本发明涉及药物及营养补充剂领域,尤其涉及一种儿童全功能型营养补充剂。本发明儿童全功能型营养补充剂是由无水葡萄糖、脱脂乳粉、全脂乳粉、乳清蛋白、低聚半乳糖、低聚果糖、水苏糖、乳糖、DHA藻油、固体玉米糖浆、抗坏血酸钠、酪蛋白酸钠、柠檬酸钠、磷脂、L‑抗坏血酸、β‑胡萝卜素、烟酸、核黄素、醋酸维生素A、D‑泛酸钙、叶酸、生育酚、盐酸吡哆醇、盐酸硫胺素、氰钴胺、维生素D3、牛磺酸、碳酸钙、碳酸镁、葡萄糖酸锌、乙二胺四乙酸铁钠和酪蛋白磷酸肽制成的,优选冲剂和咀嚼片剂。本发明全功能型营养补充剂配方合理、作用全面、食用安全,经试验,科学服用本发明儿童全功能型营养补充剂可显著提高儿童免疫力。

本发明涉及药物及营养补充剂领域,尤其涉及一种儿童全功能型营养补充剂。
背景技术
营养补充剂,又称膳食补充剂,是指含有某些特定营养素,补充人体所需的氨基酸、微量元素、维生素、矿物质等的营养剂。营养补充剂作为日常饮食的一种辅助手段,弥补人类正常膳食中可能摄入不足,同时又是人体所必需的营养素,并最终实现调节人体正常生理活动的目的。
不同年龄、性别、机体状况的人群由于其生理条件的差异,所需营养素的种类和数量往往明显有别,因此,针对特定人群研制特定配方组成的营养补充剂已成为医药食品行业的普遍共识。
目前,研究人员已开发出了一些专门针对儿童的营养补充剂产品。例如,专利申请CN201610440876.7“一种用于增强免疫力的儿童冲剂”中公开了一种用于增强免疫力的儿童冲剂,由全脂乳粉、酶解骨粉、乳铁蛋白、果糖、菊粉、异麦芽酮糖、蛋黄粉、速溶椰粉、初乳碱性蛋白、水解蛋黄粉、复配营养素制得。专利申请CN201610012502.5“一种适用于10岁以下儿童的复合维生素片”中公开了一种适用于儿童的复合维生素片,其成分包括:β-胡萝卜素、维生素C、维生素D、维生素B1、核黄素、烟酸、泛酸、维生素B6、生物素、叶酸、维生素B12、钙、铁、锌及辅料。专利申请CN02124007.8“一种全元营养素补充剂”中公开了一种全元营养补充剂,是将酪蛋白,牛磺酸,赖氨酸,多不饱和脂肪酸,膳食纤维,金属离子吸肽,含钙、镁、钾、铁、锌、铜、铬、锰、硒、铜的矿物质及微量元素,复合维生素按比例混合均匀后加水稀释,喷雾干燥,高温灭菌制得的。
本发明在上述产品的基础上,通过反复的配比试验,进一步优化了儿童营养补充剂的配方组成及成分含量配比,最终制得了一种非常适宜于儿童食用的全功能型营养补充剂,本发明营养补充剂配方合理、作用全面、食用安全,科学服用可显著提高儿童免疫力、辅助大脑及身体发育、预防儿童贫血。

我们研究发现,不同类别的营养素之间在吸收和作用发挥过程中存在着互相依存、互相制约、互相促进的关系。如果进行单一种类的营养补充,一方面会破坏体内的营养环境,引起营养失衡,另一方面,其吸收和作用效果也往往并不理想。比如:
(1)据调查,我国缺铁性贫血的儿童中有近一半是补钙过量造成的,原因是钙与铁同属二价离子,过量的钙会阻碍铁的吸收。因此我们的配方在补钙的同时添加了特定量的铁,使其恰好能够抵消所补钙质对正常铁元素的吸收所产生的阻碍作用。
(2)补钙产品中没有维生素D配合,补钙效果差,甚至会引起小儿佝偻病。
(3)单一补铁,没有维生素C的参与,铁的吸收效果很不理想。
(4)单纯使用DHA,补脑效果非常有限。我们将其与卵磷脂、牛磺酸配合使用,使其促进大脑发育的作用明显增强。
