Abstract
本发明涉及含鱼眼、茶氨酸和γ‑氨基丁酸的缓解视疲劳的组合物,属于医药健康领域。本发明提供了缓解视疲劳的组合物,其活性成分包括鱼眼球组织或其提取物、鱼眼窝组织或其提取物、茶氨酸和γ‑氨基丁酸。人群试用结果表明,本发明组合物不仅针对视疲劳眼睛局部症状,更能够明显改善不同程度视疲劳引起的精神神经症状,如注意力不集中、记忆力下降等,缓解视疲劳效果显著,具有广阔的市场前景。

技术领域
本发明涉及含鱼眼、茶氨酸和γ-氨基丁酸的缓解视疲劳的组合物,属于医药健康领域。
背景技术
视疲劳又称眼疲劳,是以患者眼的自觉症状为基础,眼或全身器质性因素与精神(心理)因素相互交织的综合征,并非独立的眼病,因而常常被称为眼疲劳综合征(参见:1、李凤鸣.中华眼科学[M].北京:人民卫生出版社,2006,7~8;2、渠鸿颖等.视疲劳病因及其治疗的研究进展[J].中国中医眼科杂志,2003,14(1):59~61)。轻度视疲劳会在用眼后出现眼部酸、胀等眼部症状,休息后很快恢复;中度视疲劳会有明显的眼部症状,影响学习和工作;重度视疲劳除有明显的眼部症状和视作业不能持久外,还有记忆力减退、失眠等全身不适症状,干扰患者的视觉和生活质量,严重影响学习、工作和生活(参见:马越.花青素饮料缓解视疲劳作用的人体试验观察.中国食品学报[J].2015,8(15):8)。
大量的临床实践证明,屈光不正是视疲劳的主要原因之一,如远视、散光、近视患者,在未予矫正屈光不正时,看远或看近时动用较大的调节使睫状肌持续紧张造成患者的调节性视疲劳。另外,长时间近距离工作、工作或阅读环境照明不足等因素都可以导致人眼调节紧张,产生视疲劳。患者的精神状态和全身情况也与视疲劳的发生密切相关,精神压力大、体质衰弱的患者易产生视疲劳。
随着手机、平板电脑广泛进入人们的生活,人群全体用眼时间不断延长,近距离用眼强度增大,各年龄阶层视疲劳的发生率急速增多。能够改善视疲劳,并缓解由视疲劳带来的注意力、记忆力下降等问题的产品越来越受到消费者青睐。
目前,市面上改善视疲劳的产品通常仅针对蓝光损伤本身,对眼睛局部症状的改善效果不明显,而且并不能解决不同程度视疲劳引起的其他症状,如注意力不集中,记忆力下降等。因此,提供一种对视疲劳眼睛局部症状及精神神经症状缓解效果明显的产品,成为了一个亟待解决的问题。
发明内容
本发明的目的在于提供缓解视疲劳的组合物。
本发明提供了缓解视疲劳的组合物,活性成分包括鱼眼球组织或其提取物、鱼眼窝组织或其提取物、茶氨酸和γ-氨基丁酸。
进一步地,所述活性成分的重量配比为:鱼眼球组织10~1000份、鱼眼窝组织10~1000份、茶氨酸20~1000份、γ-氨基丁酸10~1000份。
进一步地,所述活性成分的重量配比为:鱼眼球组织100~800份、鱼眼窝组织100~800份、茶氨酸100~400份、γ-氨基丁酸70~500份。
优选地,所述活性成分的重量配比为:鱼眼球组织300~600份、鱼眼窝组织300~600份、茶氨酸200~400份、γ-氨基丁酸100~300份。
进一步优选地,所述活性成分的重量配比为:鱼眼球组织:鱼眼窝组织:茶氨酸:γ-氨基丁酸=1:1:(1~2):1。
