背景技术:
糖尿病是一组以高血糖为主要特征的代谢性疾病。高血糖则是由于胰岛素分泌缺陷或其生物作用受损,或两者兼有引起。糖尿病时长期存在的高血糖,导致各种组织,特别是眼、肾、心脏、血管、神经的慢性损害、功能障碍。免疫是指机体免疫系统识别自身与异己物质,并通过免疫应答排除抗原性异物,以维持机体生理平衡的功能。免疫是人体的一种生理功能,人体依靠这种功能识别“自己”和“非己”成分,从而破坏和排斥进入人体的抗原物质,或人体本身所产生的损伤细胞和肿瘤细胞等,以维持人体的健康。随着社会和经济的进步以及人们生活水平的提高,人们已开始从重治疗转向重预防,这将极大地推进研究降血糖、提高免疫力的药品、食品等。
海参,属棘皮动物门海参纲(Holothurioidea),自古就是一种高蛋白质的营养品,《本草纲目》中指出海参有主补元气、滋益五脏六腑和祛虚损的养生功效,在历史上被称作“海八珍”,与鲍鱼、鱼翅齐名。现代科学研究发现,海参含有许多重要的生理活性物质,如海参多肽、海参多糖、海参皂甙、甾醇、脂肪酸和毒素等,这些生理活性物质具有增强免疫力、抗肿瘤、抗凝血、保护神经组织、镇痛、抗真菌和降低血粘度等作用。
石斛,为兰科石斛属多种植物新鲜或干燥茎的统称,是中国历史最悠久的中药养生品之一,在我国现存最早的药物学专著,成书于东汉的《神农本草经》就有记载:“石斛-味甘平。主伤中,除痹,下气,补五脏虚劳,羸瘦,强阴。久服厚肠胃,轻身延年。一名林兰(御览引云,一名禁生,观本,作黑字),生山谷。”,《本草纲目》、《名医别录》等古代医学名著更是推崇有佳,被历代名医奉为“补五脏虚劳,羸瘦,强阴,厚肠胃,轻身延年”的上医良药,有极高的药用价值。

