背景技术
视疲劳又称眼疲劳,是以患者眼的自觉症状为基础,眼或全身器质性因素与精神(心理)因素相互交织的综合征,并非独立的眼病,因而常常被称为眼疲劳综合征。轻度视疲劳会在用眼后出现眼部酸、胀等眼部症状,休息后很快恢复;中度视疲劳会有明显的眼部症状,影响学习和工作;重度视疲劳除有明显的眼部症状和视作业不能持久外,还有疲劳、失眠等全身不适症状,干扰患者的视觉和生活质量,严重影响学习、工作和生活。
大量的临床实践证明,屈光不正是视疲劳的主要原因之一,如远视、散光、近视患者,在未予矫正屈光不正时,看远或看近时动用较大的调节使睫状肌持续紧张造成患者的调节性视疲劳。另外,长时间近距离工作、工作或阅读环境照明不足等因素都可以导致人眼调节紧张,产生视疲劳。患者的精神状态和全身情况也与视疲劳的发生密切相关,精神压力大、体质衰弱的患者易产生视疲劳。
随着便携式终端性能改善和通信技术发达及文化产业的成长,导致现代人的眼睛除了睡觉时间之外不能休息而负担过重,长时间读书、使用电脑、看电视等长时间用眼使得眼科相关疾病持续增长。中国互联网络发展状况统计2017报告,手机网民达7.24亿,手机网民占比达96.3%!手机使用习惯调查报告显示:近五成的用户每天使用手机达5小时以上,比例高达47.17%,28.08%的用户每天使用手机在3-5小时。长时间看手机等视屏终端会对眼睛的健康造成严重威胁,调查显示,有90%的使用者在长时间面对手机以后感到视物模糊、眼睛酸疼,甚至会出现疲劳、头痛、头晕等精神症状。
因此,研发对视疲劳具有显著缓解作用的产品,具有重要意义。

本发明提供了缓解视疲劳的组合物,是活性成分包括鱼眼球组织或其提取物和鱼眼窝组织或其提取物的食品、保健品或药品,其中,活性成分还包括茶氨酸和/或花青素。

本发明包含上述鱼类眼窝、眼球的提取物作为有效成分,在此,获得鱼类眼窝、眼球的提取物的方法没有特别限定,能够使用本发明所属的技术领域中通常使用的各种提取方法。将例如、精制水、甲醇、乙醇、甘油、乙酸乙酯、丁二醇、丙二醇、二氯甲烷、己烷中的一种单独作为溶剂或将其中的2种以上混合后的溶剂作为提取溶剂使用,进行提取。
另外,本发明如上所述优选利用从精制水、甲醇、乙醇、甘油、乙酸乙酯、丁二醇、丙二醇、二氯甲烷、己烷及其混合物选择的提取溶剂,将经过超高压处理的鱼类的眼球提取物进行回流提取、冷浸之后,进行过滤,将过滤后的过滤液转送到浓缩槽,在60℃以下进行加压浓缩及冻结干燥。
在本发明的一个实施方式中,鱼类眼窝、眼球提取物采用如下方式获得:在鱼类眼窝和眼球粉碎物中加入1倍-20倍、优选为1倍-10倍左右的水、甲醇或乙醇等低级(C1-C4)醇的极性溶剂或它们的混合溶剂,在5℃-40℃提取温度下,利用冷浸提取、回流冷却提取、热水提取、超声波提取、超高压提取等提取方法,获得提取液,将提取液过滤、加压浓缩或冻结干燥,由此获得加入于极性溶剂的提取物。
在本发明的一个实施方式中,上述鱼类眼窝、眼球提取物是在压力50MPa~3200MPa、温度30℃~95℃下处理3分钟~80分钟提取的。
另外,本发明的鱼类眼窝、眼球的提取物不仅包括采用上述的提取溶剂提取的提取物,还包括采用其他方法提取的提取物。例如,采用超高压提取法进行提取,利用酶解提取法进行提取,超声波或微波进行提取,SC-CO2提取法进行提取,利用具备预定分子量截止值的限外过滤膜进行的分离,各种色谱法,基于为了根据例如大小、电荷、疏水性或亲和性进行分离而制作的色谱图进行的分离等,通过附加地实施的各种精制方法而获得的分划,也属于本发明的鱼类眼窝、眼球的提取物。
另外,本发明的鱼类眼窝、眼球的提取物包括:将上述提取物的一部分或全部浓缩而得到的浓缩物;或者,再次将该浓缩物干燥而制造的提取精华物;以及提取物中含有的主要发挥效果的化学物质本身。
