In an exemplary embodiment, each kilogram of bird's nest raw material is soaked in about 80-200 liters of water or aqueous medium for swelling for at least 3 hours, after which the water or aqueous medium is removed by filtration, and about 20-40 liters of water or aqueous medium per kilogram of bird's nest raw material is added, then heated to about 85 degrees celsius or above, cooked for at least about 6 hours, after which the cooked bird's nest is removed by filtration.
Removing nidus Collocaliae after stewing, optionally adding water or aqueous medium to a constant volume of about 0.001-0.1g crude drug/ml, such as about 0.001-0.1g crude drug/ml, about 0.002-0.09g crude drug/ml, about 0.003-0.08g crude drug/ml, about 0.004-0.07g crude drug/ml, about 0.005-0.06g crude drug/ml, about 0.006-0.05g crude drug/ml, about 0.007-0.04g crude drug/ml, about 0.008-0.03g crude drug/ml, about 0.009-0.02g crude drug/ml, or about 0.01g crude drug/ml.
The bird's nest raw material can be pretreated by adopting a pretreatment method which is conventional in the field. For hairpins, the surface dirt and the surface attached feathers can be removed by repeatedly rinsing. For the treated cubilose, water can be directly added for soaking and expansion.
In a specific embodiment, the water extract of bird's nest is prepared by the following steps:
a) soaking nidus Collocaliae in water or aqueous medium at about 50-150 times, such as about 60, 70, 80, 90, 100, 110, 120, 130 or 140 times, preferably 100 times, weight of nidus Collocaliae for about 1-5 hours, preferably about 3 hours, e.g. at room temperature, and filtering to obtain nidus Collocaliae residue; and
b) decocting the residue with water or aqueous medium about 20-40 times, such as about 25-35 times, preferably 30 times, the weight of nidus Collocaliae for about 4-8 hr, such as about 5-7 hr, preferably about 6 hr, and filtering to obtain nidus Collocaliae extractive solution, i.e. water extract.
The obtained cubilose extract can be converted into other forms by any means known in the art. For example, by freeze-drying into powder form, to suit different specific uses.
Figure 1 illustrates an exemplary process flow for preparing the composition of the present invention.
Composition comprising a metal oxide and a metal oxide
The composition of the invention is based on the combination of saussurea involucrate extract and cubilose extract.
The saussurea involucrate extract can be extracted by water or alcohol, and the cubilose extract can be water extract in the invention.
In one aspect, the weight ratio of saussurea involucrate extract to cubilose extract contained in the present composition is in the range of about 5:1 to 1:2, preferably about 5:1 to 1:1, e.g. about 5:1, 4.5:1, 4:1, 3.5:1, 3.4:1, 3.3:1, 3.2:1, 3.1:1, 3:1, 2.9:1, 2.8:1, 2.7:1, 2.6:1, 2.5:1, 2:1, 1.5:1, 1:1.5 and 1:2, e.g. 3: 1.
In one aspect, the concentration of the saussurea involucrate extract is 0.001-0.1g crude drug/ml, such as about 0.001-0.1g crude drug/ml, about 0.002-0.09g crude drug/ml, about 0.003-0.08g crude drug/ml, about 0.004-0.07g crude drug/ml, about 0.005-0.06g crude drug/ml, about 0.006-0.05g crude drug/ml, about 0.007-0.04g crude drug/ml, about 0.008-0.03g crude drug/ml, about 0.009-0.02g crude drug/ml, or about 0.01g crude drug/ml.
In one aspect, the concentration of the bird's nest extract is about 0.001-0.1g crude drug/ml, such as about 0.001-0.1g crude drug/ml, about 0.002-0.09g crude drug/ml, about 0.003-0.08g crude drug/ml, about 0.004-0.07g crude drug/ml, about 0.005-0.06g crude drug/ml, about 0.006-0.05g crude drug/ml, about 0.007-0.04g crude drug/ml, about 0.008-0.03g crude drug/ml, about 0.009-0.02g crude drug/ml, or about 0.01g crude drug/ml.
In a specific embodiment, the saussurea involucrate extract is aqueous extract of saussurea involucrate with concentration of about 0.01g crude drug/ml, the bird's nest extract is aqueous extract of bird's nest with concentration of about 0.01g crude drug/ml, and the weight ratio of the saussurea involucrate extract to the bird's nest extract is about 3: 1.
