本发明属于生命科学中的治疗医学与预防医学领域,尤其涉及一种不仅具有核基因损伤恢复、干细胞增殖、线粒体增殖及提高线粒体膜电位等功效,还具有提高骨髓间充质干细胞端粒酶活性及增加神经干细胞等自噬小体数量、提高抗氧化力及免疫力的延缓衰老的营养组合物及应用。
背景技术
美国食品和药物管理局现已核准进行“用二甲双胍对抗衰老”的临床试验,期待证明二甲双胍能延缓衰老并阻止疾病的发生与发展,但迄今没有结果。日本庆应义塾大学已经宣布和美国华盛顿大学合作,在全球首次开展抗衰老物质烟酰胺单核苷酸(NMN)的人体临床试验。因NMN参与细胞内烟酰胺腺嘌呤二核苷酸(NAD)的合成,而NAD是细胞能量转化的重要辅酶,如果能够通过这项研究确认摄入NMN是安全和有效的,将来有可能用NMN来预防和治疗伴随衰老发生的疾病。

中科院神经科学研究所、脑科学与智能技术卓越创新中心蔡时青课题组以秀丽线虫为模式生物,首次揭示了个体之间衰老速度差异的遗传基础,发现了一条新的信号通路调控动物衰老,阐明了神经肽介导的胶质细胞—神经元信号在衰老速度调控中的重要作用。9日,《自然》杂志发表了这一成果。
那么,从线虫实验到延缓人体衰老还有多远。蔡时青研究员介绍说:“衰老遗传机制太复杂了,用一个机制解释所有衰老现象是不可能的。”但毫无疑问的是,该工作为抗衰老研究提供一个全新视角,进一步解析个体之间衰老差异将有助于系统地理解健康衰老的调控机制。
大脑干细胞可减缓小鼠衰老
据美国《科学美国人》月刊网站2017年7月26日报道,以前的研究表明,下丘脑与衰老相关,但是最新研究显示,这一区域的干细胞可能会减缓这个过程。纽约爱因斯坦医学院的神经内分泌学家蔡东升称,这是讲得通的,因为下丘脑参与了许多身体功能。

MEDIPOST株式会社吴元一等《含有人脐带血衍生的间充质干细胞的组合物诱导神经前体细胞分化和增殖为神经细胞的用途》(申请号:CN200780044097.4),所述组合物对于治疗神经损伤性疾病是有效的。
“长寿”是人类有史以来的共同追求,但能活到“天年”(120岁)者甚微,大多随着年龄增长或疾病的困扰, 使全身器官与功能衰竭而去。为此,人们正在研制延缓衰老的相关药物。
但是迄今为止,还没有关于即具有核基因损伤修复、干细胞增殖、线粒体增殖及提高线粒体膜电位等功效,又具有提高骨髓间充质干细胞端粒酶活性及增加自噬小体数量、提高抗氧化力及免疫力的延缓衰老的营养组合物及应用的相关报道。
发明内容
本发明是为了解决现有技术所存在的上述技术问题,提供一种不仅具有核基因损伤修复、干细胞增殖、线粒体增殖及提高线粒体膜电位等功效,还具有提高骨髓间充质干细胞端粒酶活性及增加神经干细胞等的自噬小体数量、提高抗氧化力及免疫力的延缓衰老的营养组合物及应用。
本发明的技术解决方案是:一种延缓衰老的营养组合物,其特征在于各组分质量比如下:

本发明是以孵化3日的鸡胚胎与其它营养物相配伍,不仅具有核基因损伤修复、干细胞增殖、线粒体增殖及提高线粒体膜电位等功效,还可提高骨髓间充质干细胞端粒酶活性及增加自噬小体数量、提高抗氧化力及免疫力,可显著延缓衰老。
实验表明:
本发明营养组合物对肝脏组织、骨髓、脑组织中的DNA重组修复(双链断裂修复)基因损伤具有明显的恢复作用;对肝脏、骨髓组织中的DNA的SOS修复(错误倾向修复)基因损伤具有恢复作用,使DNA在受到严重损伤时,保证DNA复制能够正常进行;对脑组织DNA的切除修复(核苷酸切除修复)基因损伤具有一定的恢复作用;对骨髓、脑组织的切除修复(碱基切除修复)基因损伤有一定的恢复作用;对肝脏组织和骨髓中的DNA错配修复基因损伤具有一定的恢复作用。
本发明营养组合物对肠黏膜干细胞的增殖有显著的促进作用;对神经干细胞、骨髓造血干细胞、胃干细胞、胰腺干细胞、肾脏干细胞的增殖具有一定的促进作用。
本发明营养组合物能够促进肝细胞线粒体数目明显增多、线粒体膜电位明显增高,肝细胞浆中未见脂滴;促进脑海马部神经元细胞内线粒体明显增多与线粒体膜电位明显增高;促进肾小管上皮细胞内线粒体明显增多与线粒体膜电位明显增高;促进脾脏B细胞内线粒体明显增多与线粒体膜电位明显增高;促进了骨髓造血细胞内线粒体膜电位明显增高。
本发明营养组合物能够促进骨髓间充质干细胞端粒酶的活性,有利于其基因的表达,增强了向成骨细胞分化的能力,降低了向脂肪细胞分化的趋势;能够明显增多脑海马部神经元细胞自噬小体数量,自噬小体的增加将对脑损伤的恢复发挥重要作用;能够显著增多肾小球上皮细胞自噬小体数量;对肾小球基底膜足突的损伤具有恢复作用;能够促进脾脏B细胞自噬小体数量的增加。
本发明营养组合物可显著提高人体过氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH—PX)、过氧化物氢酶(CAT)的水平,明显降低肝脏、脑组织中脂褐素的含量;
本发明营养组合物具有抑制衰老脾脏、胸腺组织机构的退化作用;能够改善T、B细胞增殖功能;能够显著升高白细胞介素—2(IL—2)、白细胞介素—4(IL—4)、干扰素(IFN—γ),显著降低转化生长因子-β(TGF-β)含量。
本发明营养组合物对于衰老机体,能够明显增加食欲和体重,改善精神状态;增强活动能力,提高记忆能力;对肝、肾、脑、肠、肌肉、皮肤损伤具有明显恢复作用。

实验:
第一部分 不同孵育天数鸡胚蛋的干细胞分泌因子检测
一、鸡胚干细胞分泌因子的检测
实验分组:根据孵育天数第3、6、9天以及取材的部位分为:①第3天蛋清组;②第3天蛋黄组;③第3天鸡胚组;④第6天蛋清组;⑤第6天蛋黄组;⑥第6天鸡胚组;⑦第9天蛋清组;⑧第9天蛋黄组;⑨第9天鸡胚组。
实验流程:将受精鸡蛋放置于温度为37°C~38°C,湿度为60%~70% 的孵蛋器中进行孵育,选出孵育第三天、第六天、第九天胚龄的鸡蛋(如图1所示),取出蛋清、蛋黄、鸡胚。样本处理方式:取200ul的蛋清、蛋黄、研磨好的鸡胚加800ul PBS混匀,离心2000r/min10min,取上清,用样本稀释液10倍稀释上述样本,随后进行干细胞分泌因子检测。鸡胚、蛋清、蛋黄的干细胞分泌因子包括:干细胞因子(SCF)、神经细胞生长因子(NGF)、上皮细胞生长因子(EGF)、白介素(IL-2、IL-4)。
二、实验方法
(一)本次试验所采用的试剂盒:Chicken KITLG/SCF ELISA Kit、Chicken NGF ELISAKit、Chicken IL-4 ELISA Kit、Chicken IL-2 ELISA Kit、Chicken EGF ELISA Kit均来自lifespan公司,Chicken Tau ELISA KIT来自上海酶联生物公司。
(二)实验操作均按照说明书进行。
1、标准品的制备:将1ml标准品稀释液加入到冻干的标准品粉末中,室温轻轻混匀10min,避免产生气泡。
2、标准品的稀释如表2。

3、加样:分别设空白孔(空白对照孔不加样本品,其余各步骤操作相同)、标准孔、待测样品孔,在酶标包被板上待测样品孔准确加样100ul,空白孔准确加100ul样本稀释液。加样时应将样品加于酶标板孔底部,尽量不触及孔壁。
4、温育:用封板膜封板后置于37°C温育90min。
5、加抗体:加入100ul生物素标记抗体,封板后置于37°C温育1h。
6、配液:将25倍浓缩洗涤液用蒸馏水或去离子水25倍稀释。
7、洗涤:小心揭掉封板膜,弃去液体,甩干,加入洗涤液,静置90s后弃去,如此重复3次。
8、加酶:每孔加入100ul酶标试剂,封板后置于37℃温育30min。
9、洗涤:小心揭掉封板膜,弃去液体,甩干,加入洗涤液,静置90s后弃去,如此重复5次。
10、显色:每孔加入90ulTMB底物溶液,封板后置于37°C温育15min。
11、终止:每孔加入50ul终止液。
12、测定:以空白孔调零,450nm波长依次测定各孔的吸光度(OD值)。
三、实验结果
1. 干细胞因子 (SCF)检测