本发明鱼类眼窝、眼球或其组合物作为药品、食品、尤其功能性食品组合物制造。本发明的功能性食品组合物包含在食品制造时通常添加的成分,包含例如、蛋白质、碳水化合物、脂肪、营养素及调味剂。例如,在作为饮料制造的情况下,除了作为有效成分的鱼类眼窝组织和眼球组织粉碎物或提取物之外,作为附加成分,还可以包含香味剂或天然碳水化合物。例如,天然碳水化合物可以使用单糖(例如、葡萄糖、果糖等);双糖(例如、麦芽糖、蔗糖等);低聚糖;多糖(例如、糊精、环式糊精等);及糖醇(例如、木糖醇、山梨糖醇、赤藻糖醇等)。作为香味剂,可以使用天然香味剂(例如、祝马丁、甜叶菊提取物等)及合成香味剂(例如、糖精、阿斯巴特等)。作为糖果制造的情况下,可以与食糖、糖浆、甜味剂、食用胶或淀粉、油脂、乳制品、可可制品等形成组合。

其中,本发明的食品组合物中,相对于组合物的总重量,优选上述鱼类眼窝、眼球的粉碎物或提取物的含量为0.0001~95%重量百分比。在含量<0.0001%的重量百分比的情况下,难以期待效果;在含量>95.0%重量百分比的情况下,也未能看到功效有显著提高。
其中,本发明的食品组合物中还可以含有食品领域通常使用的辅助剂,溶剂、抗氧化剂、芳香剂、润滑剂等,这些多种辅助剂的量是本发明所属领域通常使用量,例如,相对于组合物总重量,为0.01~30%(w/v)。另外,在任何情况下,都以辅助剂及其比例不能给本发明的食品组合物的优异性质造成影响为前提。
在本发明中,鱼类眼窝、眼球粉碎物具有眼睛营养物质的补充作用,但是为了使口感或口味更佳,并提高视疲劳治疗及预防功效,优选以将有效成分提取的提取物形态使用。
本发明所述茶氨酸即N-乙基-L-谷氨酰胺。茶氨酸具有降压安神、改善睡眠、促进大脑功能等功效,已被用作保健食品和药品原料。有研究发现,人体摄入50mg茶氨酸,α-脑电波会逐渐增加,协助精神放松并保持头脑灵活。茶氨酸能使大脑细胞中线粒体内神经递质—多巴胺显著增加,具有抗疲劳作用,可通过调节神经传达物质来改善学习能力和记忆力。茶氨酸缓解视疲劳和干眼症及蓝光损伤所致的其他症状,如:失眠、情绪低落、头晕头痛、记忆衰退、内分泌紊乱、学习障碍等。
花青素是一类水溶性生物黄酮,在自然界中多以花色苷的形式存在,大多数花青素具有如下基本结构:

本发明所述花青素优选蓝莓来源,蓝莓果实含有防止脑神经衰老、增强心脏功能、明目及抗癌等独特功效物质,因此被国际粮农组织列为人类五大健康食品之一。欧洲广泛运用的医药制品标准以25%花青素含量为标准。每天80-160mg提取物被推荐用于改善眼部健康,而160-240mg则可治疗眼部疾病。蓝莓提取物没有任何已知副作用,而且过多的摄入会迅速随尿液和胆汁排出。蓝莓中含的花青贰色素,具有活化视网膜功效,可以强化视力,防止眼球疲劳。蓝莓主要功效:保护视力,防止失明、青光明、白内障、视网膜出血、改善近视、黄斑退化、糖尿病性网膜症、色素性视网膜炎及夜盲症等。蓝莓中的花青素可促进视网膜细胞中视紫质的再生成,可预防重度近视及视网膜剥离,并可增进视力。法国多年使用花青素治疗糖尿病性视网膜病,显著减少了眼睛毛细血管出血,改善了视力。花青素也用来防止糖尿病患者白内障手术后的并发症。花青素能减轻自由基对眼睛晶状体的损害,缓解因长时间操作电脑或驾驶等带来的眼部疲劳和损害。
本发明所述低聚果糖的来源包括但不限于菊芋、菊苣。以菊芋粉为原料,用菊糖内切酶水解作用,经精制低聚果糖浆。聚果糖是指2~5个果糖基为链节,以一个葡萄糖基为链的端基,以果糖基→果糖连接键为主体骨架连结形成的碳水化合物,是一种天然活性物质。甜度为蔗糖的0.3-0.6倍。既保持了蔗糖的纯正甜味性质,又比蔗糖甜味清爽。是具有调节肠道菌群,增殖双歧杆菌,促进钙的吸收,调节血脂,免疫调节,抗龋齿等保健功能的新型甜味剂,低聚果糖不易被人体吸收,热量值很低,不会导致肥胖,被誉为继抗生素时代后最具潜力的新一代添加剂——促生物质;已在乳制品、乳酸菌饮料、固体饮料、糖果、饼干、面包、果冻、冷饮等多种食品中应用。在糖果、饮料中的应用作为独特的低糖、低热值、难消化的功能性甜味剂。
