Abstract
本发明公开了一种原花青素抗衰老组合物,包括以重量份计算的原花青素2‑10份、红景天提取物1‑5份、黑木耳提取物2‑8份、巴戟天提取物1‑5份、淫羊藿提取物1‑5份、薏苡仁提取物2‑8份、葡萄籽油10‑25份。本发明还公开了一种原花青素抗衰老组合物的制备方法和其保健品、护肤品中的应用。本发明所述的原花青素抗衰老组合物通过原花青素与其他组份之间的协同增效作用,有效抑制和清除自由基,具有良好的抗氧化抗衰效果。
Description
技术领域
本发明涉及保健品技术领域,更具体地说,涉及一种原花青素抗衰老组合物及其制备工艺及其制备工艺和应用。
背景技术
衰老是一种自然的过程,生物分子自然交联学说在论证生物体衰老的分子机制时指出:生物体是一个不稳定的化学体系,属于耗散结构,体系中各种生物分子具有大量的活泼基团,它们必然相互作用发生化学反应使生物分子缓慢交联以趋向化学活性的稳定。随着时间的推移,交联程度不断增加,生物分子的活泼基团不断消耗减少,原有的分子结构逐渐改变,这些变化的积累会使生物组织逐渐出现衰老现象。衰老可分为两类:生理性衰老和病理性衰老。前者指成熟期后出现的生理性退化过程,后者是由于各种外来因素(包括各种疾病)所导致的老年性变化。随着人们物质生活水平的提高,抗衰老已经成为人们生活中的一个重要议题,尤其是对于女性,为了保持年轻的体态和容貌,在抗衰老的产品中也是不惜重金。
原花青素,简称OPC,是一种有着特殊分子结构的生物类黄酮,由儿茶素、表儿茶素及表儿茶素没食子酸酯通过C4-C6或C4-C8连接而成的不同聚合度的混合物;一般为红棕色粉末,气微、味涩,溶于水和大多有机溶剂。原花青素水溶性好,极易被机体吸收,生物利用度达90%以上,是目前为止所发现的最强效的抗氧化剂和自由基清除剂。原花青素在体内抗氧化能力是维生素E的50倍、维生素C的20倍,主要存在存在于黑枸杞、葡萄籽、银杏叶、柏树、松树皮、黑米种皮、玫瑰花等多种植物中。

针对现有技术的上述缺陷,本发明的目的在于提供一种原花青素抗衰老组合物。
本发明的另一目的在于提供一种原花青素抗衰老组合物的制备工艺。
为实现上述目的,本发明所采用的技术方案如下:

进一步的,本发明所述的原花青素抗衰老组合物包括以重量份计算的
原花青素5-8份、
红景天提取物2-3份、
黑木耳提取物3-5份、
巴戟天提取物2-3份、
淫羊藿提取物2-4份、
薏苡仁提取物3-5份、
葡萄籽油15-20份。
进一步的,本发明所述的原花青素为单倍体、寡聚体、二聚体、三聚体、四聚体中一种或两种以上混合。
进一步的,本发明所述的红景天提取物通过以下方法提取:称取干红景天粉碎样品以石油醚脱脂三次后,用50-70%的乙醇回流提取5-8h,趁热过滤,重复提取三次合并滤液,减压回收溶剂,干燥后得到红景天提取物。
进一步的,本发明所述的黑木耳提取物通热水浸提得到的提取物,提取物中的黑木耳多糖包括以质量百分数计算的葡萄糖60-65%,甘露糖30-33%和半乳糖5-8%。
进一步的,本发明所述的所述巴戟天提取物通过以下方法提取:巴戟天粉碎后用50-60%乙醇提取一次,然后用水提取两次,合并提取液并浓缩,浓缩液过大孔树脂,依次用水、乙醇洗脱,得到水洗脱部分和乙醇洗脱部分,水洗部分经醇沉,得沉淀部位过滤后干燥,得到巴戟天提取物;
所述淫羊藿提取物通过以下方法提取:将淫羊藿粉末用65-75%乙醇为提取液,按料液比为1∶20-40(g/mL)在45-55℃下浸泡20-50min,然后超声提取30min,抽滤并收集提取液,提取液用旋转蒸发仪浓缩,依次用石油醚、三氯甲烷、乙酸乙酯和正丁醇进行纯化,纯化液除去溶剂后即得到淫羊藿提取物。在淫羊藿提取物中主要的活性物质是淫羊藿素总黄酮,此外还含有淫羊藿苷、淫羊藿素、脱水淫羊藿素、去甲淫羊藿素;淫羊藿素总黄酮有良好的免疫促进作用,通过促进成骨细胞增长、抑制破骨细胞分化来治疗和预防骨质疏松症,保护心肌缺血缺氧、降低耗氧,改善血液流变性,保护生殖系统,同时具有抗炎、抗肿瘤、抗衰老等多种药理作用。
所述薏苡仁提取物通过以下方法制备而成:薏苡仁粉碎后用10倍体积的蒸馏水浸泡4-8小时,继续加热提取1-2小时,提取完成后过滤,滤液加热浓缩、挥干,即得到薏苡仁提取物。
一种原花青素抗衰老组合物的制备工艺,包括
组分提取步骤:分别提取红景天提取物、薏苡仁提取物、巴戟天提取物、淫羊藿提取物、黑木耳提取物;
调制步骤:按比例将原花青素加入到葡萄籽油中,搅拌均匀后一次加入红景天提取物、薏苡仁提取物、巴戟天提取物、淫羊藿提取物、黑木耳提取物,搅拌均匀后即成。
一种原花青素抗衰老复合饮料,其包含本发明所述的原花青素抗衰老组合物。
一种原花青素抗衰老软胶囊,包括壁材和芯材,所述芯材为本发明所述的原花青素抗衰老组合物。
一种皮肤抗氧化外用制剂,包括本发明所述的原花青素抗衰老组合物。
相比现有技术,本发明具有以下有益效果:
1.本发明所述的原花青素抗衰老组合物通过原花青素与其他组份之间的协同增效作用,有效抑制和清除自由基,具有良好的抗氧化抗衰效果;
2.本发明所述的原花青素抗衰老组合物采用纯天然的提取物,无毒无副作用,既可以作为护肤品的添加成分,也可以作为口服保健品。