干细胞因子 (SCF) 又称肥大细胞生长因子,能诱导干细胞进入细胞周期,促进干细胞分裂,增强干细胞的自我更新能力,并提高干细胞对其他细胞因子的敏感性,是一种多效应的干细胞分泌因子,它在造血及维持肥大细胞、胚芽细胞、黑色素母细胞和精原细胞的存活、增殖和分化过程中起着重要作用。
结果如下:
蛋清组,蛋黄组及鸡胚组SCF的表达量见图2。结果显示,三组之间SCF浓度有显著差异(F=293.825,P=0.000),鸡胚组SCF浓度明显高于其它两组;不同天数之间SCF浓度有显著差异(F=114.646,P=0.000),第3天和第6天SCF浓度三组均有显著性差异。分组和天数的交互效应显著(F=101.703,P=0.000)。以上结果表明,第3天鸡胚组SCF浓度最高。
2. 神经细胞生长因子 (NGF) 检测
神经细胞生长因子 (NGF) 是神经营养因子家族中最早发现的神经生长调节因子,具有营养神经元和促突起生长双重生物学功能。广泛分布于外周神经系统和中枢神经系统如海马、大脑、小脑、下丘脑和中脑运动神经元,通过胆碱能神经逆行运输至前脑基底核,维持胆碱能神经元的存活和功能。在脑组织受损时,内源性NSC可增殖并分化,补偿缺失的神经细胞,修复部分神经功能。在胚胎早期,NGF能显著促进神经前体细胞的增殖与分化。
结果如下:
蛋清组,蛋黄组及鸡胚组NGF的表达量见图3。结果显示,三组之间NGF浓度有显著差异(F=634.186,P=0.000),鸡胚组NGF浓度明显高于其它两组;不同天数之间NGF浓度三组有显著差异 (F=372.627,P=0.000)。分组和天数的交互效应显著(F=380.099,P=0.000)。以上结果表明,第3天鸡胚组NGF浓度最高。
3、上皮细胞生长因子 (EGF) 检测
上皮细胞生长因子(EGF)为多肽生长因子,EGF具有促进多种细胞增殖的作用,可以诱导神经脊母细胞分化成黑色素细胞和神经元细胞来参与外周神经系统的发育。此外,EGF与雌雄性动物的生殖有关,它主要参与黄体生成素刺激卵巢的过程、促进精原细胞的有丝分裂和精原细胞集落的形成,促进胚胎早期鸡胚绒毛尿囊膜血管的新生。
结果如下:
蛋清组,蛋黄组及鸡胚组EGF的表达量见图4。结果显示,三组之间EGF浓度有显著差异(F=66.198,P=0.000),鸡胚组EGF浓度显著高于其它两组;不同天数之间EGF浓度有显著差异 (F=20.710,P=0.000),且第3天EGF浓度高于其它两组。分组和天数的交互效应显著(F=79.007,P=0.000)。以上结果表明,第3天鸡胚组EGF浓度最高。
4、白细胞介素-2 (IL-2) 检测
白细胞介素2是T淋巴细胞分泌的一种重要淋巴因子,在免疫反应过程中可促进 T、B细胞的增殖、分化,增强单核细胞及 NK 细胞的杀伤活性,在转录、翻译及受体信号传导过程中发挥着重要作用。在鸡的胚胎早期,对病毒入侵就有了一定的免疫调节能力。
结果如下:
蛋清组,蛋黄组及鸡胚组IL-2的表达量见图5。结果显示,三组之间IL-2浓度有显著差异 (F=434.426,P=0.000),鸡胚组IL-2浓度显著高于其它两组。不同天数之间IL-2浓度无显著差异 (F=2.698,P=0.074)。分组和天数的交互效应显著 (F=38.376,P=0.000)。以上结果表明,第3天鸡胚组IL-2浓度最高。
5、白细胞介素-4 (IL-4) 检测
IL-4是由Th2细胞分泌的细胞因子,能刺激B细胞分化,生成免疫球蛋白,激活免疫应答,刺激肥大细胞增殖,促进巨噬细胞提呈抗原和杀伤肿瘤细胞的功能,IL-4协同集落刺激因子(CSF)刺激造血细胞增殖。
结果如下:
蛋清组,蛋黄组及鸡胚组IL-4的表达量见图6。结果显示,三组之间IL-4浓度有显著差异 (F=158.71,P=0.000),鸡胚组IL-4浓度显著高于其它两组。不同天数之间IL-4浓度有显著差异 (F=24.396,P=0.000),且第3天IL-4浓度高于其它两组。分组和天数的交互效应显著 (F=24.712,P=0.000)。以上结果表明,第3天鸡胚组IL-4浓度最高。
小结:第3天鸡胚组的SCF、NGF、EGF、IL-2及IL-4浓度均为最高浓度。
第二部分 本发明营养组合物对延缓衰老作用的实验
一、D-半乳糖衰老大鼠模型的建立
实验分组:根据动物药理学的药品剂量和综合试验的设计分析,分为:①正常对照组;②衰老模型组;③本发明实施例2组(鸡胚营养组合物组);④鸡胚对照组,选取第3天胚龄的鸡蛋,每个鸡蛋取3ml鸡胚液,冻干成粉;⑤营养组合物对照组,即背景技术所述现有营养组合物;⑥蛋清对照组,选取未受精的鸡蛋,每个鸡蛋取3ml蛋清,冻干成粉。
实验流程:本实验采用D-半乳糖建立衰老动物模型,每日1次颈背部皮下注射 D-半乳糖500mg/(kg-d),连续造模90天,空白对照组颈背部皮下注射等剂量的生理盐水注射液。同时,从建模第1天起,鸡胚营养组合物组、鸡胚组、营养组合物组、蛋清组,分别以相应剂量对大鼠进行灌胃,1~15天以0.3816g/d 对大鼠进行灌胃,16~30天以0.7632g/d 对大鼠进行灌胃,30~90天以1.1448g/d 对大鼠进行灌胃。每天一次,直至第90天,拉颈处死大鼠,采样进行检测。
二、实验方法
1. 外观检测
观察各组大鼠的一般状态,摄食及饮水情况、活动度、毛色、光泽度等方面的改变情况。
2. 形态学观察
将大鼠部分肝、脑、脾、肾、胃肠、皮肤(背部)、心脏、骨骼肌置于10%甲醛中固定,依次经过常规脱水、石蜡浸泡包埋、切片、HE 染色,显微镜观察各组织细胞的形态结构。
3. 免疫组化干细胞标志物检测
石蜡切片脱蜡、水化;PBS洗2~3次各5min;3% H2O2(80%甲醇)滴加在玻片上,室温静置10min;PBS洗2~3次各5min;抗原修复;PBS洗2~3次各5min;滴加正常山羊血清封闭液,室温20min。甩去多余液体,滴加一抗50μl,4°C过夜。过夜后需在37°C孵育45min。PBS洗3次各5分钟;滴加二抗40~50μl,室温静置,或37°C 1h;PBS洗3次各5min;DAB显色5~10min,在显微镜下掌握染色程度;PBS或自来水冲洗10min;苏木素复染1min,盐酸酒精分化;自来水冲洗10~15min;脱水、透明、封片、镜检。
4. 免疫系统功能检测
(1) 脾脏、胸腺指数测定
动物称重后,颈椎脱臼处死,无菌条件下剖取胸腺和脾脏称重,计算指数。
(2) MTT 法检测淋巴细胞增殖情况
无菌制备大鼠脾细胞悬液,RPMI1640 培养液调整细胞密度为2 ×106个/ml。每组设实验孔和对照孔,均设3 个复孔。取100 μl脾细胞悬液加入96 孔板,实验孔加入100μl 含ConA ( T 淋巴细胞增殖反应) 或LPS(B淋巴细胞增殖反应)的RPMI1640 培养液,使终浓度为5 μg/ml,对照孔只加等量的RPMI1640 培养液,将培养板置于37°C,5% CO2培养箱中培养48 h或72h,加入20ul CCK-8,37°C 5% CO2培养箱中孵育4h,酶标仪于450nm 波长处读取吸光值 (A),并计算刺激指数SI,SI = ( 实验孔A-对照孔A) / 对照孔A。
(3) 细胞因子IL-2、IL-4 、IFN-γ和 TGF-β含量检测
眼球后静脉丛取血,静置后离心取血清,ELISA 法测定血清中IL-2、IL-4 、IFN-γ和TGF-β含量。
5. 抗氧化防御系统观察
眼球后静脉丛取血,静置后离心取血清,ELISA 法测定:血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、活性氧(ROS)、铜蓝蛋白(CER)、金属硫蛋白(MT)、丙二醛(MDA)含量。
6. 修复系统检测
大鼠肝、脑、骨髓细胞DNA切除修复、重组修复(双链断裂修复)、错配修复、倾向差错修复(SOS修复)。
