Abstract
本发明属于菌种制备技术领域,公开了一种动物双歧杆菌及其制备方法,所述动物双歧杆菌的保藏编号为:CGMCC No:12944。本发明的动物双歧杆菌NMC分离自健康百岁老人肠道,菌株具有极强的耐酸性,能抵抗胃酸顺利到达肠道实验证明菌株具有显著缓解便秘、改善肠道菌群、缓解溃疡性结肠炎、提高II性糖尿病动物胰岛功能及脂肪代谢能力、降低血中甘油三酯胆固醇水平等益生功能,是优良益生菌菌株;可以治疗便秘、肿瘤防治、保护肝脏、心血管疾病、改善乳糖消化、延缓衰老。

技术领域
本发明属于菌种制备技术领域,尤其涉及一种动物双歧杆菌及其制备方法。
背景技术
菌株(Strain)又称品系,表示同种微生物不同来源的纯种培养,从自然界中分离得到的每一个微生物纯培养都可称一个菌株。菌株是指从不同来源的标本中分离而得的相同菌种,叫菌株。如培养伤寒杆菌时可从血液中、骨髓中、粪便中、胆汁中进行分离培养,而以胆汁中者最为良好,称为标准株。益生菌是一类对宿主有益的活性微生物,是定植于人体肠道、生殖系统内,能产生确切健康功效从而改善宿主微生态平衡、发挥有益作用的活性有益微生物的总称。人体、动物体内有益的细菌或真菌主要有:酪酸梭菌、乳杆菌、双歧杆菌、放线菌、酵母菌等。目前世界上研究的功能最强大的产品主要是以上各类微生物组成的复合活性益生菌,其广泛应用于生物工程、工农业、食品安全以及生命健康领域。现有的益生菌菌株功能单一,益生功能差。

针对现有技术存在的问题,本发明提供了一种动物双歧杆菌及其制备方法。
本发明是这样实现的,一种动物双歧杆菌,所述动物双歧杆菌的保藏编号为:CGMCC No:12944。
本发明的另一目的在于提供一种所述动物双歧杆菌的制备方法,所述动物双歧杆菌的制备方法包括:
步骤一,将107量级的菌株接种于双歧杆菌培养基;
步骤二,在厌氧或好氧条件下,在15-38℃的温度下培养9-72小时;
步骤三,得到培养液。
本发明的优点及积极效果为:动物双歧杆菌NMC分离自健康百岁老人肠道,菌株具有极强的耐酸性,能抵抗胃酸顺利到达肠道实验证明菌株具有显著缓解便秘、改善肠道菌群、缓解溃疡性结肠炎、提高II性糖尿病动物胰岛功能及脂肪代谢能力、降低血中甘油三酯胆固醇水平等益生功能,是优良益生菌菌株;可以保护心血管疾病、改善乳糖消化、延缓衰老。

下面结合附图对本发明的应用原理作详细的描述。
本发明实施例提供的动物双歧杆菌的保藏编号为:CGMCC No:12944。保藏人谢小俐、西藏高原之宝牦牛乳业股份有限公司于2016年9月5日送至中国微生物菌种保藏管理委员会普通微生物中心,并登记入册,由2016年9月5日起保藏三十年;中国微生物菌种保藏管理委员会普通微生物中心于2016年9月5日检测,结果为存活,建议的分类命名为动物双歧杆菌;中国微生物菌种保藏管理委员会普通微生物中心地址为:北京市朝阳区北辰西路1号院3号,中国科学院研究所。
本发明的双歧杆菌分离自西藏牦牛奶中。
本发明提供了一种动物双歧杆菌,保藏编号为:CGMCC No:12944。分类命名:动物双歧杆菌,Bifidobacterium animalis。
本发明实施例提供的动物双歧杆菌NMC来自天湖纳木措,打尔古藏布河的注入处。当地人制作酸奶的方法是先燃起一堆牛粪火,把新鲜的牦牛奶倒在大锅里,慢慢把奶煮沸,然后倒入特质的木桶中,加少量的旧酸奶作为引子,再存放在30摄氏度至40摄氏度的地方,经过1-2天时间凝固,就能制成白嫩醇厚的牦牛酸奶。当地主妇把酸奶当做主食,身材曼妙,采集团队想要破解美丽的秘密,提取了纳木措动物双歧杆菌。

S101:将107量级的菌株接种于双歧杆菌培养基;
S102:在厌氧或好氧条件下,在15-38℃的温度下培养9-72小时;
S103:得到培养液。

本发明实施例提供的功能食品组合物含有如上所述的动物双歧杆菌的活菌体。
本发明实施例提供的功能食品组合物含有如上所述的动物双歧杆菌在制备保护心血管疾病、改善乳糖消化、延缓衰老的功能食品组合物中的应用
本发明可以进一步分离上述培养液中的双歧杆菌的活菌体,例如可以通过离心和/或过滤的方法实现。
实施例2
本发明提供了一种由如上所述的方法制备的功能食品组合物。
本发明提供了一种功能食品组合物,该功能食品组合物含有如上所述的动物双歧杆菌的活菌体。
在优选情况下,以功能食品组合物的总重量为基准,动物双歧杆菌的活菌体的含量为105-1010CFU/g,进一步优选为107-109CFU/g。
符合上述要求的功能食品组合物可以包括动物双歧杆菌的培养液(例如经该动物双歧杆菌发酵制得的发酵乳制品)、分离的动物双歧杆菌的活菌体等。

步骤二:在厌氧或好氧条件下,在15-38℃的温度下培养9-72小时;
步骤三:得到培养液。
3.一种由权利要求1所述动物双歧杆菌制备的益生菌保健品。
