技术领域
本发明属于保健口服液领域,具体涉及一种雪莲培养物口服液及其制备方法。
背景技术
由于雪莲具有很好的药效价值,在市场上使用越来越广泛,但由于其自然生长周期长,以及人们的过度开发,导致野生雪莲资源的匮乏,而人工种植虽然可短暂缓解这种情况,但是人工种植的环境要求苛刻,成活率低,而且药效成分的含量不理想。随着雪莲培养物生物技术的提高,雪莲培养物具有与野生天山雪莲相似的成分和药效,而且雪莲培养物中各成分含量均高于野生天山雪莲,更适用于保健品市场。
芒草是快生植物,生长环境要求不高而且成本低,目前的利用领域比较窄,多是作为能源作物,用于生产生物燃料或合成一些化学产品,利用率不高,由于芒草中也含有多种氨基酸和黄酮,而黄酮可阻止氧化,改善血液循环,具有一定的药效成分,但是由于芒草的青味以及其中黄酮在水中溶解度较差,没有很好的提取方法,因此目前芒草还未广泛地应用在食品或保健品领域。荷叶中含有荷叶碱和荷叶黄酮,可以在食品及保健品领域加以利用,现有技术中运用在食品行业提取黄酮的方法都不是太理想。
发明内容
为了克服上述技术问题,本发明提供了一种雪莲培养物口服液及其制备方法,通过芒草、荷叶提取液,并混合雪莲培养物,阿拉伯糖,低聚玉米肽,决明子,起到降脂、间接阻糖,进而达到降压的实际功效。在调节三高的同时,还可以改善肠道功能,提高机体免疫力,从而到达标本兼治的最终效果。
目前芒草基本还没有应用于食品领域,因此对其的制备方法的技术基本没有,而针对荷叶的提取液已有一些方法,但是基本是采用在热水中多次提取的方法,由于黄酮在水中的溶解度较差,因此提取液中黄酮的提取效果不理想。针对这个问题,本发明在热水中加入磷酸氢二钾,形成弱碱性溶液环境并辅以超声提取可得到较好的提取效果。
本发明所采用的技术方案是:一种雪莲培养物口服液,由下列按重量份计算的原料制备而成:雪莲培养物粉20-30份、芒草18-22份、荷叶10-12份、磷酸氢二钾2-4份、低聚玉米肽5-12份、L阿拉伯糖1-3份、决明子速溶粉5-10份、树莓浓缩汁2-5份、聚葡萄糖1-2份、木糖醇1-5份和黄原胶2-5份。
雪莲培养物口服液的制备方法包括以下步骤:
(1)将芒草超声清洗后切小块,隔水蒸2小时,然后进行榨汁,过滤得到芒草汁和芒草滤渣;将荷叶超声清洗后切小块,榨汁,过滤得到荷叶汁和荷叶滤渣;
(2)将芒草滤渣烘箱中干燥1-2h,用气流粉碎机粉碎投入浸提罐中,加入10-15倍的饮用纯化水,45℃下浸泡1h,加入磷酸氢二钾总质量的70%,在85-90℃下超声辅助提取40min,滤膜过滤;滤渣第二次加入5-10倍的饮用纯化水,加入磷酸氢二钾总质量的20%,在85-90℃下超声辅助提取40min,滤膜过滤;将两次滤液合并,减压浓缩至原来体积的1/2,即得芒草提取液;
(3)向荷叶滤渣中加入5倍质量的体积分数为75%的乙醇,80℃下搅拌30min后,微波辅助回流提取15min,继续超声辅助80℃回流1h,减压抽滤得第一次滤渣和第一次滤液,向第一次滤渣中加入3倍质量的体积分数为75%的乙醇,80℃下微波辅助回流提取15min,继续超声辅助80℃回流1h,减压抽滤得第二次滤渣和第二次滤液;将第一次滤液和第二次滤液合并减压浓缩回收乙醇,得到浓缩液,加入2倍质量的水,搅拌10min后静置10min,滤布过滤,即得荷叶提取液;
