自动化构建方案
第一步,向96孔板的目标孔底部中注入1μl模板,后将96孔板置于96板架上,探头伸入96孔板上部,切换通道至管道A使得管道中充满第一轮PCR反应液,开启通道,向目标孔中注入29μl的第一轮PCR反应液。取下96孔板,贴上密封膜,震荡混匀,离心后置于PCR仪上进行反应。反应液组分如表1,反应条件如表2。
第二步,切换管道至管道B使得管道中充满磁珠混合液,从PCR仪上取出第一轮PCR结束的96孔板置于板架上,探头将PCR管顶部刺入停于管上部,注入15μL磁珠,混匀静置5分钟使其充分接触后,置于磁力架上2分钟后使磁珠吸附于底部,吸取上清转移至96孔纯化板。
第三步,将96孔板置于板架上,探头将PCR管顶部刺入停于管上部,注入27μL磁珠,开启磁力架电源并反复切换正负极,使得磁珠与反应液混匀,关闭磁力架静置3分钟。再次开启磁力架电源上,使得磁珠吸附于底部一侧,吸取探头深入另一侧,吸取上清。
第四步,切换至管道C,使得管道中充满BW13,探头向孔道中注入40μL溶液,悬浮混匀并静置2min。开启磁力架磁力使得磁珠被吸附于管壁一侧,吸附3min,开启吸取探头,于另一侧吸取上清。
第五步,切换至管道D,使得管道中充满70%乙醇,开启注入探头向纯化管中注入100μL70%乙醇,反复10次切换磁力架正负极,使得在管壁两侧来回吸附磁珠以保证充分悬浮磁珠。最后磁力停止于一侧,保证磁珠充分吸附于管壁,开启吸取探头,将上清吸取。重复一次。
第六步,切换管道E,使得管道中充满空气,开启注射探头使得空气不断注入纯化管,使得磁珠干燥。
第七步,切换管道F,使得管道中充满二轮PCR反应液,开启注射探头,注入30μL反应液,切换磁力架正负极,使得磁珠与反应液充分混合。取下96孔板,加封封口膜置于PCR仪上。反应液组分如表3,反应条件如表4。
第八步,切换管道至管道B,使得管道中充满磁珠悬浮液,探头刺入密封膜,开启注入探头,注入30μL磁珠悬浮液,切换磁力架正负极,使得磁珠与液体充分混匀后关磁力电源,静置3分钟使得磁珠充分吸附目标核酸片段。开启磁力电源,使得磁珠吸附于一侧管壁。开启吸取探头,将上清吸取。
第九步,切换管道至管道F,开启探头向管中注入40μL BW10,切换正负极,使得磁珠与BW10充分混匀,关磁力架电源,静置3min。开启磁力架电源,使得磁珠吸附于一侧管壁,开启吸取探头将上清吸去。
第十步,切换管道至管道D,注入70%乙醇溶液100μL,开启磁力架电源并切换正负极使得磁珠与乙醇溶液混匀,最后使得磁珠吸附于一侧管壁,开启吸取探头将上清吸去。
第十一步,切换至管道G,开启注入探头,使得空气进入管中,将磁珠干燥。
第十二步,切换至管道H,开启注入探头,注入20μL洗脱液,开启磁力架电源,使得洗脱液与洗脱液充分混匀,关磁力架电源静置3分钟后,再次开启磁力架电源,使得磁珠吸附于管壁一侧。开启封口膜,吸取上清,此时上清即为构建好的测序文库。
注:如不使用电磁,可以通过移动强力磁铁的距离来完成磁性的切换。
