Description

Traditional Chinese medicine compound preparation for inhibiting blue ear virus and preparation method thereof
Technical Field
The invention belongs to the field of traditional Chinese medicine preparations, and particularly relates to a traditional Chinese medicine compound preparation for inhibiting a blue ear virus and a preparation method thereof.
Background
The porcine reproductive and respiratory syndrome is a second type infectious disease in China, has high contagious property and has the local epidemic characteristic. The porcine reproductive and respiratory syndrome virus only infects pigs, other animals do not infect the pigs, and pigs of various ages and breeds of days can be infected with the virus, wherein piglets and pregnant sows with the ages of days within 1 month are the most susceptible pig groups.
The porcine reproductive and respiratory syndrome is mainly characterized by dysgenesis, dyspnea, blue purple of ears and other infectious diseases. The pigs can be infected at different ages and varieties, the incidence rate is high, the incidence rate accounts for 50-80% of the whole pig group, the sick pigs die quickly, and the death rate is as high as 80%. Sepsis is mostly a change after death. Pregnant sows and piglets are most susceptible to infection, and late-stage abortion, stillbirth and mummy of pregnant sows occur.
Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) is a reproductive and respiratory syndrome virus, the primary target cells of PRRSV are the monocyte-macrophage system such as alveolar macrophages, alveolar type ii pneumocytes, peripheral monocytes, epithelial germ cells of the seminiferous tubules and macrophages in the stroma, macrophages in ovarian follicles. Of these, porcine alveolar macrophages are the most sensitive.
The sick sows are mainly characterized by depression, reduced or abolished appetite, fever, dyspnea of different degrees, and abortion, premature birth, stillbirth, mummy and weak piglets of the sows at the later stage of pregnancy (105-107 days). The sow abortion rate can reach 50-70%, the mortality rate can reach more than 35%, the mummy can reach 25%, part of newborn piglets show symptoms such as dyspnea, dyskinesia and paresis, and the mortality rate is obviously increased (40-80%) within 1 week after delivery. A few sows show postpartum agalactia, retained fetal membranes and increased vaginal secretions.
Piglets of 1 month of age show typical respiratory symptoms, dyspnea, sometimes abdominal breathing, loss or abolition of appetite, body temperature rise above 40 ℃, diarrhea. Coarse and disorganized hair, ataxia, progressive wasting, edema of the eyelids. The ear and body surface skin of a small part of piglets can be purple, the death rate of the piglets before weaning can reach 80-100%, the weight gain of the piglets after weaning is reduced, the daily weight gain can be reduced by 50-75%, and the death rate is increased (10-25%). The pig can resist the slow growth and is easy to have other secondary diseases.
Growing and finishing pigs show mild clinical symptoms with different procedures of respiratory symptoms, and few cases can show cough and deep purple color on the back, edge, abdomen and tail skin of ears. Infected pigs are susceptible to secondary infection and the corresponding symptoms.
The disease rate of breeding boars is low and mainly shows general clinical symptoms, but the semen quality of boars is reduced, the semen is malformed, and the semen can carry virus.
The traditional Chinese medicine is safe and effective in treatment, and has no residue and drug resistance. Therefore, if a traditional Chinese medicine compound preparation with good curative effect can be developed, the traditional Chinese medicine compound preparation has obvious economic and social significance for the pig breeding industry.

From the perspective of traditional Chinese veterinary medicine, porcine reproductive and respiratory syndrome is analyzed, and the 'deficiency-evil coordination' is a cause of morbidity, particularly the morbidity and mortality of suckling piglets are serious, namely the five internal organs in the traditional Chinese medicine are in deficiency, and the virus enters in the deficiency to cause exuberance and decline of pathogenic factors and disease resistance; the disease is frequent in summer, and it is usually caused by exogenous dampness-heat transforming into fire, resulting in failure of lung qi to disperse, abnormal ascending and descending, and imbalance of yin and yang. The porcine reproductive and respiratory syndrome belongs to warm diseases, and the treatment is preferably 'heat clearing and detoxifying', 'lung heat clearing and cough relieving', 'body resistance strengthening and evil eliminating', 'antibacterial and anti-inflammation'. Accordingly, the inventor of the invention determines the formula of the invention, and the compatibility mechanism is as follows: the astragalus membranaceus has the effects of tonifying qi, invigorating yang, expelling toxin, expelling pus, enhancing the immune function of the organism and improving the immune function; rhubarb, radix et rhizoma Rhei, purging pathogenic heat and promoting bowel movement, cooling blood and removing toxic substance, activating blood stasis and dredging channels; folium Isatidis, fructus forsythiae, and fructus Gardeniae have effects of cooling blood, removing toxic substances, relieving swelling, resolving hard mass, purging pathogenic fire, relieving restlessness, and inducing diuresis for treating stranguria; sweet wormwood herb, which is used for clearing deficiency heat, improving eyesight and relieving summer heat; honeysuckle flower, flos Lonicerae, heat-clearing and detoxicating, anti-inflammatory, tonifying deficiency and treating wind; radix bupleuri; the bupleurum can enhance the functions of natural killer cells and phagocytic function and improve the titer of virus specific antibodies; the houttuynia cordata has the effects of resisting bacteria and diminishing inflammation and improving the immunity; the mulberry leaves can disperse wind and heat, clear lung heat and moisten dryness, calm liver yang, clear liver and improve vision, cool blood and stop bleeding; the medicines are combined together, so that the traditional Chinese medicine can regulate yin and yang, treat both symptoms and root causes, balance yin and yang in physique and immunity, and achieve an ideal breeding state of 'having few diseases and being died'.
The preparation method comprises the following steps: weighing the raw materials according to a proportion, adding ethanol for extraction at room temperature, filtering, concentrating the filtrate under reduced pressure until the relative density at 60 ℃ is 1.1-1.3, then adding silicon dioxide, uniformly mixing, drying and crushing to obtain the traditional Chinese medicine compound preparation, wherein each 1000g of the traditional Chinese medicine compound preparation is equivalent to 2800-3000 g of crude drugs.
Preferably, the ethanol is 50v% ethanol.
Preferably, ethanol is extracted twice, filtered, and the two filtrates are combined; during the first extraction, the adding amount of ethanol is 5-8 times of the weight of the raw material medicine, and the extraction is carried out for 1.5-2 h; during the second extraction, the amount of ethanol added is 3-5 times of the weight of the raw materials, and the extraction lasts for 1-1.5 h.