常规应用Trizol法提取各标本的总RNA,紫外分光光度计测定A260和A280,以检测RNA含量和纯度,并置放于-70°C保存。
反转录体系20μl,包含样品RNA 1μl,按照TaKaRa的反转录试剂盒提供的说明书进行操作。反转录反应条件为:65°C 1min,30°C 5min,65°C 15min,98°C 5min,5°C 5min。
PCR反应体系50 μl,包含反转录液10 μl。PCR反应条件:94°C预变性1min;97°C变性 20s,64°C退火20s, 72°C 20s延伸,循环30次;72°C延伸5min,4°C恒定。取5μl PCR扩增产物经1%琼脂糖凝胶电泳,紫外灯下凝胶成像。
7. 线粒体、自噬体损伤修复观察
(1) 透射电镜观察
取大鼠部分肝、脾、脑、肾组织标本上不同部位的3张切片进行透射电镜(JEM-100CXE型) 超微结构的观察,每张切片分别随机观察5~6个部位,并按相同放大倍数摄片,每组随机抽取l0张照片,采用Epson微机和Summa Sketch PIus数字化仪,并使用Sigmascan软件,对各组照片作电镜图像的形态进行分析,同时计数线粒体、自噬体的平均数目。
(2) 线粒体膜电位测定
细胞(5×105个)接种于25cm2培养瓶中, 37℃培养24 h后,加入不同浓度的姜黄素分别作用1 h和 6 h 后,收获细胞,PBS洗两遍,重悬于2ml 含1.0 µM 罗丹明123的新鲜培养液中,37°C水浴振摇孵育10 min,离心去掉细胞,荧光比色计测定培养液中罗丹明123的强度,激发波长490 nm,发射波长520 nm,结果以细胞吸收的罗丹明123的荧光强度表示。
8. 端粒酶检测
取大鼠股骨干骨髓细胞进行端粒酶活性分析。Trizol法提取细胞中总RNA,检测所提取的RNA的浓度,并通过A260/A280值检测其纯度。将提取的RNA根据操作说明进行反转录。反转录体系为20μl,反转录反应条件为:25°C 5min,42°C 60min,70°C 5min。将产物cDNA根据扩增体系说明书加入EP管中,放于RT-PCR仪进行实时定量扩增。扩增体系总体积为25μl,反应条件为:95°C预变性30s,95°C变性5s、60°C退火、延长30s,共循环40次。
9. 脂褐素检测
将大鼠断头处死,取出脑组织,肝脏组织在生理盐水中浸泡,用滤纸沥干表面血液污渍,取100mg组织,加入2ml氯仿:甲醇( V/V = 2: l) 混合液,充分振摇3 min,3000r/min离心10 min,吸出氯仿层,再加入2ml氯仿∶甲醇( V/V = 2:1) 混合液,400ul蒸馏水,充分振摇3 min,3000r/min 离心10min,吸出氯仿层,再加氯仿定容至5ml。用荧光分光光度计测定其荧光强度( 激发光波长360 nm,发射光波长420nm,狭缝10nm) 。以新配制的硫酸奎宁标准液( 1 μg/ml) 荧光强度为50,计算每克大脑皮层中所含相当于标准荧光强度的荧光物质微克数,即为脂褐素含量。
10. 自噬检测指标:
脑组织自噬蛋白(LC3-II,Beclin1)的表达量。用RIPA细胞裂解液裂解脑组织,BAC法对其进行浓度测定,采用上样缓冲液将细胞浓度调齐,每孔上样量为40μg。120V电压电泳直至Marker到达浓缩胶与分离胶交界处,转换电压至150V,直至Marker完全分开。根据Marker的指示,切下对应目的蛋白位置的凝胶及GAPDH相对应的凝胶。在湿转仪器中,恒流300mA,将蛋白转至PVDF膜上。将PVDF膜放入5%的封闭液 (5%的脱脂牛奶) 中37°C封闭2h。LC3-II一抗稀释比为1/500、Beclin-1一抗稀释比为1/800,封闭好的PVDF膜与一抗4°C孵育过夜。次日,在摇床中37°C震摇2h,使蛋白与一抗充分混匀。TBST洗膜4次,每次8 min,将膜浸泡于二抗(1/5000稀释)中,于37°C摇床震摇1.5h。TBST洗膜4次,每次8 min。按ECL发光试剂说明书在ImageQuant LAS 500 机器中检测结果。
11. 行为学检测
游泳水迷宫试验采用环形水池,水池中放一低于水面1cm左右的逃避平台。模型小鼠预先训练4天,然后开始正式试验,每只小鼠每天早晚各训练2次,每次持续60s或者当小鼠跳到水中的逃避平台时立即结束。
检测指标:①逃避时间,每只小鼠从放入水中开始,到跳到逃避平台所用时间;②120s内穿越逃避平台次数。
12. 间充质干细胞(BMSCs)的成脂诱导分化
成脂诱导法:将分离纯化的骨髓间充质干细胞常规培养,消化传代,以1×105/well的密度,400ul/well接种于预先放置有盖玻片的6孔培养板中,制备细胞爬片,当细胞生长达80% 融合时,加入10%的胎牛血清、1μmol/L地塞米松、0.5 mmol/L IBMX、10 mg/L牛胰岛素的H-DMEM诱导3天,再用10% 的胎牛血清、10 mg/L牛胰岛素的H-DMEM处理1天,如此循环3次后,用10%的胎牛血清、10 mg/L牛胰岛素的H-DMEM处理7天,每隔三四天换液1次。对照组始终加入含体积分数为10% 的胎牛血清、10 mg/L牛胰岛素的H-DMEM,每隔三四天换液1次。诱导后细胞用PBS洗涤2~3次,10% 甲醛室温固定40min。PBS洗2~3次,饱和油红O染液用一蒸水以3:2稀释,室温染色30min,PBS洗数次直至无肉眼观察到沉渣,光镜观察。
13. 间充质干细胞(BMSCs)成骨细胞培养及鉴定
取P2代骨髓间充质干细胞,按2×105个/孔接种于12孔板,当细胞贴壁生长达80% 融合时,吸除孔中培养液,换为成骨细胞诱导液 (含高糖DMEM、体积分数为10% 胎牛血清、10- 8mol/L地塞米松、10-2 mol/L β-甘油磷酸钠、50 mg/L抗坏血酸),每3天更换诱导液1次。培养14天后,吸除培养液,PBS洗2次,10%甲醛室温固定40 min,PBS洗3次,加入0.1% 茜红素(3:2稀释),室温染色10min,去除染液,PBS洗3次后,显微镜下观察。
14. 流式细胞仪分析
取大鼠左胫骨,去除表面肌肉组织,修剪骨垢,用18 号针插入胫骨踝端,用PBS 5mL 冲洗入试管内,以200目筛网过滤。1000 r/min 离心5min,弃上清液。将细胞加入5mL Percoll分离液(相对比重1.073g/L),2000r/min,离心25min,取中间单个核细胞层。PBS洗涤后调整单个核细胞数为l×106/管,每样本3管,测定管加入FITC标记的CD34或CD45抗体,37°C孵育2小时。然后PBS洗涤一次。用流式细胞仪检测表达CD34/45阳性细胞数量变化,以标记抗体呈阳性的细胞百分率作为表达CD34/CD45 蛋白(骨髓造血干细胞标志物)的计量标准。同时以只加标记荧光素的羊抗兔IgG和不加抗体的细胞作阴性对照。
15. mRNA表达谱芯片分析
取大鼠股骨干骨髓细胞进行mRNA表达谱芯片分析。
16. 统计学分析
所有数据均用SPSS 17.0软件进行统计分析。每一项分析至少有三次结果。数值用均数±SD表示,采用t检验。P<0.05认为有显著性差异,并在统计学上有意义。
三、实验结果
(一)对衰老大鼠全身情况的影响
衰老模型组连续注射D-半乳糖45天后,与正常对照组相比,毛色逐渐枯槁无光泽,易脱落,皮肤弹性差,精神萎靡,倦怠嗜睡,群体喜扎堆,进食量明显减少,体重增加缓慢,呈现出明显衰老体征。第90天,衰老模型组大鼠体重(379.53±49.34g)与正常对照组(495.08±64.37g,#P<0.05)相比明显下降(见图7,表3)。

本发明实施例2组与衰老模型组比较,大鼠的精神状态得到了程度改善,行动活泼,进食量明显增大,体重增加显著(见图7,表3,*P<0.05)。与衰老模型组相比,鸡胚组、营养组合物组以及蛋清组大鼠的体重略增加,但无显著差异。
以上结果表明,本发明的鸡胚营养组合物可以改善衰老大鼠的精神状态及体重。
