Description

Scalp nourishing composition, preparation method and application thereof, and scalp nourishing shampoo
Technical Field
The invention relates to the technical field of daily necessities, in particular to a scalp nourishing composition, a preparation method and application thereof, and scalp nourishing shampoo.
Background
Hair washing and caring products become necessities of daily life of people and are used for cleaning and caring hair. At present, shampoo, hair conditioner and other washing and protecting products in the market are various in variety, but most products attach importance to the effect of hair, neglect the protection of scalp, cannot improve the hair quality fundamentally and destroy the ecological environment of the scalp.
The ecological environment of the scalp is maintained mainly by three major balances: lipid, flora and metabolic balance. Wherein, the imbalance of the grease secretion of the scalp can cause the scalp to be greasy, and further cause the hair to be greasy; when the scalp flora environment is unbalanced and a large amount of harmful bacteria are bred, the phenomenon of itching can be caused; an imbalance in scalp stratum corneum metabolism can lead to the production of dandruff.
Some products are added with partial powerful chemical reagents for enhancing the effects of cleaning, softening and relieving itching, and the substances easily cause scalp injury and can cause serious imbalance of scalp ecological environment after long-term use.
For example, some products are added with silicone oil in order to make washed hair more beautiful and smooth, but the silicone oil is synthetic polymer which is extremely difficult to degrade, and excessive use of the silicone oil can cause silicone oil accumulation on scalp, so that not only can the secretion of grease be unbalanced, but also the substances can stimulate a large amount of proliferation of harmful bacteria to cause phenomena such as itching of the head; some products are added with antifungal agents such as climbazole and piroctone olamine in order to increase the effect of relieving itching, and although the addition of the antifungal agents can inhibit the propagation of bacteria to a certain extent and achieve the effect of relieving itching, the products easily cause scalp allergy, easily cause imbalance of scalp grease secretion, and even aggravate the occurrence of alopecia, hair breakage and the like.
Therefore, in order to improve the hair quality fundamentally and return the scalp and hair to a healthy state, it is necessary to maintain the scalp ecological environment.

Performance detection
(1) Expression of TLR9 (Toll-like receptor 9)
The principle is as follows: toll-like receptors (TLRs) are an important class of protein molecules involved in nonspecific immunity (innate immunity) and also serve as a bridge for connecting nonspecific immunity and specific immunity. TLRs are single transmembrane non-catalytic proteins that can recognize molecules of conserved structure derived from microorganisms. Studies have shown that malassezia furfur and malassezia globosa can be recognized by TLR9 in keratinocytes.
Test samples: examples 1 to 7 and comparative examples 1 to 6 are scalp moisturizing compositions;
sample pretreatment: the scalp moisturizing compositions in examples 1 to 7 and comparative examples 1 to 6 were prepared into a 1 μ M solution with DMSO;
the experimental method comprises the following steps: scalp explants from 1 female donor (36 years old) were divided into 14 groups of 13 for testing samples, 1 group was used as a blank, and the scalp explants (5mm diameter) of each group were treated as follows: contacting malassezia furfur suspension in culture medium for 3 days, removing the malassezia furfur suspension, adding pre-treated sample or placebo (pure DMSO), incubating for 3 days, rinsing, cutting the skin and labelling only TLR-9 on these sections, determining the rate of change relative to TLR9 when untreated; the results are shown in Table 3 below.
TABLE 3
Figure GDA0002499508680000151
*=p<0.01
As can be seen from the blank in Table 3, the TLR-9 labelling of the explants was improved upon contact with Malassezia furfur.
As can be seen from examples 1 to 7 and comparative examples 1 to 6 in table 3, the addition of the compositions of examples 1 to 7 and comparative examples 1 to 6 after contact with malassezia furfur reduced the labeling of TLR9 to a different extent, wherein the effect of the compositions of examples 1 to 7 was more significant and significantly higher than that of comparative examples 1 to 6. It is illustrated that the nourishing scalp compositions of examples 1 to 7 have the effect of regulating the ecological balance of the scalp.
The nourishing scalp composition of comparative example 1 lacks celery seed extract and has a greater concentration of willowherb extract than example 3. as can be seen from table 3, the TLR9 marker of example 3 is reduced by a greater amount than in comparative example 1. Also, the composition of comparative example 2 lacks willowherb extract and has a greater concentration of celery seed extract than in example 3, and it can be seen from table 3 that the TLR9 marker of example 3 is reduced by a greater magnitude than in comparative example 2. It is demonstrated that the absence of celery seed extract or willowherb extract both reduce the bacteriostatic effect of the composition, and that celery seed extract and willowherb extract have a synergistic effect in the above scalp nourishing compositions.
In addition, as can be seen from comparative examples 3-5, the lack of hydrolyzed keratin and/or allantoin all contribute to the effectiveness of the composition. As can be seen from comparative example 6, when the composition reaches a certain concentration, even if the concentration of each component is increased, the bacteriostatic effect is not greatly influenced.
(2) Market research
And (3) testing a product: shampoos of examples 1-7, comparative example 1, comparative example 2, and comparative example 5
The tester: 200 persons, between 23 and 60 years of age, 100 male, 100 female, divided into 10 groups of 20 persons each, 10 persons each, oily scalp and dry scalp;
examples 1 to 5 correspond to groups 1 to 7, respectively; comparative example 1 was group 8, comparative example 2 was group 9, and comparative example 5 was group 10;
the test method comprises the following steps:
firstly, all testers take a small amount of samples to be tested to coat the samples on ears, the samples are kept stand for 20min, and if discomfort symptoms exist, red swelling and eruption symptoms exist, the test results show that all testers do not have adverse symptoms, and the subsequent test can be carried out;
the cleaning agent is used once every two days, is continuously used for three months, is not used by other similar products during the test period, and evaluates the cleaning effect, the softening effect, the itching relieving effect, the dandruff removing effect and the irritation of the product; and the corresponding shampoo was used for cleaning two days before evaluation, and no product was used thereafter. And counting the average score of each group after scoring.
Evaluation criteria:
very preferably 10 minutes, preferably 7 to 9 minutes, preferably 4 to 6 minutes, by 1 to 3 minutes;
TABLE 4
Figure GDA0002499508680000171
After the test of the groups 8 to 10, the shampoo of example 3 was used again once every two days for three months, and the test results are shown in table 5.
TABLE 5
Figure GDA0002499508680000172
As can be seen from groups 1-7 of Table 4, the shampoos of examples 1-7 had better cleansing, softening, anti-dandruff, and anti-itch effects, and were non-irritating.
As can be seen by combining the groups 8 and 9 of tables 4 and 5, the anti-dandruff and anti-itching effects of example 3 were significantly better than those of comparative examples 1 and 2, excluding the individual differences.
As can be seen by combining the 10 th groups of tables 4 and 5, the cleaning and softening effects of example 3 are significantly better than those of comparative example 5, excluding individual differences.
In addition, fig. 1 is a photograph taken at a scalp microspur of a volunteer (female 32 years old, oily scalp) in group 8 using the shampoos of comparative example 1 and example 3, a is a photograph taken three months after using the shampoo of comparative example 1, and B is a photograph taken three months after using the shampoo of example 3, and it can be seen that the dandruff condition was significantly improved after using the shampoo of example 3.
