Abstract
本发明提供了一种虾青素软胶囊,其中胶囊内容物包括以下重量份数的组分:雨生红球藻提取物180~220份、天然维生素E 140~160份、红花籽油40~60份;所述虾青素软胶囊的胶囊皮由以下组分组成:明胶108份、甘油54份、纯化水108份。同时提供了该虾青素软胶囊的制备方法。本发明虾青素软胶囊内容物中具有多种天然抗氧化成分,能够清除自由基,提高人体抗衰老能力,且富含人体必需的营养成分亚油酸、大豆卵磷脂和维生素E;能够增加人体细胞活性,提高免疫力,且组成简单,没有毒副作用。

技术领域
本发明属于保健食品领域,具体涉及一种虾青素软胶囊及其制备方法。
背景技术
虾青素是类胡萝卜素的一种,为一种较强的天然抗氧化剂,最主要的功能是清除自由基,提高人体抗衰老能力,与其他类胡萝卜素一样,虾青素属于一种脂溶性及水溶性的色素,存在于虾、蟹、鲑鱼、藻类等海洋生物中。
雨生红球藻是一种单细胞绿藻,在逆境条件下可积累大量的虾青素,被公认为自然界天然虾青素的主要来源。雨生红球藻提取物除富含有天然抗氧化剂虾青素外,还含有众多人体必需的营养成分。
由此可见,雨生红球藻提取物中含有大量对人体有益和人体必需的功能成分,其用于药品、保健品原料,应用前景十分广阔。但是虾青素对光、热、氧、湿敏感不稳定,容易被氧化,目前已有的雨生红球藻提取物口服制剂产品的稳定性还有待进一步提高。
发明内容
本发明针对现有技术中虾青素的不稳定性,加入抗氧化剂保护剂,以便提高虾青素的吸收利用率,同时对组合物的成分进行适当调整,增强改善抗氧化的效果。
本发明的第一目的是提供一种虾青素软胶囊。
本发明的第二目的是提供上述虾青素软胶囊的制备方法。
本发明的第一目的,通过以下技术方案来实现的:
一种虾青素软胶囊,其中胶囊内容物包括以下重量份数的组分:雨生红球藻提取物180~220份、天然维生素E 140~160份、红花籽油40~60份。
优选地,所述胶囊内容物包括以下重量份数的组分:雨生红球藻提取物200份、天然维生素E 150份、红花籽油50份。
所述虾青素软胶囊的胶囊皮由以下组分组成:明胶108份、甘油54份、纯化水108份。
所述雨生红球藻提取物中天然虾青素含量为4~8%。
维生素E是一种脂溶性维生素,水解产物为生育酚,是重要的抗氧化剂,可以改善脂质代谢,抑制脂质过氧化及形成自由基,保护机体细胞免受自由基的毒害,抗衰老和抗癌,预防器质性衰退疾病。
红花籽油主要成份含饱和脂肪酸7%,油酸20%,亚油酸73%,其中亚油酸为人体必需但又不能在体内自行合成的不饱和脂肪酸,红花籽油它能有效溶解胆固醇,防止人体血清胆固醇在血管壁里沉积,具有降血脂、清除血管内壁沉积物以及降血压的作用。
进一步地,所述胶囊内容物组分还包括大豆卵磷脂,所述大豆卵磷脂的重量份数为30~50份。优选地,所述大豆卵磷脂的重量份数为40份。
卵磷脂是细胞膜的主要构成部分,补充卵磷脂,可以修补被损伤的细胞膜,改善细胞膜功能,使细胞膜软化和年轻化,增加细胞活性;同时其可以与维生素相容,从而改善维生素E的溶解分散性能,提高抗氧化作用效果。
进一步地,所述胶囊内容物组分还包括绿茶提取物,所述绿茶提取物的重量份数为50~70份。优选地,所述绿茶提取物的重量份数为60份。
所述绿茶提取物中含有茶多酚质量百分比为15%~25%。
绿茶提取物是从绿茶叶片中提取的活性成分,主要包括茶多酚(儿茶素)、咖啡碱、芳香油、水分、矿物质、色素、碳水化合物、蛋白质、氨基酸、维生素等。茶多酚具有抗氧化、清除自由基等作用,能明显降低高脂血症的血清总胆固醇、甘油三脂、低密度脂蛋白胆固醇含量,同时具有恢复和保护血管内皮功能的作用。茶多酚可阻断脂质过氧化过程,提高人体内酶的活性,从而起到抗突变、抗癌症的功效。茶多酚通过提高人体免疫球蛋白总量并使其维持在高水平,刺激抗体活性的变化,从而提高人的总体免疫能力,并可促进人体的自身调理功能。