所述重量配比以鱼眼球组织、鱼眼窝组织鲜品重量计。

所述原花青素来源于葡萄籽、葡萄、山楂、花生、银杏、蓝莓、黑豆、油菜、油茶、稻、高粱、可可、蚕豆、扁豆、荔枝、沙棘、苹果、黑莓、野草莓、桂圆、越橘、沙枣、酸果蔓、柿子、山竹、石榴、柏树、松树、银合欢、白桦树、桉树、钩藤毛白杨、肉桂、莽草、茶叶、葡萄藤、山玄参、贯叶连翘、月桂、桉树、玫瑰、牵牛、紫斑蕹菜、红车轴草、睡莲、巴豆、地榆、紫甘薯、野葛、大黄、异叶木属植物中一种或两种以上。

具体实施方式
本发明具体实施方式中使用的原料、设备均为已知产品,通过购买市售产品获得。
本发明提供了缓解视疲劳的组合物,其活性成分包括鱼眼球组织或其提取物、鱼眼窝组织或其提取物、茶氨酸和γ-氨基丁酸。
鱼眼睛的构造较为简单,大多数鱼无眼睑而眼球完全裸露,鱼眼眼球外观呈椭球状,位于由眶上骨、若干眶小骨及软颅围成的眼眶内。光镜下一般海鱼的鱼眼由外至内分为角膜-巩膜层、虹膜-脉络膜层、晶状体及视网膜层。眼球前面为透明的角膜,其余为棕黑色的巩膜。眼球内侧由肌肉、视神经、血管及软骨(眼柄)构成。鱼眼睛的大小和它日常所接触的光线很有关系。生活于水的上层或中层的鱼,它们都有正常的眼睛,如鲤鱼、鲫鱼、黄鱼等鱼类。而生活在300~1500米深处的鱼类,因这里的光线极弱,一般眼睛都比较大,如大眼鲷鱼。鱼眼睛富含维生素、活性多肽、氨基酸、糖类、多种微量元素(钙、磷、铁、锌等)以及包括ω-3脂肪酸在内的多种不饱和脂肪酸,这些物质对眼睛的新陈代谢有综合性有益影响。
鱼眼窝组织是位于鱼眼睛周围的肌肉组织和脂肪组织,其中,鱼眼窝脂肪组织(眼窝脂肪是处于眼球背后的结构脂肪的一种,为保护视神经而存在,起到保护眼球免遭高水压伤害的缓冲器作用,同时又成为脑、视网膜所必需的DHA供给源)油脂含量高,尤其是海鱼鱼眼窝脂肪组织中含有大量的多不饱和脂肪酸,其中的ω-3高度不饱和脂肪酸系列(主要是DHA、EPA)具有特殊的生理功效。DHA是神经系统生长及维持的一种主要元素,是大脑和视网膜的重要构成成分,在人体大脑皮层中含量高达20%,在眼睛视膜中所占比例最大,约占50%。DHA被誉为“脑黄金”和“聪明的原动力”,但人体不能自身合成DHA,只能从食物中摄取。
目前,仅见鱼眼球组织、鱼眼窝组织或其提取物单独应用的报道,用途主要为抗血栓、增视明目等,但尚未见鱼眼球组织或鱼眼窝组织缓解视疲劳作用的相关研究,更未见将两者组合使用的报道。
本发明通过人群试用证明,鱼眼球组织、鱼眼窝组织均具有显著的缓解视疲劳的作用,对视疲劳引起的眼酸痛、眼胀、畏光、视物模糊、异物感、流泪等症状都能够起到一定程度的缓解作用,而且,将鱼眼球组织和鱼眼窝组织组合使用后,两者富含的益眼成分在功效上得到了互补,能够发挥协同增效的作用,使得缓解视疲劳的效果得到进一步提升。
另外,本发明组合物中还加入活性成分茶氨酸和γ-氨基丁酸。
茶氨酸可以抑制谷氨酸的毒性,保护神经元;具有促进脑波中α波产生的功能,从而引起轻松、愉快的感觉,缓解抑郁和焦虑情绪;降血压;促进注意力集中,增进反应速度和准确性。