(二)鸡胚营养组合物对衰老大鼠各器官的影响(光镜观察)
肝的病理切片HE染色显示,与正常对照组相比,衰老模型组大鼠肝脏组织中肝小叶结构不明显,肝索排列混乱,中央静脉数量减少,肝细胞核固缩,染色质深染,肝细胞内可见大量脂褐素颗粒(见图8);与衰老模型组相比,鸡胚营养组合物组大鼠肝脏组织中肝小叶结构清晰,肝索排列整齐,肝细胞内未见脂褐素颗粒,细胞形态规整;鸡胚组和营养组合物组大鼠肝脏组织的损伤有一定恢复,而蛋清组大鼠肝脏组织的损伤无明显恢复。
肾的病理切片HE染色显示,与正常对照组相比,衰老模型组大鼠肾小球明显固缩,体积大小不一,出现代偿性肾小球肥大,肾小管明显扩张(见图8);与衰老模型组相比,鸡胚营养组合物组大鼠肾小球体积大小均一,结构清晰,肾小管未见扩张;鸡胚组和营养组合物组大鼠肾脏组织的损伤有一定恢复,而蛋清组大鼠肾脏组织的损伤无明显恢复。
脑的病理切片HE染色显示,与正常对照组相比,衰老模型组大鼠脑组织海马区神经元细胞排列紊乱,大小不一,细胞出现轻微肿胀,周围间隙增宽(见图8);与衰老模型组相比,鸡胚营养组合物组大鼠脑组织海马区神经元细胞排列较为整齐,核深染,核膜清晰,核仁明显;鸡胚组和营养组合物组大鼠脑组织的损伤有一定恢复,而蛋清组大鼠脑组织的损伤无明显恢复。
肠的病理切片HE染色显示,与正常对照组相比,衰老模型组大鼠肠绒毛部分脱落缺失、排列稀疏,肠固有层腺体萎缩、数量减少,肌层变薄;与衰老模型组相比,鸡胚营养组合物组大鼠肠绒毛结构完整、排列规则整齐、长度较一致,腺体较丰富、大小均匀;鸡胚组和营养组合物组大鼠肠组织的损伤有一定恢复,而蛋清组大鼠肠组织的损伤无明显恢复(见图8)。
肌肉的病理切片HE染色显示,与正常对照组相比,衰老模型组大鼠肌纤维断裂,出现波浪样改变(见图8);与衰老模型组相比,鸡胚营养组合物组大鼠肌纤排列整齐、细密,肌束方向一致;鸡胚组和营养组合物组大鼠肌肉组织的损伤有一定恢复,而蛋清组大鼠肌肉组织的损伤无明显恢复。
皮肤的病理切片HE染色显示,与正常对照组相比,衰老模型组大鼠皮下真皮组织疏松,胶原含量极少,表皮细胞层可见大量色素沉着;与衰老模型组相比,鸡胚营养组合物组大鼠皮下真皮组织致密,胶原含量丰富,表皮细胞层色素沉着明显减少;鸡胚组和营养组合物组大鼠皮肤组织的损伤有一定恢复,而蛋清组大鼠皮肤组织的损伤无明显恢复(见图8)。
(三)鸡胚营养组合物对衰老大鼠干细胞的影响
1. 骨髓造血干细胞
CD34抗原属于钙黏蛋白家族成员,是一种阶段特异性白细胞分化抗原,选择性的表达于造血干细胞表面。CD45是I型跨膜蛋白,通过激活蛋白酪氨酸磷酸酶调节信号传导,维持T淋巴细胞正常活化,参与多种免疫功能,广泛存在于造血干细胞表面。采用流式细胞仪检测各组大鼠骨髓造血干细胞标志物CD34、CD45的表达情况。结果显示,与正常对照组相比,衰老模型组大鼠骨髓造血干细胞CD34、CD45的表达量明显减少(见图9,#P<0.05 衰老组与正常组;* P<0.05 鸡胚营养组合物组、鸡胚组、营养组合物组与衰老组),表明衰老大鼠骨髓造血干细胞数量减少;与衰老模型组相比,鸡胚营养组合物组、鸡胚组、营养组合物组大鼠骨髓造血干细胞CD34、CD45的表达量增多,且鸡胚营养组合物组大鼠骨髓造血干细胞CD34、CD45的表达量最多,而与衰老模型组相比,蛋清组大鼠骨髓造血干细胞CD34、CD45的表达量无明显变化。以上结果表明鸡胚营养组合物组具有促进衰老大鼠骨髓造血干细胞增殖的作用。
2. 肝脏干细胞
CD90又名Thy-1抗原,为糖基磷脂酰肌醇连接糖蛋白,是细胞黏附分子免疫球蛋白超家族的成员,主要参与细胞之间以及细胞与基质之间的相互作用,是肝干细胞的表面标志物。CK19是一种低分子角蛋白,是细胞骨架蛋白的一种亚型,其特异性表达于肝卵圆细胞表面(卵圆细胞为肝脏干细胞的子代细胞)。采用免疫组化检测衰老大鼠肝干细胞标志物CD90、CK19的表达情况。结果显示,与正常对照组相比,衰老模型组大鼠肝干细胞CK19的表达量显著减少(见图10,#P<0.05 衰老组与正常组;* P<0.05 鸡胚营养组合物组、鸡胚组、营养组合物组与衰老组),鸡胚营养组合物处理衰老大鼠后,大鼠肝干细胞CK19的表达量显著增加;与衰老模型组相比,鸡胚组、营养组合物组和蛋清组大鼠肝干细胞CK19的表达量无明显变化。以上结果表明鸡胚营养组合物对衰老大鼠肝干细胞的增殖有一定作用。
3. 肾脏干细胞
CD133又称Prominin-1,是细胞表面蛋白超家族中的一员,最初发现是造血干细胞表面抗原标志,近年来也被认为是肾脏干细胞表面分子标志。OCT4作为肾脏干细胞的标记物,对于维持干细胞的多能性和自我更新具有极其重要的作用。采用免疫组化检测衰老大鼠肾脏干细胞标志物CD133、OCT4的表达情况。结果显示,与正常对照组相比,衰老模型组大鼠CD133、OCT4的表达量显著降低(见图11,#P<0.05 衰老组与正常组;* P<0.05 鸡胚营养组合物组、鸡胚组、营养组合物组与衰老组),鸡胚营养组合物处理衰老大鼠后,大鼠CD133、OCT4的表达量明显增高;与衰老模型组相比,鸡胚组、营养组合物组大鼠CD133、OCT4的表达量增高,而蛋清组大鼠CD133、OCT4的表达量无明显变化。以上结果表明鸡胚营养组合物能够显著促进衰老大鼠肾脏干细胞的增殖。
4. 肠粘膜干细胞
Musashi-1在神经干细胞中选择性地表达,而目前研究显示,Musashi-1在肠道干细胞中也呈高表达。Lgr5是小肠干细胞的特异性表面分子标记物,在小肠的稳态维持中发挥重要的作用。采用免疫组化检测衰老大鼠肠道干细胞标志物Musashi-1、Lgr5的表达。结果显示,与正常对照组相比,衰老模型组大鼠肠道干细胞Musashi-1、Lgr5的表达量显著降低(见图12,#P<0.05 衰老组与正常组;* P<0.05 鸡胚营养组合物组、鸡胚组、营养组合物组与衰老组),鸡胚营养组合物处理衰老大鼠后,大鼠肠道干细胞Musashi-1、Lgr5的表达量明显增高;与衰老模型组相比,鸡胚组、营养组合物组大鼠肠道干细胞Musashi-1、Lgr5的表达量增高,而蛋清组大鼠肠道干细胞Musashi-1、Lgr5的表达量无明显变化。以上结果表明鸡胚营养组合物能够显著促进衰老大鼠肠黏膜干细胞的增殖。
5. 神经干细胞
Nestin是神经干细胞的特异性标记蛋白,具有促进细胞结构重构,修复神经损伤的作用。CD133是一种独特的干细胞标志,最新研究表明,CD133不仅作为造血干细胞的表面标志,还可以作为其他非造血干细胞的表面标志,如神经干细胞。采用免疫组化检测衰老大鼠神经干细胞标志物Nestin、CD133的表达情况。结果显示(见图13,#P<0.05 衰老组与正常组;* P<0.05 鸡胚营养组合物组、鸡胚组、营养组合物组与衰老组),与正常对照组相比,衰老模型组大鼠神经干细胞Nestin、CD133的表达量明显减少,鸡胚营养组合物处理衰老大鼠后,大鼠神经干细胞Nestin、CD133的表达量明显增高;与衰老模型组相比,鸡胚组、营养组合物组大鼠神经干细胞Nestin、CD133的表达量增高,而蛋清组大鼠神经干细胞Nestin、CD133的表达量无明显变化。以上结果表明鸡胚营养组合物能够显著促进衰老大鼠神经干细胞的增殖。
6.胃干细胞
胃Lgr5干细胞作为维持胃粘膜上皮细胞自我更新的组织干细胞,平均每天分裂一次,是胃粘膜上皮干细胞的标记物。Musashi-1作为一种RNA结合蛋白在不对称分裂过程中起重要作用,与胃肠道干细胞标记Lgr5在正常胃粘膜中存在共表达。采用免疫组化检测衰老大鼠肾脏干细胞标志物Lgr5、Musashi-1的表达情况,结果显示,与正常对照组相比,衰老模型组大鼠胃干细胞Lgr5的表达量明显减少(见图14,#P<0.05 衰老组与正常组;* P<0.05 鸡胚营养组合物组、鸡胚组、营养组合物组与衰老组),鸡胚营养组合物处理衰老大鼠后,大鼠胃干细胞Lgr5的表达量明显增高;与衰老模型组相比,鸡胚组、营养组合物组大鼠胃干细胞Lgr5的表达量增高,而蛋清组大鼠胃干细胞Lgr5的表达量无明显变化,表明鸡胚营养组合物能够促进衰老大鼠胃干细胞的增殖。以上结果表明鸡胚营养组合物对衰老大鼠胃干细胞的增殖有一定作用。