进一步地,制得本发明儿童全功能型营养补充剂的原料还包括下述重量份的:大蒜提取物1.05-1.20。
优选地,所述大蒜提取物通过下述方法制得:取新鲜大蒜去皮后榨汁,所得榨汁液经离心后上清液利用截流分子量为500-600的超滤膜进行超滤,所得滤液经冷冻干燥后即得到大蒜提取物。
众所周知,大蒜提取物具有增强免疫力的作用,但采用常规方法制备的大蒜提取物会对儿童胃肠道产生一定的刺激作用,因此以往的儿童营养补充剂中从未添加过大蒜提取物,经试验,我们发现加入冷冻干燥制得的大蒜汁冻干粉可显著提高产品增强免疫力的作用,并且不会产生任何刺激胃肠道的不良反应。
作为一种优选,本发明儿童全功能型营养补充剂是由下述重量份的原料制成的:

此外,本发明还涉及一种儿童全功能型营养补充剂在制备儿童食品中的应用。
此外,本发明还涉及一种儿童全功能型营养补充剂在制备儿童保健品中的应用。
综上,本发明儿童全功能型营养补充剂具有下述特点:
(1)将DHA、磷脂、牛磺酸按照一定配比联合使用,明显提高补脑效果;
(2)维生素B是人体代谢不可或缺的微量营养素,但其比例不当时,吸收率很差,本发明配方中B1、B2、B6采用特定比例,改善了吸收效果;
(3)预防贫血,在配方中使用了两组预防贫血的组方:①铁+VC预防缺铁性贫血;②叶酸+B12预防巨幼细胞性贫血;
(4)维生素C不仅能将三价铁还原成二价铁,而且能与铁螯合形成可溶性小分子络合物,有利于铁吸收,本发明配方中铁与维生素C按照特定配比组合使用,使铁吸收率提高3-6倍;
(5)本发明儿童全功能型营养补充剂作用全面均衡,包含下述作用因子:
①基础因子(乳粉、乳清蛋白组合),奠定必需氨基酸基础;
②补脑因子(DHA、磷脂、牛磺酸组合),优先保证婴幼儿大脑发育;
③代谢因子(4大B族维生素),保证少儿新陈代谢需求,促进生长发育;
④生长因子(CPP+钙、铁、锌),提高生物利用度,满足生长发育需求;
⑤益生因子(低聚半乳糖、低聚果糖、葡萄糖),促益生菌增殖;
⑥免疫因子(VA、VC、钙、铁、β-胡萝卜素、大蒜提取物),提高免疫力;

药效学实验
1.实验目的
观察本发明儿童全功能型营养补充剂对小鼠免疫力的影响。
2.实验材料
2.1受试样品
实施例1-2的儿童全功能型营养补充剂。
配制方法:以水分别配制成浓度为100.0mg/ml的混悬液。
2.2实验动物
BALB/c小鼠,18-22g,雄性。
3.实验分组及受试样品给予时间
实验共设四个组,分别为实施例1组、实施例2组、市售样品组和空白对照组,每组10只小鼠。
各组样品给予时间均为45天。
4.实验方法
4.1原理
当T淋巴细胞受ConA刺激后发生母细胞增殖反应,活细胞特别是增殖细胞中的线粒体水解酶可将MTh(一种淡黄色的唑氮盐)分解为兰紫色结晶,其光密度值能反映细胞的增殖情况。
4.2仪器和材料
RPMI1640细胞培养液、小牛血清、2-巯基乙醇、青霉素、链霉素、刀豆蛋白A(ConA)、盐酸、异丙醇、MTT、Hank,s液、PBS缓冲液(PH7.2-7.4);
纱布或200目筛网、24孔培养板、96孔培养板(平底)、手术器械、二氧化碳培养箱、酶标仪、721分光光度计、超净工作台、高压灭菌器、无菌滤器。
4.3实验步骤
4.3.1试剂配制
完全培养液:RPMI1640细胞培养液过滤除菌,用前加入10%小牛血清,1%谷氨酰胺(200mmol/L),青霉素(100U/ml),链霉素(100μg/L)及5×10-5mol/L的2-巯基乙醇,用无菌的1mol/L的HCl或1mol/L的NaOH调节PH至7.0-7.2,即得到完全培养液。