在油脂中添加茶多酚,能阻止和延缓不饱和脂肪酸的自动氧化分解,有效地抑制油脂的氧化酸败,延长其贮藏期。本发明中绿茶提取物与红花籽油共同作用,两者具有很好的协同作用,绿茶提取物中的茶多酚,可以有效保护红花籽油中的不饱和脂肪酸——亚油酸,提高其稳定性,防止脂肪氧化,使红花籽油发挥更好的作用。
本发明的第二目的,通过以下技术方案来实现的:
一种上述虾青素软胶囊的制备方法,包括以下步骤:
(1)按配方比例称取胶囊内容物各组分,搅拌使其混合均匀,抽真空,过筛,得内容物料液;
(2)按配方比例称取甘油和纯化水混合均匀,升温后加入明胶,搅拌溶融,抽真空脱气后过筛,得胶液,将胶液保温放置,备用;
(3)将内容物料液与胶液进行压丸,得软胶囊,对软胶囊进行干燥定型后,清洗胶囊,干燥即得成品虾青素软胶囊。
所述步骤(1)中抽真空真空度为0.06~0.08MPa。优选地,真空度为0.07MPa。
所述步骤(1)中过筛为过100目筛。
所述步骤(2)中升温为温度升至70~80℃;抽真空脱气的真空度为0.06~0.08MPa。优选地,升温温度为75℃,真空度为0.07MPa。
所述步骤(2)中过筛为过100目筛。
所述步骤(2)保温温度为55~65℃。优选地,保温温度为60℃。
所述步骤(3)中压丸控制内容物装量为0.4g/粒。
所述步骤(3)中压丸过程中室温控制在18-26℃,相对湿度为30%~40%。优选地,压丸过程中室温控制在22℃,相对湿度为35%。
所述步骤(3)中干燥定型为在18-26℃,相对湿度为30%-40%,定型3~5小时。优选地,干燥定型温度为22℃,相对湿度为35%,定型时间为4小时。
所述步骤(3)中清洗胶囊为用体积分数为95%的乙醇清洗胶囊。
所述步骤(3)中干燥为在24~28℃,相对湿度为20%~30%的条件下,干燥24~36h。优选地,干燥为在26℃,相对湿度为25%的条件下,干燥30h。
与现有技术相比,本发明具有以下有益效果:
(1)本发明虾青素软胶囊内容物中具有多种天然抗氧化成分,能够清除自由基,提高人体抗衰老能力,且富含人体必需的营养成分亚油酸、大豆卵磷脂和维生素E。
(2)本发明虾青素软胶囊中大豆卵磷脂与维生素E协同作用,能提高维生素E与各组分的相容性,绿茶提取物可以保护红花籽油中不饱和脂肪酸亚油酸,防止其氧化酸败,延长其贮藏期,维生素E、茶多酚均具有抗氧化效果,可以有效保护虾青素,提高其储存过程的稳定性;
(3)本发明虾青素软胶囊能够增加人体细胞活性,提高免疫力,且组成简单,没有毒副作用。
具体实施方式
下面结合具体的实施例,详细说明本发明的技术方案,以便本领域技术人员更好理解和实施本发明的技术方案。下面实施例中雨生红球藻提取物中含天然虾青素质量百分比为5%,绿茶提取物中含茶多酚的质量百分比为15%。
实施例一
一种虾青素软胶囊,其中胶囊内容物包括以下重量的组分:雨生红球藻提取物200g、天然维生素E 150g、红花籽油50g,胶囊皮由以下组分组成:明胶0.108g、甘油0.54g、纯化水0.108g。
上述虾青素软胶囊的制备方法,包括以下步骤:
(1)按配方比例称取虾青素油、天然维生素E和红花籽油,搅拌使其混合均匀后,抽真空0.07Mpa,过100目筛,得内容物料液,备用;
(2)按配方比例称取明胶、甘油和纯化水混合均匀,加热至75℃,加入明胶,搅拌溶融,抽真空脱气0.07Mpa,过100目筛,将胶液保温放置,保温温度为60℃,且肉眼检验无气泡,备用;
(3)将内容物料液与胶液进行压丸,压丸过程中控制内容物装量为0.4g/粒,室温控制在22℃,相对湿度为35%,得软胶囊,对软胶囊进行干燥定型,定型温度在22℃,相对湿度为35%,定型4小时,定型完后,用体积分数为95%的乙醇清洗胶囊,在26℃,相对湿度为25%的条件下,干燥30h,即得成品虾青素软胶囊。
食用方法及食用量:每天1次,每次1粒。
实施例二