7. 胰腺干细胞
CK19是导管上皮细胞向干细胞转化的标志,在胰腺干细胞中阳性表达。研究表明,人及大鼠胰管、胰岛组织中存在表达nestin的胰腺干细胞。本研究采用免疫组化检测衰老大鼠胰腺干细胞标志物CK19、Nestin的表达情况,结果显示(见图15,#P<0.05 衰老组与正常组;*P<0.05 鸡胚营养组合物组、鸡胚组、营养组合物组与衰老组),鸡胚营养组合物组、鸡胚组、营养组合物组及蛋清组的CK19高于衰老组,甚至高于正常组,以上结果表明鸡胚营养组合物对衰老大鼠胰腺干细胞的增殖有一定作用。
(四) 鸡胚营养组合物对衰老大鼠骨髓间充质干细胞的影响
1. 鸡胚营养组合物对衰老大鼠骨髓间充质干细胞生长的影响
各组骨髓间充质干细胞增殖能力结果见图16,衰老模型组大鼠骨髓间充质干细胞生长速度显著低于正常对照组,蛋清组大鼠骨髓间充质干细胞生长速度与衰老模型组相比无明显差异,鸡胚营养组合物组、鸡胚组、营养组合物组大鼠骨髓间充质干细胞生长速度与衰老模型组相比有明显差异,且鸡胚营养组合物组大鼠骨髓间充质干细胞生长速度仅低于正常组而明显高于其它组,以上结果表明,鸡胚营养组合物能够促进衰老大鼠骨髓间充质干细胞的增殖。
2. 鸡胚营养组合物对衰老大鼠骨髓间充质干细胞分化趋势的影响
2.1 脂肪细胞的诱导及成脂细胞相关基因的表达
正常组、衰老组、鸡胚营养组合物组、鸡胚组、营养组合物组、蛋清组P2代骨髓间充质干细胞分别用成脂细胞诱导培养液进行诱导培养,培养初期细胞逐渐变圆,少数细胞的细胞质内有高折光性的小脂滴出现,晶莹透亮,主要集中在细胞核周围。继续诱导培养第19d后细胞形态逐渐向类圆形转变,体积变大,整个细胞内从胞膜到胞核之间充满大小较均一的脂滴,成葡萄串珠状,脂肪滴被油红O染色为明亮的红色(见图17);显微镜下观察,衰老模型组骨髓间充质干细胞成脂细胞分化诱导能力高于正常对照组,鸡胚营养组合物组处理衰老大鼠后,骨髓间充质干细胞成脂细胞分化诱导能力显著下降;与衰老模型组比,鸡胚组、营养组合物组大鼠骨髓间充质干细胞成脂细胞分化诱导能力降低,而蛋清组大鼠骨髓间充质干细胞成脂细胞分化诱导能力无明显变化。
为了更客观地比较鸡胚营养组合物对大鼠骨髓间充质干细胞成脂细胞的影响,进一步检测了成脂细胞相关基因Pparr mRNA,Fabp4 mRNA的表达水平。结果显示,Fabp4和Pparr的表达水平与鸡胚营养组合物干预有显著相关性(见图17,#P<0.05 衰老组与正常组;* P<0.05 鸡胚营养组合物组、鸡胚组、营养组合物组与衰老组)。以上结果表明,鸡胚营养组合物可以降低衰老大鼠骨髓间充质干细胞成脂细胞分化诱导能力。
2.2成骨细胞的诱导及成骨细胞相关基因的表达
正常组、衰老组、鸡胚营养组合物组、鸡胚组、营养组合物组、蛋清组P2代骨髓间充质干细胞分别用成骨细胞诱导培养液诱导培养,诱导7d左右,可见细胞表面有较多棕褐色细小颗粒出现。随着诱导时间延长,棕褐色颗粒明显增加, 14天后茜红素染色(见图18)。衰老模型组骨髓间充质干细胞黑色沉积物出现较晚,钙结节数量明显少于正常对照组。与衰老模型组相比,鸡胚营养组合物组骨髓间充质干细胞成骨细胞分化诱导能力显著增强;鸡胚和营养组合物分别处理衰老大鼠后,骨髓间充质干细胞成骨细胞分化诱导能力增强,而蛋清组大鼠骨髓间充质干细胞成骨细胞分化诱导能力无明显变化,表明鸡胚营养组合物能增强衰老大鼠间充质干细胞成骨细胞分化诱导能力。
为比较鸡胚营养组合物对衰老大鼠骨髓间充质干细胞成骨细胞过程的影响,进一步检测了成骨细胞相关基因mRNA和Bglap mRNA的表达水平。结果显示,Alp和Bglap的表达水平与鸡胚营养组合物干预有明显的相关性(见图18,#P<0.05 衰老组与正常组;* P<0.05鸡胚营养组合物组、鸡胚组、营养组合物组与衰老组)。
以上结果表明衰老模型组与正常照组相比,衰老模型组大鼠骨髓间充质干细胞向成骨细胞分化能力减弱;而与衰老模型组相比,鸡胚营养组合物组、鸡胚组、营养组合物组骨髓间充质干细胞向成骨细胞分化能力均增强,但鸡胚营养组合物组骨髓间充质干细胞向成骨细胞分化能力更强。
3. 鸡胚营养组合物对衰老大鼠骨髓间充质干细胞端粒酶的影响
端粒酶是目前发现的唯一由RNA和蛋白质构成的核糖核蛋白酶,具有逆转录活性,能够以自身的RNA为模板合成端粒酶DNA。端粒酶的主要功能是对端粒DNA的延伸以及修复断裂的染色体末端,通过修复,可使染色体保持完整,防止染色体的相互融合、重组及其他损害,从而保持基因组的完整和稳定。端粒酶的长短和稳定决定了细胞的寿命,并与衰老和癌变密切相关。端粒序列长度在细胞生长的过程中不断减少,当缩短到一定程度,细胞基因组的稳定性就会遭到破坏,细胞停止分裂并退出细胞周期,开始出现衰老。因此,可以通过减缓端粒酶缩短的速率,实现细胞衰老的延缓。
在骨髓间充质干细胞中,与正常对照组相比,衰老模型组大鼠端粒酶活性明显降低(见图19,#P<0.05);与衰老模型组相比,鸡胚营养组合物组、鸡胚组、营养组合物组大鼠端粒酶活性均升高,而鸡胚营养组合物组大鼠端粒酶活性最高(见图15,*P<0.05),蛋清组大鼠端粒酶活性无明显变化(见图15,P>0.05)。
以上结果表明,鸡胚营养组合物能够提高衰老大鼠骨髓间充质干细胞端粒酶活性。
(五)鸡胚营养组合物对衰老大鼠线粒体、自噬体的影响
1. 线粒体、自噬体形态观察
肝组织透射电镜显示,与正常对照组相比,衰老模型组大鼠肝细胞内线粒体、自噬小体数目明显减少,脂滴大量沉积在肝细胞胞浆中;与衰老模型组相比,鸡胚营养组合物组大鼠肝细胞内线粒体数目明显增多,自噬小体数量显著增多,肝细胞胞浆中未见脂滴;鸡胚组、营养组合物组大鼠肝细胞内线粒体、自噬小体数目略有增多,而蛋清组大鼠肝细胞内线粒体、自噬小体数目无明显变化(见图20)。
脑组织透射电镜显示,与正常对照组相比,衰老模型组大鼠脑海马部神经元细胞内线粒体、自噬小体数目明显减少;与衰老模型组相比,鸡胚营养组合物组大鼠脑海马部神经元细胞内线粒体、自噬小体数目明显增多;鸡胚组、营养组合物组大鼠脑海马部神经元细胞内线粒体、自噬小体数目略有增多,而蛋清组大鼠脑海马部神经元细胞内线粒体、自噬小体数目无明显变化(见图21)。
肾脏透射电镜显示,与正常对照组相比,衰老模型组大鼠肾小球基底膜足突大面积融合;与衰老模型组相比,鸡胚营养组合物组大鼠肾小球基底膜足突融合程度明显降低,排列趋于正常;鸡胚组、营养组合物组大鼠肾小球基底膜足突融合程度略有降低,而蛋清组大鼠肾小球基底膜足突融合程度无明显变化(见图22)。
肾小管上皮细胞与正常对照组相比,衰老模型组大鼠肾小管上皮细胞内线粒体、自噬小体数目明显减少;与衰老模型组相比,鸡胚营养组合物组大鼠肾小管上皮细胞内线粒体、自噬小体数目明显增多;鸡胚组、营养组合物组大鼠肾小管上皮细胞内线粒体、自噬小体数目略有增多,而蛋清组大鼠肾小管上皮细胞内线粒体、自噬小体数目无明显变化(见图23)。
脾脏透射电镜显示,与正常对照组相比,衰老模型组大鼠脾脏B细胞形态不整,局部胞膜缺失,线粒体数量明显减少;与衰老模型组相比,鸡胚营养组合物组大鼠脾脏B细胞形态恢复正常,胞膜完整,线粒体数目明显增多;鸡胚组、营养组合物组大鼠B细胞形态恢复正常,胞膜完整,线粒体数目略有增多,而蛋清组大鼠B细胞形态不整,胞膜碎裂,线粒体数量减少(见图24)。
2. 线粒体膜电位测定
