背景技术
白芥子又名辣菜子。辛,温。归肺、胃。具有温肺化痰,利气散结,通络止痛的作用。白芥子在临床上应用广泛,常用于治疗胰腺癌、食癌、胃癌、肠癌、肺癌、肝癌、乳腺癌、膀胱癌、皮癌等属“痰湿证”者,也有部分止痛的功效。现代药理研究表明白芥子具有:细胞毒作用,诱导癌细胞凋亡、抑制癌基因表达、影响癌细胞代谢周期等癌药理作用。白芥子油对皮肤、黏膜有刺激用,能引起充血、灼痛,甚至起疱,内服过量可引起吐、腹痛、腹泻,因此用量不宜过大。
薄荷是中华常用中药之一。它是辛凉性发汗解热药,治流行性感冒、头疼、目赤、身热、咽喉、牙床肿痛等症。外用可治神经痛、皮肤瘙痒、皮疹和湿疹等。平常以薄荷代茶,清心明目。
决明子,中药名。是豆科植物决明或小决明的干燥成熟种子,以其有明目之功而名之。秋季采收成熟果实,晒干,打下种子,除去杂质。决明子味苦、甘、咸,性微寒,入肝、肾、大肠经;润肠通便,降脂明目,治疗便秘及高血脂,高血压。清肝明目,利水通便,有缓泻作用,降血压降血脂。
牡丹皮,中药名。为毛茛科植物牡丹干燥根皮。产于安徽、四川、河南、山东等地。苦、辛,微寒,归心、肝、肾经,具有清热凉血、活血化淤、退虚热等功效。
发明内容
本发明的目的在于提供一种以抗癌中草药为原料的除痰散结药物制剂及其制备方法,将白芥子、薄荷、决明子和牡丹皮共同提取制备得到的中药组合物不仅消除了白芥子的刺激作用,还能对白芥子起到协同作用,显著提高白芥子的除痰散结功效。
为实现上述目的,本发明采用如下技术方案:
一种以抗癌中草药为原料的除痰散结药物制剂,其原料按重量份计包括:白芥子10-20份、薄荷9-10份、决明子3-10份、牡丹皮3-10份。
优选的,其原料按重量份计包括:白芥子15份、薄荷10份、决明子5份、牡丹皮5份。
制备方法包括以下步骤:
1)首先将白芥子、薄荷、决明子和牡丹皮按重量配比称重,粉碎后,过10-30目筛,得到粗粉,然后将上述粗粉置于体积浓度为70-90%的乙醇中,回流提取3-5小时后,过滤,得滤液和滤渣;
2)向上述滤液中加入相当于其体积1-6倍量的乙酸乙酯,萃取,得到有机相,重复萃取一次,合并有机相并将其减压浓缩,回收乙酸乙酯和乙醇后得到在25℃时相对密度为1.20-1.35的稠膏1;
3)将步骤1)提取后的滤渣置于纯水中,在功率为800-1000W,频率为40-60Hz的条件下超声处理10-30分钟,然后煎煮1-2次,每次煎煮1-2小时,合并煎煮后的煎液并过滤,滤液浓缩后得到在25℃时相对密度为1.05-1.2的稠膏2;
4)向稠膏2中加入相当于其重量0.8-1.2倍量的体积浓度为85-95%的乙醇,边加入边搅拌,静置沉淀8-10小时后过滤,得到滤液,将滤液进行浓缩、干燥后得到在80℃时相对密度为1.28-1.4的稠膏3;
5)合并稠膏1和稠膏3后即得到中药组合物的提取物;
6)将步骤5)制得的中药组合物的提取物制备成临床上常用的剂型。