本发明所述虾青素的来源包括但不限于雨生红球藻。雨生红球藻被公认为自然界中生产天然虾青素的最好生物,采收雨生红球藻孢子,经破壁、干燥等工艺制成,将雨生红球藻藻粉经过乙醇和乙酸乙酯的混合溶剂提取后,经过皂化,得到雨生红球藻提取物(虾青素提取物)。以全反式虾青素计,含量为:1.5%-3%。虾青素是世界上最强的天然抗氧化剂之一,有效清除细胞内的氧自由基,增强细胞再生能力,维持机体平衡和减少衰老细胞的堆积。虾青素可以透过血脑屏障,对大脑、中枢神经系统及双眼都可起到保护作用。人类视网膜和中枢神经系统(脑)富含不饱和脂肪酸,视频终端和蓝光会氧化所致的自由基促进发生过氧化损伤。有研究表明,虾青素很容易通过血脑屏障和细胞膜,能有效地防止视网膜的氧化和感光细胞的损伤,以及对中枢神经系统尤其是对大脑起到保护作用,缓解眼疲劳症状,提高视觉灵敏度。
本发明所述(3R,3'R)-二羟基-β-胡萝卜素亦称玉米黄素,以万寿菊花来源的万寿菊油树脂为原料,经皂化、离心、过滤、干燥等工艺制成橙红色至红色粉末。玉米黄质存在于眼睛的视网膜黄斑区,是眼睛中发现的两种类胡萝卜素之一。人体无法自身合成玉米黄质,必须通过食物或补充剂获得。具有特异性吸收视网膜最具损伤性的蓝色光线的作用,是一种很有效的蓝光过滤器。除此之外,玉米黄质具有抗氧化性,可以预防和缓解老年性黄斑变性(AMD),防止白内障的形成。
叶黄素是一种含氧的类胡萝卜素,广泛存在于蔬菜、花卉、水果与某些藻类生物中的天然色素,其结构如下所示:
本发明所述叶黄素酯,以万寿菊花为原料,经过脱水粉碎、溶剂提取、低分子量醇纯化和真空浓缩等步骤生产而成深红棕色细小颗粒。在眼睛中,二者是唯一发现的类胡萝卜素,但是二者的集中位置不同,玉米黄质主要集中在视网膜的黄斑区中心,叶黄素则遍及视网膜。研究显示,在视网膜中心区域,叶黄素与玉米黄质的比例是1:2;而在视网膜周围区域,它们的比例是2:1-3:1之间。叶黄素和玉米黄质是人类视网膜中发现的主要膳食类胡萝卜素,可以保护黄斑免受蓝光损伤,提高视力,清除有害的活性氧。叶黄素和玉米黄质以及它们常见的代谢物内消旋玉米黄质,通常称为黄斑色素(MP)。证据表明叶黄素和玉米黄质是预防和减少白内障和AMD的重要膳食类胡萝卜素。美国涉及五个眼科中心的多中心眼病病例对照研究表明,摄入较多的类胡萝卜素,特别是叶黄素和玉米黄质的摄入量与降低的AMD风险有关。
本发明所述锌是人体必需的微量元素之一,在人体生长发育、生殖遗传、免疫、内分泌等重要生理过程中起着极其重要的作用。促进人体的生长发育、维持人体正常食欲、增强人体免疫力、促进伤口和创伤的愈合、维持男性正常的生精功能、调节影响大脑生理功能的各种酶及受体、影响维生素A的代谢和正常视觉。
本发明提供了缓解视疲劳的组合物。人群试用结果表明,鱼眼球组织、鱼眼窝组织均具有显著的缓解视疲劳的作用,对视疲劳引起的眼酸痛、眼胀、畏光、视物模糊、异物感、流泪等症状都能够起到缓解作用,而且,将鱼眼球组织和鱼眼窝组织组合使用后,两者富含的益眼成分在功效上得到了互补,能够发挥协同增效的作用,使得缓解视疲劳的效果得到进一步提升。鱼眼具有良好保健作用,本发明利用现代技术方法对鱼眼进行深加工,生产相应的功能性食品、药品,不仅减少鱼类加工的废弃物的产生,还能变废为宝,增加渔业资源的产出,具有广阔的市场发展前景。
具体实施方式
本发明具体实施方式中使用的原料、设备均为已知产品,通过购买市售产品获得。
本发明提供了缓解视疲劳的组合物,该组合物是活性成分包括鱼眼球组织或其提取物和鱼眼窝组织或其提取物的食品、保健品或药品,其中,活性成分还包括茶氨酸或花青素。
鱼眼睛的构造较为简单,大多数鱼无眼睑而眼球完全裸露,鱼眼眼球外观呈椭球状,位于由眶上骨、若干眶小骨及软颅围成的眼眶内。光镜下一般海鱼的鱼眼由外至内分为角膜-巩膜层、虹膜-脉络膜层、晶状体及视网膜层。眼球前面为透明的角膜,其余为棕黑色的巩膜。眼球内侧由肌肉、视神经、血管及软骨(眼柄)构成。鱼眼睛的大小和它日常所接触的光线很有关系。生活于水的上层或中层的鱼,它们都有正常的眼睛,如鲤鱼、鲫鱼、黄鱼等鱼类。而生活在300~1500米深处的鱼类,因这里的光线极弱,一般眼睛都比较大,如大眼鲷鱼。鱼眼睛富含维生素、活性多肽、氨基酸、糖类、多种微量元素(钙、磷、铁、锌等)以及包括ω-3脂肪酸在内的多种不饱和脂肪酸,这些物质对眼睛的新陈代谢有综合性有益影响。