ConA液:用双蒸水配制成100μg/ml的溶液,过滤除菌,在低温冰箱(-20℃)保存。
无菌Hank,s液:用前以3.5%的无菌NaHCO3调节PH至7.2-7.4。
MTT液:将5mg MTT溶于1ml PH7.2的PBS中,现配现用。
酸性异丙醇溶液:96ml异丙醇中加入4ml 1mol/L的HCl,临用前配制。
4.3.2脾细胞悬液制备
无菌取脾,置于盛有适量无菌Hank,s液平皿中,用镊子轻轻将脾磨碎,制成单个细胞悬液。经200目筛网过滤,或用4层纱布将脾磨碎,用Hank,s液洗2次,每次离心10min(1000r/min)。然后将细胞悬浮于1ml的完全培养液中,用台酚兰染色计数活细胞数(应在95%以上),调整细胞浓度为3×106个/ml。
4.3.3淋巴细胞增殖反应
将每一份脾细胞悬液分两孔加入24孔培养板中,每孔1ml,其中一孔加75μl ConA液(相当于7.5μg/ml),另一孔作为对照,置于5%CO2,37℃孵箱中培养72h。培养结束前4h,每孔轻轻吸去上清液0.7ml,加入0.7ml不含小牛血清的RPMI1640培养液,同时加入MTT(5mg/ml)50μl/孔,继续培养4h。培养结束后,每孔加入1ml酸性异丙醇,吹打混匀,使紫色结晶完全溶解。然后分装到96孔培养板中,每个孔作3个平行孔,用酶标仪以570nm波长测定光密度值。也可将溶解液直接移入2ml比色杯中,721分光光度计上在波长570nm测定OD值。
4.3.4结果判定
用加ConA液孔的光密度值减去不加ConA液孔的光密度值代表淋巴细胞的增殖能力,受试样品组的光密度差值显著高于对照组的光密度差值,可判定该项实验结果阳性。
5.实验结果
动物活体观测指标
一般临床观察:包括但不限于死亡情况、精神状态、行为活动、发病、呼吸、分泌物、粪便以及饮食和饮水情况等。
详细临床观察:包括但不限于精神状态、行为活动、皮肤、被毛、眼睛、耳朵、鼻、腹部、外生殖器、肛门、四肢、足和呼吸。
体重:所有动物在接收后、分组前和濒死前测定体重。
各实验组光密度差值(用加ConA液孔的光密度值减去不加ConA液孔的光密度值,取各组10只小鼠的差值平均值)如下表1所示:

从表1中可以看出,与空白对照组相比,其余各组光密度差值明显增高,提示各组小鼠的淋巴细胞增殖能力均有所增强。相对而言,本发明儿童全功能型营养补充剂(实施例1和2)对小鼠淋巴细胞增殖能力的改善效果要明显优于市售营养补充剂产品。尤其需要指出的是,实施例2配方中包含了大蒜提取物,其提高免疫力效果最为突出,从而证实了加入大蒜汁冻干粉对儿童免疫力的改善大有裨益,同时又避免了常规大蒜提取物对胃肠道强烈的刺激作用。
以上对本发明优选的具体实施方式和实施例作了详细说明,但是本发明并不限于上述实施方式和实施例,在本领域技术人员所具备的知识范围内,还可以在不脱离本发明构思的前提下作出各种变化。
Claims (10)
