背景技术
“俄色叶”(藏语)是甘孜州藏族自治州藏族民间的一味习用药材,来源于蔷薇科植物变叶海棠Malustoringoides(Rehd.)Hughes.和花叶海棠Malustiansitoria(Batal.)Schneid.的干燥叶,已收录于四川省藏药材标准中。始载于《晶镜本草》:“俄色,花白如月光一般漂亮,果如宝石一般美丽”。在我国主要分布于四川西部、甘肃南部、青海和西藏部分地区。经调查发现,“俄色叶”在甘孜州藏族自治区为一较常用的民族民间药,使用历史悠久。当地居民经常用“俄色叶”泡水服用治疗消化不良,脘腹胀痛等症;当地藏医还用“俄色叶”与酥油茶同煮治疗高血压,高血脂。其果实中还含有较丰富的维生素,其中维生素B的含量高于多种普通水果种类及其品种的含量,具有很高的营养价值。近年,四川炉霍县林业部门已经开发变叶海棠及花叶海棠嫩叶制茶。该茶叶富含蛋白质、碳水化合物、多种维生素和有益人体的微量元素;具有降血压、降血糖、降血脂、抗缺氧、养身、养颜、活血化瘀、增加食欲等功效。变叶海棠和花叶海棠属于灌木至小乔木,花为白色,叶片嫩绿,果实成熟后红果满树,是理想的观赏绿化树种,可作为盆景。

变叶海棠大叶:是7-8月月采摘的蔷薇科植物变叶海棠Malustoringoides(Rehd.)Hughes.或花叶海棠Malustiansitoria(Batal.)Schneid.的干燥叶;
变叶海棠嫩叶:是5-6月采摘的蔷薇科植物变叶海棠Malustoringoides(Rehd.)Hughes.或花叶海棠Malustiansitoria(Batal.)Schneid.的干燥叶。

优选地,所述的俄色叶的乙酸乙酯提取物是由下述方法制备:取俄色叶,粉碎,每次称取一定质量,提取3次,分别加20倍95%的乙醇加热回流提取,过滤,收集续滤液,减压回收,得乙醇部位浸膏;将乙醇部位的浸膏加水溶解,然后加入2倍的石油醚(60~90)萃取至石油醚无色,再加入2倍乙酸乙酯反复萃取直至乙酸乙酯溶液无色,合并乙酸乙酯溶液,减压回收得溶剂后得乙酸乙酯提取物。

具体实施方式
实验例1本发明俄色叶提取物的保肝作用
1.1实验材料
1.1.1药材收集
变叶海棠和花儿海棠均为2013年5月23日采自甘孜州炉霍县,赶黄草购自荷花池中药材市场,均由成都中医药大学李敏教授鉴定为蔷薇科植物变叶海棠Malustoringoides(Rehd.)Hughes.和花叶海棠Malustransitoria(Batal.)Schneid.的干燥叶,赶黄草为虎耳草科植物扯根菜PenthorumchinensePursh的干燥地上部分。
1.1.2实验药物
变叶海棠、花叶海棠及赶黄草提取物:以20倍水为提取溶剂,加热回流提取3次,每次时间1h,滤过,合并滤液,得水提物,用生理盐水溶解,配制成0.5g生药/ml备用。根据参考文献,实验时再用生理盐水将提取物分别设成3个剂量组(赶黄草1个剂量组),小鼠的低、中、高剂量组依次为:0.5g/kg·d、1g/kg·d、2g/kg·dg。
不同入药部位含量(%)
根皮苷、根皮素:为自制对照品,纯度在99.0%以上,根据参考文献,试验时以生理盐水分散将分别设成3个剂量组,小鼠的低、中、高剂量组依次为:5mg/kg·d、10mg/kg·d、15mg/kg·d。
联苯双酯:由浙江医药股份有限公司新昌制药厂提供,生产批号:110805,实验时以生理盐水分散作为阳性药对照组60mg/kg。
1.1.3实验动物
KM小鼠,清洁级,18~22g,雌雄各半,合格证号为:SCXK(川)2008-19,由四川省中医药科学院实验动物中心提供。
动物购回后饲养3天,观察各鼠的一般外观、行为活动、饮水进食情况、大小便性状,并进行称重,符合实验要求的动物供试验用。
1.1.4其他试剂及药品
实验试剂:四氯化碳(CCl4),成都市科龙试剂有限公司;橄榄油,成都市科龙试剂有限公司;谷丙转氨酶试剂盒(GPT/ALT,赖氏法),南京建成生物工程研究所,生产批号:20131106;谷草转氨酶试剂盒(GOT/AST,赖氏法),南京建成生物工程研究所,生产批号:20131107;超氧化物歧化酶试剂盒(SOD,WST法),南京建成生物工程研究所,生产批号:20131104;丙二醛试剂盒(MDA,TBA法),南京建成生物工程研究所,生产批号:20131108;总蛋白试剂盒(考马斯亮蓝法),南京建成生物工程研究所,生产批号:20131224。
实验仪器:恒温水浴锅,Thermo冷冻离心机,酶标定量测定仪(美国ThermoFisher公司VarioskanTM)。
1.2试验方法
1.2.1试验分组及处理
将小鼠随机分成空白对照组、模型对照组、联苯双酯对照组、赶黄草对照组、变叶海棠低、中、高剂量组、花叶海棠低、中、高剂量组、根皮素低、中、高剂量组、根皮苷低、中、高剂量组。共16个组,空白对照组和模型对照组灌胃生理盐水,其余组按照表1剂量分别给予相应药物,灌胃体积均为0.2ml/10g,连续灌胃给药4天,末次给药1h后,将CCl4溶于橄榄油中,配成0.6%的CCl4橄榄油溶液,按0.1ml/10g的剂量进行皮下注射建立小鼠急性肝损伤模型。空白对照组皮下注射等量的橄榄油,禁食不禁水,20h后摘眼球取血,分离血清,-4℃低温保存,用以测定血清生化指标(ALT、AST)。处死动物,剖取肝脏,称重,取0.5g肝组织,减碎,置于玻璃匀浆器中,加预冷的9被体积的0.9%生理盐水匀浆,2500rpm离心10min,上清液置-4℃低温保存备用,用以测定肝组织生化指标(SOD、MDA)。
1.2.2统计方法
运用PASW18.0软件提供的方差分析(ONE-WAYANOVA)进行处理。1.3试验结果
1.3.1俄色叶提取物对CCl4诱导肝损伤的作用
1.3.1.1俄色叶对CCl4引起的急性肝损伤小鼠血清转氨酶活性的影响
表1各组别对对CCl4引起的急性肝损伤小鼠血清转氨酶活性的影响(±s,n=10)
注:与模型对照组比较:**p<0.01,*p<0.05
与空白对照组比:##p<0.01,#p<0.05
与赶黄草对照组:△△p<0.01,△p<0.05
由表1可知,与空白对照组比较,模型对照组小鼠的血清ALT和AST水平均明显升高(p<0.01),说明CCl4所致小鼠急性肝损伤模型造模成功;与模型组相比较,俄色叶提取物的低、中、高剂量组均能明显降低CCl4致急性肝损伤小鼠的ALT、AST水平(p<0.01)。
与赶黄草对照组相比,变叶海棠组、花叶海棠组对CCl4致急性肝损伤小鼠的血清中ALT水平无显著性差异。变叶海棠高剂量组和花叶海棠低、中、高组均能明显降低对CCl4致急性肝损伤小鼠血清中AST水平(p<0.01)。
1.3.1.2俄色叶提取物对CCl4引起的急性肝损伤小鼠肝组织中SOD、MDA的影响
表2各组别对对CCl4引起的急性肝损伤小鼠肝组织中SOD活性及MDA含量的影响(±s,n=10)