鱼眼窝组织是位于鱼眼睛周围的肌肉组织和脂肪组织,其中,鱼眼窝脂肪组织(眼窝脂肪是处于眼球背后的结构脂肪的一种,为保护视神经而存在,起到保护眼球免遭高水压伤害的缓冲器作用,同时又成为脑、视网膜所必需的DHA供给源)油脂含量高,尤其是海鱼鱼眼窝脂肪组织中含有大量的多不饱和脂肪酸,其中的ω-3高度不饱和脂肪酸系列(主要是DHA、EPA)具有特殊的生理功效。DHA是神经系统生长及维持的一种主要元素,是大脑和视网膜的重要构成成分,在人体大脑皮层中含量高达20%,在眼睛视膜中所占比例最大,约占50%。DHA被誉为“脑黄金”和“聪明的原动力”,但人体不能自身合成DHA,只能从食物中摄取。
目前,仅见鱼眼球组织、鱼眼窝组织或其提取物单独应用的报道,用途主要为抗血栓、增视明目等,但尚未见鱼眼球组织或鱼眼窝组织缓解视疲劳作用的相关研究,更未见将两者组合使用的报道。
本发明通过人群试用证明,鱼眼球组织、鱼眼窝组织均具有显著的缓解视疲劳的作用,对视疲劳引起的眼酸痛、眼胀、畏光、视物模糊、异物感、流泪等症状都能够起到一定程度的缓解作用,而且,将鱼眼球组织和鱼眼窝组织组合使用后,两者富含的益眼成分在功效上得到了互补,能够发挥协同增效的作用,使得缓解视疲劳的效果得到进一步提升。
另外,本发明组合物中还加入活性成分茶氨酸和/或花青素。
茶氨酸可以抑制谷氨酸的毒性,保护神经元;具有促进脑波中α波产生的功能,从而引起轻松、愉快的感觉,缓解抑郁和焦虑情绪;降血压;促进注意力集中,增进反应速度和准确性。有利于缓解视疲劳所致的相关精神症状。
花青素具有较强的抗氧化作用。长期食用可协助清除视网膜内有毒化学物质、自由基等,增加视网膜紫红素生成,提高夜视能力,帮助眼睛适应强弱光的变化,是维护眼睛健康、预防视力受损的重要元素。花青素的主要功效为:保护视力,预防失明、青光眼、白内障、视网膜出血、改善近视、黄斑退化、糖尿病性网膜症、色素性视网膜炎及夜盲症等。
实施例1鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼粉碎物的制造
将新鲜的鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等速冷之后,与肉质分离而获取眼窝组织和眼球组织,进行了冷冻。将冷冻的眼窝组织和眼球组织用均化器或粉碎器均匀地粉碎。
实施例2鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼粉碎物的制造
将新鲜的鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等速冷之后,与肉质分离而获取眼窝组织和眼球组织,进行了冷冻。将冷冻的眼窝组织和眼球组织在低温(-40℃至-120℃)粉碎、低温研磨,将冷却到脆化点温度的物质在外力作用下破碎成粒径较小的颗粒,低温粉碎保证被粉碎物质,组织成分不受破坏。
实施例3鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼粉碎物的制造
将新鲜的鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼速冷之后,与肉质分离而获取眼球和眼窝组织,进行蒸熟,对蒸熟后的鱼眼眼球和眼窝组织控温60-80℃烘烤5-10分钟后,再碾成粉末状,备用。
实施例4鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼粉碎物的制造
将新鲜的鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼速冷之后,与肉质分离而获取眼球和眼窝脂肪组织,放入米醋中,米醋以淹没物料为度,浸泡48~72小时,捞出、烘干,粉碎至50目以下,备用。
