技术领域

背景技术
特应性是一种综合症,其中对通常暴露的无害抗原(过敏原)发生过度免疫应答(超敏反应),并且由其引起的疾病是特应性疾病。特应性皮炎、过敏性鼻炎、过敏性结膜炎、过敏性哮喘和其它特应性疾病经常在患有特应性疾病的人群中发生。至少存在与特应性的免疫学发病机制相关的1型超敏反应的过敏反应。尽管特应性以多种方式定义,但其特征在于血液中对过敏原的高含量IgE。
过敏被定义为当暴露于周围抗原时在体内发生获得性免疫应答,并且在反复暴露于抗原期间免疫应答因为对抗原的记忆而改变的病症。暴露于有害抗原时发生的过敏现象对身体具有有益效果,并通过疫苗诱导免疫反应。另一方面,如果过度诱导对无害抗原的免疫应答并且发生超敏反应,则在体内发生炎症反应以引起过敏性疾病。哮喘是一种特征在于气道慢性炎症的疾病。其是由于炎症和功能变化引起的结构变化而发生哮喘、呼吸困难和其它症状的疾病。作为哮喘中发现的慢性炎症的病因学,对周围环境中过敏原的超敏反应是一种重要的免疫机制,尤其是已知对源自屋尘螨的过敏原的超敏反应是哮喘的重要原因。
屋尘螨是属于居住在人类居住地的节肢动物的动物。屋尘螨吃食人类皮屑,大量生活在城市住宅床垫、地毯、家具等中,并且已知是世界上最重要的过敏原,过敏性疾病(诸如过敏性哮喘)的病因。屋尘螨的消化器官有各种消化酶,其通过从屋尘螨排出的粪便扩散到空气中。当其被吸入时,患有过敏性哮喘的人出现超敏反应,并且重复暴露于屋尘螨过敏原,出现打喷嚏、鼻炎症状(诸如流鼻涕)、哮喘症状(诸如喘息)、呼吸困难以及特应性皮炎诸如瘙痒。欧洲屋尘螨(Dermatophagoides pteronyssinus)和北美屋尘螨(Dermatophagoides farinae)是造成过敏性疾病的重要屋尘螨的常见示例。Der p1、Derp2、Der p3、Der p4、Der p6、Der p9和Der p15被称为欧洲屋尘螨过敏原。Der f1、Der f2、Der f11和Der f18是源自北美屋尘螨的过敏原,已知这是重要的。根据国内流行病学研究的结果,源自北美屋尘螨而不是欧洲屋尘螨的过敏原主要存在于大城市的室内灰尘中,并且已经报道了在源自北美屋尘螨的过敏原中,Der f2过敏原是室内灰尘中最多的(SonJong Ryul et al.A Survey of Mite Allergens Contamination in House.Journal ofKorean Society for Atmospheric Environment 2006;22(5):719-23)。
免疫疗法可被分类为主动免疫疗法,其直接诱导引起疾病的抗原物质进入体内以诱导体内免疫细胞活性,以及被动免疫疗法,其通过施用体外制备的物质(诸如单克隆抗体免疫疗法)来调节免疫应答。与被动免疫疗法相比,从成本和给药频率方面来看,主动免疫疗法被认为是作为预防疾病的方法的更有效方法。主动免疫疗法通过使用诱导获得性免疫应答的策略来诱导防御性免疫,所述获得性免疫应答具有诱导抗原特异性长期防御记忆的能力,这是获得性免疫的特征。控制过敏反应的关键是控制对过敏原的过敏原特异性免疫超敏反应,诸如体液(或抗体介导的)和细胞(T细胞介导的)免疫。