本发明的目的是提供一种能降血糖和提高免疫力的营养品及其制备方法。
本发明的能降血糖和提高免疫力的营养品的制备方法,包括以下步骤:将海参乙酸乙酯提取物和石斛乙酸乙酯提取物混合均匀,即得能降血糖和提高免疫力的营养品。
优选,所述的将海参乙酸乙酯提取物和石斛乙酸乙酯提取物混合均匀是将海参乙酸乙酯提取物和石斛乙酸乙酯提取物按照质量比为3~5:1混合均匀。
优选,所述的海参乙酸乙酯提取物是通过以下方法制备的:将干海参粉碎或新鲜海参去肠,洗净,干燥,粉碎;按海参质量的5~10倍,加入体积分数70%~80%的乙醇水溶液,于常温下浸泡或于50℃~80℃下回流提取2~6小时,将提取液固液分离,上清液经减压浓缩去除乙醇至浸膏,加入0.5~2mol/L的硫酸或盐酸水溶液,于50℃~100℃下水解5~10小时,用碱液调pH至中性,再用乙酸乙酯萃取,乙酸乙酯萃取液经减压浓缩干燥,得到海参乙酸乙酯提取物。
优选,所述的石斛乙酸乙酯提取物是通过以下方法制备的:将石斛干燥茎粉碎或石斛新鲜茎洗净,干燥,粉碎;按石斛质量的5~10倍,加入体积分数80%~95%的乙醇水溶液,于常温下浸泡或于50℃~80℃下回流提取2~6小时,将提取液固液分离,上清液经减压浓缩去除乙醇至浸膏,将浸膏悬浮于水中,依次用石油醚和乙酸乙酯萃取,乙酸乙酯萃取液经减压浓缩干燥,得到石斛乙酸乙酯提取物。
所述的海参优选为刺参、花刺参、梅花参、黑乳参、糙海参、海地瓜、辐肛参或白尼参。
所述的石斛优选为铁皮石斛。
所述的碱液优选为2mol/L的氢氧化钠溶液。
本发明的另一个目的是提供一种按照所述的制备方法制备得到的能降血糖和提高免疫力的营养品。
本发明能降血糖和提高免疫力的营养品还可以与食品或药品上可以接受的辅料一起使用。所述的辅料可为调味剂,如阿斯巴甜、蔗糖等或填充剂,如淀粉、糊精等。其含量没有严格限制,按照常规方法,根据工艺和需要进行选择。还可以在能降血糖和提高免疫力的营养品中添加填充剂,如淀粉、糊精等,还可以加入调味剂例如阿斯巴甜等,按常规方法制成颗粒或其它剂型。
本发明能降血糖和提高免疫力的营养品服用方法:每日2~3次,每次0.1~0.3g,可长期服用。
能降血糖和提高免疫力的营养品的药效试验:
一、本发明的营养品提高免疫力的药效试验:
选取健康NIH雄性小鼠50只,体重18~22g,随机分为5组,分别为空白对照组、营养品(实施例1制备的营养品)高、低剂量组、海参乙酸乙酯提取物(实施例1制备的海参乙酸乙酯提取物)组、石斛乙酸乙酯提取物(实施例1制备的石斛乙酸乙酯提取物)组,每组10只小鼠。营养品高剂量组给药量为300mg/kg,营养品低剂量组给药量为100mg/kg,海参乙酸乙酯提取物组给药量为300mg/kg、石斛乙酸乙酯提取物组给药量为300mg/kg,将实施例1制备的营养品、海参乙酸乙酯提取物、石斛乙酸乙酯提取物分别用蒸馏水重悬,各组按0.2mL/10g体重灌胃给药,连续15天,每天1次,空白对照组灌胃等体积的蒸馏水。首次给药即于小鼠腹部皮肤(预先用硫化钡脱毛)滴1%DNCB丙酮溶液50μL/只,次日加强一次。末次给药后1h,以1%DNCB丙酮溶液滴于小鼠右耳两面,10μL/只,24h后颈椎处死小鼠,剪下小鼠左右耳,用8mm打孔器取相同部位耳片,以左右耳重量差为肿胀度(mg)。肿胀度越大,反应越强。结果见表1。

由表1可知,本发明的营养品能显著增加小鼠耳肿胀度,说明本发明的营养品能够增强小鼠迟发超敏反应的强度,具有显著提高免疫力的功效。且营养品高剂量组小鼠的耳肿胀度显著高于海参乙酸乙酯提取物组和石斛乙酸乙酯提取物组,说明本发明的营养品将海参乙酸乙酯提取物和石斛乙酸乙酯提取物复配,具有明显协同增效作用。
二、本发明的营养品对糖尿病模型小鼠的降血糖作用:
选取健康昆明种雄性小白鼠150只,体重18~22g,喂养7天后,随机选10只作为空白对照组,其余140只小鼠用来构建糖尿病模型小鼠,建模前将小白鼠禁食(不禁水)24h,一次性腹腔注射四氧嘧啶160mg/kg,5天后禁食4h眼球取血检测空腹血糖,以空腹血糖值高于11.1mmol/L为建模成功。选取60只糖尿病模型小鼠,随机分为6组:模型对照组、阳性(盐酸二甲双胍)对照组、营养品(实施例1制备的营养品)高、低剂量组两个剂量组、海参乙酸乙酯提取物(实施例1制备的海参乙酸乙酯提取物)组、石斛乙酸乙酯提取物(实施例1制备的石斛乙酸乙酯提取物)组,每组10只。营养品高剂量组灌胃实施例1制备的营养品,给药量为300mg/kg;营养品低剂量组灌胃实施例1制备的营养品,给药量为100mg/kg;海参乙酸乙酯提取物组灌胃实施例1制备的海参乙酸乙酯提取物,给药量为300mg/kg;石斛乙酸乙酯提取物组灌胃实施例1制备的石斛乙酸乙酯提取物,给药量为300mg/kg;阳性对照组灌胃盐酸二甲双胍,给药量500mg/kg;将实施例1制备的营养品、海参乙酸乙酯提取物、石斛乙酸乙酯提取物分别用蒸馏水重悬,各组按0.2mL/10g体重灌胃给药,连续15天,每天1次,空白对照组和模型对照组灌胃等体积的蒸馏水。禁食24h,称重并测空腹血糖。结果见表2和3。

