Abstract

本发明属于饲料添加剂技术领域,具体公开了一种提高水产动物免疫能力的天然饲料添加剂,包括如下按重量份数计的组分:紫锥菊25~35份,黄芩12~25份,连翘12~30份,金银花25~35份,鱼腥草25~35份,大青叶10~20份,苦参10~20份。本发明提供的天然饲料添加剂成分安全有效,制备简单,抑菌效果显著,尤其是针对迟缓爱德华氏菌(Edwardsiella tarda)和舒氏气单胞菌(Aermonas schubertii)抑菌效果显著,同时添加到饲料中,能够显著增强水产动物的免疫能力,应用前景好。

Description

一种提高水产动物免疫能力的天然饲料添加剂及其制备方法
技术领域
本发明属于饲料添加剂技术领域,更具体地,涉及一种提高水产动物免疫能力的天然饲料添加剂及其制备方法。
背景技术
现代水产养殖中存在很多问题,例如:1、水产集约化程度不断发展、水产动物疾病逐步呈现多发性、复杂化、高危害的特点,养殖水环境恶化;2、泼洒施用大量渔用杀虫药、滥用多种抗生素、泼洒外用杀菌消毒药物等;3、造成致病微生物的耐药性、药物残留,甚至造成整个水体生态环境平衡破坏。药物残留增加,经生物链也会影响到人类健康。
迟缓爱德华氏菌(Edwardsiella tarda)和舒氏气单胞菌(Aermonas schubertii)都是水产动物常见病原菌,其中迟缓爱德华氏菌病属于肠杆菌科革兰氏阴性病原菌,是我国水产动物三类疫病。随着水产养殖规模的迅猛发展,疾病的发生率也在增加,需要寻找更好的途径来预防控制疾病。
中草药是一种天然药物,具有高效、残留少、毒副作用小、不易产生耐药性以及性能多样化等优点,并且我国资源丰富。更重要的是中草药提取液对鱼类的免疫系统进行调整,提高水产动的抗应激能力和免疫机能。
面对这些问题,目前各国都在致力于发展有机农业,寻找、开发、改进和替代抗生素等药物,开发绿色新型饲料添加剂,实现无抗养殖,迫在眉睫。
发明内容
本发明的目的在于根据现有技术中的不足,提供了一种提高水产动物免疫能力的天然饲料添加剂及其制备方法。
紫锥菊作为饲料添加剂在畜牧和人类方面有不少研究,但在水产方面报道还很少,另外紫锥菊在国际上有着“免疫”草药的称号。本发明提供了一种天然饲料添加剂,以紫锥菊作为主要成分,不仅能够提高水产动物免疫能力,同时极大地减少抗生素及药物的使用量。
本发明的目的通过以下技术方案实现:
本发明提供了一种提高水产动物免疫能力的天然饲料添加剂,包括如下按重量份数计的组分:紫锥菊25~35份,黄芩12~25份,连翘12~30份,金银花25~35份,鱼腥草25~35 份,大青叶10~20份,苦参 10~20份 。
优选地,包括如下按重量份数计的组分:紫锥菊30份,黄芩18 份,连翘18 份,金银花30 份,鱼腥草30 份,大青叶16 份,苦参 16 份。
本发明同时提供所述的天然饲料添加剂的制备方法,将所述天然饲料添加剂的各原料混合,加水浸泡,煮沸,滤出药液,重复煮沸2~4次,合并滤液,得到提取液。
优选地,将所述提取液浓缩至生药含量为1g/mL,备用。
本发明保护所述的天然饲料添加剂在制备水产动物抑菌剂中的应用,所述抑菌剂为抑制迟缓爱德华氏菌和/或舒氏气单胞菌。
本发明同时保护所述的天然饲料添加剂在制备水产动物免疫剂中的应用
与现有技术相比,本发明具有以下优点及有益效果:
本发明提供的天然饲料添加剂成分安全有效,制备简单,抑菌效果显著,尤其是针对迟缓爱德华氏菌(Edwardsiella tarda)和舒氏气单胞菌(Aermonas schubertii)抑菌效果显著,同时添加到饲料中,能够显著增强水产动物的免疫能力,应用前景好。