In a particular embodiment, the composition comprises an aqueous extract of saussurea involucrate and an aqueous extract of cubilose in a weight ratio of about 3.5:1 to 2.5:1 (e.g., 3.4:1, 3.3:1, 3.2:1, 3.1:1, 3:1, 2.9:1, 2.8:1, 2.7:1, 2.6:1), and the concentration of herba Saussureae Involueratae extract is about 0.006g crude drug/ml to 0.015g crude drug/ml (such as about 0.007-0.014g crude drug/ml, 0.008-0.013g crude drug/ml, 0.009-0.012g crude drug/ml, 0.010-0.011g crude drug/ml), the nidus Collocaliae water extract has a concentration of about 0.006g crude drug/ml to 0.015g crude drug/ml (e.g. about 0.007-0.014g crude drug/ml, 0.008-0.013g crude drug/ml, 0.009-0.012g crude drug/ml, 0.010-0.011g crude drug/ml).
Those skilled in the art will appreciate that the volume ratio of the two can be adjusted moderately with the concentration of the extract, and that the composition so adjusted can be expected to achieve synergistic effects in terms of antioxidant/anti-aging and improved whitening effects relative to the saussurea involucrate extract alone.
For example, when the volume of the prepared herba saussureae involucratae water extract is about 0.01g crude drug/ml, and the volume of the prepared nidus Collocaliae water extract is 0.01g crude drug/ml, the two are combined together by mixing, for example, in a weight/volume ratio of 3: 1. In accordance with the weight ratio, when the prepared herba saussureae involucratae water extractive solution has a constant volume of about 0.03g crude drug/ml and the prepared nidus Collocaliae water extractive solution has a constant volume of 0.01g crude drug/ml, the two are mixed together at a volume ratio of 1:1, for example.
As used herein, the unit "g crude drug/ml" refers to the number of g of raw materials contained per ml of extract.
As used herein, the term "about" when used to define a numerical value means a range of ± 5% of the indicated numerical value, e.g., ± 4%, ± 3%, ± 2% or ± 1%.
Those skilled in the art will appreciate that details not described in detail herein relating to the compositions of the present invention and products and processes of manufacture thereof may be resorted to by those skilled in the art. For example, when referring to mixing the bird's nest extract with the saussurea involucrate extract, mixing means commonly used in the art, such as mechanical mixing, stirring mixing, automatic mixing suitable for an automated process, and the like, may be employed. For example, the water used for preparing the water extract may be tap water, spring water, mineral water, distilled water, etc., which are common in the art. For another example, the saussurea involucrate powder can be prepared by ordinary techniques in the field, such as grinding.
The composition according to the present invention may contain only the saussurea involucrate extract and the bird's nest extract in the weight ratio within the scope of the present invention, and may further contain other beneficial components according to other desired effects, and may further contain auxiliary components such as acceptable carriers according to the use of the composition (e.g., for cosmetics, foods, health products, etc.) to achieve other effects in addition to oxidation resistance and whitening.
For example, the compositions of the present invention may also be used to formulate medicaments with pharmaceutical carriers and/or other pharmaceutically active ingredients, depending on the actual requirements. The compositions of the invention may also be formulated into cosmetic products with cosmetic carriers and/or other active ingredients; can be used as food additive for health products, foods, beverages, etc.; can be used as nutrition enhancer for nutritional products.
It will also be appreciated by those skilled in the art that the compositions of the present invention can be used directly in different applications, or as a feedstock material for different products, different applications, which are further processed into different products and used widely.
The composition or product according to the invention may be presented in various forms, such as a pharmaceutical, nutraceutical, food, beverage, food additive, dietary supplement or cosmetic product.
The compositions or products of the invention may be consumed directly or otherwise administered to a subject in need thereof, e.g., a human, and may, for example, be provided to the subject in vivo, e.g., orally, or in vitro, e.g., topically to the skin.
