DPPH free radical scavenging and ABTS free radical scavenging experiments for a plant antioxidant composition:

The samples were added to a 96-well plate, 3 replicate wells were set, 2 dilution gradients per sample, 10-fold dilution was performed (4 dilution gradients per sample after primary screening if the samples had significant DPPH free radical scavenging effect, 2-fold dilution was performed for replicate wells), 100 μ L per well, 0.1M DPPH 100 μ L per well. The negative control used 50% ethanol instead of sample, and the blank used 50% ethanol instead of DPPH, and the rest of the procedure was the same. The reaction solution was mixed well and incubated for 30 minutes at room temperature in the dark. And reading the absorbance value at 517nm by using a microplate reader, wherein the lower the delta OD value is, the higher the scavenging activity of the sample on free radicals is. 1.2 ABTS free radical scavenging experiments
The experiment was performed by absorptiometry. Adding samples into a 96-well plate, setting 3 repeated wells, diluting each sample by 2 dilution gradients, performing 10-fold dilution (after preliminary screening, if the samples have obvious ABTS free radical scavenging effect, each sample has 4 dilution gradients, performing 2-fold dilution, performing secondary screening), each well has 100 μ L,100 μ L of 0.1M ABTS was added to each well. The negative control uses 50% ethanol instead of sample, the blank uses 50% ethanol instead of ABTS, and the rest of the procedure is the same. The reaction solution was mixed well and incubated for 30 minutes at room temperature in the dark. trolox (Sigma, cat # 238813-1G, lot # BCBP2767V), DPPH (Sigma, cat # D9132-1G, lot # STBF52 5255V), ABTS (Sigma, cat # A1888-1G, lot # SLBN7932V), K2S2O8(Sigma, cat # P112194-25G, lot # F1508005), sodium dihydrogen phosphate (Sjogren Kagaku K.K.: 1202011), disodium hydrogen phosphate (Sjogren Kagaku K.K.: 111021), absolute ethanol (Rianlongbo (Tianjin) pharmaceutical Co., Ltd.: D4540-100ML, lot # 20150205), DMSO (volume fraction of DMSO in the final reaction system)<1 percent), Multiskan GO enzyme-linked immunosorbent assay (Thermo), PRACTUM224-1CN electronic balance (Satorius), GNP-9270 water-proof constant-temperature incubator (Shanghai sperm macroexperimental equipment Co., Ltd.), 30-300 mu L of discharge gun (Thermo Co., Ltd.), and liquid-transfering gun (Dalong Co., Ltd.) with various measuring ranges.
3. The experimental results are as follows:

Claims (1)
