技术领域
本发明涉及一种中药制剂,具体涉及一种治疗慢性肾衰竭的中药组合物及其制备方法。
背景技术
西医认为“肾衰竭是一种慢性、进行性、不可逆的疾病,预后严重”,尚无有效的药物治疗方法,用透析延长生命,换肾治疗。透析对肾脏没有治疗作用,只是代替肾脏排出一些代谢废物,如肌酐、尿素氮等,根据“用进废退”法则,透析时间长了,肾脏就会萎缩废用。换肾可以治疗肾衰竭,但是,肾源奇缺,我国只有1%的患者可获得肾源。还有排异问题很棘手。
按现代的肾衰竭分型,血肌酐132-177μmol/L为1期(代偿期),血肌酐178-442μmol/L为2期(失代偿期)疗效评定标准是:
临床治愈(简称:痊愈):血肌酐下降到132μmol/L以下,临床症状消失,恢复正常工作和生活。
肾功能显著改善(简称:显效):血肌酐下降30μmol/L以上,临床症状明显改善,可以从事不重的工作和日常生活。
肾功能稳定(简称:有效):血肌酐下降不到30μmol/L,临床症状有所改善。
无效:血肌酐不下降或上升,临床症状无明显改善,或恶化。
中医治疗肾脏病已经有几千年的历史,只是病名与西医不一样,如急性肾小球肾炎,中医名著《金匮要略》中称“风水”、“血尿”,中医辨证又分为风寒、风热、湿毒三大病症,各个证型又有专门的处方,并且记载有病案;肾衰竭属中医的“关格”,“癃闭”,中医辨证分为脾肾气虚、脾肾阳虚、肝肾阴虚、阴阳两虚等,各种证型不同的医家各有不同的治疗方药,疗效也各异。如何寻找一些中草药制剂来治疗该病,并且药物不会残留在人体内,成为了亟待解决的问题
发明内容
为解决上述问题,本发明提供了一种治疗慢性肾衰竭的中药组合物及其制备方法,所得中药制剂对治疗慢性肾衰竭具有较好的治疗作用,安全无毒、疗效显著,不产生抗药性。
为实现上述目的,本发明采取的技术方案为:
一种治疗慢性肾衰竭的中药组合物,由以下重量份的组分制备而成:
奶蓟草15-25份、菟丝子11-15份、牡蛎15-25份、黑枸杞10-15份、番茄提取物10-15份、葛仙米10-15份、田基黄8-10份、橄榄叶5-10份、何首乌10-15份、韭菜籽10-20份。
本发明还提供了一种治疗慢性肾衰竭的中药组合物的制备方法,包括如下步骤:
S1、按上述的配方称取各组分;
S2、将称取的奶蓟草洗净后,进行CO2超临界萃取,流体萃取压力为30-50MPa,萃取温度30-40℃,CO2流体流量为750-850L/h,萃取时间为3-5h,得奶蓟草提取物;;
S3、将称取的菟丝子经超微粉碎后,先用无水乙醇渗漉提取两次,每次24h,过滤,滤渣再用水提取两次,每次24h,合并四次所得的提取液,并浓缩,得菟丝子提取物;
S4、将称取的韭菜籽用闪式提取器处理,所得提取液先用武火加热,沸腾后用文火加热1-1.5h,中空纤维超滤膜过滤;药渣加15-25倍水煎煮两次,中空纤维超滤膜过滤,合并三次滤液,浓缩至1.4g/ml;缓慢加入90-100%乙醇,直至乙醇浓度为50-60%,冷藏8-9小时,2500-4500r/min离心10-20min后倒去上清液,沉淀烘干,得韭菜籽提取物;
S5、将称取的葛仙米按照接种量1.5-1.7%的比例接种微生物发酵剂,保持温度20-30℃,发酵46-58小时后,高温灭菌,喷雾干燥,得葛仙米发酵粉;
S6、将番茄用超微粉碎机粉碎后,加入6倍体积的90%甲醇或乙醇溶液,处理1-3分钟,固液分离后,回收甲醇或乙醇溶液,浓缩,使得提取物呈稠膏状,再用水饱和正丁醇对提取物萃取3次,回收溶剂后,将提取物真空浓缩干燥,得番茄提取物;
S7、将称取的牡蛎、黑枸杞、田基黄、橄榄叶、何首乌粉碎后过与所得的奶蓟草提取物、菟丝子提取物、大韭菜籽提取物、番茄提取物、葛仙米发酵粉混合搅拌均匀后按质量百分比为1-2%加入风味蛋白酶,30-50℃下酶解1-2h,过滤,得酶解液;
S8、将所得的发酵液迅速冷冻后再自然解冻,使提取物中淀粉老化沉淀;收集上清液,剩余部分离心,收集离心液体并合并于上清液中;
S9、将上清液以3-7BV/h的流速通过大孔树脂柱,动态吸附饱和后,用去离子水以5-8BV/h的流速淋洗上述大孔树脂柱至流出液无色,再用体积分数为50%-70%的有机溶剂以8-12BV/h的流速进行洗脱,收集洗脱液,冷冻干燥,得中药组合物。
优选地,所述微生物发酵剂由纳豆菌、酵素菌、谷氨酸棒杆菌按质量比3∶2∶1混合所得。
本发明具有以下有益效果:
所得中药组合物具有有效的补脾益肾,补肾阳的功效,从而可以纠正肾之阴阳,补肾之虚损,补脾健胃,使脾肾调衡至正常的状态,脾健肾强,身体强健,对治疗慢性肾衰竭具有较好的治疗作用,安全无毒、疗效显著,不产生抗药性。