Saussurea involucrate and bird's nest are traditional rare and precious Chinese medicinal materials. In a high-cold area with 3,000-4,000 meters of the original Tianshan mountain of the Tianshan snow lotus, because a large amount of manual mining is carried out, wild resources are gradually exhausted, and the yield of the Tianshan snow lotus with high quality is not large due to the difficulty of artificial culture. The invention can reduce the dosage of the saussurea involucrate and generate unexpected synergistic effect on the aspects of oxidation resistance and aging resistance by combining the saussurea involucrate and the cubilose. The compatibility can not only obviously enhance the antioxidation/anti-aging effect of the saussurea involucrate, but also enhance the whitening effect of the composition containing the compatibility, and can be widely applied to various related products such as medicines, health products, foods and beauty products. The preparation method of the composition has simple steps and easy operation, greatly simplifies the preparation process and is beneficial to industrial production.
The saussurea involucrate and the edible bird's nest are the breakthrough of the combination of the north and south medicines except the synergic complementation of the efficacies. The inventor combines the saussurea involucrate produced from the top of Tianshan mountain in the arctic-arctic cold region with the cubilose in the cave on java island in Indonesia, and the breakthrough combination becomes an example of the development of new traditional Chinese medicine resources later.
Examples
The present invention will be described in detail with reference to specific examples. However, those skilled in the art will appreciate that the specific embodiments are merely illustrative of the invention more clearly and are not to be considered in any way as limiting the invention. Unless otherwise indicated, all reagents and equipment are commercially available and all amounts are by weight.
Example 1: preparation of the extract
Taking 10g of Tianshan saussurea involucrate of the natural Tianshan saussurea involucrate farm in Xinjiang, grinding into powder, and sieving with a No. 3 sieve. Ultrasonic extracting with water (400mL) 40 times the weight of herba Saussureae Involueratae for 30 min; centrifuging at 5000 Xg for 10min, and collecting supernatant; and repeating the steps. Collecting the supernatant, adding water to a constant volume of 0.010g crude drug/ml (namely constant volume of 1L), and obtaining the saussurea involucrate extract.
Soaking 10g of Indonesia bigelovii nest in 100 times of water (1L) at room temperature for 3 hr, and filtering to obtain filter residue; stewing and boiling filter residues of the cubilose for 6 hours by using water (300mL) with the weight 30 times that of the cubilose; filtering to remove residue, adding water to constant volume of 0.010g crude drug/ml (constant volume of 1L), and collecting nidus Collocaliae extractive solution.
Example 2: test for antioxidant Effect
A standard curve is established based on the copper ion oxidation resistance of TROLOX (an analogue of vitamin E), and the total oxidation resistance of different nidus Collocaliae extracts and herba Saussureae Involueratae extracts is calculated according to the standard curve. In this test, the higher the TROLOX equivalent, the better the antioxidant capacity of the tested extract (see FIG. 2).
Example 3: synergistic effect of saussurea involucrate extract and cubilose extract
This example shows the synergistic effect of saussurea involucrate extract and cubilose extract in antioxidant capacity.
The saussurea involucrate extract, the bird's nest extract and the composition prepared according to example 1 in the weight ratio of 1:1, 3:1 and 5:1 were respectively weighed. Antioxidant capacity tests were performed in the following experimental groups to compare the effect of the compositions relative to the individual components.
Control group: water without any test substance as blank
Group 1: 1:1 group
a: composition of 12 weight parts (1 g) saussurea involucrate extract and 12 weight parts (1 g) cubilose extract
b: 12 parts by weight (i.e. 1 g) of saussurea involucrate extract and 12 parts by weight (i.e. 1 ml) of water
c: 12 parts by weight (i.e. 1 g) of cubilose extract supplemented with 12 parts by weight (i.e. 1 ml) of water
Group 2: 3:1 group
a: composition of 18 weight parts (1.5 g) saussurea involucrate extract and 6 weight parts (0.5 g) cubilose extract
b: 18 parts by weight (i.e., 1.5 g) of saussurea involucrate extract and 6 parts by weight (i.e., 0.5 ml) of water
c: 6 parts by weight (i.e. 0.5 g) of cubilose extract supplemented with 18 parts by weight (i.e. 1.5 ml) of water
Group 3: 5:1 group
a: composition of 20 weight parts (1.67 g) saussurea involucrate extract and 4 weight parts (0.33 g) cubilose extract
b: 20 parts by weight (i.e., 1.67 g) of saussurea involucrate extract and 4 parts by weight (i.e., 0.33 ml) of water
c: 4 parts by weight (i.e. 0.33 g) of cubilose extract supplemented with 20 parts by weight (i.e. 1.67 ml) of water
Three replicates were set up for each set of experiments and the average of three replicates was taken as the test result.
