背景技术
“三高症”是指高血压、高血糖(糖尿病)和高脂血症。是现代社会所派生出来的“富贵病”,可能单独存在,也可能相互关联。由于长时间饮食习惯问题,饮食中脂类过多,醇类过多。同时又没有合理的运动促进脂类醇类的代谢,导致体内脂类醇类物质逐渐增多,掺杂在血液中,使毛细血管堵塞,随着时间的推移,脂类醇类物质容易和体内游离的矿物质离子结合,形成血栓。血栓容易在血管的拐角处,或者瓶颈处堆积,钙化。同时血栓越来越多,使血管直径缩小。心脏为了保持足够的供血量,就增加血压,造成高血压疾病。血脂主要是指血清中的胆固醇和甘油三酯。无论是胆固醇含量增高,还是甘油三脂的含量增高,或是两者皆增高,统称为高脂血症。高血脂可以引起血管栓塞;高血压可以引起脑出血和脑血管破裂;高血糖可以引起糖尿病。目前三高人群数量在不断增加,而治疗方法主要是依靠中西医药物,辅助饮食控制和运动。
如中国专利CN201410324784.3,CN201710609596 .9,CN201710744781 .9,CN201710777929 .9,CN201710883481 .9均公开了具有降三高效果的保健品,公知的中医方面主要有效的成分有雪莲果,甜菜碱,杜仲,葛根,茯苓,玉米,苹果,牛蒡,苦荞等,医药结合食物控制仅能针对某一种进行控制,而且经常引起低血压、低血糖等营养不足的问题。
发明内容
本发明的目的是解决上述难于一种药物三高一起调节,无副作用的问题,提供一种辅助降三高组合物,将综合调理与精准靶向治疗相结合,中医与高科技结合,能全面调节三高,做到“糖脂一体化,三高一起调”;同时还能改善肠道功能,提高机体免疫力,而且无激素、无药物依赖、食用更安全,从而达到标本兼治的最终效果。
本发明为解决上述问题所采用的技术方案是:一种辅助降三高组合物,由以下重量组分的原料制成:海参低聚肽5-20份、玉米低聚肽5-20份、决明子提取物3-9份、山楂提取物5-10份、人参提取物2-6份、山药提取物3-9份、葛根提取物5-10份、石斛提取物2-6份、茯苓提取物3-9份、桃仁提取物3-9份、L-阿拉伯糖3-9份、低聚木糖5-10份、膳食纤维5-10份、茉莉花2-6份、木糖醇2-6份、营养酵母粉1-2份。

海参低聚肽的制备方法为:取海参粉碎制浆、加1-2倍纯水稀释,加入1.0%-5.0%的复合蛋白酶,在50℃-60℃、pH8-9条件下酶解3-6h,灭酶、离心后,上清液采用1000Da-8000Da的纳滤膜分离,收集分子量小于1000Da透过液,采用电渗析脱盐,然后加入β环状糊精和活性炭精制,精制液通过膜浓缩设备浓缩,最后经低温喷雾干燥得到海参低聚肽。
玉米低聚肽的制备方法为:取玉米蛋白粉加纯水调浆至底物浓度为3%-5%的溶液,加入1.0%-5.0%的复合蛋白酶,在50℃-60℃、pH8-9条件下酶解3-6h,灭酶、离心后,上清液采用1000-8000Da的纳滤膜分离,收集分子量小于1000Da透过液,加入β环状糊精和活性炭精制,精制液通过膜浓缩设备浓缩,最后经低温喷雾干燥得到玉米低聚肽。
决明子提取物的制备方法为:取决明子原料,粉碎后置于提取罐,加入50%-60%的乙醇回流提取2-3次,每次1小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到决明子提取物。
山楂提取物的制备方法为:取山楂原料,粉碎后置于提取罐,加5-10倍纯水回流提取2次,每次提取1-2小时,合并提取液后过滤,滤液减压浓缩,浓缩液加入乙醇醇沉,使含乙醇量为60%-70%,静置后离心,取上清液浓缩,经喷雾干燥得山楂提取物。