原花青素具有极强的抗氧化活性,是一种良好的自由基清除剂和脂质过氧化抑制剂,其作用机制是原花青素结构中有多个酚性羟基在体内释放H+,竞争性地与自由基结合,从而保护脂质不被氧化,进一步的,在本发明采用的原花青素为单倍体、寡聚体、二聚体、三聚体、四聚体中一种或两种以上混合。
红景天气微香、味苦、涩、寒;清热解毒、燥湿,用于肺热、脉热、瘟病、四肢肿胀。可用于活血、止血、理气养血、扶正固本、健脑益智、滋补强身,治疗老年性心衰,疲惫、阳痿、消渴病、肝脏疾患。现代医学研究指出:红景天为环境适应性药物,其重要功能有抗缺氧、抗疲劳、对中枢神经的双向调节、延缓衰老作用、抗毒作用、抗辐射作用和防癌作用、对内分泌系统的双向调节作用等。进一步的,本发明采用的红景天提取物可以通过购买方式获得,也可以通过以下方法提取:称取干红景天粉碎样品以石油醚脱脂三次后,用50-70%的乙醇回流提取5-8h,趁热过滤,重复提取三次合并滤液,减压回收溶剂,干燥后得到红景天提取物。
进一步的,本发明采用的黑木耳提取物是通过热水浸提得到的提取物,提取物中的黑木耳多糖包括以质量百分数计算的葡萄糖60-65%,甘露糖30-33%和半乳糖5-8%。黑木耳提取物中的主要成分黑木耳多糖与原花青素组合能显著的增加细胞的存活能力和增殖能力,进而协同提升了机体的免疫活性;表明黑木耳提取物与原花青素组合能够通过降低氧化产物量,提高抗氧化物质的含量,减少辐射细胞DNA的损伤,从而起到了有效的协同辐射防护作用。
进一步的,本发明所述的所述巴戟天提取物通过以下方法提取:巴戟天粉碎后用50-60%乙醇提取一次,然后用水提取两次,合并提取液并浓缩,浓缩液过大孔树脂,依次用水、乙醇洗脱,得到水洗脱部分和乙醇洗脱部分,水洗部分经醇沉,得沉淀部位过滤后干燥,得到巴戟天提取物;
所述淫羊藿提取物通过以下方法提取:将淫羊藿粉末用65-75%乙醇为提取液,按料液比为1∶20-40(g/mL)在45-55℃下浸泡20-50min,然后超声提取30min,抽滤并收集提取液,提取液用旋转蒸发仪浓缩,依次用石油醚、三氯甲烷、乙酸乙酯和正丁醇进行纯化,纯化液除去溶剂后即得到淫羊藿提取物。
所述薏苡仁提取物通过以下方法制备而成:薏苡仁粉碎后用10倍体积的蒸馏水浸泡4-8小时,继续加热提取1-2小时,提取完成后过滤,滤液加热浓缩、挥干,即得到薏苡仁提取物。
一种原花青素抗衰老组合物的制备工艺,包括
组分提取步骤:分别提取红景天提取物、薏苡仁提取物、巴戟天提取物、淫羊藿提取物、黑木耳提取物;
调制步骤:按比例将原花青素加入到葡萄籽油中,搅拌均匀后一次加入红景天提取物、薏苡仁提取物、巴戟天提取物、淫羊藿提取物、黑木耳提取物,搅拌均匀后即成。
一种原花青素抗衰老复合饮料,其包含本发明所述的原花青素抗衰老组合物。
一种原花青素抗衰老软胶囊,包括壁材和芯材,所述芯材为本发明所述的原花青素抗衰老组合物。