本发明的显著优点在于:本发明的以抗癌中草药为原料的除痰散结药物制剂,采用白芥子、薄荷、决明子和牡丹皮为主要原料,白芥子可以用于胰腺癌、食癌、胃癌、肠癌、肺癌、肝癌、乳腺癌、膀胱癌、皮癌等癌症的治疗,同时其具有除痰散结功效,白芥子苷本身无刺激作用,但它遇水后经芥子酶的水解作用生成白芥子油,对皮肤、黏膜有刺激用,能引起充血、灼痛,甚至起疱,内服过量可引起吐、腹痛、腹泻,本发明人发现,将白芥子、薄荷、决明子和牡丹皮共同提取制备得到的中药组合物不仅消除了白芥子的刺激作用,还能对白芥子起到协同作用,显著提高白芥子的除痰散结功效。本发明的除痰散结药物制剂可以用于治疗多种癌瘤中属于痰浊邪毒凝结,或痰热、热毒,或寒痰凝滞于肌腠、经络、脏腑之中者,本型癌瘤患者临床常伴见湿困痰多,苔浊脉滑等痰浊凝结的证候。
具体实施方式
为进一步公开而不是限制本发明,以下结合实例对本发明作进一步的详细说明。
实施例1
一种以抗癌中草药为原料的除痰散结药物制剂,其原料按重量份计包括:白芥子15份、薄荷10份、决明子5份、牡丹皮5份。
制备方法包括以下步骤:
1)首先将白芥子、薄荷、决明子和牡丹皮按重量配比称重,粉碎后,过20目筛,得到粗粉,然后将上述粗粉置于体积浓度为80%的乙醇中,回流提取4小时后,过滤,得滤液和滤渣;
2)向上述滤液中加入相当于其体积3倍量的乙酸乙酯,萃取,得到有机相,重复萃取一次,合并有机相并将其减压浓缩,回收乙酸乙酯和乙醇后得到在25℃时相对密度为1.25的稠膏1;
3)将步骤1)提取后的滤渣置于纯水中,在功率为900W,频率为50Hz的条件下超声处理20分钟,然后煎煮2次,每次煎煮2小时,合并煎煮后的煎液并过滤,滤液浓缩后得到在25℃时相对密度为1.1的稠膏2;
4)向稠膏2中加入相当于其重量1倍量的体积浓度为90%的乙醇,边加入边搅拌,静置沉淀9小时后过滤,得到滤液,将滤液进行浓缩、干燥后得到在80℃时相对密度为1.35的稠膏3;
5)合并稠膏1和稠膏3后即得到中药组合物的提取物;
6)将步骤5)制得的中药组合物的提取物制备成临床上常用的剂型。
实施例2
一种以抗癌中草药为原料的除痰散结药物制剂,其原料按重量份计包括:白芥子10份、薄荷9份、决明子3份、牡丹皮3份。
制备方法包括以下步骤:
1)首先将白芥子、薄荷、决明子和牡丹皮按重量配比称重,粉碎后,过10目筛,得到粗粉,然后将上述粗粉置于体积浓度为70%的乙醇中,回流提取3小时后,过滤,得滤液和滤渣;
2)向上述滤液中加入相当于其体积1倍量的乙酸乙酯,萃取,得到有机相,重复萃取一次,合并有机相并将其减压浓缩,回收乙酸乙酯和乙醇后得到在25℃时相对密度为1.20的稠膏1;
3)将步骤1)提取后的滤渣置于纯水中,在功率为800W,频率为40Hz的条件下超声处理10分钟,然后煎煮1次,每次煎煮1小时,合并煎煮后的煎液并过滤,滤液浓缩后得到在25℃时相对密度为1.05的稠膏2;
4)向稠膏2中加入相当于其重量0.8倍量的体积浓度为85%的乙醇,边加入边搅拌,静置沉淀8小时后过滤,得到滤液,将滤液进行浓缩、干燥后得到在80℃时相对密度为1.28的稠膏3;
5)合并稠膏1和稠膏3后即得到中药组合物的提取物;
6)将步骤5)制得的中药组合物的提取物制备成临床上常用的剂型。
实施例3
一种以抗癌中草药为原料的除痰散结药物制剂,其原料按重量份计包括:白芥子20份、薄荷10份、决明子10份、牡丹皮10份。
制备方法包括以下步骤:
1)首先将白芥子、薄荷、决明子和牡丹皮按重量配比称重,粉碎后,过30目筛,得到粗粉,然后将上述粗粉置于体积浓度为90%的乙醇中,回流提取5小时后,过滤,得滤液和滤渣;