人参提取物的制备方法为:取人参原料,粉碎后置于提取罐,加入60%-70%的乙醇回流提取2-3次,每次1.5小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到人参提取物。
山药提取物的制备方法为:取山药原料,去皮、切块、磨浆后置于提取罐,加入5-10倍纯水回流提取2次,每次提取1-2小时,合并提取液后过滤,滤液减压浓缩,浓缩液加入乙醇醇沉,使含乙醇量为70%-80%,静置后离心,取沉淀冷冻干燥、粉碎、过80目筛即得山药提取物。
葛根提取物的制备方法为:取葛根原料,粉碎后置于提取罐,加入60%-70%的乙醇回流提取2-3次,每次1.5小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到葛根提取物。
石斛提取物的制备方法为:取石斛原料,粉碎后置于提取罐,加入5-10倍纯水回流提取2次,每次提取1-2小时,合并提取液后过滤,滤液减压浓缩,浓缩液加入乙醇醇沉,使含乙醇量为50%-60%,静置后离心,取上清液浓缩,经喷雾干燥得石斛提取物。

L-阿拉伯糖是2008年国家卫生部批准的新资源食品,是一种新型的低热量甜味剂,可以抑制人体肠道内蔗糖酶的活性,从而具有了抑制蔗糖吸收的功效;此外,L-阿拉伯糖还可以抑制身体脂肪堆积,可被用于防治肥胖、高血压、高血脂等疾病。日本的厚生省将L-阿拉伯糖列入“调节血糖的专用特殊保健食品添加剂”,美国医疗协会将L-阿拉伯糖列入“抗肥胖的营养补充剂或非处方药”。
低聚木糖是由2-7个木糖分子以β-1,4糖苷键结合而成的功能性聚合糖,是聚合糖类中增殖双歧杆菌功能最强的品种之一,它的功效性是其他聚合糖类的近20倍。低聚木糖可以显著的使双歧杆菌大量增殖,从而达到使益生菌成为肠道优势菌种,排除有害菌的目的,具有减少有毒发酵产物及有害细菌酶的产生,抑制病原菌和腹泻,防止便秘,保护肝脏功能,降低血清胆固醇和血压,增强机体免疫力的保健功效。而且低聚木糖很难为人体消化酶所分解,既不影响血糖浓度,也不增加血糖中胰岛素水平,并且不会形成脂肪沉积,因此糖尿病人、肥胖病人和低血糖病人均可放心食用。
膳食纤维被营养学界补充认定为第七类营养素,食用后具有保持肠胃通畅的功能,增强通便、降低肠道癌的风险;还具有结合清除体内的胆酸,明显降低血清胆固醇,更容易引致饱腹感,明显降低饭后血糖水平等功能。
本发明有益效果显著:本发明采用“降三高”效果显著的海参低聚肽、玉米低聚肽,辅以决明子提取物、山楂提取物、人参提取物、山药提取物、葛根提取物、石斛提取物、茯苓提取物、桃仁提取物等滋阴润燥、健脾利湿、调理肝肾与气血的传统“药食同源”原料,改善肠道健康的低聚木糖、膳食纤维,以及L-阿拉伯糖、茉莉花、木糖醇、营养酵母粉配制而成,将综合调理与精准靶向治疗相结合,中医与高科技结合,能全面调节三高,做到“糖脂一体化,三高一起调”;同时还能改善肠道功能,提高机体免疫力,从而达到标本兼治的最终效果。本发明组方采用低聚肽与药膳同源的食品原料进行科学配伍,达到高效利用,见效快,辅助降三高(血糖、血压、血脂)效果显著,而且无激素、无药物依赖、食用更安全。

具体实施方式
下面结合具体实施例对本发明做详细说明,但本发明并不局限于具体实施例:
