本发明属于植物的深加工技术领域,具体涉及一种具有酪氨酸酶激活作用的铁皮石斛花提取物及其制备方法与应用。
背景技术
酪氨酸酶(EC1.14.18.1,tyrosinase)又称多酚氧化酶,是一种含铜的金属酶,广泛存在于微生物、动植物和人体中。酪氨酸酶(tyrosinase)具有多种特征性催化活性,既可以作为酪氨酸羟化酶、多巴氧化酶,又可以作为5,6-二羟基吲哚氧化酶,在黑色素生成过程中起着至关重要的作用。酪氨酸酶的异常表达可直接或间接导致人类疾病,如恶性黑色素瘤、皮肤斑点和白癜风。另外,目前研究发现黑色素异常是白发的产生原因,因此激活酪氨酸酶的活性可能可促进毛发黑素的生成,从而达到乌发的目的。总之,酪氨酸酶激活物质在医药、化妆品等领域具有重要的应用前景。
虽然对于抑制酪氨酸酶的物质研究和相关发明较多,但对激活酪氨酸酶的物质研究甚少。前期一些研究已从药材中筛选酪氨酸酶激活物质,如补骨脂、葛根、丹参等(参见文献:涂彩霞等,47种中药对酪氨酸酶活性的影响及酶动力学的研究.中国麻风皮肤病杂志,2006(06):第456-458页;丁翔云,对酪氨酸酶活性有激活作用的中药的筛选,2012,湖南中医药大学;王锡元,酪氨酸酶抑制剂和激活剂的研究,2007,江南大学)。公告号为CN103585216A的专利从中药白首乌中提取、分离得到的一种C21甾苷(C21甾苷A)对蘑菇酪氨酸酶二酚酶活力有很强的激活作用;公告号为CN106420678A的专利,发明了具有酪氨酸酶激活作用的含有酚羟基的芳基化合物。
铁皮石斛(Dendrobium catenatum)是兰科(Orchidaceae)石斛属(Dendrobium)的多年生草本植物,是我国传统名贵中药材。铁皮石斛每年6月开花,且持续时间长,开花量较大,近年来随着铁皮石斛种植面积的增加,铁皮石斛花资源量巨大。在民间该花作为观赏或食用,鉴于地方具有食用习惯,2018年6月21日国家卫计委将铁皮石斛花列入新食品原料的终止审查名单,建议按照食品管理。铁皮石斛花含有黄酮类、氨基酸、多糖、挥发油等成分,并具有滋阴护肝、缓解精神紧张之功效。随着作为食品管理,可以预期铁皮石斛花的开发与加工将得到极大地发展。
目前已有一些专利文章报道铁皮石斛花的利用,但主要作为茶或饮料。如,公告号为CN106260316A、CN107897426A、CN107751475A、CN107751509A、CN107982434A的专利都发明了经过采摘、干燥等工艺的铁皮石斛花加工方法;公告号CN108450179A的专利发明了富硒铁皮石斛花的制备方法;公告号CN108041211A的专利将铁皮石斛花与橘皮、铁皮石斛叶、红茶、显齿蛇葡萄、罗汉果、甘草等提取物混合,发明了一种铁皮石斛花茶产品;公告号为CN108029828A的专利将石斛花与桑葚、苹果、哈密瓜、猕猴桃、火龙果、椰子等水果混合,发明了一种含有石斛花的冲泡型果茶;公告号为CN108522734A的专利,将铁皮石斛花浸提液加入桂花浸提液、绿茶浸提液等发明了种铁皮石斛花茶饮料。
已有报道相比,无论是从已有对于酪氨酸酶激活物质的研究与发明来看,还是从铁皮石斛花的研究与应用来看,均未开展铁皮石斛花促进酪氨酸酶活性的发明创造。
发明内容
本发明的目的是为了解决现有技术的不足,提供一种具有酪氨酸酶激活作用的铁皮石斛花提取物及其制备方法与应用。本发明制备方法提取的两种铁皮石斛花提取物,二者都具有很好的酪氨酸酶激活活性,在医药、化妆品和食品加工领域都具有广泛的应用前景。本发明以铁皮石斛花为原料进行精深加工与应用,促进了该资源的高效利用。
为实现上述目的,本发明采用的技术方案如下:
一种具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,包括如下步骤:
干燥的铁皮石斛花,经过水或乙醇提取,过滤,滤液减压浓缩,后冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物。
进一步,优选的是,所述的具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法如下:
选择称取无氧化、霉烂的干燥石斛花,加入质量是石斛花质量20-30倍的蒸馏水沸煮20-30分钟,过滤;重复上述提取步骤1次或多次,合并滤液,40-75℃减压浓缩至粘稠状,冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物,该提取物为红褐色,为水提取物。
