃5min,将获得cDNA模板-20℃保存备用。
2.聚合酶链式反应扩增:
采用25μl反应体系:PCR反应管中,加入10μM的引物F1、R1各加1μL,再加入2.5mMdNTPs 2μL,25mM MgCl21.5μL,5U Taq聚合酶0.5μL,模板cDNA 1μl,10×ThermoPolReaction Buffer 2.5μl,用灭菌双蒸水将体系均补齐至25μL,进行PCR反应。
以该反应产物为模板,取1μl,10μM的引物F2、R2各加1μL,再加入2.5mM dNTPs2μL,25mM MgCl21.5μL,5U Taq聚合酶0.5μL,10×ThermoPol Reaction Buffer 2.5μl,用灭菌双蒸水将体系均补齐至25μL,进行PCR反应。
PCR反应条件,95℃预变性5min;95℃30s、48℃30s、72℃30s,30个循环;72℃延伸10min,4℃保存。

聚合酶链式反应扩增产物分析:
取聚合酶链式反应扩增的最终反应产物4μL,加入至含有0.5μg/mL溴化乙锭(EB)染料的2%的琼脂糖凝胶中,于100V电压下电泳30min,在凝胶成相系统上成像观察扩增产物的图谱。若巢式RT-PCR能扩增出972bp(SEQ ID NO.2所示)特征性条带,则该样品为阳性;如无任何条带,则为阴性。
实施例4:
黄鳝弹状病毒RT-PCR检测引物特异性试验
利用实施例3所述的方法,对表1中的待检病毒样品进行检测,同时设置阴性对照组(双蒸水)和阳性对照组(CrERV),待检病毒样品具体为:鲤春病毒血症病毒(SVCV),草鱼呼肠孤病毒873(GCRV-873),鳜鱼弹状病毒(SCRV),传染性造血器官坏死病毒(IHNV)。

结果如表1所示,仅阳性对照组(CrERV)和鳜鱼弹状病毒(SCRV)可检测到972bp特征性条带,其余组均无特征条带,说明黄鳝弹状病毒CrERVRT-PCR检测引物具备很强的特异性。通过DNAStar序列分析结果,可将鳜鱼弹状病毒和黄鳝弹状病毒区分开来,其中黄鳝弹状病毒在扩增的产物序列中能够分析出一个大的ORF(864bp);而在扩增鳜鱼弹状病毒的产物中能够获得两个小的ORF,其中较大的ORF为849bp,较小的ORF为219bp,为了使比对序列过程中操作简单,在使用DNAStar软件比对中仅需区分两株弹状病毒P基因中ORF大小即可,如PCR产物测序结果中出现864bp大小的ORF为黄鳝弹状病毒,当阳性结果序列中出现849bp则划分为鳜鱼弹状病毒。
实施例5:
黄鳝弹状病毒RT-PCR检测引物的灵敏度
将浓度为1500ng/μL的CrERVcDNA模板10倍梯度稀释,利用实施例3的方法,检测利用本发明提供的引物PCR的检测限,结果显示105倍稀释还可检测到目的条带,也就是最低可检测15.00pg/μL的cDNA模板。
实施例6:
黄鳝弹状病毒RT-PCR检测引物在制备黄鳝弹状病毒检测试剂盒中的应用:
1.黄鳝组织总RNA的提取及cDNA的合成:采用提取感染黄鳝弹状病毒的黄鳝的脾肾等内脏组织的总RNA。配置逆转录反应体系:3μL总RNA,Oligo(dT)18primer 1μL,灭菌双蒸水8μL,混匀65℃5min后立刻冰浴;将5×Reaction Buffer 4μl,RNase抑制剂1μl,5mM dNTPs 2μL,M-MLV逆转录酶1μL加入冰浴的反应液中,混匀后,42℃1h,95℃5min,将获得cDNA模板-20℃保存备用。
2.聚合酶链式反应扩增:
采用25μl反应体系:PCR反应管中,加入10μM的引物F1、R1各加1μL,再加入2.5mMdNTPs 2μL,25mM MgCl21.5μL,5U Taq聚合酶0.5μL,模板cDNA 1μl,10×ThermoPolReaction Buffer 2.5μl,用灭菌双蒸水将体系均补齐至25μL,进行PCR反应。
以该反应产物为模板,取1μl,10μM的引物F2、R2各加1μL,再加入2.5mM dNTPs 2μL,25mM MgCl21.5μL,5U Taq聚合酶0.5μL,10×ThermoPol Reaction Buffer2.5μl,用灭菌双蒸水将体系均补齐至25μL,进行PCR反应。PCR反应条件,95℃预变性5min;95℃30s、48℃30s、72℃30s,30个循环;72℃延伸10min,4℃保存。

3.聚合酶链式反应扩增产物分析:
取聚合酶链式反应扩增的最终反应产物4μL,加入至含有0.5μg/mL溴化乙锭(EB)染料的2%的琼脂糖凝胶中,于100V电压下电泳30min,在凝胶成相系统上成像观察扩增产物的图谱。电泳图片显示972bp特征性条带,则结果为阳性;如无任何条带,则结果为阴性。
结果显示,阳性对照(黄鳝弹状病毒CrERV)显示972bp特征性条带,阴性(水)对照无明显条带,1-5号感染了黄鳝弹状病毒CrERV的黄鳝组织样品显示972bp特征性条带,说明样品1-5为黄鳝弹状病毒阳性,与实际情况相符。
实施例7:
黄鳝弹状病毒CrERV在制备黄鳝弹状病毒的实验室对照品中的应用:
即将本发明提供的黄鳝弹状病毒CrERV作为对照品,将待确认的其他黄鳝弹状病毒与本发明的病毒进行生理生化上的比较,并且将待测样品跟SEQ ID NO.1所示序列进行比较。

1.一种分离自黄鳝的弹状病毒,所述的弹状病毒为黄鳝弹状病毒(Chinese rice-field eel rhabdovirus)CrERV,保藏编号为:CCTCC NO:V201819。
2.权利要求1所述的弹状病毒的基因,所述的基因为SEQ ID NO.1所示的多核苷酸。
3.权利要求1所述弹状病毒的检测引物,所述的引物为:F1:5'-GAGATGAAGGAAGGACTGAGGAGAA-3',R1:5'-CCCGACACCACTAAAGTATAAAA-3';F2:5'–GAGGAGCATTAGTTACTTAAGGGTC-3',R2:5'-TCACAATTTGAATCGTCATGACATA-3'。
4.权利要求1所述的黄鳝弹状病毒CrERV在制备黄鳝弹状病毒病疫苗中的应用。
5.权利要求1所述的黄鳝弹状病毒CrERV或权利要求2所述的多核苷酸在制备黄鳝弹状病毒对照品中的应用。
6.权利要求2所述的多核苷酸或权利要求3所述的引物在制备黄鳝弹状病毒检测试剂盒中的应用。