奶蓟草25份、菟丝子15份、牡蛎25份、黑枸杞15份、番茄提取物15份、葛仙米15份、田基黄10份、橄榄叶10份、何首乌15份、韭菜籽20份。

一种治疗慢性肾衰竭的中药组合物,由以下重量份的组分制备而成:
奶蓟草20份、菟丝子13份、牡蛎20份、黑枸杞12.5份、番茄提取物12.5份、葛仙米12.5份、田基黄9份、橄榄叶7.5份、何首乌12.5份、韭菜籽15份。

S1、按实施例1-实施例3所述的配方称取各组分;
S2、将称取的奶蓟草洗净后,进行CO2超临界萃取,流体萃取压力为30-50MPa,萃取温度30-40℃,CO2流体流量为750-850L/h,萃取时间为3-5h,得奶蓟草提取物;;

S1、按权利要求1所述的配方称取各组分;
S2、将称取的奶蓟草洗净后,进行CO2超临界萃取,流体萃取压力为30-50MPa,萃取温度30-40℃,CO2流体流量为750-850L/h,萃取时间为3-5h,得奶蓟草提取物;;
S3、将称取的菟丝子经超微粉碎后,先用无水乙醇渗漉提取两次,每次24h,过滤,滤渣再用水提取两次,每次24h,合并四次所得的提取液,并浓缩,得菟丝子提取物;
S4、将称取的韭菜籽用闪式提取器处理,所得提取液先用武火加热,沸腾后用文火加热1-1.5h,中空纤维超滤膜过滤;药渣加15-25倍水煎煮两次,中空纤维超滤膜过滤,合并三次滤液,浓缩至1.4g/ml;缓慢加入90-100%乙醇,直至乙醇浓度为50-60%,冷藏8-9小时,2500-4500r/min离心10-20min后倒去上清液,沉淀烘干,得韭菜籽提取物;
S5、将称取的葛仙米按照接种量1.5-1.7%的比例接种微生物发酵剂,保持温度20-30℃,发酵46-58小时后,高温灭菌,喷雾干燥,得葛仙米发酵粉;
S6、将番茄用超微粉碎机粉碎后,加入6倍体积的90%甲醇或乙醇溶液,处理1-3分钟,固液分离后,回收甲醇或乙醇溶液,浓缩,使得提取物呈稠膏状,再用水饱和正丁醇对提取物萃取3次,回收溶剂后,将提取物真空浓缩干燥,得番茄提取物;
S7、将称取的牡蛎、黑枸杞、田基黄、橄榄叶、何首乌粉碎后过与所得的奶蓟草提取物、菟丝子提取物、大韭菜籽提取物、番茄提取物、葛仙米发酵粉混合搅拌均匀后按质量百分比为1-2%加入风味蛋白酶,30-50℃下酶解1-2h,过滤,得酶解液;
S8、将所得的发酵液迅速冷冻后再自然解冻,使提取物中淀粉老化沉淀;收集上清液,剩余部分离心,收集离心液体并合并于上清液中;
S9、将上清液以3-7BV/h的流速通过大孔树脂柱,动态吸附饱和后,用去离子水以5-8BV/h的流速淋洗上述大孔树脂柱至流出液无色,再用体积分数为50%-70%的有机溶剂以8-12BV/h的流速进行洗脱,收集洗脱液,冷冻干燥,得中药组合物。
3.根据权利要求2所述的一种治疗慢性肾衰竭的中药组合物的制备方法,其特征在于,所述微生物发酵剂由纳豆菌、酵素菌、谷氨酸棒杆菌按质量比3∶2∶1混合所得。