γ-氨基丁酸是哺乳动物中枢神经系统中最重要的抑制性神经递质,在生物体内参与多种神经生理活动,具有调节血压、改善脑机能、抗惊厥、促进精神安定、营养神经细胞等生理功能。在人脑中,虽然γ-氨基丁酸可由脑部的谷氨酸在脱羧酶的作用下转化而成,但是年龄的增长和精神压力加大使γ-氨基丁酸的积累异常困难,而通过日常饮食补充可有效的改善这种状况。
目前市售产品只有针对眼睛局部症状的抗氧化剂或叶黄素和/或3R,3'R)-二羟基-β-胡萝卜素,起效慢(1~2月)对中重度视疲劳带来的全身症状没有有效的解决方案。同时,由于消费者很难感受到效果而不易坚持服用。
本发明通过将茶氨酸和γ-氨基丁酸作为活性成分添加到含有鱼眼球和鱼眼窝的产品中,能够明显加速鱼眼对眼睛局部症状的缓解效果,服用1天后即可显效,而且长期服用后对眼睛局部症状的缓解效果较单用鱼眼更佳。此外,加入茶氨酸和γ-氨基丁酸后还能明显改善注意力和记忆力等视疲劳全身症状,包括可以减轻或缓解注意力及记忆力下降问题,试用者普遍反映使用后情绪焦虑缓解明显,由眼疲劳引发的睡眠问题也明显缓解。
本发明在提高缓解视疲劳功效同时,使消费者能最快体验到效果,显著提高消费者使用意愿,从而使视疲劳的缓解变得轻松简单。
实施例1鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼球提取物(以DHA为指标成分)的制造
原料处理:取新鲜海鱼鱼眼(眼球组织)用水洗净,沥干后,在0.05-0.1%的柠檬酸溶液中浸泡10-30min,于40-70℃烘干至含水分小于4%,再粉碎至100-200目的海鱼粉;
酶解:将经处理后的海鱼粉加缓冲液用6mol/L盐酸调pH值为4-6,加入质量分数0.1-1.0%,酶活3.0×104u/g的木瓜蛋白酶和质量分数0.5-1.5%,酶活1.0×104u/g的纤维素酶,于40-60℃搅拌酶解1-2h,之后用0.5mo1/L氢氧化钠调pH至中性;
真空提取:将得到的海鱼酶解液置于真空提取设备中,控制真空度85-90KPa,温度40-60℃提取1-2小时,同时灭酶,得到提取液;
分离:将提取液于6000rpm离心20分钟,收集上清液;
浓缩:将得到的液体置于真空浓缩设备中,控制真空度85-90KPa,温度40-60℃浓缩至原体积的l/5-1/3,得到DHA浓缩液。
实施例2鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼等鱼眼窝提取物(以DPA、EPA和DHA为指标成分)的制造
将海鱼加工后的眼窝洗净,以固液重量比是1:1-1.2的比例加入水,再磨成浆状得到匀浆;取匀浆用有机溶剂萃取1-3次,有机溶剂与匀浆体积之比为1:1.2-1.5,收集萃取液,得到含DPA、EPA和DHA油脂的萃取液备用,所述有机溶剂指的是石油醚或乙醚;萃取液回收有机溶剂,得到含DPA、EPA和DHA的鱼油,取与鱼油等体积的0.3mol/L甲醇钠混合,室温条件下充氮搅拌10分钟后,调节pH值到7.0-7.2,回收甲醇,得到甲酯化的鱼油;对甲酯化鱼油进行分子蒸馏,包括脱气、预热蒸馏及蒸馏三个技术步骤,其中脱气步骤在温度20-30℃、系统压力在1-2mbar的条件下进行脱气;预热蒸馏步骤在鱼油