(4)将雪莲培养物粉先经超微粉碎机粉碎,加入相同重量的引用纯化水,在50℃下搅拌20min,保温的状态下加入磷酸氢二钾总质量的10%,超声辅助提取1h,滤膜过滤,得到第一次雪莲滤液和滤渣,在滤渣中加入相同重量的饮用纯化水保持在50℃,然后加入0.5-1%的质量比2:1的纤维素酶和果胶酶,酶解2h,升至90℃灭酶,滤膜过滤,得到第二次雪莲滤液,将两次雪莲滤液合并得到雪莲液;
(5)将芒草汁、荷叶汁、芒草提取液、荷叶提取液、雪莲液、低聚玉米肽、L阿拉伯糖、决明子速溶粉加入到混合提取液中,在40℃下搅拌混合2h,然后再加入树莓浓缩汁、聚葡萄糖、木糖醇和黄原胶搅拌混合30min;然后将混合料液抽入管式微滤膜中,调节压力至0.2MPa,收集通过管式微滤膜的滤液;
(6)90℃杀菌,冷却至55℃,进入高压均质机均质,均质压力为25MPa,均质后的料液即雪莲培养物口服液。
芒草先蒸2h,去除其青味,产生香气,然后榨汁,得到的芒草汁味道更好。芒草和荷叶榨汁后,虽然很多营养成分都在汁里了,但是芒草渣和荷叶渣中残存有黄酮,为了更充分的利用,把芒草渣和荷叶渣一起进行提取。
在对芒草渣、荷叶渣进行提取时,45℃下先浸泡1h,然后加入磷酸氢二钾,在85-90℃下超声辅助提取40min,由于加入磷酸氢二钾使得溶液呈弱碱性以及超声辅助提取,提取效率较之单纯热水的提取效率高,因此首次提取只需加10-15倍的水,相对于现有技术,加水量大大减少,降低了能耗。
将雪莲培养物粉先进行提取,然后对雪莲滤渣进行酶解,能尽量多的得到雪莲中的有效成分,而且果胶酶的加入使得雪莲液较为澄清,成色较好;最后将芒草汁、荷叶汁、雪莲液、低聚玉米肽、L阿拉伯糖、决明子速溶粉加入到混合提取液中,在40℃下搅拌混合2h,各组分能更好的相互融合,药效更好,同时得到的口服液的口感更加细腻,而且低聚玉米肽、L阿拉伯糖、决明子速溶粉都可以很好的溶解于水中,因此本发明得到的雪莲培养物口服液毫无颗粒感。
雪莲培养物具有显著的调血脂功能,阿拉伯糖可抑制蔗糖的吸收,低聚玉米肽降压效果明显、决明子对于原发性的高血压有很好的疗效
本发明的有益效果:高效提取芒草、荷叶中黄酮,结合雪莲培养物、低聚玉米肽、L阿拉伯糖、决明子速溶粉等成分,能起到降脂、间接阻糖,进而达到降压的实际功效。本发明对原料的有效成分进行最大的利用,而且在调节三高的同时,还可以改善肠道功能,提高机体免疫力,口感好而细腻,毫无颗粒感,营养和保健效果双保证,更适用于工业化生产。
具体实施方式
实施例1
一种雪莲培养物口服液,由下列按重量份计算的原料制备而成:雪莲培养物粉20份、芒草22份、荷叶12份、磷酸氢二钾4份、低聚玉米肽12份、L阿拉伯糖3份、决明子速溶粉10份、树莓浓缩汁5份、聚葡萄糖2份、木糖醇5份和黄原胶5份。
制备工艺为:(1)将22份芒草超声清洗后切小块,隔水蒸2小时,然后进行榨汁,过滤得到芒草汁和芒草滤渣;将12份荷叶超声清洗后切小块,榨汁,过滤得到荷叶汁和荷叶滤渣;
(2)将芒草滤渣烘箱中干燥1h,用气流粉碎机粉碎投入浸提罐中,加入12倍的饮用纯化水,浸泡1h,加入2.8份磷酸氢二钾,在88℃下超声辅助提取40min,滤膜过滤;滤渣第二次加入7倍的饮用纯化水,加入0.8份磷酸氢二钾,在88℃下超声辅助提取40min,滤膜过滤;将两次滤液合并,减压浓缩至原来体积的1/2,即得芒草提取液;