进一步,优选的是,所述的具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法如下:
选择称取无氧化、霉烂的干燥石斛花,加入质量是石斛花质量20-30倍体积浓度为20-80%的乙醇水溶液,室温浸提3-5天,过滤,30-60℃减压浓缩至原体积的1/9-1/11,冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物,该提取物为红褐色,为醇提取物。
进一步,优选的是,冷冻干燥温度为-60℃。
本发明同时保护上述具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法制得的具有酪氨酸酶激活作用的铁皮石斛花提取物。
本发明再提供上述具有酪氨酸酶激活作用的铁皮石斛花提取物作为制备具有激活酪氨酸酶活性的化妆品。
本发明所述的化妆品是指以涂抹、喷洒或者其他类似方法,散布于人体表面的任何部位,如皮肤、毛发、指趾甲、唇齿等,以达到清洁、保养、美容、修饰和改变外观,或者修正人体气味,保持良好状态为目的的化学工业品或精细化工产品。包括但不限于此具有美黑或乌发功能的精华素、洗发水及化妆品等。
本发明还提供上述具有酪氨酸酶激活作用的铁皮石斛花提取物作为制备具有激活酪氨酸酶活性的药物。
本发明所述的药物以具有酪氨酸酶激活作用的铁皮石斛花提取物为活性成分,可制备用于治疗黑色素缺失性疾病如白癜风、白发以及其它需要激活酪氨酸酶活性的疾病的药物。所述的药物还可以从本发明提取物中进一步分离纯化,使用其分离纯化的具体成分。
本发明另外提供上述具有酪氨酸酶激活作用的铁皮石斛花提取物作为制备具有激活酪氨酸酶活性的保健品。
本发明所述的保健品包括但不限于以下几种形式:茶、压片糖果、饮料。本发明所述的保健品可以为具有美黑、乌发功能的保健品。

本发明首次从铁皮石斛花中提取得到具有酪氨酸酶激活作用的铁皮石斛花提取物,该制备方法简单,可靠,所得到的铁皮石斛花提取物具有很好的酪氨酸酶激活活性,在医药、化妆品和食品加工领域都具有广泛的应用前景。

实施例1
一种具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,步骤如下:
选择称取无氧化、霉烂的干燥石斛花,加入质量是石斛花质量20倍的蒸馏水沸煮20分钟,过滤;重复上述提取步骤1次,合并滤液,40℃减压浓缩至粘稠状,冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物。
实施例2
一种具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,步骤如下:
选择称取无氧化、霉烂的干燥石斛花,加入质量是石斛花质量30倍的蒸馏水沸煮30分钟,过滤;重复上述提取步骤3次,合并滤液,75℃减压浓缩至粘稠状,-60℃冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物。
实施例3
一种具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,步骤如下:
选择称取无氧化、霉烂的干燥石斛花,加入质量是石斛花质量25倍的蒸馏水沸煮28分钟,过滤;滤液于50℃下减压浓缩至粘稠状,-60℃冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物。
实施例4
一种具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,步骤如下:
选择称取无氧化、霉烂的干燥石斛花,加入质量是石斛花质量20倍体积浓度为20%的乙醇水溶液,室温浸提3天,过滤,30℃减压浓缩至原体积的1/9,冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物。
实施例5
一种具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,步骤如下:
选择称取无氧化、霉烂的干燥石斛花,加入质量是石斛花质量30倍体积浓度为80%的乙醇水溶液,室温浸提5天,过滤,60℃减压浓缩至原体积的1/11,-60℃冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物。
实施例6