细菌分泌的细胞外囊泡(EV)是直径为20-100nm的球形磷脂双层。生化和蛋白质组学研究已经示出外膜蛋白、脂多糖(LPS)、外膜脂质、DNA、RNA和细胞质中的蛋白质。最近,已经报道了革兰氏阴性菌诸如脑膜炎奈瑟菌(Neisseria meningitides)、鲍氏不动杆菌(Acinetobacter baumannii)、牙龈卟啉单胞菌(Porphyromonas gingivalis)、肠炎沙门氏菌(Salmonella enteric)、血清型鼠伤寒沙门氏菌(serovar Typhimurium)、幽门螺杆菌(Helicobacter pylori)和霍乱弧菌(Vibrio cholerae)分泌细胞外囊泡以诱导针对细菌感染的防御免疫,而革兰氏阳性细菌诸如金黄色葡萄球菌(Staphylococcus aureus)还能够通过细胞外囊泡抑制细菌感染,以诱导针对细菌感染的防御免疫。
作为抗原载体的源自革兰氏阴性细菌的囊泡具有诱导严重毒性例如败血症的问题,尽管其具有优异的免疫诱导效果。这是由于在革兰氏阴性细菌的细胞外膜中存在被称为内毒素的脂多糖(LPS),其在目前已经示出通过大量存在于血管内皮细胞中的TLR4受体诱导全身性血管炎症,从而导致败血症。作为用于解决由革兰氏阴性细菌来源的细胞外囊泡导致的毒性问题的方法,可将除去存在于细胞外囊泡中的LPS或处理抑制LPS活性的化合物的方法用作抗原载体。原生质体是植物或细菌的活细胞材料。通过机械/酶促方法除去这些细胞的细胞外膜和细胞壁,除去细菌外膜中的内毒素含量,并解决囊泡本身的毒性问题。此外,可处理药物诸如多粘菌素B以除去细胞外囊泡中存在的内毒素的活性。
到目前为止,屋尘螨的过敏原免疫疗法主要用于从屋尘螨中提取过敏原或作为重组蛋白形式,并且不存在过敏原在抗原载体诸如细胞外囊泡中表达的情况。

因此,本发明提供一种药物组合物,所述药物组合物包含表达源自屋尘螨的过敏原的细菌、来源于所述细菌的细胞外囊泡,及其制备方法。

技术方案
本发明提供一种用于预防或治疗过敏性疾病的药物组合物,所述药物组合物包含细菌来源的细胞外囊泡作为活性成分,其中所述细菌是过表达的屋尘螨来源的过敏原基因,其中所述过敏性疾病由屋尘螨来源的抗原引起。
在本发明的一个实施方案中,所述细菌为大肠杆菌或沙门氏菌(Salmonellaspp.)。
在本发明的另一个实施方案中,所述屋尘螨为欧洲屋尘螨(Dermatophagoidespteronyssinus)或北美屋尘螨。
在本发明的另一个实施方案中,所述屋尘螨来源的过敏原选自Der p1、Der p2、Der p3、Der p4、Der p6、Der p9、Der p15、Der f1、Derf2、Der f11和Der f18。
此外,本发明提供一种制备用于预防或治疗由屋尘螨来源的抗原引起的过敏性疾病的细菌来源的囊泡的方法。所述方法包括以下过程:
(a)过表达细菌中的屋尘螨来源的抗原基因;
(b)在培养基中培养细菌;
(c)将所述细菌来源的囊泡与所述培养基分离;并且

发明模式
为了开发抗原特异性免疫调节剂,重要的是选择抗原和佐剂以诱导期望的免疫应答。在本发明中,靶向Der f2过敏原,所述Der f2过敏原为用于控制对屋尘螨过敏原的超敏反应的在韩国最丰富的免疫调节剂候选抗原。
另一方面,基于来源于细菌的细胞外囊泡可诱导对宿主的各种免疫应答的事实,已经尝试使用来源于细菌的细胞外囊泡作为佐剂,但是来源于细菌的细胞外囊泡具有LPS、肽聚糖等,因此存在必须解决由于细胞外囊泡引起的毒性问题的问题。因此,在本发明中,为了消除在使用来源于细菌的细胞外囊泡作为药物递送载体时细胞外囊泡的毒性问题,除去细胞外囊泡的外膜和包含肽聚糖层的细胞壁以制备原生质体,并且制备和使用人造细胞外囊泡。还尝试了通过将多粘菌素B协同给药于培养细菌后天然分泌的囊泡来降低毒性,所述多粘菌素B抑制囊泡外膜中存在的LPS的活性。