由表2可知,在灌胃给药15天后,阳性对照组、营养品高、低剂量组小鼠的体重均显著高于模型对照组小鼠,营养品高剂量组小鼠体重显著高于海参乙酸乙酯提取物组和石斛乙酸乙酯提取物组小鼠。横向比较(实验前后体重)发现,空白对照组小鼠体重增加,模型对照组小鼠体重降低,表明糖尿病模型小鼠体重出现负增长,阳性对照组、营养品高、低剂量组小鼠体重均显著高于模型对照组小鼠,且均低于空白对照组小鼠,说明本发明的营养品能减缓糖尿病模型小鼠体重降低趋势。因此,本发明的营养品对糖尿病模型小鼠体重下降具有减缓作用,且效果优于盐酸二甲双胍。

由表3可知,灌胃给药15天后,阳性对照组、营养品高剂量组、营养品低剂量组、海参乙酸乙酯提取物组和石斛乙酸乙酯提取物组小鼠的血糖值均极显著低于模型对照组小鼠。横向比较(实验前后血糖值)发现,灌胃给药15天后,阳性对照组、营养品高剂量组、营养品低剂量组、海参乙酸乙酯提取物组和石斛乙酸乙酯提取物组小鼠的血糖值都明显降低,营养品高剂量组小鼠血糖值显著低于阳性对照组、海参乙酸乙酯提取物组和石斛乙酸乙酯提取物组小鼠。因此,本发明的营养品具有较强的降血糖作用,且效果优于盐酸二甲双胍,营养品的降血糖效果显著高于单用(海参乙酸乙酯提取物或石斛乙酸乙酯提取物),两者复配具有协同增效作用。

将新鲜刺参去肠,洗净,干燥,粉碎,取10kg置于80kg体积分数75%的乙醇水溶液中,于70℃下回流提取5小时,将提取液于6000rpm离心10min分离除渣,上清液经减压浓缩去除乙醇至浸膏,加入3L的2mol/L的盐酸水溶液,于90℃下水解8小时,用2mol/L氢氧化钠溶液调pH至中性,再用等体积乙酸乙酯萃取,乙酸乙酯萃取液经减压浓缩去除乙酸乙酯至浸膏,真空冷冻干燥,得到海参乙酸乙酯提取物;将铁皮石斛新鲜茎洗净,干燥,粉碎,取10kg置于80kg体积分数90%的乙醇水溶液中,于70℃下回流提取5小时,将提取液于6000rpm离心10min分离除渣,上清液经减压浓缩去除乙醇至浸膏,将浸膏悬浮于3L水中,依次用石油醚和乙酸乙酯各3L萃取,乙酸乙酯萃取液经减压浓缩去除乙酸乙酯至浸膏,真空冷冻干燥,得到石斛乙酸乙酯提取物;将海参乙酸乙酯提取物和石斛乙酸乙酯提取物按照质量比为5:1混合均匀,即得能降血糖和提高免疫力的营养品。经动物实验证明,该营养品具有降血糖和提高机体免疫力的作用。

将干黑乳参粉碎,取10kg置于100kg体积分数80%的乙醇水溶液中,于80℃下回流提取2小时,将提取液于6000rpm离心10min分离除渣,上清液经减压浓缩去除乙醇至浸膏,加入3L的2mol/L的盐酸水溶液,于100℃下水解5小时,用2mol/L氢氧化钠溶液调pH至中性,再用等体积乙酸乙酯萃取,乙酸乙酯萃取液经减压浓缩去除乙酸乙酯至浸膏,真空冷冻干燥,得到海参乙酸乙酯提取物;将铁皮石斛干燥茎粉碎,取10kg置于100kg体积分数95%的乙醇水溶液中,于80℃下回流提取2小时,将提取液于6000rpm离心10min分离除渣,上清液经减压浓缩去除乙醇至浸膏,将浸膏悬浮于3L水中,依次用石油醚和乙酸乙酯各3L萃取,乙酸乙酯萃取液经减压浓缩去除乙酸乙酯至浸膏,真空冷冻干燥,得到石斛乙酸乙酯提取物;将海参乙酸乙酯提取物和石斛乙酸乙酯提取物按照质量比为4:1混合均匀,即得能降血糖和提高免疫力的营养品。经动物实验证明,该营养品具有降血糖和提高机体免疫力的作用。