实施例四
一种虾青素软胶囊,其中胶囊内容物包括以下重量的组分:雨生红球藻提取物180g、天然维生素E 160g、红花籽油60g,大豆卵磷脂50g,胶囊皮由以下组分组成:明胶0.108g、甘油0.54g、纯化水0.108g。
上述虾青素软胶囊的制备方法,包括以下步骤:

以下对本发明营养制剂的抗氧化性进行进一步的临床研究
试验1组:本发明实施例1所制得的胶囊。
试验2组:本发明实施例6所制得的胶囊产品。
对照组:服安慰剂,剂量与试验组相同。
1.方法
(1)受试者排除标准:
选择年龄在30-60岁,身体健康状况良好,无明显脑、心、肝、肺、肾等疾病,无长期服药史,志愿受试保证配合的人群作为纳入标准。排除标准:妊娠或哺乳期妇女;合并有严重疾病患者;短期内服用与受试功能有关的物品,影响到对结果的判断者;不符合纳入标准,未按规定服用受试样品,无法判断功效或资料不全影响功效或安全性判断者。
(2)试验设计及分组:
180例受试者,按过氧化脂质含量(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)水平随机分为试验1组60例、试验2组60例、安慰剂组60例,尽可能考虑影响结果的主要因素如年龄、性别、生活饮食习惯等,进行均衡性检验,以保证组间的可比性。
(3)受试物的计量和使用方法:
每日1次,每次1粒,连续服用30天。试验期间不改变原来的生活习惯,正常饮食。
2.实验结果
观察试验前后MDA的变化及MDA下降百分率,SOD的变化及SOD升高百分率,GSH-Px变化及GSH-Px升高百分率,各项功效指标情况见下表1。

从抗氧化试验结果可知,安慰剂组三项指标使用后均无明显变化。过氧化物歧化酶(SOD)试验组1上升了7.20%,试验组2上升了11.21%。谷胱甘肽过氧化物酶(GSH-Px)活性试验组1上升了5.47%,试验组2上升了8.44%。过氧化脂质含量(MDA)试验组1下降了5.35%,试验组2下降了9.85%。试验结果分析,证明了本发明组合物具有较好的抗氧化作用。

3.根据权利要求1所述的虾青素软胶囊,其特征在于,所述胶囊内容物组分还包括大豆卵磷脂,所述大豆卵磷脂的重量份数为30~50份。
4.根据权利要求1所述的虾青素软胶囊,其特征在于,所述胶囊内容物组分还包括绿茶提取物,所述绿茶提取物的重量份数为50~70份。
5.一种权利要求1-4任一项所述的虾青素软胶囊的制备方法,其特征在于,包括以下步骤:
(1)按配方比例称取胶囊内容物各组分,搅拌使其混合均匀,抽真空,过筛,得内容物料液;
(2)按配方比例称取甘油和纯化水混合均匀,升温后加入明胶,搅拌溶融,抽真空脱气后过筛,得胶液,将胶液保温放置,备用;
(3)将内容物料液与胶液进行压丸,得软胶囊,对软胶囊进行干燥定型后,清洗胶囊,干燥即得成品虾青素软胶囊。
6.根据权利要求5所述的虾青素软胶囊的制备方法,其特征在于,所述步骤(2)中升温为温度升至70~80℃;抽真空脱气的真空度为0.06~0.08MPa。
7.根据权利要求5所述的虾青素软胶囊的制备方法,其特征在于,所述步骤(2)保温温度为60±5℃。
8.根据权利要求8所述的虾青素软胶囊的制备方法,其特征在于,所述步骤(3)中压丸控制内容物装量为0.4g/粒。
9.根据权利要求5所述的虾青素软胶囊的制备方法,其特征在于,所述步骤(3)中压丸过程中室温控制在18-26℃,相对湿度为30%~40%。