具体实施方式
以下结合具体实施例来进一步说明本发明,但实施例并不对本发明做任何形式的限定。除非特别说明,本发明采用的试剂、方法和设备为本技术领域常规试剂、方法和设备。
除非特别说明,本发明所用试剂和材料均为市购。
实施例1: 一种天然饲料添加剂,包括:紫锥菊 30 份,黄芩18 份,连翘 18 份,金银花 30 份,鱼腥草 30 份,大青叶 16 份,苦参 16 份。
对比例1:紫锥菊单方抑菌效果。提取方法与紫锥菊复方相同。
实施例2:
取实施例1中原料混合,得到紫锥菊复方158g,剪碎,放入煎药壶中,加蒸馏水约500mL淹没中草药即可,浸泡30分钟,滤出蒸馏水,重新加干净蒸馏水,大火煮沸后,文火一个半小时,滤出药液,重复三次,合并三次过滤液,浓缩至生药含量为1g/mL,用0.22μL无菌滤膜抽滤除菌后放置在4℃冰箱保存待用。(煎煮法)
实施例3:
取紫锥菊复方158g,剪碎,放于烧杯中,加蒸馏水高于中草药即可,用保鲜膜封口,浸泡30分钟后滤出,然后加60%的乙醇约500mL,保鲜膜封口,浸泡2h,滤出提取液,重复一次,合并两次滤液,浓缩至生药含量为1g/mL,用0.22μL无菌滤膜抽滤除菌后放置在4℃冰箱保存待用。(60%乙醇提取)
实施例4:
取紫锥菊复方158g,用高速粉碎机将其粉碎1min,过60目筛,分别取1g放入15mL离心管中,加10mL蒸馏水,静置72h,期间每隔12小时震荡一次,高速离心机8000*g离心10min,用移液枪取上清液,用0.22μL无菌滤膜抽滤除菌,制得生药含量为1g/mL药液,放置在4℃冰箱保存待用。(水提法)
效果验证:
用纸片扩散法和微量稀释法测定实施例2~4中方法提取得到的紫锥菊复方中草药对迟缓爱德华氏菌和舒氏气单胞菌的体外抑菌作用。
纸片扩散法测抑菌圈大小:
其中,图1~图4中字母相同表示差异不显著;不同表示差异显著(P<0.05);下同。
结果表明,实施例2中煎煮法提取紫锥菊复方抑制效果显著好于60%乙醇和水浸泡提取法(图3和图4可以对比看出);并且紫锥菊复方效果显著好于紫锥菊单方(紫锥菊单方见图1和图2)。实施例2中方法对迟缓爱德华氏菌最小抑菌浓度(MIC)﹤3.125mg/mL,抑菌圈大于14mm,对舒氏气单胞菌最小抑菌浓度(MIC)﹤6.25mg/mL,抑菌圈大于15mm。对提高罗非鱼和杂交鳢的生长、免疫也有显著的作用。
经实验证明,本发明紫锥菊复方中草药可以作为提高水产动物免疫能力的饲料添加剂,减少抗生素和药物的使用。

Claims (6)

1.一种提高水产动物免疫能力的天然饲料添加剂,其特征在于,包括如下按重量份数计的组分:
紫锥菊25~35份,黄芩12~25份,连翘12~30份,金银花25~35份,鱼腥草25~35 份,大青叶10~20份,苦参 10~20份 。
2.根据权利要求1所述的天然饲料添加剂,其特征在于,包括如下按重量份数计的组分:
紫锥菊30份,黄芩18 份,连翘18 份,金银花30 份,鱼腥草30 份,大青叶16 份,苦参16 份。
3.一种权利要求1或2所述的天然饲料添加剂的制备方法,其特征在于,将所述天然饲料添加剂的各原料混合,加水浸泡,煮沸,滤出药液,重复煮沸2~4次,合并滤液,得到提取液。
4.根据权利要求1所述的制备方法,其特征在于,将所述提取液浓缩至生药含量为1g/mL,备用。
5.权利要求1或2所述的天然饲料添加剂在制备水产动物抑菌剂中的应用,其特征在于,所述抑菌剂为抑制迟缓爱德华氏菌和/或舒氏气单胞菌。
6.权利要求1或2所述的天然饲料添加剂在制备水产动物免疫剂中的应用。