线粒体是促进能量转换,参与细胞凋亡的重要细胞器,由于膜上各种生物泵的作用,使膜内外维持着不同梯度的离子浓度,产生线粒体膜电位,线粒体膜电位的大小直接反应线粒体功能及数量。采用荧光探针罗丹明123(Rh123)测定鸡胚营养组合物对线粒体膜电位的影响。Rh123是一种可透性细胞膜的阳离子荧光染料,是一种线粒体跨膜电位的指示剂,它能够透过活细胞的细胞膜,并在细胞的线粒体内聚集,从而发出黄绿色荧光。Rh123对线粒体膜电位非常敏感,其荧光增强说明线粒体数量的增多和线粒体内电负性的增强,反之则减弱。
与正常对照组相比,衰老模型组大鼠骨髓造血细胞、肝细胞、脾细胞、脑细胞、肾细胞内线粒体膜电位呈明显下降趋势,表明线粒体功能缺失(见表4及图25);与衰老模型组相比,鸡胚营养组合物组大鼠骨髓造血细胞、肝细胞、脾细胞、脑细胞、肾细胞内线粒体膜电位呈明显增高的趋势;与衰老模型组相比,鸡胚组、营养组合物组大鼠骨髓造血细胞、肝细胞、脑细胞内线粒体膜电位增高,有显著差异,脾细胞、肾细胞内线粒体膜电位无明显差异,而蛋清组大鼠骨髓造血细胞、肝细胞、脾细胞、脑细胞、肾细胞内线粒体膜电位无明显变化(见表4及图25,#P<0.05 衰老组与正常组;* P<0.05 鸡胚营养组合物组、鸡胚组、营养组合物组与衰老组)。
以上结果表明鸡胚营养组合物可促进衰老大鼠骨髓造血细胞、肝细胞、脾细胞、脑细胞、肾细胞线粒体膜电位升高。

3. 鸡胚营养组合物对衰老大鼠脑组织自噬体的影响
自噬是存在于真核细胞中的一种自我保护机制,能够通过自噬体与溶酶体的融合去除受损细胞器从而维持细胞稳态。随着年龄的增长,细胞内自噬作用开始减弱,导致细胞适应外界环境和自身防御反应的能力下降,损伤的细胞结构及大量氧自由基等活性氧化合物不能有效地被清除,细胞稳态发生变化,加速细胞老化。因此,衰老与细胞自噬体存在着密切的关系。
调控自噬分子机制很复杂,整个过程需要有众多分子的参与,如微管相关蛋白轻链3(LC3)和Beclin-1,二者被确认为自噬基因,在自噬体的形成过程中扮演着重要角色。LC3是自噬泡形成过程中的关键蛋白,其具有两种形式:LC3-I和其蛋白水解衍生物LC3-II。LC3-I定位于包浆,LC3-II定位于自噬双层膜上。一旦自噬体与溶酶体融合,自噬体内的LC-II便被溶酶体中的水解酶降解,所以LC3-II含量或LC-II/LC-I比值的大小反映了自噬体活性的强弱。同样,Beclin-1也是自噬形成过程中的关键基因,代表了自噬活性。因此,本实验选用这两个自噬蛋白LC3-II、Beclin-1。
与正常对照组相比,衰老模型组大鼠脑组织中LC3-II、Beclin-1蛋白表达量显著降低(见图26,#P<0.05);鸡胚营养组合物处理衰老大鼠后,大鼠脑组织中LC3-II、Beclin-1蛋白表达量明显升高(见图26,*P<0.05);与衰老模型组相比,鸡胚组和营养组合物组大鼠脑组织中LC3-II、Beclin-1蛋白表达量升高,而蛋清组大鼠脑组织中LC3-II、Beclin-1蛋白表达量无明显变化。
以上结果表明,鸡胚营养组合物在调节自噬水平对抗衰老大鼠脑组织损伤过程中发挥重要的作用。
(六)鸡胚营养组合物对衰老大鼠基因损伤修复的影响
1. 切除修复
1.1核苷酸切除修复
核苷酸切除修复 (NER) 是人类重要的DNA修复系统,可识别并修复多种结构不相关的DNA损伤,主要修复嘧啶二聚体、DNA加合物和各种引起DNA螺旋扭曲变形的损伤。本实验选取了上述 3 个 NER 基因 (Ercc1,Ercc2,Xpc),从转录水平检测了其在实验组中肝脏、骨髓以及脑中的表达变化(见图27)。
在脑组织中,与正常对照组相比,衰老模型组大鼠脑组织中核苷酸切除修复基因Xpc表达量下调;与衰老模型组相比,鸡胚营养组合物组、鸡胚组、营养组合物组大鼠脑组织中Xpc表达量明显增加,但鸡胚营养组合物组大鼠脑组织中Xpc表达量最高,蛋清组大鼠脑组织中Xpc表达量无明显变化。
以上结果表明鸡胚营养组合物对衰老大鼠核苷酸切除修复基因损伤具有恢复作用。
1.2碱基切除修复
碱基切除修复 (base excision repair, BER) 是指切除和替换由内源性烷化剂和外源性致癌剂作用产生的DNA碱基损伤,DNA糖基化酶参与此过程,随后糖-磷酸键断裂,切去碱基残基,在修复酶作用下DNA链连接修复损伤。本实验选取了上述 4 个 BER 基因(Apex1,Ogg1,Polb,MTH1),从转录水平检测了其在各组中的表达变化(见图28)。
在骨髓中,与正常对照组相比,衰老模型组大鼠碱基切除修复基因MTH1表达量下调;鸡胚营养组合物处理衰老大鼠后,鸡胚营养组合物组、鸡胚组和营养组合物组大鼠骨髓中碱基切除修复基因MTH1表达量均增加,但鸡胚营养组合物组大鼠骨髓中碱基切除修复基因MTH1表达量最高;与衰老模型组相比,蛋清组大鼠骨髓中碱基切除修复基因MTH1表达量无明显变化。
在脑组织中,与正常对照组相比,衰老模型组大鼠脑组织中碱基切除修复基因MTH1表达量下调;与衰老模型组相比,鸡胚营养组合物组、鸡胚组、营养组合物组大鼠脑组织中MTH1表达量明显增加,但鸡胚营养组合物组大鼠脑组织中MTH1表达量最高,而蛋清组大鼠脑组织中MTH1表达量无明显变化。
以上结果表明鸡胚营养组合物对衰老大鼠碱基切除修复基因损伤具有一定的恢复作用。
2. 错配修复
DNA错配修复系统 (MMR) 广泛存在于生物体中,是进化上保守的生化通路,由一系列特异性修复DNA碱基错配的酶分子 (错配修复基因产物) 组成,主要修复DNA复制过程中产生的错配,从而保证遗传物质的完整性和稳定性,避免遗传物质发生突变。本实验选用最常用的4个错配修复基因PMS2、MLH1、MSH3、MSH2(见图29)。
在肝脏和骨髓中,与正常对照组相比,衰老模型组大鼠中错配修复基因MSH2表达量均下调;鸡胚营养组合物处理衰老大鼠后,大鼠肝脏组织和骨髓中错配修复基因MSH2表达量增加;与衰老模型组相比,鸡胚组和营养组合物组大鼠肝脏组织和骨髓中错配修复基因MSH2表达量均增加,但增加量低于鸡胚营养组合物组,而蛋清组大鼠肝脏组织和骨髓中错配修复基因MSH2表达量无明显变化。
以上结果表明鸡胚营养组合物对衰老大鼠错配修复基因损伤具有一定的恢复作用。
3. 重组修复(双链断裂修复)
双链断裂是真核生物DNA损伤中最严重的类型,由于缺乏一条完整的互补链作为修复模板,DNA序列难以完全恢复,易造成遗传信息的丢失。准确的DNA修复是维持染色体完整性的前提,同源重组则在 DNA修复过程中发挥关键作用。本实验选用最常用的4个同源重组修复基因 Rad51、Rad52、Xrcc1、Xrcc2(见图30)。
在肝脏组织中,与正常对照组相比,衰老模型组大鼠同源重组修复基因Xrcc1、Xrcc2表达量均下调;鸡胚营养组合物处理衰老大鼠后,大鼠肝脏组织中同源重组修复基因Xrcc1、Xrcc2表达量均显著增加;与衰老模型组相比,鸡胚组和营养组合物组大鼠肝脏组织中同源重组修复基因Xrcc1、Xrcc2表达量增加,但增加量低于鸡胚营养组合物组,而蛋清组大鼠肝脏组织中同源重组修复基因Xrcc1、Xrcc2表达量无明显变化。
在骨髓中,与正常对照组相比,衰老模型组大鼠同源重组修复基因Rad51表达量下调;与衰老模型组相比,鸡胚营养组合物组、鸡胚组、营养组合物组大鼠骨髓中同源重组修复基因Rad51表达量均明显增加,但鸡胚营养组合物组大鼠骨髓中同源重组修复基因Rad51表达量最高,蛋清组大鼠骨髓中同源重组修复基因Rad51表达量无明显变化。
在脑组织中,与正常对照组相比,衰老模型组大鼠同源重组修复基因Rad51、Xrcc1表达量均下调。鸡胚营养组合物处理衰老大鼠后,大鼠肝脏组织中同源重组修复基因Rad51、Xrcc1表达量均显著增加;与衰老模型组相比,鸡胚组和营养组合物组大鼠肝脏组织中同源重组修复基因Rad51、Xrcc1表达量增加,而蛋清组大鼠肝脏组织中同源重组修复基因Rad51、Xrcc1表达量均无明显变化。
以上结果表,鸡胚营养组合物对衰老大鼠同源重组修复基因的损伤具有恢复作用。
4. SOS修复(错误倾向修复)
