糖尿病(diabetes)是以高血糖为特征的全面代谢紊乱性疾病,与冠心病、癌症并称为人类生命的三大杀手,严重影响人们的身心健康。随着人们生活水平的不断提高及饮食的不规律,我国糖尿病患病率逐年升高,并且呈现年轻化的趋势,糖尿病已成为重大的公共卫生问题。

雪莲培养物1.6-3.4份、人参7-27份、苦瓜50-80份、玉米须36-74份、茯苓28-52份、山楂12-18份、枸杞15-21份、平卧菊三七35-50份、抗性糊精16-30份。
雪莲培养物:是指在无菌条件下,将雪莲离体的植物器官、组织、细胞、胚胎或原生质体培养在人工配制的培养基上,在人工控制的条件下进行培养,使其组织或细胞增值进而按照需要进行培养的技术;或使其脱分化产生愈伤组织,并再逐步分化出器官并长成完整的植株而生产的培养物。
平卧菊三七:为菊科菊三七属平卧菊三七的干燥根茎,其味甘淡,性平,可通经活络、消炎止咳、散瘀消肿、活血生肌。做菜食用或药用,具有降血脂、降血压、降血糖作用,可以补钙、抗流感和乙肝病毒、抗感冒、提高人体免疫力等功效。

雪莲培养物2-3份、人参9-25份、苦瓜55-75份、玉米须45-65份、茯苓35-45份、山楂15份、枸杞18份、平卧菊三七45份、抗性糊精20-25份。

其中,所述雪莲培养物的蛋白质≥20%、总黄酮≥7%、水分≤10%、灰分≤10%。
其中,所述雪莲培养物购自大连普瑞康生物技术有限公司。
其中,它是由所述原料的原生药粉,或水或有机溶剂提取物为活性成分,加上药物、保健品或食品领域可接受的辅料或者辅助性成分制备成常用的口服制剂。
其中,所述口服制剂是口服液、膏剂、颗粒剂、粉剂、片剂、胶囊剂、饮料。
本发明还提供了上述组合物在制备具有降血糖作用的药品、保健食品及高血糖患者食品中的用途。
其中,所述药品为治疗糖尿病的药物;
其中,所述糖尿病为1型糖尿病和/或2型糖尿病。
本发明提供的组合物可以显著降低糖尿病及高血糖患者的血糖,而且可有效降低糖尿病患者的胰岛素用量,治疗效果显著;另外,本发明组合物适合作为糖尿病及高血糖患者的药膳食品,且口感佳,应用前景良好。