(3)向荷叶滤渣中加入5倍质量的体积分数为75%的乙醇,80℃下搅拌30min后,微波辅助回流提取15min,继续超声辅助80℃回流1h,减压抽滤得第一次滤渣和第一次滤液,向第一次滤渣中加入3倍质量的体积分数为75%的乙醇,80℃下微波辅助回流提取15min,继续超声辅助80℃回流1h,减压抽滤得第二次滤渣和第二次滤液;将第一次滤液和第二次滤液合并减压浓缩回收乙醇,得到浓缩液,加入2倍质量的水,搅拌10min后静置10min,滤布过滤,即得荷叶提取液;
(4)将20份雪莲培养物粉先经超微粉碎机粉碎,加入20份的饮用纯化水,在50℃下搅拌20min,保温的状态下加入0.4份磷酸氢二钾质量,超声辅助提取1h,滤膜过滤,得到第一次雪莲滤液和滤渣,在滤渣中加入相同重量的饮用纯化水保持在50℃,然后加入滤渣重量0.8%的质量比2:1的纤维素酶和果胶酶,酶解2h,升至90℃灭酶,滤膜过滤,得到第二次雪莲滤液,将两次雪莲滤液合并得到雪莲液;
(5)将芒草汁、荷叶汁、雪莲液、低聚玉米肽、L阿拉伯糖、决明子速溶粉加入到混合提取液中,在40℃下搅拌混合2h,然后再加入树莓浓缩汁、聚葡萄糖、木糖醇和黄原胶搅拌混合30min;然后将混合料液抽入管式微滤膜中,调节压力至0.2MPa,收集通过管式微滤膜的滤液;
(6)90℃杀菌,冷却至55℃,进入高压均质机均质,均质压力为25MPa,均质后的料液及时灌装于无菌的干净干燥瓶中(10ml/支),加盖密闭,100℃流通蒸汽灭菌30min,制得雪莲培养物口服液。
实施例2
一种雪莲培养物口服液,由下列按重量份计算的原料制备而成:雪莲培养物粉26份、芒草20份、荷叶11份、磷酸氢二钾3份、低聚玉米肽8份、L阿拉伯糖2份、决明子速溶粉8份、树莓浓缩汁4份、聚葡萄糖1份、木糖醇3份和黄原胶3份。
制备工艺为:(1)将20份芒草超声清洗后切小块,隔水蒸2小时,然后进行榨汁,过滤得到芒草汁和芒草滤渣;将11份荷叶超声清洗后切小块,榨汁,过滤得到荷叶汁和荷叶滤渣;
(2)将芒草滤渣烘箱中干燥2h,用气流粉碎机粉碎投入浸提罐中,加入15倍的饮用纯化水,浸泡1h,加入2.1份磷酸氢二钾,在90℃下超声辅助提取40min,滤膜过滤;滤渣第二次加入10倍的饮用纯化水,加入0.6份磷酸氢二钾,在90℃下超声辅助提取40min,滤膜过滤;将两次滤液合并,减压浓缩至原来体积的1/2,即得芒草提取液;
(3)向荷叶滤渣中加入5倍质量的体积分数为75%的乙醇,80℃下搅拌30min后,微波辅助回流提取15min,继续超声辅助80℃回流1h,减压抽滤得第一次滤渣和第一次滤液,向第一次滤渣中加入3倍质量的体积分数为75%的乙醇,80℃下微波辅助回流提取15min,继续超声辅助80℃回流1h,减压抽滤得第二次滤渣和第二次滤液;将第一次滤液和第二次滤液合并减压浓缩回收乙醇,得到浓缩液,加入2倍质量的水,搅拌10min后静置10min,滤布过滤,即得荷叶提取液;
(3)将26份雪莲培养物粉先经超微粉碎机粉碎,加入26份的饮用纯化水,在50℃下搅拌20min,保温的状态下加入0.3份磷酸氢二钾质量,超声辅助提取1h,滤膜过滤,得到第一次雪莲滤液和滤渣,在滤渣中加入相同重量的饮用纯化水保持在50℃,然后加入滤渣重量1%的质量比2:1的纤维素酶和果胶酶,酶解2h,升至90℃灭酶,滤膜过滤,得到第二次雪莲滤液,将两次雪莲滤液合并得到雪莲液;
(4)将芒草汁、荷叶汁、雪莲液、低聚玉米肽、L阿拉伯糖、决明子速溶粉加入到混合提取液中,在40℃下搅拌混合2h,然后再加入树莓浓缩汁、聚葡萄糖、木糖醇和黄原胶搅拌混合30min;然后将混合料液抽入管式微滤膜中,调节压力至0.2MPa,收集通过管式微滤膜的滤液;