SOS修复是指DNA受到严重损伤、细胞处于危急状态时所诱导的一种DNA修复方式,它能提高复制对模板损伤的容忍能力,即降低了复制对DNA模板的要求,使DNA复制能够正常进行,保持DNA双链的完整性。本实验选用最常用的两个SOS修复基因RecA、LexA(见图31)。
在肝脏,骨髓组织中,与正常对照组相比,衰老模型组大鼠SOD修复基因RecA表达量均下调;鸡胚营养组合物处理衰老大鼠后,大鼠肝脏、骨髓组织中SOD修复基因RecA表达量均显著增加;与衰老模型组相比,鸡胚组和营养组合物组大鼠肝脏、骨髓组织中SOD修复基因RecA表达量有一定的增加;蛋清处理衰老大鼠后,大鼠肝脏,骨髓组织中SOD修复基因RecA表达量无变化。
以上结果表明鸡胚营养组合物对衰老大鼠SOS修复基因的损伤具有恢复作用。
(七)鸡胚营养组合物对衰老大鼠行为学的影响
Morris水迷宫(Morris water maze, MWM)实验是一种强迫实验动物(大鼠、小鼠)游泳,学习寻找隐藏在水中平台的一种实验,主要用于测试实验动物对空间位置觉和方向觉(空间定位)的学习记忆能力,由于对年龄相关性空间记忆损害有可靠的敏感性,Mirros水迷宫是判断老年小鼠空间学习记忆能力的有效工具,将实验动物的学习记忆障碍和感觉、运动缺陷等分离开来。
设计的水迷宫主要有一圆柱型水池和一可移动位置的站台组成。水池高120cm,直径150cm,站台直径8cm,水池上空通过一个数字摄像机和计算机相连接。预先在水池中注入清水,水深70cm,水池下方为黑色,便于摄像头采集大鼠的运动轨迹,站台表面为黑色,使大鼠不能看到,水面高出站台表面0.5-1cm。水温控制在22±0.5°C。
实验操作:
(1)将动物(大鼠或小鼠)头朝池壁放入水中,东、西、南、北四个位置随机选取一个作为起始位置,记录动物找到水下平台的时间(s)。在前几次训练中,如果该时间超过60s,则引导动物到平台,让动物在平台上停留10s。
(2)将动物移开、擦干。必要时将动物(尤其是大鼠)放在150W的白炽灯下烤5min,放回笼内。每只动物每天训练4次,两次训练之间间隔15~20min,连续训练5d。
(3)最后一次获得性训练结束后的第二天,将平台撤除,开始120s的探查训练。将动物由原先平台象限的对侧放入水中。记录动物在目标象限(原先放置平台的象限)所花的时间和进入该象限的次数,以此作为空间记忆的检测指标。
(4)测定动物的工作记忆(working memory)。探查训练结束后的第二天,开始维持4天的对位训练。将平台放在原先平台所在象限的对侧象限,方法与获得性训练相同。每天训练4次。连续训练5d,检测时记录每次找到平台的时间和游泳距离以及游泳速度。
与正常对照组相比,衰老模型组大鼠找到藏在水下的逃避平台,所需的时间明显延长(见表5),而撤掉平台后,穿越平台的次数明显缩短;与衰老模型组相比,鸡胚营养组合物组、鸡胚组、营养组合物组大鼠找到藏在水下的逃避平台,所需的时间明显缩短,撤掉平台后,穿越平台的次数明显增加(见表6);蛋清组与衰老模型组相比较,大鼠找到藏在水下的逃避平台,所需的时间略缩短,撤掉平台后,穿越平台的次数减少,但差异无统计学意义。

以上结果显示,鸡胚营养组合物可以改善衰老大鼠学习记忆能力。
(八)鸡胚营养组合物对衰老大鼠免疫系统功能的影响
1. 脾脏、胸腺指数测定
随着机体渐进性衰老,免疫器官,免疫细胞、免疫相关因子的数量和功能会发生明显变化。脾脏是最大的外周免疫器官,其为各种成熟的淋巴细胞提供定居场所以及发生免疫应答。而胸腺是免疫调节的中枢器官,主要为T淋巴细胞的分化、成熟提供场所,对于机体的细胞免疫功能以及免疫反应的发生具有重要作用。脾脏和胸腺均会随着机体的渐进性衰老而质量变轻以及功能下降。
与正常对照组相比,衰老模型组大鼠脾脏指数、胸腺指数均明显降低,其差异具有统计学意义(见图29,#P<0.05),表明衰老模型组大鼠的脾脏、胸腺发生萎缩;与衰老模型组相比,鸡胚营养组合物组、鸡胚组、营养组合物组大鼠脾脏指数、胸腺指数均明显升高(见图33,*P<0.05),但鸡胚营养组合物组大鼠脾脏指数、胸腺指数最大,而蛋清组大鼠脾脏指数、胸腺指数无明显变化。
以上结果提示,鸡胚营养组合物有抑制脾脏、胸腺组织结构退化的作用。
2. MTT 法检测淋巴细胞的增殖
细胞增殖是动物生长、发育和繁殖的基础,哺乳动物都是以细胞增殖的方式产生新的细胞,替代或补充体内衰老和死亡的细胞。细胞经过一系列的增殖传代后将进入一种停滞状态(衰老),此时,由于细胞表面有丝分裂原受体表达量减少,外界给予有丝分裂源刺激,细胞的增殖能力仍将下降。
与正常对照组相比,衰老模型组大鼠脾脏组织中,48h T、B淋巴细胞转化刺激指数(SI)明显降低(见图34,#P<0.05);与衰老模型组相比,鸡胚营养组合物组、鸡胚组、营养组合物组大鼠脾脏组织中,48h T、B淋巴细胞转化刺激指数(SI)均升高,但鸡胚营养组合物组大鼠脾脏组织48h T、B淋巴细胞转化刺激指数(SI)最高(见图34,*P<0.05),而蛋清组大鼠脾脏组织中,48hT、B淋巴细胞转化刺激指数(SI)无明显变化。
与正常对照组相比,衰老模型组大鼠脾脏组织中,72h T、B淋巴细胞转化刺激指数(SI)显著降低(见图34,#P<0.05);鸡胚营养组合物处理衰老大鼠后,大鼠脾脏组织中,72hT、B淋巴细胞转化刺激指数明显升高(见图34,*P<0.05);与衰老模型组相比,鸡胚组、营养组合物组大鼠脾脏组织中,72h T、B淋巴细胞转化刺激指数均升高,但升高幅度低于鸡胚营养组合物组,而蛋清组大鼠脾脏组织中,48h T、B淋巴细胞转化刺激指数(SI)无明显变化(见图34,P>0.05)。
以上结果表明鸡胚营养组合物能够改善衰老大鼠T、B细胞增殖功能,增强衰老大鼠的机体免疫力。
3. 细胞因子IL-2,IL-4、 IFN-γ和TGF-β 含量检测
白细胞介素-2(IL-2)主要由Th1细胞产生,具有促进 T 细胞增殖分化,增强T细胞、NK细胞活性,诱导干扰素生成的作用,是反映机体免疫功能强弱的重要指标,IL-2的产生减少可加速衰老进程。白细胞介素-4(IL-4)主要由Th2细胞产生,其主要功能是促进B细胞活化、增殖和分化,诱导B细胞发生Ig类别转换,产生Ig E或Ig G 类抗体,具有较强的免疫调节功能。γ干扰素(IFN-γ )主要由Th1细胞产生,能激活巨噬细胞,促进其杀伤病原体和肿瘤的能力,增强抗原提呈细胞提呈抗原能力,活化NK细胞,是抗病毒的主要细胞因子。转化生长因子-β(TGF-β)是一种调节性T细胞分泌抑制性细胞因子,属于TH2型细胞因子,具有抑制免疫活性细胞的增殖、抑制淋巴细胞的分化和抑制细胞因子产生的作用。在正常生理条件下,机体的TH1细胞和TH2细胞处于动态平衡,维持机体正常的免疫应答反应,IL-2、IL-4 、IFN-γ和TGF-β 水平的高低是反应机体免疫功能强弱的重要指标。
与正常对照组相比,衰老模型组大鼠血清中IL-2、IL-4、IFN-γ含量显著下降(见图35,#P<0.05);与衰老模型组相比,鸡胚营养组合物组大鼠血清中IL-2、IL-4、IFN-γ含量显著升高(见图35,*P<0.05),鸡胚组大鼠血清中IL-2、IFN-γ含量明显升高(见图35,*P<0.05),营养组合物组血清中IFN-γ含量明显升高(见图35,*P<0.05),IL-2、IL-4含量略升高,但无明显变化,蛋清组血清中IL-2、IL-4、IFN-γ含量无明显变化(见图35,P>0.05)。
与正常对照组相比,衰老模型组大鼠血清中TGF-β含量显著升高(见图35,#P<0.05);与衰老模型组相比,鸡胚营养组合物组大鼠血清中TGF-β含量显著下降(见图35,*P<0.05),鸡胚组大鼠血清中TGF-β含量下降(见图35,*P<0.05),营养组合物组血清中TGF-β含量下降(见图35,*P<0.05),而蛋清组血清中TGF-β含量无明显变化(见图31,P>0.05)。