实施例5鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼粉碎物的制造
将新鲜的鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等速冷之后,与肉质分离而获取眼窝组织和眼球组织,进行低温干燥或喷雾干燥,将干燥的眼窝组织和眼球组织用均化器或粉碎器或低温粉碎均匀地粉碎。
实施例6鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼提取物的制造
将新鲜的鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼球速冷之后,将与肉质分离而获取的冷冻眼窝组织和眼球组织用70%乙醇水溶液于100~120℃提取2~3小时。
实施例7鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼提取物的制造
将新鲜的鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼球速冷之后,将与肉质分离而获取的冷冻眼球用超高压处理机在压力800MPa下将温度升温到60℃,处理30分钟之后,冷却到室温。将经过超高压处理的金枪鱼眼球提取物,利用精制水进行5次各3个小时的回流提取,进行冷浸之后,过滤。过滤液转送到浓缩槽,在50℃以下加压浓缩及冻结干燥。使用精制水、乙醇、丁二醇、丙二醇中的1种以上的溶剂,将得到的加压浓缩物及冻结干燥物以0.001~80.0%重量百分比的含量溶解,制备出最终提取物。
实施例8鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼球提取物(以DHA为指标成分)的制造
原料处理:取新鲜海鱼鱼眼(眼球组织)用水洗净,沥干后,在0.05-0.1%的柠檬酸溶液中浸泡10-30min,于40-70℃烘干至含水分小于4%,再粉碎至100-200目的海鱼粉;
酶解:将经处理后的海鱼粉加缓冲液用6mol/L盐酸调pH值为4-6,加入质量分数0.1-1.0%,酶活3.0×104u/g的木瓜蛋白酶和质量分数0.5-1.5%,酶活1.0×104u/g的纤维素酶,于40-60℃搅拌酶解1-2h,之后用0.5mo1/L氢氧化钠调pH至中性;
真空提取:将得到的海鱼酶解液置于真空提取设备中,控制真空度85-90KPa,温度40-60℃提取1-2小时,同时灭酶,得到提取液;
分离:将提取液于6000rpm离心20分钟,收集上清液;
浓缩:将得到的液体置于真空浓缩设备中,控制真空度85-90KPa,温度40-60℃浓缩至原体积的l/5-1/3,得到DHA浓缩液。
实施例9鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼窝提取物(以DPA、EPA和DHA为指标成分)的制造
将海鱼加工后的眼窝洗净,以固液重量比是1:1-1.2的比例加入水,再磨成浆状得到匀浆;取匀浆用有机溶剂萃取1-3次,有机溶剂与匀浆体积之比为1:1.2-1.5,收集萃取液,得到含DPA、EPA和DHA油脂的萃取液备用,所述有机溶剂指的是石油醚或乙醚;萃取液回收有机溶剂,得到含DPA、EPA和DHA的鱼油,取与鱼油等体积的0.3mol/L甲醇钠混合,室温条件下充氮搅拌10分钟后,调节pH值到7.0-7.2,回收甲醇,得到甲酯化的鱼油;对甲酯化鱼油进行分子蒸馏,包括脱气、预热蒸馏及蒸馏三个技术步骤,其中脱气步骤在温度20-30℃、系统压力在1-2mbar的条件下进行脱气;预热蒸馏步骤在鱼油以15-20公斤/小时的进料速度送入预热级蒸馏器中,在温度170-200℃、系统压力0.001-0.01mbar、刮膜转子速度150-250转/分钟的条件下蒸馏;蒸馏步骤以5-20公斤/小时的进料速度送入蒸馏器中,在温度250-350℃、系统压力0.001-0.01mbar、刮膜转子速度150-250转/分钟的条件下蒸馏,收集290-310℃时的馏分,即得到较高纯度的含EPA、DHA以及DPA的复合物。
