背景技术
医用胶原蛋白海绵是一种用途广泛的常用外科手术材料,主要用于手术中的止血、防止 组织间相互粘连等,术后可被组织完全降解吸收。具有诱导成纤维细胞衍生、聚集,刺激胶原生成、排列,刺激毛细血管的形成,保障肉芽组织生长需要的供血、供氧,调节不同细胞的功能,加速胶原的吸收和分泌,促进创面修复、愈合,预防术后并发症等。医用胶原蛋白海绵还可以进一步加工成为人造皮肤。现有医用胶原蛋白海绵绝大部分是采用哺乳动物(猪、牛等)的胶原蛋白如牛跟腱等为原料制造的,用日本黄姑鱼为原料制造医用胶原蛋白海绵至今未见文献报道。
发明内容
本发明的目的在于克服现有技术存在的不足,而提供一种制备工艺简单,使用可靠安全,成本低,使用效果好的日本黄姑鱼鱼皮胶原蛋白医用海绵的制备方法。
本发明的目的是通过如下技术方案来完成的,一种日本黄姑鱼鱼皮胶原蛋白医用海绵的制备方法,所述的制备方法是:先将日本黄姑鱼鱼皮除杂蛋白、脱脂,然后先用5%乙酸结合胃蛋白酶提取胶原蛋白,离心干燥后得到日本黄姑鱼鱼皮胶原蛋白,再加入食品级的谷氨酰胺转氨酶进行交联,冷冻干燥后获得胶原蛋白海绵。
作为优选:所述的除杂蛋白、脱脂的过程是:采用0.4—0.7mol/L的Na2CO3溶液与0.02—0.03mol/L的十二烷基苯磺酸钠溶液联用,并以脉冲的方式进行超声波辅助,进行0.8—1.3h的去杂蛋白处理;所述脱脂处理的脱脂条件为:料液比1:74—88g/mL,采用无水乙醚作为脱脂试剂,鱼片浸泡14—18h去脂肪,每3—4h进行溶液更换;
所述除杂蛋白、脱脂处理之后,先在处理后的鱼皮中加入0.5—1mol/L的5%乙酸,体积比为1:15—20,均质5—10min;然后加入胃蛋白酶,加酶量1—1.5%,料液比为1:25—35,酶解温度为3.5—4.5℃,酶解时间为30—35h,灭酶,酶解初期采用超声波辅助泵搅拌提取25—35min;提取胶原蛋白,离心干燥后得到日本黄姑鱼鱼皮胶原蛋白;
作为优选:在得到的日本黄姑鱼鱼皮胶原蛋白中加入酸度调节剂,调整pH值为5.5-6.5;向调整pH值后的日本黄姑鱼鱼皮胶原蛋白中加入日本黄姑鱼鱼皮胶原蛋白的质量百分比为0.05%-0.3%的谷氨酰胺转氨酶,并在40—50℃的温度下保持40min—80min,促使日本黄姑鱼鱼皮胶原蛋白产生交联反应。

作为优选的实施例:所述的除杂蛋白、脱脂的过程是:采用0.4—0.7mol/L的Na2CO3溶液与0.02—0.03mol/L的十二烷基苯磺酸钠溶液联用,并以脉冲的方式进行超声波辅助,进行0.8—1.3h的去杂蛋白处理;所述脱脂处理的脱脂条件为:料液比1:74—88g/mL,采用无水乙醚作为脱脂试剂,鱼片浸泡14—18h去脂肪,每3—4h进行溶液更换;
所述除杂蛋白、脱脂处理之后,先在处理后的鱼皮中加入0.5—1mol/L的5%乙酸,体积比为1:15—20,均质5—10min;然后加入胃蛋白酶,加酶量1—1.5%,料液比为1:25—35,酶解温度为3.5—4.5℃,酶解时间为30—35h,灭酶,酶解初期采用超声波辅助泵搅拌提取25—35min;提取胶原蛋白,离心干燥后得到日本黄姑鱼鱼皮胶原蛋白;