(5)90℃杀菌,冷却至55℃,进入高压均质机均质,均质压力为25MPa,均质后的料液及时灌装于无菌的干净干燥瓶中(10ml/支),加盖密闭,100℃流通蒸汽灭菌30min,制得雪莲培养物口服液。
实施例3
一种雪莲培养物口服液,由下列按重量份计算的原料制备而成:雪莲培养物粉30份、芒草18份、荷叶10份、磷酸氢二钾2份、低聚玉米肽6份、L阿拉伯糖1份、决明子速溶粉6份、树莓浓缩汁3份、聚葡萄糖1份、木糖醇2份和黄原胶2份。
制备工艺为:(1)18份芒草超声清洗后切小块,隔水蒸2小时,然后进行榨汁,过滤得到芒草汁和芒草滤渣;将10份荷叶超声清洗后切小块,榨汁,过滤得到荷叶汁和荷叶滤渣;
(2)将芒草滤渣烘箱中干燥2h,用气流粉碎机粉碎投入浸提罐中,加入10倍的饮用纯化水,45℃浸泡1h,加入1.4份磷酸氢二钾,在85℃下超声辅助提取40min,滤膜过滤;滤渣第二次加入5倍的饮用纯化水,加入0.4份磷酸氢二钾,在85℃下超声辅助提取40min,滤膜过滤;将两次滤液合并,减压浓缩至原来体积的1/2,即得芒草提取液;
(3)向荷叶滤渣中加入5倍质量的体积分数为75%的乙醇,80℃下搅拌30min后,微波辅助回流提取15min,继续超声辅助80℃回流1h,减压抽滤得第一次滤渣和第一次滤液,向第一次滤渣中加入3倍质量的体积分数为75%的乙醇,80℃下微波辅助回流提取15min,继续超声辅助80℃回流1h,减压抽滤得第二次滤渣和第二次滤液;将第一次滤液和第二次滤液合并减压浓缩回收乙醇,得到浓缩液,加入2倍质量的水,搅拌10min后静置10min,滤布过滤,即得荷叶提取液;
(4)将30份雪莲培养物粉先经超微粉碎机粉碎,加入30份的饮用纯化水,在50℃下搅拌20min,保温的状态下加入0.2份磷酸氢二钾质量,超声提取1h,滤膜过滤,得到第一次雪莲滤液和滤渣,在滤渣中加入相同重量的饮用纯化水保持在50℃,然后加入0.5%的质量比2:1的纤维素酶和果胶酶,酶解2h,升至90℃灭酶,滤膜过滤,得到第二次雪莲滤液,将两次雪莲滤液合并得到雪莲液;
(5)将芒草汁、荷叶汁、雪莲液、低聚玉米肽、L阿拉伯糖、决明子速溶粉加入到混合提取液中,在40℃下搅拌混合2h,然后再加入树莓浓缩汁、聚葡萄糖、木糖醇和黄原胶搅拌混合30min;然后将混合料液抽入管式微滤膜中,调节压力至0.2MPa,收集通过管式微滤膜的滤液;
(6)90℃杀菌,冷却至55℃,进入高压均质机均质,均质压力为25MPa,均质后的料液及时灌装于无菌的干净干燥瓶中(10ml/支),加盖密闭,100℃流通蒸汽灭菌30min,制得雪莲培养物口服液。
为表明本发明的保健效果,本发明对50例高血压、50例高血脂患者作了系统的临床观察:
受试人群:选择病史、病情、年龄、性别及原治疗方法相当的高血压及高血脂患者,年龄在45-55岁之间,无心血管、肝、肾等严重原发性疾病及精神病。高血压:在未使用抗高血压药物情况下,收缩压≥139mmHg和/或舒张压≥90mmHg,按照血压水平将高血压分为1、2、3级。高血脂:以成年人空腹血清总胆固醇超过5.72毫摩尔/升,甘油三酯超过1.70毫摩尔/升,诊断为高血脂症,将总胆固醇在5.2-5.7毫摩尔/升称为边缘性升高。受试者在原有治疗基础上,停用降压降脂药,服用本发明口服液。定期检查血压和空腹血脂,观察期两个月,30天为一疗程。