实施例10鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼窝提取物(以活性油脂为指标成分)的制造
新鲜(或冷冻保存,流水解冻)的鱼头,获得眼窝组织,匀浆。
超声辅助复合酶酶解水提法制备鱼眼窝油脂:向所述匀浆液中加入0.5~1.5倍体积的水;用300-800瓦超声波处理所述匀浆液10-30分钟;用1-3摩尔/升盐酸或氢氧化钠调pH为6-9;加入中性蛋白酶(EC 3.4.23.6),酶加量为鱼脑匀浆重量的0.1-2%,30-60℃酶解1-3小时;90-100℃灭酶(优选10分钟或10钟以上);用1-3摩尔/升盐酸或氢氧化钠调pH为7-11;加入碱性蛋白酶(EC3.4.21.14),酶加量为鱼脑匀浆重量的0.1-2%,30-60℃酶解1-3小时;酶解后在90-100℃灭酶(优选10分钟或10分钟以上),于>=5000×g离心10-30分钟,得游离油、乳状层、水层、沉淀层。收集游离油层得鱼眼窝油;水层与乳状层合并,经干燥得干粉。超临界二氧化碳法萃取鱼眼窝油脂:超临界萃取条件为萃取压力15-35兆帕,萃取温度30-50℃,CO2流速5-20升/小时,萃取时间60-120分钟;CO2自下而上流经萃取器,提取液经减压阀减压后流经第一分离器,部分油脂在温度35-45℃、压力8-10兆帕沉淀于第一分离器底部;提取液经第一分离器减压后进入第二分离器,剩余油脂在温度25-35℃,压力2-4兆帕下沉淀于第二分离器底部;收集合并油脂。分离出油脂CO2,再经高压泵压缩成超临界状态,重新进入萃取器循环使用。合并上述所获油脂,得鱼眼窝油。
实施例11鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼窝提取物(以活性油脂为指标成分)的制造
新鲜(或冷冻保存,流水解冻)的鱼头,去除皮和骨头,获得眼窝组织,匀浆。
超声辅助复合酶酶解:向所述匀浆液中加入0.5倍体积的水;用500瓦超声波处理所述匀浆液10分钟;用1摩尔/升氢氧化钠调pH为8;加入中性蛋白酶(EC3.4.23.6),酶加量为鱼脑匀浆重量的1%,50℃酶解1小时;95℃灭酶15分钟;用3摩尔/升氢氧化钠调pH为9;加入碱性蛋白酶(EC3.4.21.14),酶加量为鱼脑匀浆重量的1%,50℃酶解1小时;酶解后在100℃灭酶15分钟,于15000×g离心15分钟,得游离油、乳状层、水层、沉淀层。收集游离油层得鱼眼窝油;水层与乳状层合并,经干燥得干粉。超临界二氧化碳法萃取鱼眼窝油脂,超临界萃取条件为:萃取压力25兆帕,萃取温度40℃,CO2流速15升/小时,萃取时间100分钟;CO2自下而上流经萃取器,提取液经减压阀减压后流经第一分离器,部分油脂在温度40℃、压力8兆帕下不溶于CO2沉淀于分离器底部;提取液经第一分离器减压后进入第二分离器,剩余油脂在温度35℃,压力2兆帕下不溶于CO2沉淀于分离器底;收集合并分离器中油脂,即得鱼眼窝油脂产品;分离出油脂的CO2,再经高压泵压缩成超临界状态,重新进入萃取器循环使用。合并所获油脂,得鱼眼窝油。
实施例12本发明糖果的制备
原料重量配比:鱼眼窝组织粉碎物或提取物100-800份(以鲜品重量计)、鱼眼球组织粉碎物或提取物100-800份(以鲜品重量计)、茶氨酸100-400份。
制备方法:
1、按重量配比将鱼眼窝组织粉碎物或提取物、鱼眼球组织粉碎物或提取物、茶氨酸混合均匀后加入糖浆搅拌均匀。
2、注模冷却成型后可按需制成硬质、奶糖、夹心、酥质、焦香(太妃)、充气、凝胶、胶基、压片、流质、膜片、花式等糖果类型,以及巧克力及巧克力制品、代可可脂巧克力和代可可脂巧克力制品、果冻(果汁型、果肉型、果味型、乳汁型等)等糖果制品。

制备方法:选择鱼眼窝组织粉碎物或提取物、鱼眼球组织粉碎物或提取物、茶氨酸、蓝莓提取物,(3R,3'R)-二羟基-β-胡萝卜素,搅拌均匀,臭氧灭菌,灌装,可直接冲服或作为功能性辅料加入到其它食品中。