原花青素2份、
红景天提取物5份、

分别提取红景天提取物、薏苡仁提取物、巴戟天提取物、淫羊藿提取物、黑木耳提取物;
按比例将原花青素加入到葡萄籽油中,搅拌均匀后一次加入红景天提取物、薏苡仁提取物、巴戟天提取物、淫羊藿提取物、黑木耳提取物,搅拌均匀后即成。

分别提取红景天提取物、薏苡仁提取物、巴戟天提取物、淫羊藿提取物、黑木耳提取物;
按比例将原花青素加入到葡萄籽油中,搅拌均匀后一次加入红景天提取物、薏苡仁提取物、巴戟天提取物、淫羊藿提取物、黑木耳提取物,搅拌均匀后即成。
效果及性能评价
1.原花青素抗衰老组合物急毒性试验
将100只Wister大鼠随机分为5组,每组雌雄各半,其中4组对应实施例1-4为实验组,一组为生理盐水对照组。
给药方式:每天早晚各一次灌胃给药,实验组给药剂量为1.0/(Kg·d),对照组按照20mL/(Kg·d)剂量,连续28天,观察记录动物的外观体征、行为和粪便等性状,每天给药前称重,再定量给药。给药28天后,实验组和对照组均无大鼠死亡,也无明显异常现象。
2.细胞试验
大鼠脾细胞的制备:大鼠:Wister雄性,由哈尔滨医科大学基础医学院动物中心提供,6~8周龄,体重150g~200g。大鼠麻醉、脱臼处死,将大鼠置于75%酒精中,浸泡5~7min,取出大鼠至无菌皿中;在超净台内,无菌取脾,放入盛有预冷PBS的培养皿中;将脾脏放在75μm筛网上,剪碎,筛网脾脏位置的下面,对应放上无菌培养皿,用无菌注射器针芯研磨脾组织,同时不断加入PBS,从无菌皿中,获得细胞悬液,轻轻吹打均匀后,1000r/min,离心10min,保留沉淀细胞,弃去上清;加3~5mL红细胞裂解液,轻轻吹打充分混匀,破碎红细胞,静止放置3~5min;1000r/min,离心5min,保留沉淀细胞,弃去上清;用PBS清洗细胞两次,每次1000r/min,离心5min,弃上清;加入1640培养基中,该培养基中含10%(v/v)胎牛血清以及终浓度均为100μg/mL青霉素和链霉素;将细胞吹打混匀,在37℃、含5%CO2的潮湿环境的培养箱中,培养2h后,取细胞悬液,再用无菌75μm筛网过滤,0.4%台盼蓝染色后在显微镜下计数,观察活细胞(大于95%),调整细胞浓度至5×106。
本实验将脾细胞分为十组,具体如下:
Normal:正常+PBS
Model:辐射+PBS
组1:辐射+实施例1的组合物(0.2μg/mL、0.5μg/mL、1.0μg/mL)、
组2:辐射+实施例2的组合物(0.2μg/mL、0.5μg/mL、1.0μg/mL)
组3:辐射+实施例3的组合物(0.2μg/mL、0.5μg/mL、1.0μg/mL)
组4:辐射+实施例4的组合物(0.2μg/mL、0.5μg/mL、1.0μg/mL)
对比1:辐射+原花青素(0.2μg/mL、0.5μg/mL、1.0μg/mL)
1)细胞活力的测定
混匀细胞,铺96孔板,每孔加入培养液95μL,其后加药5μL,使每孔液体总量达100μL。细胞培养设未辐射正常对照组、辐射模型组和辐射给药组,每组设6个平行孔,加药12h后,进行辐照前,弃加药上清,加培养基,辐照后,加药,置细胞培养箱中培养24h,加入10μLMTT(5mg/mL)溶液。细胞培养板继续培养4h后,1000g/min,10min,弃上清液,每孔加入100μLDMSO。用Bio-Rad550酶联免疫检测仪于490nm波长处,测每孔的吸光度值(A值)按下式求出细胞存活率。