具体地讲,在本发明中,将作为源自北美屋尘螨的主要过敏原的Der f2抗原在大肠杆菌中过表达,然后进行培养。然后,通过处理溶菌酶制备原生质体,并通过挤出方法制备细胞外囊泡来制备候选物质。此外,为了优化候选疫苗,克隆细菌中存在的Mic A小RNA,插入沙门氏菌并过表达,然后进行培养。然后,PMB与囊泡同时施用,通过在保持免疫原性的同时降低毒性来建立最佳浓度。
随后,在体外和体内评估加载抗原的候选细胞外囊泡的免疫原性,以确定给药途径和剂量用于功效评估。结果,发现加载抗原的细胞外囊泡是有效的疫苗。
此外,为了评估免疫调节剂在疾病模型中的功效,将北美屋尘螨提取物和明矾注入腹腔并用过敏原致敏,并且然后将单独的螨提取物溶液施用于鼻腔以制备哮喘动物模型。
在本发明中,“屋尘螨过敏性疾病”没有特别限制,只要其是可由屋尘螨过敏原引起的疾病即可,但优选选自哮喘、鼻炎和特应性皮炎。
在本发明中,“治疗或预防过敏性疾病”包括缓解和减轻过敏性疾病,和改善症状,并且还包括降低患过敏性疾病的可能性。
在本发明中,能够过表达该基因的细菌没有特别限制,但优选大肠杆菌或沙门氏菌。
在本发明中,在分离细胞外囊泡的步骤中,可使用天然分泌的细胞外囊泡。对于分离细胞外囊泡的方法没有特别限制,但其可通过利用过滤器浓缩,然后进行超速离心等来获得。
在本发明中,可人工制备细胞外囊泡,并且用于制备该囊泡的方法没有特别限制,但其可通过利用过滤器挤出获得,并且还包括诸如离心和超速离心的方法。
在本发明中,从培养基中分离细胞外囊泡的方法没有特别限制,只要包括细胞外囊泡即可。例如,可使用诸如离心、超速离心、利用过滤器过滤、凝胶过滤色谱、自由流动电泳、毛细管电泳、使用聚合物分离、以及它们的组合之类的方法,从细菌培养基或发酵食品中分离细胞外囊泡。此外,可进一步包括诸如洗涤以用于去除杂质和浓缩所获得的细胞外囊泡之类的工艺。所述细胞外囊泡包括天然或人工分泌的细胞外囊泡。
在本发明中,源自屋尘螨的过敏原在通过上述方法制备的细胞外囊泡的内腔中表达。Der f2、Der p2等作为表达蛋白质是优选的,并且Derf2是最优选的。
在本发明中,通过该方法制备的细胞外囊泡可具有10-200nm,并且优选地20-100nm的平均直径。
在本发明中,可将用于治疗或预防过敏性疾病的组合物制备成药物组合物。为了将该组合物用于治疗和预防,可根据本发明自身施用细胞外囊泡,但优选施用包含细胞外囊泡作为活性成分的药物组合物。
所述药物组合物包含作为活性成分的分离的细胞外囊泡,并且可包括药学上可接受的载体。通常可用于制剂的药学上可接受的载体包括盐水、无菌水、林格氏溶液、缓冲盐水、环糊精、右旋糖溶液、麦芽糖糊精溶液、甘油、乙醇、脂质体等,但本发明不是限于此。根据需要,还可包括其它常规添加剂,诸如抗氧化剂、缓冲剂等。另外,还可添加稀释剂、分散剂、表面活性剂、粘合剂、润滑剂等以将组合物制备成可注射制剂,诸如水溶液、悬浮液、乳液等、丸剂、胶囊、颗粒剂或片剂。根据组分,合适的药学上可接受的载体及其制剂可以优选参考Remington的文献(Remington's Pharmaceutical Science,Mack PublishingCompany,Easton PA)中公开的方法。根据本发明的药物组合物不限于制剂,而是可制备成注射剂、吸入剂、外用皮肤药物等。