以上结果提示,鸡胚营养组合物能够增强衰老大鼠的免疫功能。
(九)鸡胚营养组合物对衰老大鼠抗氧化防御系统的影响
机体在代谢过程中,能够不断产生自由基,同时机体又存在自由基清除系统。超氧化物歧化酶 (SOD) 是体内重要的抗氧化酶,能清除超氧阴离子自由基,保护细胞免受损伤,随着衰老,血清中SOD逐渐降低。谷胱甘肽过氧化物酶(GSH-PX)是机体内广泛存在的一种重要的催化过氧化氢分解的酶,在线粒体内含量较高,可以保护细胞膜的结构和功能,使其免受过氧化氢的氧化损伤。随着衰老,血清中GSH-PX逐渐降低。广泛存在于机体中的过氧化氢酶(CAT) 对细胞具有保护作用,能将代谢过程中不间断产生的过氧化氢分解成水和分子氧,减轻和阻断脂质发生过氧化,保护机体免受过氧化物的侵害,延缓细胞的衰老,从而预防自由基引发的疾病。活性氧(ROS) 能与生物膜中不饱和脂肪酸形成过氧化脂质,引起神经元细胞膜损伤,导致机体衰老。铜蓝蛋白 (CER) 具有氧化酶活性,参与铜的转运,催化Fe2+氧化为Fe3+,Fe3+与血液中的转铁蛋白结合,随血液循环运送至机体的各个部位,CER的浓度可以反应机体的炎症情况,随着衰老,机体的免疫功能逐渐下降,炎性因子随之增多,CER浓度也随之升高。金属硫蛋白 (MT) 为胞浆蛋白,富含-SH,具有极强的自由基清除能力和抗脂质过氧化的功能,约为超氧化物歧化酶 (SOD) 的1000倍。MT是应激状态下的保护性因子,但随着年龄增长,血清中的含量却异常表达,这可能是机体应对持续性氧化应激的内源性反应。丙二醛 (MDA) 是机体中细胞膜受到氧自由基的攻击、细胞膜中的多不饱和脂肪酸发生脂质过氧化的产物,可反映机体内脂质过氧化的程度,间接地反映出细胞损伤的程度。
与正常对照组相比,衰老模型组血清中SOD、GSH-PX、CAT含量明显下降(见图36,#P<0.05);与衰老模型组相比,鸡胚营养组合物组血清中SOD、GSH-PX、CAT含量均显著升高(见图36,*P<0.05);与衰老模型组相比,鸡胚组大鼠血清中GSH-PX含量升高(见图36,*P<0.05),营养组合物组血清中SOD、CAT含量升高(见图36,*P<0.05),而蛋清组大鼠血清中SOD、GSH-PX、CAT含量均无明显变化(见图36,P>0.05)。
与正常对照组相比,衰老模型组血清中,ROS、CER、MT、MDA含量明显升高(见图36,#P<0.05);与衰老模型组相比,鸡胚营养组合物组血清中,ROS、CER、MT、MDA含量均显著降低(见图36,*P<0.05);与衰老模型组相比,鸡胚组大鼠血清中ROS、CER、MT、MDA含量降低(见图36,*P<0.05),营养组合物组血清中ROS、CER、MT、MDA含量降低(见图32,*P<0.05),而蛋清组大鼠血清中ROS、CER、MT、MDA含量均无明显变化(见图36,P>0.05),
以上结果表明,鸡胚营养组合物能够提高衰老大鼠抗氧化能力。
(十)鸡胚营养组合物对衰老大鼠肝脏、脑组织脂褐素的影响
脂褐素是过氧化脂质分解产物,主要积聚在大脑皮层和海马部位,其过多会破坏磷脂膜结构,导致线粒体和粗面内质网减少,从而使神经元数目也随之减少。随着年龄增长,机体内脂褐素含量逐渐增多,因其常在老年动物细胞中存在故又称之为老年色素,是目前公认的衰老标志之一。
与正常对照组相比,衰老模型组大鼠肝脏、脑组织中脂褐素含量明显增高,具有显著性差异(见图37,#P<0.05); 与衰老模型组相比,鸡胚营养组合物组、鸡胚组、营养组合物组大鼠肝脏、脑组织中脂褐素含量明显降低,但鸡胚营养组合物组大鼠肝脏、脑组织中脂褐素含量最低(见图37,*P<0.05),表明鸡胚营养组合物组能够较好地抑制脂褐素在肝脏、脑组织的积累,而蛋清组大鼠肝脏、脑组织中脂褐素含量略降低,但无显著性差异(见图37,P<0.05)。
以上结果表明,鸡胚营养组合物对衰老大鼠肝脏、脑组织脂褐素累积有一定的抑制作用。
Claims (9)
1.一种延缓衰老的营养组合物,其特征在于各组分质量比如下:
核苷酸2400~3400、赖氨酸1200~3000、甲硫氨酸800~1600、苯丙氨酸800~1600、苏氨酸200~800、色氨酸200~600、精氨酸1500~3000、组氨酸800~2600、甘氨酸200~600、天冬氨酸300~2600、亮氨酸200~600、异亮氨酸200~800、缬氨酸300~1000、丝氨酸200~600、谷氨酰胺600~2000、谷氨酸600~2600、脯氨酸200~1200、酪氨酸200~800、半胱氨酸200~1200、γ-氨基丁酸100~600、牛磺酸150~600、乳清酸200~600、大豆磷脂1200~3000、蛋黄卵磷脂700~1200、脑磷脂200~600、α-亚麻酸1000~2000、γ-亚麻酸500~1600、肌醇60~260、羟基酪醇200~600、维生素A 0.8~1.2RE、维生素B1 5~10、维生素B2 3~5、维生素B6 3~6、维生素B12 0.006~0.01、维生素C 100~200、维生素D 0.01~0.03、维生素E 10~20a-TE、烟酸20~30NE、叶酸0.4~0.9DFE、铁15~20、锌12~15、锰10~15、铜6~10、硒0.2~0.3、铬0.04~0.06、钾100~800、钙150~1200、镁200~600、胆碱500~1200、L-肉碱0.3~0.6、戊糖200~300、碳酸钠90~120、孵化三日鸡胚胎0.3~20只的冻干粉。
2.根据权利要求1所述延缓衰老的营养组合物,其特征在于各组分质量比如下:核苷酸2400、赖氨酸1200、甲硫氨酸800、苯丙氨酸800、苏氨酸200、色氨酸200、精氨酸1500、组氨酸800、甘氨酸200、天冬氨酸300、亮氨酸200、异亮氨酸200、缬氨酸300、丝氨酸200、谷氨酰胺600、谷氨酸600、脯氨酸200、酪氨酸200、半胱氨酸200、γ-氨基丁酸100、牛磺酸150、乳清酸200、大豆磷脂1200、蛋黄卵磷脂700、脑磷脂200、α-亚麻酸1000、γ-亚麻酸500、肌醇60、羟基酪醇200、维生素A 0.8RE、维生素B1 5、维生素B2 3、维生素B6 3、维生素B12 0.006、维生素C100、维生素D 0.01、维生素E 10a-TE、烟酸20NE、叶酸0.4DFE、铁15、锌12、锰10、铜6、硒0.2、铬0.04、钾100、钙150、镁200、胆碱500、L-肉碱0.3、戊糖200、碳酸钠90、孵化三日鸡胚胎0.3只的冻干粉。
3.一种如权利要求1或2所述延缓衰老的营养组合物,其特征是在制备核基因损伤恢复药物、制备干细胞增殖药物、制备线粒体增殖药物或提高线粒体膜电位药物中的应用。
4.一种如权利要求1或2所述延缓衰老的营养组合物,其特征是在制备提高线粒体膜电位药物中的应用。
5.一种如权利要求1或2所述延缓衰老的营养组合物,其特征是在制备提高骨髓间充质干细胞端粒酶活性及增加细胞自噬小体药物中的应用。
6.一种如权利要求1或2所述延缓衰老的营养组合物,其特征是在制备抗氧化药物中的应用,能够提高过氧化物歧化酶(SOD)等的水平;降低肝脏、脑组织中脂褐素的含量。
7.一种如权利要求1或2所述延缓衰老的营养组合物,其特征是在制备提高免疫功能药物中的应用。
8.一种如权利要求1或2所述延缓衰老的营养组合物,其特征是能够促进神经干细胞增殖;增加脑海马部神经元细胞自噬小体数量、促进了细胞内线粒体的增多及膜电位的增高,DNA的重组修复对脑组织中的基因损伤具有恢复作用;促进肝细胞内自噬小体数量增多;能够显著增多肾小球上皮细胞自噬小体数量;能够提升骨髓间充质干细胞端粒酶的活性;
对肠黏膜干细胞的增殖具有促进作用。
9.一种如权利要求1或2所述延缓衰老的营养组合物,其特征在于能够促进脾脏B细胞自噬小体数量的增加,具有抑制衰老脾脏、胸腺组织的退化作用。
