Abstract

本发明涉及一种止血粉的制备,特别是一种白芨多糖复合止血粉及其制备方法。白芨多糖复合止血粉,其特征在于该复合止血粉是由海藻酸钙多孔微球与白芨多糖粉按重量比1:1—3配置而成。制备白芨多糖复合止血粉的方法,其制备工艺为:海藻酸钙多孔微球的制备、复合止血粉的制备。本发明技术方案是利用海藻酸钙多孔微球所具有的较大的比表面积和孔洞结构这一特点,利用白芨多糖具有的收敛止血、促进血小板聚集的特性,将白芨多糖置于海藻酸钙多孔微球中,既发挥了白芨多糖收敛止血、促进血小板聚集的特性,又利用了海藻酸钙多孔微球作为载体,使本发明技术方案所述的复合止血粉具有吸水率高、止血效果好、使用方便、不受创面、伤口大小的限制等特点。

所属技术领域
本发明涉及一种止血粉的制备,特别是一种白芨多糖复合止血粉及其制备方法。
背景技术
白芨是一种传统的中药,属于兰科植物,具有收敛止血、清热利湿、消肿生肌之功效,尤其是对于内脏出血、皮肤烧烫伤和外伤出血具有显著性的效果。
经研究证实,真正具有止血功效的是白芨中的一种多糖,其性黏,具有收敛止血及生肌的作用,并能促进红细胞和血小板的聚集,明显缩短凝血时间和凝血酶原时间,降低血管的通透性,改善血管脆性,使血管收缩。
海藻酸盐是一种天然的高分子多糖,广泛存在于各类棕色海藻中,具有生物相容性好、可降解、止血、抑菌、促愈合等特性。而海藻酸钙敷料易发生离子交换,转变成水溶性的海藻酸钠,形成凝胶覆盖于伤口之上,为伤口的愈合提供一个湿润的环境促进伤口愈合。常见的具有多孔结构的止血剂主要通过物理吸附止血,起到分子筛的作用,止血剂与血液接触的瞬间会选择性吸收血液中的水分,促进血液中有形成分在颗粒周围高密度粘附聚集,导致血小板和凝血因子浓缩,从而凝血系统被激活,纤维蛋白沉积,可以达到快速凝血的目的。
发明内容
本发明的目的是提供一种止血效果好、使用方便的白芨多糖复合止血粉及其制备方法。
本发明的技术方案是:一种白芨多糖复合止血粉,其特征在于该复合止血粉是由海藻酸钙多孔微球与白芨多糖粉按重量比1:1—3配置而成。
上述海藻酸钙多孔微球的吸水率为230—290%。
制备上述权利要求所述白芨多糖复合止血粉的方法,其制备工艺为:
步骤一:海藻酸钙多孔微球的制备:称取0.25—1克的氯化钠溶于97—97.75克去离子水中,然后再加入2克海藻酸钠配成海藻酸钠混合溶液备用;
在100克液体石蜡中加入0.5克质量百分比浓度为3%的Span80,在温度为45℃状态下加入上述海藻酸钠混合溶液20g,并搅拌20—40分钟,然后在搅拌状态下加入质量百分比浓度为5%CaCl2溶液10克进行交联反应20—40分钟后停止搅拌,离心、洗涤、冷冻干燥,即得海藻酸钙多孔微球;
步骤二:复合止血粉的制备:将上述步骤获得的海藻酸钙多孔微球和白芨多糖按重量比为1:1—3加入去离子水中混合均匀后,冷冻干燥,得到复合止血粉。
上述步骤三中海藻酸钙多孔微球和白芨多糖在去离子水中混合均匀是采用漩涡混合器进行混合,混合时间为5min。
本发明技术方案是利用海藻酸钙多孔微球所具有的较大的比表面积和孔洞结构这一特点,利用白芨多糖具有的收敛止血、促进血小板聚集的特性,将白芨多糖置于海藻酸钙多孔微球中,既发挥了白芨多糖收敛止血、促进血小板聚集的特性,又利用了海藻酸钙多孔微球作为载体,使本发明技术方案所述的复合止血粉具有吸水率高、止血效果好、使用方便、不受创面、伤口大小的限制等特点,尤其适合烧烫伤和皮肤擦伤患者的治疗。
具体实施方式
实施例1:
步骤一:海藻酸钙多孔微球的制备:称取0.5gNaCl溶于97.5g去离子水中,然后再加入2g海藻酸钠配成海藻酸钠混合溶液备用;
在100g液体石蜡中加入0.5g质量百分比浓度为3%的Span80,用于调节HLB值作为混合油相,然后加入上述海藻酸钠混合溶液20g,在转速470rpm、45℃恒温水浴中乳化30分钟后,加入质量百分比浓度为5%的CaCl2溶液10g进行交联反应,搅拌30分钟后,离心、洗涤、冷冻干燥,即得海藻酸钙多孔微球;该海藻酸钙多孔微球的吸水率为290%
步骤二:复合止血粉的制备:将上述2g海藻酸钙多孔微球与2g白芨多糖加入100mL去离子水中,用漩涡混合器混合均匀5min后,冷冻干燥,得到复合止血粉4g。
实施例2—5:
在实施例一的基础上,改变步骤一中NaCl的质量百分比浓度,其余条件不变。

在上述实施例2中,搅拌时间均为40分钟,制备的海藻酸钙多孔微球吸水率为287%。
在上述实施例3中,搅拌时间均为20分钟,制备的海藻酸钙多孔微球吸水率为225%。
在上述实施例4中,搅拌时间均为36分钟,制备的海藻酸钙多孔微球吸水率为240%。
在上述实施例5中,搅拌时间均为28分钟,制备的海藻酸钙多孔微球吸水率为235%。
实施例6—8:
在上述任意实施例的基础上,改变海藻酸钙多孔微球与白芨多糖的质量比,其余条件不变。

对比云南白药的BCI84.92%,止血时间178.3±17.89s。
上述BCI为体外凝血指数,凝血指数越小,说明材料的凝血效果越好。
上述止血时间越小,说明材料止血效果越好。

Claims (4)

1.一种白芨多糖复合止血粉,其特征在于该复合止血粉是由海藻酸钙多孔微球与白芨多糖粉按重量比1:1—3配置而成。
2.根据权利要求1所述的白芨多糖复合止血粉,其特征在于上述海藻酸钙多孔微球的吸水率为230—290%。
3.制备上述权利要求所述白芨多糖复合止血粉的方法,其制备工艺为:
步骤一:海藻酸钙多孔微球的制备:称取0.25—1克的氯化钠溶于97—97.75克去离子水中,然后再加入2克海藻酸钠配成海藻酸钠混合溶液备用;