A0—未辐射组细胞吸光值。
各组细胞存活率的计算结果参见表1。

表1的结果显示:正常对照组比较对比1中原花青素组能提高脾细胞存活率,而组1-组4的本发明的组合物处理组显著增加了脾细胞的存活能力。
2)细胞增殖能力的测定
混匀细胞,铺96孔板,每孔加入培养液95μL,其后加药5μL,使每孔液体总量达100μL。细胞培养设空白组和药物组,每组设6个平行孔,加药后培养72h时,加入10μL MTT(5mg/mL)溶液。细胞培养板继续培养4h,然后将培养板,1000g/min,10min,弃上清液,每孔加入100μL DMSO。用Bio-Rad 550酶联免疫检测仪于490nm波长处,测每孔的吸光度值(A值)。按下式求出细胞存活增殖率。

试验结果:与正常对照组相比(120.01±10.11%),辐射模型组中脾淋巴细胞增殖显著的降低(15.45±2.01%,p<0.01)。原花青素够以剂量依赖方式显著的增加小鼠脾淋巴细胞增殖活性(p<0.05)。组1-组4的小鼠,在ConA的作用下对脾淋巴细胞增殖具有最为显著的刺激作用(85.17±4.09%,p<0.01),并且CI值为0.37,表明在组合物的作用效果下,协同效果最强。
对本领域的技术人员来说,可如以上描述的技术方案以及构思,做出其它各种相应的改变以及形变,而所有的这些改变以及形变都应该属于本发明权利要求的保护范围之内。

1.一种原花青素抗衰老组合物,其特征在于,包括以重量份计算的
原花青素2-10份、
红景天提取物1-5份、
黑木耳提取物2-8份、
巴戟天提取物1-5份、
淫羊藿提取物1-5份、
薏苡仁提取物2-8份、
葡萄籽油10-25份。
2.如权利要求1所述的原花青素抗衰老组合物,其特征在于,包括以重量份计算的
原花青素5-8份、
红景天提取物2-3份、
黑木耳提取物3-5份、
巴戟天提取物2-3份、
淫羊藿提取物2-4份、
薏苡仁提取物3-5份、
葡萄籽油15-20份。
3.如权利要求1或2所述的原花青素抗衰老组合物,其特征在于,所述原花青素为单倍体、寡聚体、二聚体、三聚体、四聚体中一种或两种以上混合。
4.如权利要求1所述的原花青素抗衰老组合物,其特征在于,所述红景天提取物通过以下方法提取:称取干红景天粉碎样品以石油醚脱脂三次后,用50-70%的乙醇回流提取5-8h,趁热过滤,重复提取三次合并滤液,减压回收溶剂,干燥后得到红景天提取物。
5.如权利要求1所述的原花青素抗衰老组合物,其特征在于,所述黑木耳提取物是通过热水浸提得到的提取物,提取物中的黑木耳多糖包括以质量百分数计算的葡萄糖60-65%,甘露糖30-33%和半乳糖5-8%。
6.如权利要求5所述的原花青素抗衰老组合物,其特征在于,
所述巴戟天提取物通过以下方法提取:巴戟天粉碎后用50-60%乙醇提取一次,然后用水提取两次,合并提取液并浓缩,浓缩液过大孔树脂,依次用水、乙醇洗脱,得到水洗脱部分和乙醇洗脱部分,水洗部分经醇沉,得沉淀部位过滤后干燥,得到巴戟天提取物;
所述淫羊藿提取物通过以下方法提取:将淫羊藿粉末用65-75%乙醇为提取液,按料液比为1∶20-40(g/mL)在45-55℃下浸泡20-50min,然后超声提取30min,抽滤并收集提取液,提取液用旋转蒸发仪浓缩,依次用石油醚、三氯甲烷、乙酸乙酯和正丁醇进行纯化,纯化液除去溶剂后即得到淫羊藿提取物。