实施例1:一种辅助降三高组合物,由以下重量组分的原料制成:海参低聚肽5-20份、玉米低聚肽5-20份、决明子提取物3-9份、山楂提取物5-10份、人参提取物2-6份、山药提取物3-9份、葛根提取物5-10份、石斛提取物2-6份、茯苓提取物3-9份、桃仁提取物3-9份、L-阿拉伯糖3-9份、低聚木糖5-10份、膳食纤维5-10份、茉莉花2-6份、木糖醇2-6份、营养酵母粉1-2份。
将上述原料按照比例分别进行处理获得。
海参低聚肽的制备方法为:取海参粉碎制浆、加1-2倍纯水稀释,加入1.0%-5.0%的复合蛋白酶,在50℃-60℃、pH8-9条件下酶解3-6h,灭酶、离心后,上清液采用1000Da-8000Da的纳滤膜分离,收集分子量小于1000Da透过液,采用电渗析脱盐,然后加入β环状糊精和活性炭精制,精制液通过膜浓缩设备浓缩,最后经低温喷雾干燥得到海参低聚肽。
玉米低聚肽的制备方法为:取玉米蛋白粉加纯水调浆至底物浓度为3%-5%的溶液,加入1.0%-5.0%的复合蛋白酶,在50℃-60℃、pH8-9条件下酶解3-6h,灭酶、离心后,上清液采用1000-8000Da的纳滤膜分离,收集分子量小于1000Da透过液,加入β环状糊精和活性炭精制,精制液通过膜浓缩设备浓缩,最后经低温喷雾干燥得到玉米低聚肽。
决明子提取物的制备方法为:取决明子原料,粉碎后置于提取罐,加入50%-60%的乙醇回流提取2-3次,每次1小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到决明子提取物。
山楂提取物的制备方法为:取山楂原料,粉碎后置于提取罐,加5-10倍纯水回流提取2次,每次提取1-2小时,合并提取液后过滤,滤液减压浓缩,浓缩液加入乙醇醇沉,使含乙醇量为60%-70%,静置后离心,取上清液浓缩,经喷雾干燥得山楂提取物。
人参提取物的制备方法为:取人参原料,粉碎后置于提取罐,加入60%-70%的乙醇回流提取2-3次,每次1.5小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到人参提取物。
山药提取物的制备方法为:取山药原料,去皮、切块、磨浆后置于提取罐,加入5-10倍纯水回流提取2次,每次提取1-2小时,合并提取液后过滤,滤液减压浓缩,浓缩液加入乙醇醇沉,使含乙醇量为70%-80%,静置后离心,取沉淀冷冻干燥、粉碎、过80目筛即得山药提取物。
葛根提取物的制备方法为:取葛根原料,粉碎后置于提取罐,加入60%-70%的乙醇回流提取2-3次,每次1.5小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到葛根提取物。
石斛提取物的制备方法为:取石斛原料,粉碎后置于提取罐,加入5-10倍纯水回流提取2次,每次提取1-2小时,合并提取液后过滤,滤液减压浓缩,浓缩液加入乙醇醇沉,使含乙醇量为50%-60%,静置后离心,取上清液浓缩,经喷雾干燥得石斛提取物。
茯苓提取物的制备方法为:取茯苓原料,粉碎后置于提取罐,加入60%-70%的乙醇回流提取2-3次,每次1-2小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到茯苓提取物。
桃仁提取物的制备方法为:取桃仁原料,粉碎后置于提取罐,加入70%-80%的乙醇回流提取2-3次,每次1-2小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到桃仁提取物。
按照表1中指定的原料组分含量进行实验实施例1至实施例4中组分的制作方法,分别得到不同组分含量的组合物1至组合物4。