用于施用根据本发明的药物组合物的方法没有特别限制,而是根据期望的方法,可包括胃肠外给药,例如肌肉注射、皮下施用、吸入施用、鼻腔施用、舌下施用、皮肤施用或口服施用。
剂量范围可根据患者的体重、年龄、性别和健康状况、饮食、给药持续时间、给药方式、排泄率、疾病严重程度等而有所不同。每日使用的剂量是指足够量的本发明的治疗性物质,其被施用于需要治疗的受试者以便缓解疾病。治疗性物质的有效剂量可根据具体化合物、疾病的状态和严重性以及需要治疗的受试者而有所不同,并且一般可由熟练的从业者确定。作为非限制性示例,用于人体的根据本发明的组合物的剂量可根据患者的年龄、体重和性别、给药方式、健康状况和疾病的严重程度而有所不同。
在下文中,将描述本发明的优选示例性实施例以促进对本发明的理解。然而,以下实施例应当被认为仅是描述性意义,并且本发明的范围不限于实施例。
实施例
实施例1:制备加载Der f2抗原的细胞外囊泡(参见图1)
通过用于Der f2克隆的RT-PCR方法合成cDNA的Der f2基因(参考序列:基因库AB195580),北美屋尘螨的主要过敏原。将PCR产物插入T-平端PCR克隆试剂盒(Solgent)中,并用EcoRI(OmpA,ABC转运蛋白)和BglII(FepA)切割每个质粒。片段通过电泳分离,插入pET-30a质粒中,通过热休克法转化到大肠杆菌DH5a中,最后克隆该基因。
将上述实施例中获得的Der f2克隆体在Luria Bertani肉汤(Merch)中于37℃、200rpm下培养12小时,并且然后进行Expre质粒mini prep。使用试剂盒(GeneAll)来分离每种质粒并转染到大肠杆菌BL21(Real Biotech)中。
在200rpm和37℃下,将BL21在100ml LB肉汤(1mM IPTG)中培养3小时直至OD值达到0.3,并且通过在4℃下以3,000×g离心10分钟获得的细菌沉淀用Tris(0.01M TrisHCl,0.5M蔗糖)洗涤,在4℃下以3,000×g离心10分钟,并重悬于Tris缓冲液中。在37℃下,用0.01M EDTA以100rpm将重悬的细菌处理30分钟,在4℃下沉淀,以3,000×g处理10分钟,并且然后用Tris缓冲液洗涤以用于沉淀。
接着将细菌沉淀重悬浮于Tris缓冲液中,并在37℃、100rpm下用1mg/ml溶菌酶(Sigma)处理2小时以获得原生质体,并且然后利用LiposoFast挤出机(Avestin)依次通过孔径为10μM、5μM和1μM的膜挤出。最后,通过用10%和50%opti-prep密度梯度培养基(OptiPrep)超速离心获得加载Der f2抗原的细胞外囊泡。
实施例2:加载Der f2抗原的细胞外囊泡的物理特性
将实施例1中获得的加载Der f2抗原的细胞外囊泡用PBS(50μg/ml)稀释,并将10μl加载到300目铜网格(EMS)中。将乙酸双氧铀(2%)滴在网格上进行阴性染色,并用JEM1011电子显微镜(JEOL)观察。结果,如图2所示,证实细胞外囊泡是球形的并且被脂质双层包围。
为了测量在实施例1中获得的加载Der f2抗原的细胞外囊泡的尺寸,用PBS(500ng/ml)稀释细胞外囊泡,并通过使用Zetasizer Nano ZS(Malvern Instruments)的动态光散射来测量直径大小分布并使用Dynamic V6软件分析结果。结果,如图3所示,发现细胞外囊泡的直径为100-300nm。
实施例3:加载Der f2抗原的囊泡的先天性免疫应答