由表2可知,与空白对照组比较,模型对照组小鼠的肝组织中SOD酶活力明显降低(p<0.01),MDA含量明显升高(p<0.01),说明CCl4所致小鼠急性肝损伤模型造模成功;与模型组相比较,俄色叶提取物的低、中、高剂量组均能明显升高CCl4致急性肝损伤小鼠肝组织中SOD酶活力(p<0.01),降低MDA含量(p<0.01)。
与赶黄草对照组相比,变叶海棠组和花叶海棠高、低剂量组均能显著地升高对CCl4致急性肝损伤小鼠的肝组织中SOD酶活力(p<0.01)。变叶海棠组、花叶海棠组均能明显升高对CCl4致急性肝损伤小鼠肝组织中MDA含量(p<0.01)。

实验例3本发明俄色叶乙醇提取物的保肝功效
一、药物制备和实验分组
变叶海棠大叶为2014年7月16日采自甘孜州炉霍县,变叶海棠嫩叶为2015年5月23日采自甘孜州炉霍县,均由由成都中医药大学李敏教授鉴定为蔷薇科植物变叶海棠Malustoringoides(Rehd.)Hughes.的干燥叶。
取干燥叶,粉碎,每次称取一定质量,提取3次,分别加20倍95%的乙醇加热回流提取,过滤,收集续滤液,减压回收,得乙醇部位浸膏。将乙醇部位的浸膏加水溶解,然后加入2倍的石油醚(60~90)萃取至石油醚无色,再加入2倍乙酸乙酯反复萃取直至乙酸乙酯溶液无色,合并乙酸乙酯溶液,减压回收得溶剂后得部位浸膏。
石油醚(60~90),购自成都市科龙化工试剂厂。
根皮苷、根皮素:为自制对照品,纯度在99.0%以上。
联苯双酯:由浙江医药股份有限公司新昌制药厂提供。
二、俄色叶提取物及其单体对CCl4体外诱导肝细胞株HL-7702HL-7702损伤的影响。
(1)HL-7702细胞的培养、传代、冻存、复苏、收集足够数量的细胞
人肝HL-7702细胞为贴壁细胞。将细胞接种于培养瓶中,加入含10%灭火胎牛血清(FBS)的RPMI1640培养基5ml,置37℃、5%CO2饱和湿度孵育箱内培养(传代、冻存、复苏)。镜下观察细胞贴壁生长情况。收集对数生长期细胞进行各项实验。
(2)CCl4损伤HL7702细胞模型的建立
正交试验设计选择二因素三水平确定细胞损伤模型。A-CCl4损伤时间(h);B-CCl4浓度(mmol/L)。根据L9(34)正交表进行试验设计。在96孔培养板中加入1×105个/mL细胞悬液,毎孔100μL。待细胞完全贴壁后,试验孔分别加入相应的含CCl4mmol/L无血清培养基,100μL,每组4孔,正常对照孔加入含0.1%DMSO的无血清培养液100μL。培养箱中孵育相应的时间后,毎孔加入5mg/mLMTT20μL,培养4h,吸弃上清液,毎孔加入150μLDMSO,摇床上振摇10min。在酶标仪490nm波长处测定各孔吸光度OD值,并按下式计算肝细胞的存活率:
存活率(%)=实验组A490nm/正常对照组A490nm*100%,以确定CCl4对肝细胞的损伤条件。