以15-20公斤/小时的进料速度送入预热级蒸馏器中,在温度170-200℃、系统压力0.001-0.01mbar、刮膜转子速度150-250转/分钟的条件下蒸馏;蒸馏步骤以5-20公斤/小时的进料速度送入蒸馏器中,在温度250-350℃、系统压力0.001-0.01mbar、刮膜转子速度150-250转/分钟的条件下蒸馏,收集290-310℃时的馏分,即得到较高纯度的含EPA、DHA以及DPA的复合物。
实施例3本发明组合物的制备
原料重量配比:鱼眼窝组织粉碎物或提取物300~600份(以鲜品重量计)、鱼眼球组织粉碎物或提取物300~600份(以鲜品重量计)、茶氨酸200~400份、γ-氨基丁酸100~300份、花青素50~200份(或者虾青素:4~12份和或原花青素1-200份)、(3R,3'R)-二羟基-β-胡萝卜素1-5份、叶黄素和/或叶黄素酯5~15份。
制备方法:
1、按重量配比将鱼眼窝组织粉碎物或提取物(以鲜品重量计)、鱼眼球组织粉碎物或提取物(以鲜品重量计)、茶氨酸、γ氨基丁酸、花青素(或虾青素和或原花青素)、(3R,3'R)-二羟基-β-胡萝卜素、叶黄素酯混合均匀后加入糖浆搅拌均匀。
2、注模冷却成型后可按需制成硬质、奶糖、夹心、酥质、焦香(太妃)、充气、凝胶、胶基、压片、流质、膜片、花式等糖果类型,以及巧克力及巧克力制品、代可可脂巧克力和代可可脂巧克力制品、果冻(果汁型、果肉型、果味型、乳汁型等)等糖果制品。
实施例4本发明组合物的制备
原料重量配比:鱼眼窝组织粉碎物或提取物200-500份(以鲜品重量计)、鱼眼球组织粉碎物或提取物200-500份(以鲜品重量计)、茶氨酸200~400份、γ氨基丁酸100~300份、花青素50~200份(或者虾青素:4~12份和或原花青素1-300份)、(3R,3'R)-二羟基-β-胡萝卜素1-10份、叶黄素和/或叶黄素酯1~25份。
制备方法:选择鱼眼窝组织粉碎物或提取物、鱼眼球组织粉碎物或提取物、茶氨酸、γ氨基丁酸、花青素(或原花青素和或雨生红球藻提取物)、(3R,3'R)-二羟基-β-胡萝卜素、叶黄素酯的原料混合,加入所需的处理水和30-45液态奶进行充分搅拌、溶解、均质,均质压力为25Mpa,再进行超高温灭菌,无菌包装,经检验合格后入库。
实施例5本发明组合物的制备
原料重量配比:鱼眼窝组织粉碎物或提取物300-600份(以鲜品重量计)、鱼眼球组织粉碎物或提取物300-600份(以鲜品重量计)、γ氨基丁酸100~400份、花青素50~200份(或者虾青素:4~12份和或原花青素1-400份)、(3R,3'R)-二羟基-β-胡萝卜素0.2-10份、叶黄素和/或叶黄素酯0.6~25份。
制备方法:选择鱼眼窝组织粉碎物或提取物、鱼眼球组织粉碎物或提取物、茶氨酸、蓝莓提取物,搅拌均匀,臭氧灭菌,灌装,可直接冲服或作为功能性辅料加入到其它食品中。
以下通过试验例证明本发明的有益效果。以下所述鱼眼球组织提取物使用根据实施例1方法制备的罗非鱼眼球组织提取物,鱼眼窝组织提取物使用根据实施例2方法制备的罗非鱼眼窝组织提取物。
试验例1鱼眼球组织和/或鱼眼窝组织缓解视疲劳及其相关症状的试用观察