加载Der f2抗原的细胞外囊泡应当能够诱导免疫应答以便将细胞外囊泡用作免疫调节剂。因此,为了评价加载Der f2抗原的细胞外囊泡诱导先天性免疫应答的水平,使用树突细胞(骨髓来源的树突细胞,BMDC)进行体外实验。
首先,将BMDC(5×105个细胞/孔)在包含10%FBS和抗生素(100单位/ml青霉素和100μg/ml链霉素)的DMEM中于37℃下在24孔组织培养板(TPP)中培养24小时。在除去培养基后,用以0.1和1μg/ml补充的无血清DMEM培养基替换加载Der f2抗原的细胞外囊泡。15小时后,通过BMDC中的CD86表达程度和使用培养基的ELISA测定,来测量作为T细胞分化细胞因子的IL-12和IL-6的表达水平。
结果,如图4所示,加载Der f2抗原的细胞外囊泡极大地增加了CD86在抗原呈递细胞中的表达(参见图4)。另外,如图5所示,用加载Der f2抗原的细胞外囊泡处理的抗原呈递细胞显著增加对于分化成Th1细胞而言重要的IL-12的分泌,并显著增加对于分化成Th17细胞而言重要的IL-6的分泌(图5)。
实施例4:源自MicA sRNA过表达菌株的囊泡的物理分析
使用具有优异的Hfq结合sRNA保守性的sRNA将包含与大肠杆菌sRNA碱基序列相同的Mic AsRNA(CATCTCTGAATTCAGGGATGATGATAACAAATGCGCTTCTTT)的pBRplac-sRNA质粒引导到沙门氏菌中。具体地讲,通过电穿孔将pBRplac-sRNA引入鼠伤寒沙门氏菌(SalmonellaTyphimurium)14028S菌种中。
在上述实施例中,体外培养sRNA过表达菌株,并分离囊泡,并且通过动态光散射和透射电子显微镜确认囊泡的物理性质。结果,如表1和图7所示,确认了来源于Mic A sRNA过表达菌株的细胞外囊泡的大小为约20nm,与对照组pBRplac相似,并且具有相似的形状(图7)。

实施例5:源自过表达Mic A sRNA的沙门氏菌的囊泡的免疫原性
已知沙门氏菌菌株诱导肠炎并诱导Th1免疫应答。因此,证实了由sRNA过表达获得的细胞外囊泡诱导Th1免疫应答的能力。具体地,每组5只小鼠(C67BL/6)以2天的间隔通过腹腔接受100ug/hd的细胞外囊泡。在第5天,从腹膜腔的淋巴结取出细胞,并使用抗CD3和CD28进行T细胞再刺激12小时,并测量由T细胞分泌的细胞因子量。
结果,如图8所示,与对照组相比,当施用源自sRNA过表达菌株的细胞外囊泡时,与Th1和Th17免疫反应性相关的细胞因子IFN-γ和IL-17增加,并且当施用源自sRNA过表达菌株的细胞外囊泡时,与Th2免疫应答相关的细胞因子IL-4与对照组没有区别(图8)。
由此,证实了通过使用源自过表达sRNA的沙门氏菌菌株的细胞外囊泡,可诱导与活细菌感染相似的Th1和Th17免疫反应性。
实施例6:源自过表达Mic A sRNA的沙门氏菌的囊泡的优化
沙门氏菌来源的囊泡本身具有良好的免疫原性,但由于存在于细胞外膜中的LPS导致的毒性是一个问题。为了解决该问题,通过将抑制LPS活性的PMB与源自过表达Mic AsRNA的沙门氏菌的囊泡混合来评价先天免疫应答的诱导水平。
具体地,将炎性细胞(巨噬细胞RAW264.7,5×105个细胞/孔)在包含10%FBS和抗生素(100单位/ml青霉素和100μg/ml链霉素)的DMEM中于37℃下在24孔组织培养板(TPP)中培养24小时。除去培养基后,将包含PMB的细胞外囊泡改变成为以1ug/ml的浓度添加的无血清DMEM培养基。15小时后,测定炎症细胞中IL-6和TNF-α的分泌。