作为进一步的优选实施例:本发明在得到的日本黄姑鱼鱼皮胶原蛋白中加入酸度调节剂,调整pH值为5.5-6.5;向调整pH值后的日本黄姑鱼鱼皮胶原蛋白中加入日本黄姑鱼鱼皮胶原蛋白的质量百分比为0.05%-0.3%的谷氨酰胺转氨酶,并在40—50℃的温度下保持40min—80min,促使日本黄姑鱼鱼皮胶原蛋白产生交联反应。
本发明所述的冷冻干燥步骤为:预冻:-30—-25℃预冻2—4h;冷冻干燥:-40—-32℃,1 .5—6Pa冷冻干燥5—8h;升华干燥:5—9℃,45—57Pa升华干燥2 .4—3.1h;解析干燥:1—4℃,4—6Pa解析干燥1—3h,得胶原蛋白医用海绵材料。
本发明的其它实施例可以在上述公开的实施例基础上,进行具体数值和特征的简单替换,可以获得无数个实施例,且本领域技术人员在了解本发明内容的基础上,结合公知技术,能够方便地实施本发明,且不需要作出任何创造性的劳动。
Claims (4)
1.一种日本黄姑鱼鱼皮胶原蛋白医用海绵的制备方法,其特征在于:所述的制备方法是:先将日本黄姑鱼鱼皮除杂蛋白、脱脂,然后先用5%乙酸结合胃蛋白酶提取胶原蛋白,离心干燥后得到日本黄姑鱼鱼皮胶原蛋白,再加入食品级的谷氨酰胺转氨酶进行交联,冷冻干燥后获得胶原蛋白海绵。
2.根据权利要求1所述的日本黄姑鱼鱼皮胶原蛋白医用海绵的制备方法,其特征在于:所述的除杂蛋白、脱脂的过程是:采用0.4—0.7mol/L的Na2CO3溶液与0.02—0.03mol/L的十二烷基苯磺酸钠溶液联用,并以脉冲的方式进行超声波辅助,进行0.8—1.3h的去杂蛋白处理;所述脱脂处理的脱脂条件为:料液比1:74—88g/mL,采用无水乙醚作为脱脂试剂,鱼片浸泡14—18h去脂肪,每3—4h进行溶液更换;
所述除杂蛋白、脱脂处理之后,先在处理后的鱼皮中加入0.5—1mol/L的5%乙酸,体积比为1:15—20,均质5—10min;然后加入胃蛋白酶,加酶量1—1.5%,料液比为1:25—35,酶解温度为3.5—4.5℃,酶解时间为30—35h,灭酶,酶解初期采用超声波辅助泵搅拌提取25—35min;提取胶原蛋白,离心干燥后得到日本黄姑鱼鱼皮胶原蛋白。
3.根据权利要求1或2所述的日本黄姑鱼鱼皮胶原蛋白医用海绵的制备方法,其特征在于:在得到的日本黄姑鱼鱼皮胶原蛋白中加入酸度调节剂,调整pH值为5.5-6.5;向调整pH值后的日本黄姑鱼鱼皮胶原蛋白中加入日本黄姑鱼鱼皮胶原蛋白的质量百分比为0.05%-0.3%的谷氨酰胺转氨酶,并在40—50℃的温度下保持40min—80min,促使日本黄姑鱼鱼皮胶原蛋白产生交联反应。
4.根据权利要求1所述的日本黄姑鱼鱼皮胶原蛋白/藻酸盐复合海绵的制备方法,其特征在于:所述的冷冻干燥步骤为:
预冻:-30—-25℃预冻2—4h;冷冻干燥:-40—-32℃,1 .5—6Pa冷冻干燥5—8h;升华干燥:5—9℃,45—57Pa升华干燥2 .4—3.1h;解析干燥:1—4℃,4—6Pa解析干燥1—3h,得胶原蛋白医用海绵材料。