The test results are shown in fig. 3. Therefore, the tested combination of the cubilose extract and the saussurea involucrate extract with different proportions shows synergistic antioxidant capacity.
Alternative experiments were performed with other sources of bird's nests and found that the synergy was not limited to the specific source of bird's nests (data not shown). Without being bound to a particular theory, the inventors speculate that this is related to the consistency of the bird's nest in its antioxidant efficacy, and thus the consistency of the active ingredients that exert antioxidant efficacy.
Example 4: whitening effect test of composition of saussurea involucrate extract and cubilose extract
The test for tyrosinase activity inhibition is a conventional test method for testing whitening effects. Wherein tyrosine is converted into levodopa (L-DOPA) and Dopachrome (Dopachrome) by tyrosinase. The DOPAchrome has absorbance at 492nm, so that the activity of the tyrosinase can be quantified and calculated. Dopachrome is only a temporary storage and it is converted to Melanin (Melanin). As mentioned above, melanin on human skin is transformed from dopachrome produced by Melanocytes (Melanocytes).
This example employs a tyrosinase activity inhibition assay as is conventional in the art, wherein 0.5mM levodopa is placed in a 50mM sodium phosphate solution at pH 6.8, the tyrosinase concentration is 0.2mg/mL, and the control or sample volume for the assay is 200. mu.L. As a blank control, water without any added extract was used, and 24 parts by weight of each of the compositions of group 1a, group 2a and group 3a in example 3 and 24 parts by weight of the saussurea involucrate extract prepared according to example 1 alone (i.e., 2g of saussurea involucrate extract) were used as different test samples.
And placing the control and different test samples in a 96-well plate, standing for 20 minutes at room temperature under the condition of no light, reading the absorbance at the wavelength of 492nm by using a plate reader, and calculating the inhibition rate of the different test samples on tyrosinase.
Three replicates were set for each set of tests and the average of three replicates was taken as the test result. The test results are shown in fig. 4. Therefore, compared with the single saussurea involucrate extract, the whitening effect of the composition with different proportions is obviously improved. This shows that the composition of the present invention can exert a sufficient whitening effect while having a synergistically beneficial antioxidant effect.
As proved by the application, the bird's nest is matched with the saussurea involucrate to be used, and the integral oxidation resistance and the whitening effect of the formula are fully exerted in a synergistic and beneficial mode. The preparation process is simple, the cost can be reduced, and increasingly deficient natural precious resources of the wild saussurea involucrate can be protected. Meanwhile, under the condition of the current shortage of traditional Chinese medicine resources, the invention is an innovative solution with foresight by combining and applying the very rare compatibility of north-south cross-border of the saussurea involucrate and the cubilose.
As previously mentioned, the present invention has been specifically explained in detail in connection with one or more embodiments. However, it is not to be understood that the scope of the present invention is limited thereby. It should be noted that, for those skilled in the art, various changes and modifications can be made without departing from the spirit and substance of the present invention, and these changes and modifications are within the scope of the present invention.

Claims (2)

1. A product for resisting oxidation and/or whitening, which consists of saussurea involucrate extract and cubilose extract, and is characterized in that:
wherein the weight ratio of the saussurea involucrate extract to the cubilose extract is 3: 1;
wherein the saussurea involucrate extract is a saussurea involucrate water extract, and the cubilose extract is a cubilose water extract;
wherein the water extract of saussurea involucrate is prepared by the following steps:
a. providing saussurea involucrate;
b. performing ultrasonic extraction in water or aqueous medium; and
c. removing residue to obtain herba Saussureae Involueratae water extract;
wherein the water extract of cubilose is prepared by the following steps:
a. soaking nidus Collocaliae in water or aqueous medium;
b. removing the liquid;
c. stewing the soaked cubilose in water or an aqueous medium; and
d. removing the stewed bird's nest to obtain the water extract of bird's nest.
2. A method of making the composition of claim 1, comprising:
a) providing saussurea involucrate extract and cubilose extract; and
b) mixing the saussurea involucrate extract and the cubilose extract in a weight ratio of 3: 1.