结果,如图9所示,当以1μg/ml或10μg/ml的PMB浓度将细胞外囊泡混合并施用时,IL-6分泌减少。并且TNF-α分离的量由浓度为10μg/ml的PMB抑制。这意味着通过上述方法制备的囊泡的最佳PMB稀释浓度为1μg/ml至10μg/ml。
为了确定最佳PMB稀释浓度,在上述方法中,将PMB浓度细分为1、2、4、8和10ug/ml,并与囊泡一起施用于巨噬细胞。结果,发现超过2μg/ml的浓度,巨噬细胞中IL-6的分泌不再减少,因此证实通过稀释PMB浓度可优化候选物质(图10)。
实施例7:由加载Der f2抗原的囊泡引起的抗体免疫应答
为了证实加载Der f2抗原的细胞外囊泡在体内诱导抗体免疫反应性的能力和确定加载Der f2抗原的细胞外囊泡的有效量,向小鼠施用加载Der f2抗原的细胞外囊泡以诱导免疫。此外,通过在诱导免疫应答后从向其施用对照(PBS)和加载Der f2抗原的细胞外囊泡的小鼠收集血液来测量Der f2特异性IgG抗体,并收集支气管肺泡灌洗液并且测量Derf2特异性sIgA抗体的浓度。
具体地,将包含100ng Der f2的100μl PBS在4℃下涂布到96孔黑色板(GreinerBio-one)上并持续24小时。涂覆有Der f2的板用PBS洗涤,利用用PBS稀释的1%BSA溶液处理1小时,并且然后封闭并再次用PBS洗涤。
从来自施用有对照(PBS)和加载Der f2抗原的细胞外囊泡的小鼠的血液、血清和细胞层分离血清。将细胞层和上清液从支气管肺泡灌洗液中分离之后,用1%BSA溶液稀释血清和上清液,并在涂有Der f2抗原的平板上反应2小时。
温育后,将平板洗涤并与200ng/ml辣根过氧化物酶缀合的山羊抗小鼠IgG和IgA(Santa Cruz Biotechnology)一起温育2小时。再次洗涤平板之后,加入ECL底物(ThermoScientific),并用Victor Wallac 1420装置(PerkinElmer)分析IgG和sIgA抗体的滴度。
结果,如图11所示,不管是否在鼻腔中施用细胞外囊泡,通过肌内注射包含加载Der f2抗原的细胞外囊泡来诱导免疫应答的小鼠血清增加了Der f2抗原特异性IgG抗体产量。另一方面,仅在肌内注射加载Der f2抗原的细胞外囊泡,之后进行鼻腔给药时,支气管肺泡灌洗液中的Der f2抗原特异性sIgA产量增加(图12A和图12B)。
实施例8:加载Der f2抗原的细胞外囊泡引起的T细胞免疫应答
在体内,施用加载Der f2抗原的细胞外囊泡以诱导免疫应答,并且然后通过测量施用对照(PBS)和加载Der f2抗原的细胞外囊泡的小鼠中的T细胞中产生的细胞因子量来评估T细胞应答。
换句话说,通过使提取的肺组织通过100μm细胞过滤器(BD Biosciences),将提取的肺组织溶于5ml注射器洗涤缓冲液(包含2.5%FBS和0.01M HEPES的DMEM)中。在4℃下用氯化铵溶液(STEM CELL)将分离的细胞处理10分钟以使红细胞细胞溶解。将细胞用洗涤缓冲液洗涤并用40μm细胞过滤器(BD)过滤,并且然后使用包含10%FBS、50μM2-ME、0.01MHEPES和抗生素(100单位/ml青霉素、100μg/ml链霉素)的RPMI 1640培养基在24孔板中培养12小时。此时,用1μg/ml抗CD3(eBioscience)和1μg/ml抗CD28(eBioscience)抗体涂覆24孔板以重新刺激T细胞。
