本发明揭示了一种创面修复壳聚糖凝胶,其按照质量包括成份:壳聚糖:1‑3份;醋酸:0.5‑1份;碳酸氢钠:0.5‑1份;聚乙烯醇:8‑12份;甘油:0.1‑1份;杀菌剂:0.01‑0.05份;乙醇:2‑3份;明胶:1‑2份;尿囊素:5‑8份;水:15‑25份。本发明一种创面修复壳聚糖凝胶的有益效果在于:抗菌性更长,双重协同协同杀菌,尿囊素加速愈合,促进愈合,缩短愈合时间。
Description
【技术领域】
本发明属于医用材料技术领域,特别是涉及一种创面修复壳聚糖凝胶。
【背景技术】
于2014年09月10日公告的中国专利第CN104027448A号揭露了一种壳聚糖凝胶,该壳聚糖凝胶包括以下重量百分比的原料:改性壳聚糖0.5%-4.5%,甘油0.5%-15%,明矾0.5%-5%,薄荷醇0.01%-0.13%,羧甲基纤维素钠0.2%-5%,卡波姆.2%-1%。该壳聚糖凝胶还包括重量百分比为1-6%的植物提取物,所述植物提取物包括以下重量份的原料:黄柏30-60份,升麻20-40份,木鳖子10-30份,白鲜皮10-30份,无花果叶0-40份,赤芍10-20份,将上述原料加纯化水提炼,过滤取滤液,浓缩、干燥,得到植物提取物。本发明的改性壳聚糖属高聚阳离子,其活性基团带正电荷,能将细菌吸附粘连在一起,阻止其扩散,吸附细菌后,破坏细菌的细胞膜,致使细胞质内的DNA和RNA流出,从而达到杀灭细菌的效果。阳离子同时还能与细菌表面产生的酸性物质如脂多糖、磷壁质酸、糖醛酸磷壁质、荚膜多糖等相互作用,形成复杂的高分子电解质,从而使细菌的细胞膜功能发生紊乱、死亡。
于2007年05月16日公告的专利第CN1961970A号揭露一种医用壳聚糖敷料,由壳聚糖、聚乙烯醇、明胶、甘油组成,上述各组份按重量份计:壳聚糖:5-10,聚乙烯醇:6-15,明胶:5-10,甘油:10-20,水:45-75,将聚乙烯醇、明胶用水充分溶胀,再水浴溶化,将壳聚糖溶于甘油水溶液中,将两种溶液混在一起充分搅匀,调节pH值至4-7,过滤、去泡,而得医用壳聚糖敷料。本发明满足创面的湿润要求、吸渗而与创面不形成积液、并且在创面溶解后重新成膜,为创面提供理想愈合环境和平台。
以上技术均通过壳聚糖为原料,通过与其他原料合成凝胶,对杀灭细菌、修复创面都是非常有效。
在大致基于上述技术方案上,做出相关的研究,从而达到了提高创面修复速度的效果。
【发明内容】
本发明的主要目的在于提供一种可以提高创面修复速度的壳聚糖凝胶。
本发明通过如下技术方案实现上述目的:一种创面修复壳聚糖凝胶,其按照质量包括成份:壳聚糖:1-3份;醋酸:0.5-1份;碳酸氢钠:0.5-1份;聚乙烯醇:8-12份;甘油:0.1-1份;杀菌剂:0.01-0.05份;乙醇:2-3份;明胶:1-2份;尿囊素:5-8份;水:15-25份。
所述杀菌剂为醋酸氯已定或者苯扎溴铵。
本发明还通过如下技术方案实现上述目的:一种创面修复壳聚糖凝胶制备方法,其特征在于,向15-25份的水中加入1-3份壳聚糖、0.5-1份醋酸、0.5-1份碳酸氢钠、8-12份聚乙烯醇、0.1-1份甘油、2-3份乙醇、1-2份明胶,搅匀,将上述混合液进行密封,通过低温冷藏,形成多孔状结构干胶后,再加入0.01-0.05醋酸氯已定或者苯扎溴铵与5-8份尿囊素,干胶自然化解为凝胶状。
其中,上述低温冷藏条件为:通过-200℃以下液氮低温冷藏5分钟以上。
与现有技术相比,本发明创面修复壳聚糖凝胶的有益效果在于:抗菌性更长,双重协同协同杀菌,尿囊素加速愈合,促进愈合,缩短愈合时间。
【具体实施方式】
本发明为一种创面修复壳聚糖凝胶,其按照质量包括成份:大分子量(20-30万)壳聚糖:1-3份、醋酸:0.5-1份、浓度为4-6%的碳酸氢钠:0.5-1份、聚乙烯醇8-12份、甘油:0.1-1份、杀菌剂:0.01-0.05份、乙醇:2-3份、明胶:1-2份及尿囊素:5-8份,水:15-25份。其中杀菌剂为醋酸氯已定或者苯扎溴铵。
下面结合实施例对本发明的技术方案做进一步的说明。
实施例一:
向试管加入25g纯净水,并向试管中加入分子量为20-30万之间的3g壳聚糖;
向壳聚糖加入1g醋酸,并搅拌均匀,对壳聚糖进行讲解;
再加入1g浓度为5%的碳酸氢钠调解,搅匀并测试;
再向试管中分别加入12g聚乙烯醇、1g甘油、3g乙醇、2g明胶,搅匀;
将该混合液进行密封,通过液氮(-200℃以下)低温冷藏5分钟以上,形成干胶,干胶形成多孔状结构,再对干胶最后加入0.05g醋酸氯已定(或苯扎溴铵)与8g尿囊素,干胶充分吸收醋酸氯已定(或苯扎溴铵)、尿囊素,待干胶自然慢慢化解为凝胶状。
实施例二:
向试管加入15g纯净水,并向试管中加入分子量为20-30万之间的1g壳聚糖;
向壳聚糖加入0.5g醋酸,并搅拌均匀,充分溶解壳聚糖;
再加入0.6g浓度为4%的碳酸氢钠调解,搅匀;
再向试管中分别加入8g聚乙烯醇、0.1g甘油、2g乙醇、1g明胶,搅匀;
将该混合液进行密封,通过液氮(-200℃以下)低温冷藏5分钟以上,形成干胶,干胶形成多孔状结构,再对干胶最后加入0.01g醋酸氯已定(或苯扎溴铵)与5g尿囊素,干胶充分吸收醋酸氯已定(或苯扎溴铵)、尿囊素,待干胶自然慢慢化解为凝胶状。
实施例三:
向试管加入20g纯净水,并向试管中加入分子量为20-30万之间的2g壳聚糖;
向壳聚糖加入0.8g醋酸,并搅拌均匀,充分溶解壳聚糖;
再加入0.8g浓度为6%的碳酸氢钠调解,搅匀;
再向试管中分别加入10g聚乙烯醇、0.5g甘油、2.5g乙醇、1.5g明胶,搅匀;
将该混合液进行密封,通过液氮(-200℃以下)低温冷藏5分钟以上,形成干胶,干胶形成多孔状结构,再对干胶最后加入0.04g醋酸氯已定(或苯扎溴铵)与6g尿囊素,干胶充分吸收醋酸氯已定(或苯扎溴铵)、尿囊素,待干胶自然慢慢化解为凝胶状。
实施例四:
向试管加入50g纯净水,并向试管中加入分子量为20-30万之间的5g壳聚糖;
向壳聚糖加入1.6g醋酸,并搅拌均匀,充分溶解壳聚糖;
再加入1.6g浓度为6%的碳酸氢钠调解,搅匀;
再向试管中分别加入20g聚乙烯醇、1g甘油、5g乙醇、3g明胶,搅匀;
将该混合液进行密封,通过液氮(-200℃以下)低温冷藏5分钟以上,形成干胶,干胶形成多孔状结构,再对干胶最后加入0.03g醋酸氯已定(或苯扎溴铵)与12g尿囊素,干胶充分吸收醋酸氯已定(或苯扎溴铵)、尿囊素,待干胶自然慢慢化解为凝胶状。
实施例五:
向试管加入30g纯净水,并向试管中加入分子量为20-30万之间的2.5g壳聚糖;
向壳聚糖加入1.5g醋酸,并搅拌均匀,充分溶解壳聚糖;
再加入1.5g浓度为5%的碳酸氢钠调解,搅匀并测试;
再向试管中分别加入18g聚乙烯醇、0.5g甘油、5g乙醇、2g明胶,搅匀;
将该混合液进行密封,通过液氮(-200℃以下)低温冷藏5分钟以上,形成干胶,再对干胶最后加入0.03g醋酸氯已定(或苯扎溴铵)与12g尿囊素,待干胶自然慢慢化解为凝胶状。
将上述多个实施例中的样品放置在同一环境下(在样品置于54℃~57℃、相对湿度>75%的恒温箱中14天后)进行测试:

通过在创伤老鼠表皮实验,提供5只老鼠,分别在每只老鼠提供5处大致相同的创伤面,分别对5只老鼠不同的创伤面进行交叉涂抹各个实施例形成的凝胶,通过观察其创伤面的愈合程度,采用标准透明方格胶片来直接测量创面的面积,并计算出12小时、24小时时各实施例的平均愈合率。

因此,本发明的凝胶为弱酸性:ph在5.2-5.9之间,相比其他产品对皮肤刺激更小,利于细胞生长,不利于细菌生长。加入适量的壳聚糖加醋酸氯已定,并与凝胶充分吸收,抗菌性更长,双重协同协同杀菌,尿囊素加速愈合,促进愈合,缩短愈合时间。添加了明胶,抑制疤痕,有效抑制疤痕的形成。
以上所述的仅是本发明的一些实施方式。对于本领域的普通技术人员来说,在不脱离本发明创造构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。

1.一种创面修复壳聚糖凝胶,其特征在于,其按照质量包括成份:

3.一种创面修复壳聚糖凝胶制备方法,其特征在于,向15-25份的水中加入1-3份壳聚糖与0.5-1份醋酸,经过溶解后,再加入0.5-1份浓度为4-6%浓度的碳酸氢钠、8-12份聚乙烯醇、0.1-1份甘油、2-3份乙醇、1-2份明胶,搅匀,将上述混合液进行密封,通过低温冷藏,形成多孔状结构干胶后,再加入0.01-0.05醋酸氯已定或者苯扎溴铵与5-8份尿囊素,干胶自然化解为凝胶状。
4.一种创面修复壳聚糖凝胶制备方法,其特征在于,上述低温冷藏条件为:-200℃以下液氮低温冷藏5分钟以上。