设置3个试验组,分别按表2所示取鱼眼球组织提取物、鱼眼窝组织提取物,括号内表示对应的新鲜鱼眼球组织、鱼眼窝组织的重量,将其装入等大不透明胶囊,供眼疲劳患者试服,连续服用60天(表2对应每日服用量),观察试用效果。
根据《视疲劳诊疗专家共识》(2014年)诊断为视疲劳患者144例,症状包括视觉障碍:近距离工作或阅读不持久,出现暂时性视物模糊或重影;眼部不适:眼胀、眼痛、眼烧灼感、流泪、眼痒、眼异物感及眼眶疼痛。全身症状:易疲劳,头痛、头晕,记忆力减退,记忆力下降,焦虑,烦躁,以及其他神经官能症的症状。年龄18-65岁,随机分3组。根据国食药监保化[2012]107号《缓解视疲劳功能评价方法》制订视疲劳评定量表,见表1,计算服用前、服用60天积分降低率:(治疗前总积分-治疗后总积分)/治疗前总积分×100%,治疗后积分降低率≥30%为有效。症状积分改善有效率统计结果见表2。

试验例2本发明组合物缓解视疲劳及其相关症状的试用观察
设置9个试验组,分别按表3所示用量取鱼眼球组织提取物、鱼眼窝组织提取物、γ-氨基丁酸和茶氨酸,括号内表示对应的新鲜鱼眼球组织、鱼眼窝组织、γ-氨基丁酸和茶氨酸的重量,将其装入等大不透明胶囊,供眼疲劳患者试服,连续服用60天(表3对应每日服用量),观察试用效果。
根据《视疲劳诊疗专家共识》(2014年)诊断为视疲劳患者234例,症状包括视觉障碍:近距离工作或阅读不持久,出现暂时性视物模糊或重影;眼部不适:眼胀、眼痛、眼烧灼感、流泪、眼痒、眼异物感及眼眶疼痛。全身症状:易疲劳,头痛、头晕,记忆力减退,记忆力下降,焦虑,烦躁,以及其他神经官能症的症状。年龄18-65岁,随机分9组。根据国食药监保化[2012]107号《缓解视疲劳功能评价方法》制订视疲劳评定量表,见表1。全身症状进行单项评分:注意力下降评价(0分:能坚持专注工作或学习30分钟;1分:偶有注意力下降;2分:时有注意力下降;3分:经常有注意力下降);记忆力下降评价(0分:昨日事情全部能回忆;1分:偶有记忆力下降;2分:时有记忆力下降;3分:经常有记忆力下降);注:“偶有”是指1-2次/2天;“时有”是指1-3次/天;“经常”是指>3次/天。计算服用1天、60天积分降低率:(治疗前总积分-治疗后总积分)/治疗前总积分×100%,治疗后积分降低率≥30%为有效。症状积分改善有效率统计结果见表3。

试验组1仅添加了鱼眼球及鱼眼窝组织提取物,试用结果表明其具有显著的缓解视疲劳眼睛局部症状的效果,服用60天后改善率达到88%,但起效较慢,服用1天有效率仅为20%。
试验组2~9在鱼眼球及鱼眼窝组织提取物的基础上添加了活性成分γ-氨基丁酸和茶氨酸,结果明显加速了对眼睛局部症状的缓解效果,服用1天后即可显效,而且长期服用后对眼睛局部症状的缓解效果较试验组1更佳(对比试验组1、2可知)。此外,加入γ-氨基丁酸和茶氨酸的试验组还能明显改善注意力和记忆力情况。