结果,如图13所示,与对照相比,加载Der f2抗原的细胞外囊泡处理组中由Th1细胞产生的主要细胞因子IFN-γ的量增加。并且在鼻腔给药时,加载Der f2抗原的细胞外囊泡剂量依赖性地降低。在IL-17(Th17中产生的主要细胞因子)的情况下,不考虑鼻腔给药,与对照组相比,加载Der f2抗原的细胞外囊泡给药组增加(图13)。
实施例9:加载Der f2抗原的细胞外囊泡在从北美屋尘螨中提取的过敏原诱导的
哮喘动物模型中的抗炎作用
为了评价加载Der f2抗原的细胞外囊泡在屋尘螨过敏原诱导的小鼠哮喘模型中的功效,进行以下实验(参见图14)。如图14所示,从北美屋尘螨中提取过敏原以制备哮喘模型,并且然后将明矾与75μg过敏原混合,并且每天两次给药于腹腔,之后将单独的50μg过敏原给药于鼻腔两次以制备哮喘模型(图14)。
为了利用加载Der f2抗原的细胞外囊泡诱导免疫应答,在制备哮喘模型之前,在肌内注射和鼻腔给药时以由实施例5的方法设定的浓度将加载Der f2抗原的细胞外囊泡预处理三次,如图9所示。并且在制备哮喘模型期间,仅将加载Der f2抗原的细胞外囊泡施用于鼻腔(图14)。
结果,如图15所示,当阳性对照小鼠未施用加载Der f2抗原的细胞外囊泡时,在由屋尘螨过敏原诱导哮喘时,支气管肺泡灌洗液中的炎性细胞数显著增加。与阳性对照相比,在哮喘模型之前利用加载Der f2抗原的细胞外囊泡诱导的小鼠在支气管肺泡灌洗液中的炎性细胞数显著减少。与阳性对照组相比,在建立哮喘模型之前和之后施用加载Der f2抗原的细胞外囊泡的小鼠具有显著减少的炎性细胞,以及加载有Der f2抗原的细胞外囊泡在建立哮喘模型之前施用(图15)。
因此,当将加载Der f2抗原的细胞外囊泡给药于鼻腔时,发现有效抑制由北美屋尘螨引起的哮喘。
本领域普通技术人员应该理解,本发明的上述说明是示例性的,并且在不脱离本发明的技术精神或本质特征的情况下,本文公开的示例性实施方案可容易修改成其它特定形式。因此,上述示例性实施方案应当被解释为说明性的且不限于任何方面。
工业实用性
本发明使用包含屋尘螨过敏原(Der f2等)的细菌来源的囊泡作为源自屋尘螨的过敏原免疫调节剂,以控制由过敏原引起的超敏反应。最终,可有效地预防或治疗由屋尘螨引起的过敏性疾病。
Claims (5)
1.一种用于预防或治疗过敏性疾病的药物组合物,其包含作为活性成分的细菌来源的细胞外囊泡,其中所述细菌为过表达的屋尘螨来源的过敏原基因,其中所述过敏性疾病由屋尘螨来源的过敏原引起,
其中所述屋尘螨是Dermatophagoides farinae,
其中所述屋尘螨来源的过敏原是Der f2,以及
其中所述细胞外囊泡加载有过敏原Der f2。
2.根据权利要求1所述的药物组合物,其中所述细菌为大肠杆菌或沙门氏菌。
3.一种制备用于预防或治疗由屋尘螨来源的过敏原引起的过敏性疾病的细菌来源的囊泡的方法,所述方法包括以下过程:
(a)在细菌中过表达屋尘螨来源的过敏原基因,其中所述屋尘螨来源的过敏原是Derf2;
(b)在培养基中培养所述细菌;
(c)将所述细菌来源的囊泡与所述培养基分离,其中所述囊泡加载有过敏原Der f2;并且
(d)移除所述细菌来源的囊泡的外膜中的内毒素,其中所述屋尘螨是Dermatophagoides farinae。
4.根据权利要求3所述的方法,其中所述细菌为大肠杆菌或沙门氏菌。
5.根据权利要求1所述的组合物在制备用于预防或治疗过敏性疾病的药剂中的用途。