以下通过试验例证明本发明的有益效果。以下所述鱼眼球组织提取物使用根据实施例8方法制备的罗非鱼眼球组织提取物,鱼眼窝组织提取物使用根据实施例9方法制备的罗非鱼眼窝组织提取物。
试验例1鱼眼球组织和/或鱼眼窝组织缓解视疲劳及其相关症状的试用观察
设置3个试验组,分别按表2所示取鱼眼球组织提取物、鱼眼窝组织提取物,括号内表示对应的新鲜鱼眼球组织、鱼眼窝组织的重量,将其装入等大不透明胶囊,供眼疲劳患者试服,连续服用60天(表2对应每日服用量),观察试用效果。
根据《视疲劳诊疗专家共识》(2014年)诊断为视疲劳患者144例,症状包括视觉障碍:近距离工作或阅读不持久,出现暂时性视物模糊或重影;眼部不适:眼胀、眼痛、眼烧灼感、流泪、眼痒、眼异物感及眼眶疼痛,年龄18-65岁,随机分3组。根据国食药监保化[2012]107号《缓解视疲劳功能评价方法》制订视疲劳评定量表,见表1,计算服用前、服用60天积分降低率:(治疗前总积分-治疗后总积分)/治疗前总积分×100%,治疗后积分降低率≥30%为有效。症状积分改善有效率统计结果见表2。

人群试用结果显示:鱼眼球组织、鱼眼窝组织均具有显著的缓解视疲劳的作用,对视疲劳引起的眼酸痛、眼胀、畏光、视物模糊、异物感、流泪等症状都能够起到一定程度的缓解作用,而且,将鱼眼球组织和鱼眼窝组织组合使用后,两者富含的益眼成分在功效上得到了互补,能够发挥协同增效的作用,使得缓解视疲劳的效果得到进一步提升。
试验例2不同配比的鱼眼球组织、鱼眼窝组织和花青素缓解视疲劳及其相关症状的试用观察
分别按表3所示取鱼眼球组织提取物、鱼眼窝组织提取物和花青素,表格中表示对应的新鲜鱼眼球组织、鱼眼窝组织、花青素的重量,将其装等大不透明胶囊,供眼疲劳患者试服,连续服用60天(表3对应每日服用量),观察试用效果。
根据《视疲劳诊疗专家共识》(2014年)诊断为视疲劳患者318例,症状包括视觉障碍:近距离工作或阅读不持久,出现暂时性视物模糊或重影;眼部不适:眼胀、眼痛、眼烧灼感、流泪、眼痒、眼异物感及眼眶疼痛,年龄18-65岁,随机分6组。按照与试验例1相同的方法统计症状积分改善有效率,结果见下表:

试用结果显示:鱼眼球组织、鱼眼窝组织和花青素在本发明限定的配比范围内(即鱼眼球组织10~1000份,鱼眼窝组织10~1000份,花青素10~1000份)组合使用(试验组1~4),均能够发挥显著的缓解视疲劳的作用,对影响视疲劳人群的眼酸痛、眼胀、畏光、视物模糊、异物感、流泪等症状均有明显疗效,症状积分改善有效率达到91%以上。若在本发明限定的配比范围外混合鱼眼球组织、鱼眼窝组织和花青素,所得组合物也有一定的缓解视疲劳的作用,但效果有所下降(试验组5、6)。
试验例3不同配比的鱼眼球组织、鱼眼窝组织和茶氨酸缓解视疲劳及其相关症状的试用观察
分别按表5所示取鱼眼球组织提取物、鱼眼窝组织提取物和茶氨酸。表格中表示对应的新鲜鱼眼球组织、鱼眼窝组织、茶氨酸的重量,将其装等大不透明胶囊,供眼疲劳患者试服,连续服用60天(表5对应每日服用量),观察试用效果。
轻度视疲劳282例、重度视疲劳36例,随机分6组。试服者(年龄18-65岁),轻度视疲劳患者(根据《视疲劳诊疗专家共识》(2014年)分类:症状局限在眼部(视觉障碍:近距离工作或阅读不持久,出现暂时性视物模糊或重影;眼部不适:眼胀、眼痛、眼烧灼感、流泪、眼痒、眼异物感及眼眶疼痛);重度视疲劳患者(根据《视疲劳诊疗专家共识》(2014年)分类:兼有全身症状(疲劳,注意力不集中,头痛,头晕,严重时甚至恶心、呕吐,并出现焦虑、烦躁、失眠以及其他神经官能症的症状则为重度视疲劳)。根据国食药监保化[2012]107号《缓解视疲劳功能评价方法》制订视疲劳评定量表,见表4,计算服用前、服用60天积分降低率:(治疗前总积分-治疗后总积分)/治疗前总积分×100%,治疗后积分降低率≥30%为有效。症状积分改善有效率统计结果见表5。