具体实施方式
下面以实施例作进一步说明,但本发明不局限于这些实施例。
雪莲培养物:大连普瑞康生物技术有限公司;
鲜品质量要求:性状:紫红色团状颗粒;蛋白质≥1%;总黄酮≥0.4%;水分≤96%;灰分≤1%;
干品质量要求:性状:紫灰色粉末;蛋白质≥20%;总黄酮≥7%;水分≤10%;灰分≤10%。
蛋白酶:购自湖南奥驰生物科技有限公司,技术指标:10万活力。
实施例1本发明组合物的制备
原料:雪莲培养物(干品)0.2kg、人参0.9kg、苦瓜5.5kg、玉米须6.5kg、茯苓4.5kg、山楂1.5kg、枸杞1.8kg、平卧菊三七4.5kg、抗性糊精2kg、木糖醇0.02kg。
制备工艺如下:
(一)人参提取
1.前处理:如果购进的是原药,还应该对其拣选,杂质及虫蛀霉变品,除去非食用部位,然后进行切制。
2.提取、过滤:将人参投于多功能提取罐内。第一次加水20倍量,65℃温浸30分钟,打开蒸汽阀门,调节提取罐内压力不得超过0.2Mpa,打开蒸汽升温至微沸后计时,保持95-100℃提取1.5小时,先过20目筛,在100目滤布过滤,放液于酶解罐内;第二次加水10倍量,按第一次的提取方法再提一次,放液于酶解罐内,至两次滤液合并于酶解罐内,加入质量百分比为3‰(w/w)的木瓜蛋白酶,在30-40摄氏度或常温下酶解6小时,然后高速离心,离心液通过陶瓷膜过滤,过滤后的液体及杂质反复离心。
3.浓缩:将滤液泵入二效浓缩器中浓缩至相对密度为1.02(65~70℃)。一效浓缩温度60~75℃,真空度0.03~0.05Mpa;二效浓缩温度50~55℃,真空度0.03Mpa-0.07Mpa,蒸汽压力0.06~0.10Mpa。浸膏收率应为100-120%,浸膏备用。
(二)苦瓜提取
1.前处理:如果购进的是原药,还应该对其拣选,杂质及虫蛀霉变品,除去非食用部位,然后进行切制。
2.提取、过滤:将人参投于多功能提取罐内。第一次加水10倍量,打开蒸汽阀门,调节提取罐内压力不得超过0.2Mpa,打开蒸汽升温至微沸后计时,保持100℃提取1小时,先过20目筛,在100目滤布过滤,放液于贮液罐内;第二次加水8倍量,按第一次的提取方法再提一次,放液于贮液罐内,至两次滤液合并于贮液罐内,高速离心,离心液通过陶瓷膜过滤,过滤后的液体及杂质反复离心。
3.浓缩:将滤液泵入二效浓缩器中浓缩至相对密度为1.01(65~70℃)。一效浓缩温度60~75℃,真空度0.03~0.05Mpa;二效浓缩温度50~55℃,真空度0.03Mpa-0.07Mpa,蒸汽压力0.06~0.10Mpa。浸膏收率应为100-120%,浸膏备用。
(三)雪莲培养物(干品)处理
1.将雪莲培养物(干品)置于酶解罐中,加入等重量的水,搅拌均匀,加入质量百分比为3‰(w/w)的蛋白酶,常温下酶解12小时。
2.浓缩:酶解液通过陶瓷膜过滤,将滤液泵入二效浓缩器中浓缩至相对密度为1.01(65~70℃)。一效浓缩温度60~75℃,真空度0.03~0.05Mpa;二效浓缩温度50~55℃,真空度0.03Mpa-0.07Mpa,蒸汽压力0.06~0.10Mpa。浸膏收率约为80%,浸膏备用。
(四)混合
将上述三种浸膏边混合边搅拌30分钟,同时加入定量的抗性糊精,得到的浸膏备用。
(五)喷雾干燥
蒸汽压力控制在0.4MPa-0.5MPa之间,将喷雾干燥机进风温度设置为150℃,出风温度设置为85℃,将第四步得到的浸膏送入喷雾干燥机中,进料速度设置在3.5—4.0Hz,塔体压差保持在-100—-200Pa,收集粉子,粉子收率为10.0-12.5%,人参总皂苷含量应≥0.1%,装入包装袋,不超过袋容积2/3,用真空机抽真空封口,称量贴签,备用。
(六)中药原料的提取
1.前处理:如果购进的是原药,还应该对其拣选,杂质及虫蛀霉变品,除去非食用部位。
2.提取:将玉米须、茯苓、山楂、枸杞、平卧菊三七投于多功能提取罐内。第一次加水10倍量,打开蒸汽阀门,调节提取罐内压力不得超过0.2Mpa,打开蒸汽升温至微沸后计时,保持100℃提取1小时,先过20目筛,在100目滤布过滤,放液于贮液罐内;第二次加水8倍量,按第一次的提取方法再提一次,放液于贮液罐内,至两次滤液合并于贮液罐内。
3.浓缩:将滤液泵入二效浓缩器中浓缩至相对密度为1.01(65~70℃)。一效浓缩温度60~75℃,真空度0.03~0.05Mpa;二效浓缩温度50~55℃,真空度0.03Mpa-0.07Mpa,蒸汽压力0.06~0.10Mpa。浸膏收率为65-70%,浸膏备用。
(七)将中药原料浓缩液送入喷雾干燥机。蒸汽压力控制在0.4MPa-0.5MPa之间,将喷雾干燥机进风温度设置为150℃,出风温度设置为180℃,进料速度设置在3.5—4.0Hz,塔体压差保持在-100—-200Pa,收集粉子,收率为10-15%,粉子装入包装袋,不超过袋容积2/3,用真空机抽真空封口,称量贴签,备用。
(八)混合、调配:将配方投料量的喷雾粉子投入到配制缸中加入等重量纯化水,蒸汽加热至50℃,边加热边搅拌至均匀,再加入木糖醇,然后缓缓加入纯化水定容1000L,调pH至6.5。
(九)灌装:20ml/瓶
(十)灭菌:108℃恒温灭菌30分钟。
(十一)包装。
实施例2本发明组合物的制备
原料:雪莲培养物(干品)0.3kg、人参2.5kg、苦瓜7.5kg、玉米须4.5kg、茯苓3.5kg、山楂1.5kg、枸杞1.8kg、平卧菊三七4.5kg、抗性糊精2.5kg、木糖醇0.02kg。
制备工艺:同实施例1的制备。
以下通过试验例具体说明本发明的有益效果。
试验例1本发明药物或保健食品的疗效考察
1、取实施例1制备的组合物,选择20例确诊糖尿病患者,进行效果验证。
具体方法:在饭后10分钟内食用,每天3次,每次20ml(折合固形物6g),然后用快速血糖仪检测。
结果见表1。
表1数据统计表(血糖值单位:毫摩尔/升)
由表1可知,统计数据显示:20例人群中,有13例餐后血糖降低比例超过10%,视为有效降低血糖;11例空腹血糖降低比例超过10%,视为有效降低血糖;有1例减少胰岛素用量,治疗效果显著。
2、取实施例2制备的组合物,选择20例确诊糖尿病患者,进行效果验证。
具体方法:在饭后10分钟内食用,每天3次,每次20ml(折合固形物6g),然后用快速血糖仪检测。
结果见表2。

由表2可知,统计数据显示:20例人群中,有18例餐后血糖降低比例超过10%,视为有效降低血糖;14例空腹血糖降低比例超过10%,视为有效降低血糖;有17例减少胰岛素用量,治疗效果显著。
综上,本发明提供的组合物可以显著降低糖尿病及高血糖患者的血糖,而且可有效降低糖尿病患者的胰岛素用量,治疗效果显著;另外,本发明组合物适合作为糖尿病及高血糖患者的药膳食品,且口感佳,应用前景良好。

1.一种具有降血糖作用的药物、保健食品或食品组合物,其特征在于:它是由含有下述重量配比的原料制备而成:
雪莲培养物1.6-3.4份、人参7-27份、苦瓜50-80份、玉米须36-74份、茯苓28-52份、山楂12-18份、枸杞15-21份、平卧菊三七35-50份、抗性糊精16-30份。
2.根据权利要求1所述的组合物,其特征在于:它是由下述重量配比的原料制备而成:
雪莲培养物2-3份、人参9-25份、苦瓜55-75份、玉米须45-65份、茯苓35-45份、山楂15份、枸杞18份、平卧菊三七45份、抗性糊精20-25份。

9.权利要求1-8任意一项所述组合物在制备具有降血糖作用的药品、保健食品及高血糖患者食品中的用途。
10.根据权利要求9所述的用途,其特征在于:所述药品为治疗糖尿病的药物;