实施例5
一种辅助降三高组合物,颗粒剂的制备:按实施例1-4配方分别进行称取,投入混合机内混合30分钟,使其均匀,加入麦芽糖醇浆水溶液作为湿润剂,混合 30 分钟,混合后的物料用15目筛制粒,放入干燥设备内干燥,干燥颗粒用50目筛网筛分,去除粗颗粒和细粉,包装,即得。
实施例6
一种辅助降三高组合物,片剂的制备:按实施例1-4配方分别进行称取,投入混合机内混合30分钟,使其均匀,加入麦芽糖醇浆水溶液作为湿润剂,混合 10分钟,混合后的物料用10目筛制粒,放入干燥设备内干燥,加入干燥颗粒总量的1份的硬脂酸镁,整粒,压片、包装,即得。
实施例7
一种辅助降三高组合物,口服液的制备:按实施例1-4配方分别进行称取,加入40倍量纯化水溶解得溶液,加入溶液量0.05份的山梨酸钾,然后过滤、分装、灭菌、包装,即得。
实施例8
本发明一种辅助降三高组合物人体试服试验。
志愿者1:刘女士,64岁,糖尿病患者,伴有高血压,甘油三酯轻微超标。服用前:血糖8.29,血压100/175,甘油三酯2.3;服用四周后,“三高”症状均有所改善,特别是血糖和血压方面,血糖5.74,血压95/155,甘油三酯2.18,而且患者感觉身体轻松,免疫力提高,肠道状况改善,便秘症状消失。
志愿者2:邵先生,43岁,糖尿病,伴有高血压,甘油三酯严重超标。服用前:血糖16.02,血压100/160,甘油三酯6.17;服用四周后,血糖13.3,血压90/130,甘油三酯3.23,而且患者的头晕、头痛、易疲劳等症状改善明显,精神状态改善极佳,免疫力也有所提高。
经 225 位不同情况、不同程度的“三高”患者试用,人体试服效果如下附图1-3所示,显效的达180位,有效的为45位,无效的有25位,总有效率达 90%。
以上已以较佳实施例公开了本发明,然其并非用以限制本发明,凡采用等同替换或者等效变换方式所获得的技术方案,均落在本发明的保护范围之内。
Claims (6)
1.一种辅助降三高组合物,其特征在于,主要由以下重量组分的原料制成:海参低聚肽5-20份、玉米低聚肽5-20份、决明子提取物3-9份、山楂提取物5-10份、人参提取物2-6份、山药提取物3-9份、葛根提取物5-10份、石斛提取物2-6份、茯苓提取物3-9份、桃仁提取物3-9份、L-阿拉伯糖3-9份、低聚木糖5-10份、膳食纤维5-10份、茉莉花2-6份、木糖醇2-6份、营养酵母粉1-2份。
2.根据权利要求1所述的一种辅助降三高组合物,其特征在于,所述的一种辅助降三高组合物的剂型为颗粒剂、胶囊、片剂或口服液。
3.根据权利要求1或2所述一种辅助降三高组合物,其特征在于,其原料按下述方法制得:
海参低聚肽:取海参粉碎制浆、加1-2倍纯水稀释,加入1.0%-5.0%的复合蛋白酶,在50℃-60℃、pH8-9条件下酶解3-6h,灭酶、离心后,上清液采用1000Da-8000Da的纳滤膜分离,收集分子量小于1000Da透过液,采用电渗析脱盐,然后加入β环状糊精和活性炭精制,精制液通过膜浓缩设备浓缩,最后经低温喷雾干燥得到海参低聚肽;

山楂提取物:取山楂原料,粉碎后置于提取罐,加5-10倍纯水回流提取2次,每次提取1-2小时,合并提取液后过滤,滤液减压浓缩,浓缩液加入乙醇醇沉,使含乙醇量为60%-70%,静置后离心,取上清液浓缩,经喷雾干燥得山楂提取物;
人参提取物:取人参原料,粉碎后置于提取罐,加入60%-70%的乙醇回流提取2-3次,每次1.5小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到人参提取物;