试用结果显示:鱼眼球组织、鱼眼窝组织和茶氨酸在本发明限定的配比范围内(即鱼眼球组织10~1000份,鱼眼窝组织10~1000份,茶氨酸20~1000)组合使用(试验组1~4),均能够发挥显著的缓解视疲劳的作用,不仅对轻、重度视疲劳的视觉障碍和眼部局部不适症状有治疗效果,对影响重度视疲劳人群的疲劳、注意力不集中、头痛、头晕、焦虑、烦躁、失眠等神经官能症的症状也有改善,症状积分改善有效率达到91%以上。若在本发明限定的配比范围外混合鱼眼球组织、鱼眼窝组织和茶氨酸,所得组合物也有一定的缓解视疲劳的作用,但效果有所下降(试验组5、6)。
Claims (20)
1.缓解视疲劳的组合物,其特征是:是活性成分包括鱼眼球组织或其提取物和鱼眼窝组织或其提取物的食品、保健品或药品,其中,活性成分还包括茶氨酸和/或花青素。
2.如权利要求1所述的组合物,其特征是:所述活性成分的重量配比为:鱼眼球组织10~1000份,鱼眼窝组织10~1000份,茶氨酸20~1000份和/或花青素10~1000份。
3.如权利要求1或2所述的组合物,其特征是:所述鱼眼球组织:鱼眼窝组织的重量配比为1:1。
4.如权利要求1~3任意一项所述的组合物,其特征是:所述鱼眼球组织或鱼眼窝组织选自鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼、魟鱼、鳕鱼、鲭鱼、坚鳞鲈、胡瓜鱼、三文鱼、玉筋鱼、金线鱼、安康鱼、大白鲨、鲂鱼、明太鱼、东方鲈、鲤鱼、鲫鱼、青鱼、枪鱼、真鲷、鲻鱼、马鲛鱼、鲇鱼、银鱼、秋刀鱼、翻车鱼、黑鲷、带鱼、桂鱼、欧洲鲈鱼、泥鳅、乌鱼、叫姑鱼、东方黄鲂鮄、剑鱼、暗纹东方鲀、黄鳍金枪鱼、灰鲭鲨、黄花鱼、河豚、鲽鱼、鼠鱼、鲥鱼、黑鮶鱼、沙丁鱼、斑鳍光鳃鱼、远东多线鱼、马头鱼、鳗鱼、鲳鱼、黄姑鱼中一种或两种以上鱼类的眼球组织或眼窝组织。
5.如权利要求1~3任意一项所述的组合物,其特征是:所述提取物为水提物或醇提物。
6.如权利要求1~3任意一项所述的组合物,其特征是:所述花青素来源于蓝莓、葡萄籽、黑加仑、紫甘薯、血橙、红球甘蓝、茄子、樱桃、红莓、草莓、桑葚、山楂、牵牛花中一种或两种以上。
7.如权利要求1~6任意一项所述的组合物,其特征是:活性成分还包括虾青素、低聚果糖、(3R,3'R)-二羟基-β-胡萝卜素、叶黄素或叶黄素酯、锌中一种或两种以上的混合物。
8.如权利要求7所述的组合物,其特征是:活性成分还包括虾青素1~250份。
9.如权利要求7所述的组合物,其特征是:活性成分还包括低聚果糖10~500份。
10.如权利要求7或9所述的组合物,其特征是:所述低聚果糖来源于菊芋和/或菊苣。
11.如权利要求7所述的组合物,其特征是:活性成分还包括(3R,3'R)-二羟基-β-胡萝卜素0.2~10份。
12.如权利要求7所述的组合物,其特征是:活性成分还包括叶黄素和/或叶黄素酯0.6~25份。
13.如权利要求7所述的组合物,其特征是:活性成分还包括锌0.5~50份。
14.如权利要求1~13任意一项所述的组合物,其特征是:是各重量配比的活性成分,加入可接受的辅料或者辅助性成分制备而成的食品、保健品或药品。
15.如权利要求14所述的组合物,其特征是:所述食品、保健品或药品为口服形式。
16.如权利要求15所述的组合物,其特征是:所述食品或保健品为糖果、饮料、饼干、面包、果冻或膳食补充剂。
17.权利要求1~16任意一项所述组合物的制备方法,其特征是:包括如下步骤:取各重量配比的活性成分,加入食品、保健品或药品中可接受的辅料或者辅助性成分,即得。
18.权利要求1~16任意一项所述组合物在制备缓解视疲劳的食品、保健品或药品中的用途。
19.如权利要求18所述的用途,其特征是:所述食品、保健品或药品具有缓解视疲劳所致眼酸痛、眼胀、畏光、视物模糊、异物感或流泪的功效。
20.如权利要求18所述的用途,其特征是:当所述组合物含有活性成分茶氨酸时,所述食品、保健品或药品还具有缓解视疲劳所致疲劳、注意力不集中、头痛、头晕、恶心、呕吐、焦虑、烦躁或失眠的功效。