方差分析表
最终选择A3B2:10mmol/LCCl4作用六小时。
(3)生化仪检测各组肝细胞培养上清液ALT(GPT)、AST(GOT)泄露量、LDH渗出率
ALT、AST、LDH可以敏感地提示肝细胞损伤及程度,因此本试验中,选择ALT、AST、LDH作为肝细胞损伤程度的评定指标。
①收集对数生长期细胞,接种人肝HL-7702细胞于24孔培养板中,每孔加入300ul完全培基,使细胞密度为1*10^6个/孔,置37℃、5%C02培养箱内孵育12小时过夜,至接近长满。
②实验孔加入不同浓度药物200μL,每组设6个重复孔,并设正常对照组和模型组。给药24h,后加入10mmol/LCCl4(注意动作快,防止挥发)于培养板中共同干预细胞6小时。
③收集培养上清液,按试剂盒说明书用酶标仪测定ALT、AST泄露量及LDH渗出率。
所有数据均以X土S来表示,由SPSS19.0统计软件完成。先进行方差齐性检验,如符合方差齐性,组间比较釆用单因素方差分析,治疗前后两组间比较采用配对样本t检验方法,确定P<0.05为差异有显著性意义。

由上表可以看出,与模型组相比,本发明俄色叶的乙酸乙酯提取物,无论是变叶海棠大叶还是变叶海棠嫩叶的提取物,高、中、低剂量组均可以有效降低LDH、AST、ALT的含量,说明其可以有效保护肝细胞免受CCl4的损害。
将本发明变叶海棠大叶、变叶海棠嫩叶的提取物的高、中剂量组,与纯的根皮苷、根皮素的高、中剂量主要相比可知,本发明提取物在给药剂量更小的情况下,对肝细胞的保护与纯的根皮苷、根皮素却相当。
综上,本发明俄色叶及提取物用于保肝药效明确、可控,为临床提供了一种新的选择,且制备方法简单,成本低廉,工业应用前景良好。
Claims (10)
1.俄色叶及其提取物在制备保肝的药物或保健食品中的用途。
2.根据权利要求1所述的用途,其特征在于:所述的俄色叶来源于蔷薇科植物变叶海棠Malustoringoides(Rehd.)Hughes.或花叶海棠Malustiansitoria(Batal.)Schneid.的干燥叶芽及叶。
3.根据权利要求2所述的用途,其特征在于:所述俄色叶为变叶海棠大叶或者变叶海棠嫩叶。
4.根据权利要求1~3任意一项所述的用途,其特征在于:所述的俄色叶的提取物为水提取物。
5.根据权利要求1~3任意一项所述的用途,其特征在于:所述的俄色叶的提取物为乙酸乙酯提取物。
6.根据权利要求5所述的用途,其特征在于:所述的俄色叶的乙酸乙酯提取物是由下述方法制备:取俄色叶95%乙醇回流提取;乙醇部位浸膏加水分散,再依次用石油醚、乙酸乙酯萃取,取乙酸乙酯萃取溶剂,除去乙酸乙酯即得乙酸乙酯提取物。
7.根据权利要求6所述的用途,其特征在于:所述的俄色叶的乙酸乙酯提取物是由下述方法制备:取俄色叶,粉碎,每次称取一定质量,提取3次,分别加20倍95%的乙醇加热回流提取,过滤,收集续滤液,减压回收,得乙醇部位浸膏;将乙醇部位的浸膏加水溶解,然后加入2倍的石油醚(60~90)萃取至石油醚无色,再加入2倍乙酸乙酯反复萃取直至乙酸乙酯溶液无色,合并乙酸乙酯溶液,减压回收得溶剂后得乙酸乙酯提取物。
8.根据权利要求1~7任意一项所述的用途,其特征在于:所述保肝的药物或保健食品是治疗和/或预防化学性肝损伤的药物或保健食品。
9.一种俄色叶提取物,其特征在于:它是俄色叶的乙酸乙酯提取物,由下述方法制备:取俄色叶95%乙醇回流提取;乙醇部位浸膏加水分散,再依次用石油醚、乙酸乙酯萃取,取乙酸乙酯萃取溶剂,除去乙酸乙酯即得乙酸乙酯提取物。