从试验组2~9可以看出,鱼眼球组织、鱼眼窝组织和茶氨酸、γ-氨基丁酸在本发明限定的配比范围内(即鱼眼球组织10~1000份,鱼眼窝组织10~1000份,茶氨酸20~1000份、γ-氨基丁酸10~1000份)组合使用,均能够发挥明显的缓解视疲劳的作用,对影响视疲劳人群的眼睛局部症状和全身症状均有明显疗效。其中,优选比例为:鱼眼球组织或其提取物300~600份、鱼眼窝组织或其提取物300~600份、茶氨酸200~400份、γ-氨基丁酸100~300份。

1.缓解视疲劳的组合物,其特征是:活性成分包括鱼眼球组织或其提取物、鱼眼窝组织或其提取物、茶氨酸和γ-氨基丁酸。
2.如权利要求1所述的组合物,其特征是:所述活性成分的重量配比为:鱼眼球组织10~1000份、鱼眼窝组织10~1000份、茶氨酸20~1000份、γ-氨基丁酸10~1000份。
3.如权利要求2所述的组合物,其特征是:所述活性成分的重量配比为:鱼眼球组织100~800份、鱼眼窝组织100~800份、茶氨酸100~400份、γ-氨基丁酸70~500份;优选地,所述活性成分的重量配比为:鱼眼球组织300~600份、鱼眼窝组织300~600份、茶氨酸200~400份、γ-氨基丁酸100~300份;进一步优选地,所述活性成分的重量配比为:鱼眼球组织:鱼眼窝组织:茶氨酸:γ-氨基丁酸=1:1:(1~2):1。
4.如权利要求1~3任意一项所述的组合物,其特征是:所述鱼眼球组织或鱼眼窝组织选自鲔鱼、金枪鱼、鲣鱼、竹荚鱼、罗非鱼、魟鱼、鳕鱼、鲭鱼、坚鳞鲈、胡瓜鱼、三文鱼、玉筋鱼、金线鱼、安康鱼、大白鲨、鲂鱼、明太鱼、东方鲈、鲤鱼、鲫鱼、青鱼、枪鱼、真鲷、鲻鱼、马鲛鱼、鲇鱼、银鱼、秋刀鱼、翻车鱼、黑鲷、带鱼、桂鱼、欧洲鲈鱼、泥鳅、乌鱼、叫姑鱼、东方黄鲂鮄、剑鱼、暗纹东方鲀、黄鳍金枪鱼、灰鲭鲨、黄花鱼、河豚、鲽鱼、鼠鱼、鲥鱼、黑鮶鱼、沙丁鱼、斑鳍光鳃鱼、远东多线鱼、马头鱼、鳗鱼、鲳鱼、黄姑鱼中一种或两种以上鱼类的眼球组织或眼窝组织。
5.如权利要求1~3任意一项所述的组合物,其特征是:所述提取物为水提物或醇提物。
6.如权利要求1~5任意一项所述的组合物,其特征是:活性成分还包含花青素、虾青素、原花青素、叶黄素、叶黄素酯、(3R,3'R)-二羟基-β-胡萝卜素中一种或两种以上的混合物。
7.如权利要求1~6任意一项所述的组合物,其特征是:是各重量配比的活性成分,加入可接受的辅料或者辅助性成分制备而成的食品、保健品或药品。
8.如权利要求7所述的组合物,其特征是:所述食品、保健品或药品为口服形式。
9.如权利要求8所述的组合物,其特征是:所述食品或保健品为糖果、饮料、饼干、面包、果冻或膳食补充剂。
10.权利要求1~9任意一项所述组合物的制备方法,其特征是:包括如下步骤:取各重量配比的活性成分,加入食品、保健品或药品中可接受的辅料或者辅助性成分,即得。
11.权利要求1~9任意一项所述组合物在制备缓解视疲劳的食品、保健品或药品中的用途。
12.如权利要求11所述的用途,其特征是:所述食品、保健品或药品具有缓解视疲劳所致眼睛局部症状和/或精神神经症状的功效。
13.如权利要求12所述的用途,其特征是:所述眼睛局部症状为眼酸痛、眼胀、畏光、视物模糊、眼干涩、异物感或流泪。
14.如权利要求12所述的用途,其特征是:所述精神神经症状为注意力下降、记忆力减退、焦虑或失眠。