服用方法:本发明口服液10ml/次,日服3次,饭前服用。观察期间,受试者需保持合理饮食结构及作息时间,禁止非观察的其他药物。

从附表1和2可以看出,受试患者服用本发明后,未使用其他降压降脂药物就可达到降压降脂的目的。试验统计:用本发明口服液对50例高血脂患者、50例高血压患者作治疗试验,对降血脂、降血压的总显效率为88%,有效率为85%。在临床试验过程中未发现该药有明显不良反应,服用安全。
以上所述,仅为本发明较佳的具体实施方式,但本发明的保护范围并不局限于此,任何熟悉本技术领域的技术人员在本发明披露的技术范围内,根据本发明的技术方案及其发明构思加以等同替换或改变,都应涵盖在本发明的保护范围之内。

Claims (2)

1.一种雪莲培养物口服液,其特征在于:由下列按重量份计算的原料制备而成:雪莲培养物粉20-30份、芒草18-22份、荷叶10-12份、磷酸氢二钾2-4份、低聚玉米肽5-12份、L阿拉伯糖1-3份、决明子速溶粉5-10份、树莓浓缩汁2-5份、聚葡萄糖1-2份、木糖醇1-5份和黄原胶2-5份;其制法包括以下步骤:
(1)将芒草超声清洗后切小块,隔水蒸2小时,然后进行榨汁,过滤得到芒草汁和芒草滤渣;将荷叶超声清洗后切小块,榨汁,过滤得到荷叶汁和荷叶滤渣;
(2)将芒草滤渣烘箱中干燥1-2h,用气流粉碎机粉碎投入浸提罐中,加入10-15倍的饮用纯化水,45℃下浸泡1h,加入磷酸氢二钾总质量的70%,在85-90℃下超声辅助提取40min,滤膜过滤;滤渣第二次加入5-10倍的饮用纯化水,加入磷酸氢二钾总质量的20%,在85-90℃下超声辅助提取40min,滤膜过滤;将两次滤液合并,减压浓缩至原来体积的1/2,即得芒草提取液;
(3)向荷叶滤渣中加入5倍质量的体积分数为75%的乙醇,80℃下搅拌30min后,微波辅助回流提取15min,继续超声辅助80℃回流1h,减压抽滤得第一次滤渣和第一次滤液,向第一次滤渣中加入3倍质量的体积分数为75%的乙醇,80℃下微波辅助回流提取15min,继续超声辅助80℃回流1h,减压抽滤得第二次滤渣和第二次滤液;将第一次滤液和第二次滤液合并减压浓缩回收乙醇,得到浓缩液,加入2倍质量的水,搅拌10min后静置10min,滤布过滤,即得荷叶提取液;
(4)将雪莲培养物粉先经超微粉碎机粉碎,加入相同重量的饮用 纯化水,在50℃下搅拌20min,保温的状态下加入磷酸氢二钾总质量的10%,超声辅助提取1h,滤膜过滤,得到第一次雪莲滤液和滤渣,在滤渣中加入相同重量的饮用纯化水保持在50℃,然后加入0.5-1%的质量比2:1的纤维素酶和果胶酶,酶解2h,升至90℃灭酶,滤膜过滤,得到第二次雪莲滤液,将两次雪莲滤液合并得到雪莲液;
(5)将芒草汁、荷叶汁、雪莲液、低聚玉米肽、L阿拉伯糖混合,决明子速溶粉加入到芒草提取液和荷叶提取液的混合提取液中,在40℃下搅拌混合2h,然后再加入树莓浓缩汁、聚葡萄糖、木糖醇和黄原胶搅拌混合30min;然后将混合料液抽入管式微滤膜中,调节压力至0.2MPa,收集通过管式微滤膜的滤液;
(6)90℃杀菌,冷却至55℃,进入高压均质机均质,均质压力为25MPa,均质后的料液即雪莲培养物口服液。
2.根据权利要求1所述的一种雪莲培养物口服液,其特征在于:由下列按重量份计算的原料制备而成:雪莲培养物粉26份、芒草20份、荷叶11份、磷酸氢二钾3份、低聚玉米肽8份、L阿拉伯糖2份、决明子速溶粉8份、树莓浓缩汁4份、聚葡萄糖1份、木糖醇3份和黄原胶3份。