将新鲜糙海参去肠,洗净,干燥,粉碎,取10kg置于60kg体积分数75%的乙醇水溶液中,于常温下浸泡提取6小时,将提取液于6000rpm离心10min分离除渣,上清液经减压浓缩去除乙醇至浸膏,加入3L的2mol/L的盐酸水溶液,于100℃下水解5小时,用2mol/L氢氧化钠溶液调pH至中性,再用等体积乙酸乙酯萃取,乙酸乙酯萃取液经减压浓缩去除乙酸乙酯至浸膏,真空冷冻干燥,得到海参乙酸乙酯提取物;将铁皮石斛新鲜茎洗净,干燥,粉碎,取10kg置于60kg体积分数95%的乙醇水溶液中,于常温下浸泡提取6小时,将提取液于6000rpm离心10min分离除渣,上清液经减压浓缩去除乙醇至浸膏,将浸膏悬浮于3L水中,依次用石油醚和乙酸乙酯各3L萃取,乙酸乙酯萃取液经减压浓缩去除乙酸乙酯至浸膏,真空冷冻干燥,得到石斛乙酸乙酯提取物;将海参乙酸乙酯提取物和石斛乙酸乙酯提取物按照质量比为5:1混合均匀,即得能降血糖和提高免疫力的营养品。经动物实验证明,该营养品具有降血糖和提高机体免疫力的作用。

1.一种能降血糖和提高免疫力的营养品的制备方法,其特征在于,包括以下步骤:将海参乙酸乙酯提取物和石斛乙酸乙酯提取物混合均匀,即得能降血糖和提高免疫力的营养品。
2.根据权利要求1所述的制备方法,其特征在于,所述的将海参乙酸乙酯提取物和石斛乙酸乙酯提取物混合均匀是将海参乙酸乙酯提取物和石斛乙酸乙酯提取物按照质量比为3~5:1混合均匀。
3.根据权利要求1所述的制备方法,其特征在于,所述的海参乙酸乙酯提取物是通过以下方法制备的:将干海参粉碎或新鲜海参去肠,洗净,干燥,粉碎;按海参质量的5~10倍,加入体积分数70%~80%的乙醇水溶液,于常温下浸泡或于50℃~80℃下回流提取2~6小时,将提取液固液分离,上清液经减压浓缩去除乙醇至浸膏,加入0.5~2mol/L的硫酸或盐酸水溶液,于50℃~100℃下水解5~10小时,用碱液调pH至中性,再用乙酸乙酯萃取,乙酸乙酯萃取液经减压浓缩去除乙酸乙酯至浸膏,干燥,得到海参乙酸乙酯提取物。
4.根据权利要求1所述的制备方法,其特征在于,所述的石斛乙酸乙酯提取物是通过以下方法制备的:将石斛干燥茎粉碎或石斛新鲜茎洗净,干燥,粉碎;按石斛质量的5~10倍,加入体积分数80%~95%的乙醇水溶液,于常温下浸泡或于50℃~80℃下回流提取2~6小时,将提取液固液分离,上清液经减压浓缩去除乙醇至浸膏,将浸膏悬浮于水中,依次用石油醚和乙酸乙酯萃取,乙酸乙酯萃取液经减压浓缩去除乙酸乙酯至浸膏,干燥,得到石斛乙酸乙酯提取物。
5.根据权利要求1~4任一项所述的制备方法,其特征在于,所述的海参为刺参、花刺参、梅花参、黑乳参、糙海参、海地瓜、辐肛参或白尼参。
6.根据权利要求1~4任一项所述的制备方法,其特征在于,所述的石斛为铁皮石斛。
7.根据权利要求3所述的制备方法,其特征在于,所述的碱液为2mol/L的氢氧化钠溶液。
8.一种按照权利要求1~4任一项所述的制备方法制备得到的能降血糖和提高免疫力的营养品。