山药提取物:取山药原料,去皮、切块、磨浆后置于提取罐,加入5-10倍纯水回流提取2次,每次提取1-2小时,合并提取液后过滤,滤液减压浓缩,浓缩液加入乙醇醇沉,使含乙醇量为70%-80%,静置后离心,取沉淀冷冻干燥、粉碎、过80目筛即得山药提取物;
葛根提取物:取葛根原料,粉碎后置于提取罐,加入60%-70%的乙醇回流提取2-3次,每次1.5小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到葛根提取物;
石斛提取物:取石斛原料,粉碎后置于提取罐,加入5-10倍纯水回流提取2次,每次提取1-2小时,合并提取液后过滤,滤液减压浓缩,浓缩液加入乙醇醇沉,使含乙醇量为50%-60%,静置后离心,取上清液浓缩,经喷雾干燥得石斛提取物;
茯苓提取物:取茯苓原料,粉碎后置于提取罐,加入60%-70%的乙醇回流提取2-3次,每次1-2小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到茯苓提取物;
桃仁提取物:取桃仁原料,粉碎后置于提取罐,加入70%-80%的乙醇回流提取2-3次,每次1-2小时,合并提取液后过滤,滤液减压浓缩,经喷雾干燥得到桃仁提取物。
4.根据权利要求1或2所述的一种辅助降三高组合物的制备方法,其特征在于,颗粒剂的制备方法如下:取海参低聚肽5-20份、玉米低聚肽5-20份、决明子提取物3-9份、山楂提取物5-10份、人参提取物2-6份、山药提取物3-9份、葛根提取物5-10份、石斛提取物2-6份、茯苓提取物3-9份、桃仁提取物3-9份、L-阿拉伯糖3-9份、低聚木糖5-10份、膳食纤维5-10份、茉莉花2-6份、木糖醇2-6份、营养酵母粉1-2份,投入混合机内混合10-30分钟,使其均匀,加入麦芽糖醇浆水溶液作为湿润剂,混合 5-10 分钟,混合后的物料用10-20目筛制粒,放入干燥设备内干燥,干燥颗粒用 20-60 目筛网筛分,去除粗颗粒和细粉,包装,即得。
5.根据权利要求1或2所述的一种辅助降三高组合物的制备方法,其特征在于,片剂的制备方法如下:取海参低聚肽5-20份、玉米低聚肽5-20份、决明子提取物3-9份、山楂提取物5-10份、人参提取物2-6份、山药提取物3-9份、葛根提取物5-10份、石斛提取物2-6份、茯苓提取物3-9份、桃仁提取物3-9份、L-阿拉伯糖3-9份、低聚木糖5-10份、膳食纤维5-10份、茉莉花2-6份、木糖醇2-6份、营养酵母粉1-2份,加入羧甲基纤维素5-15份,投入混合机内混合10-30分钟,使其均匀,加入麦芽糖醇浆水溶液作为湿润剂,混合 5-10 分钟,混合后的物料用10-20目筛制粒,放入干燥设备内干燥,加入干燥颗粒总量的1-5份的硬脂酸镁,整粒,压片、包装,即得。
6.根据权利要求1或2所述的一种辅助降三高组合物的制备方法,其特征在于,口服液的制备方法如下:取海参低聚肽5-20份、玉米低聚肽5-20份、决明子提取物3-9份、山楂提取物5-10份、人参提取物2-6份、山药提取物3-9份、葛根提取物5-10份、石斛提取物2-6份、茯苓提取物3-9份、桃仁提取物3-9份、L-阿拉伯糖3-9份、低聚木糖5-10份、膳食纤维5-10份、茉莉花2-6份、木糖醇2-6份、营养酵母粉1-2份,加入6-40倍量纯化水溶解得溶液,加入溶液量0.05-1.0份的山梨酸钾,然后过滤、分装、灭菌、包装,即得。