一种具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,步骤如下:
选择称取无氧化、霉烂的干燥石斛花,加入质量是石斛花质量25倍体积浓度为60%的乙醇水溶液,室温浸提4天,过滤,45℃减压浓缩至原体积的1/10,-60℃冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物。
实施例7
一种具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,步骤如下:
选择称取无氧化、霉烂的干燥石斛花200g,加入4000mL体积浓度为50%的乙醇水溶液,室温浸提3天,过滤,40-45℃减压浓缩至原体积的1/10,-60℃冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物20g。
实施例8
一种具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,步骤如下:
选择称取无氧化、霉烂的干燥石斛花200g,加入2500mL蒸馏水沸煮28分钟,过滤;重复上述提取步骤1次,合并滤液,60-65℃减压浓缩至粘稠状,-60℃冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物25g。
性能检测
铁皮石斛花提取物酪氨酸酶活性测定:
用PBS缓冲液将本发明提取物配制为待测液。分别设置对照组(PBS缓冲液作为待测液)和不同添加量的本发明提取物溶液(质量浓度为1.5%、3.0%、4.5%和6.0%,以PBS缓冲液为溶剂进行配制)实验组和空白组(只采用PBS缓冲液作为待测液,且测活性的时候不加酶),按如下反应检测酪氨酸酶活性:250 微升待测液加入750微升0.09 mg/mL的酪氨酸溶液于37℃下保温1h;加入25 微升酪氨酸酶溶液(1 mg/mL),于37℃下反应30 min,于450 nm下测定其吸光度值。计算公式
酪氨酸酶抑制率(%)=
其中,A0为对照组吸光度值;A1为实验组吸光度值;A2为空白组吸光度值;当样品在所检测添加浓度下抑制率为负,且小于-50%时,则认为该样品具有较好的酪氨酸酶激活性。结果如图1所示,发现质量浓度为3.0%的铁皮石斛花醇提取物溶液以及质量浓度为4.5%和6.0%的水醇提取物溶液对酪氨酸酶均具有较高的激活作用。

Claims (8)
1.一种具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,其特征在于,包括如下步骤:
干燥的铁皮石斛花,经过水或乙醇提取,过滤,滤液减压浓缩,后冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物。
2.根据权利要求1所述的具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,其特征在于:
选择称取无氧化、霉烂的干燥石斛花,加入质量是石斛花质量20-30倍的蒸馏水沸煮20-30分钟,过滤;重复上述提取步骤1次或多次,合并滤液,40-75℃减压浓缩至粘稠状,冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物。
3.根据权利要求1所述的具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,其特征在于:选择称取无氧化、霉烂的干燥石斛花,加入质量是石斛花质量20-30倍体积浓度为20-80%的乙醇水溶液,室温浸提3-5天,过滤,30-60℃减压浓缩至原体积的1/9-1/11,冷冻干燥得到具有酪氨酸酶激活作用的铁皮石斛花提取物。
4.根据权利要求2或3所述的具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法,其特征在于:冷冻干燥温度为-60℃。
5.权利要求1~3任意一项所述的具有酪氨酸酶激活作用的铁皮石斛花提取物的制备方法制得的具有酪氨酸酶激活作用的铁皮石斛花提取物。
6.权利要求5所述的具有酪氨酸酶激活作用的铁皮石斛花提取物作为制备具有激活酪氨酸酶活性的化妆品。
7.权利要求5所述的具有酪氨酸酶激活作用的铁皮石斛花提取物作为制备具有激活酪氨酸酶活性的药物。
8.权利要求5所述的具有酪氨酸酶激活作用的铁皮石斛花提取物作为制备具有激活酪氨酸酶活性的保健品。