2)向上述滤液中加入相当于其体积6倍量的乙酸乙酯,萃取,得到有机相,重复萃取一次,合并有机相并将其减压浓缩,回收乙酸乙酯和乙醇后得到在25℃时相对密度为1.35的稠膏1;
3)将步骤1)提取后的滤渣置于纯水中,在功率为1000W,频率为60Hz的条件下超声处理30分钟,然后煎煮2次,每次煎煮2小时,合并煎煮后的煎液并过滤,滤液浓缩后得到在25℃时相对密度为1.2的稠膏2;
4)向稠膏2中加入相当于其重量1.2倍量的体积浓度为95%的乙醇,边加入边搅拌,静置沉淀10小时后过滤,得到滤液,将滤液进行浓缩、干燥后得到在80℃时相对密度为1.4的稠膏3;
5)合并稠膏1和稠膏3后即得到中药组合物的提取物;
6)将步骤5)制得的中药组合物的提取物制备成临床上常用的剂型。
应用实施例
李某,男,55 岁,肺癌患者同时伴有咳嗽,痰多,咽喉肿痛,服用本发明的除痰散结药物制剂1天后,痰明显减少,咽喉肿痛症状明显减轻,继续服用1天后,咳嗽,痰多,咽喉肿痛的病情痊愈。
刘某,女,69 岁,胃癌患者同时伴有气喘,咽喉肿痛,肺热痰郁,咳嗽不止,服用本发明的除痰散结药物制剂两天后,咽喉肿痛和咳嗽不止症状明显减轻,继续服用1天后,气喘,咽喉肿痛,肺热痰郁,咳嗽不止的病情痊愈。
林某,男,45 岁,肝癌患者同时伴有咳嗽,痰多,痰黄稠粘,咽喉肿痛,服用本发明的除痰散结药物制剂两天后,咳嗽,痰多,痰黄稠粘,咽喉肿痛的症状减缓,继续服用1后,咳嗽,痰多,痰黄稠粘,咽喉肿痛的病情痊愈。

Claims (3)
1.一种以抗癌中草药为原料的除痰散结药物制剂,其特征在于,其原料按重量份计包括:白芥子10-20份、薄荷9-10份、决明子3-10份、牡丹皮3-10份。
2.根据权利要求1所述的一种以抗癌中草药为原料的除痰散结药物制剂,其特征在于,其原料按重量份计包括:白芥子15份、薄荷10份、决明子5份、牡丹皮5份。
3.一种制备如权利要求1或2所述的以抗癌中草药为原料的除痰散结药物制剂的制备方法,其特征在于,包括以下步骤:
1)首先将白芥子、薄荷、决明子和牡丹皮按重量配比称重,粉碎后,过10-30目筛,得到粗粉,然后将上述粗粉置于体积浓度为70-90%的乙醇中,回流提取3-5小时后,过滤,得滤液和滤渣;
2)向上述滤液中加入相当于其体积1-6倍量的乙酸乙酯,萃取,得到有机相,重复萃取一次,合并有机相并将其减压浓缩,回收乙酸乙酯和乙醇后得到在25℃时相对密度为1.20-1.35的稠膏1;
3)将步骤1)提取后的滤渣置于纯水中,在功率为800-1000W,频率为40-60Hz的条件下超声处理10-30分钟,然后煎煮1-2次,每次煎煮1-2小时,合并煎煮后的煎液并过滤,滤液浓缩后得到在25℃时相对密度为1.05-1.2的稠膏2;
4)向稠膏2中加入相当于其重量0.8-1.2倍量的体积浓度为85-95%的乙醇,边加入边搅拌,静置沉淀8-10小时后过滤,得到滤液,将滤液进行浓缩、干燥后得到在80℃时相对密度为1.28-1.4的稠膏3;
5)合并稠膏1和稠膏3后即得到中药组合物的提取物;
6)将步骤5)制得的中药组合物的提取物制备成临床上常用的剂型。