1.一种儿童全功能型营养补充剂,其特征在于它是由下述重量份的原料制成的:无水葡萄糖 12-30;脱脂乳粉 3.9-5.4;全脂乳粉 6.4-6.7;乳清蛋白 3.2-3.5;低聚半乳糖4.1-5.2;低聚果糖 4.65-5.80;水苏糖 3.03-3.25;乳糖 7.2-7.5;DHA藻油 1.65-1.72;固体玉米糖浆 26-33;抗坏血酸钠 0.037-0.049;酪蛋白酸钠0.65-0.82;柠檬酸钠 0.93-1.49;磷脂 2.52-2.70;L-抗坏血酸 0.16-0.20;β-胡萝卜素 0.85-1.45;烟酸 0.005-0.035;核黄素 0.001-0.006;醋酸维生素 A0.0009-0.0042;D-泛酸钙 0.002-0.075;叶酸0.0002-0.0009;生育酚 0.009-0.024;盐酸吡哆醇 0.001-0.006;盐酸硫胺素 0.001-0.006;氰钴胺 0.000001-0.000012;维生 素D3 0.000001-0.000013;牛磺酸 0.08-0.15;碳酸钙 1.6-2.7;碳酸镁0.26-0.70;葡萄糖酸锌 0.052-0.126;乙二胺四乙酸铁钠 0.105-0.625;酪蛋白磷酸肽 0.23-0.26。
2.如权利要求1所述的儿童全功能型营养补充剂,其中各原料的重量份是:无水葡萄糖20;脱脂乳粉 4.4;全脂乳粉 6.5;乳清蛋白 3.3;低聚半乳糖 4.5;低聚果糖 4.95;水苏糖3.15;乳糖 7.3;DHA藻油 1.69;固体玉米糖浆 31;抗坏血酸钠 0.039;酪蛋白酸钠 0.72;柠檬酸钠 1.21;磷脂 2.60;L-抗坏血酸0.18;β-胡萝卜素 1.05;烟酸 0.015;核黄素0.002;醋酸维生素 A 0.0022;D-泛酸钙 0.015;叶酸 0.0004;生育酚 0.012;盐酸吡哆醇0.002;盐酸硫胺素0.003;氰钴胺 0.000003;维生素 D3 0.000003;牛磺酸 0.12;碳酸钙1.9;碳酸镁 0.46;葡萄糖酸锌 0.082;乙二胺四乙酸铁钠 0.175;酪蛋白磷酸肽 0.24。
3.如权利要求1所述的儿童全功能型营养补充剂,其特征在于每100g所述营养补充剂中含有下述重量的营养成分:
蛋白质 9.5-10.2g;脂肪 5.2-5.4g;碳水化合物 50.5-53.0g;膳食纤维22.5-24.0g;钠 108-112mg;维生素A 1350-1365μg;维生素D 1.8-2.0μg;维生素E 14.85-15.05mg;维生素B1 1.65-1.72mg;维生素B2 1.66-1.70mg;维生素B6 1.66-1.70mg;维生素B12 4.64-4.70μg;维生素C 185.0-185.6mg;烟酸 20.60-20.75mg;叶酸 368-378μg;泛酸 5.45-5.55mg;镁 136-141mg;钙 990-1020mg;铁 18.4-18.7mg;锌 13.79-13.82mg;牛磺酸 119-123mg。
4.如权利要求3所述的儿童全功能型营养补充剂,每100g所述营养补充剂中含有下述重量的营养成分:
蛋白质 9.8g;脂肪 5.3g;碳水化合物 52.0g;膳食纤维 23.5g;钠 110mg;维生素A1360μg;维生素D 1.9μg;维生素E 15.00mg;维生素B1 1.68mg;维生素B2 1.68mg;维生素B61.68mg;维生素B12 4.67μg;维生素C185.4mg;烟酸 20.70mg;叶酸 376μg;泛酸 5.49mg;镁140mg;钙1000mg;铁 18.6mg;锌 13.80mg;牛磺酸 120mg。
5.如权利要求1所述的儿童全功能型营养补充剂,制得所述儿童全功能型营养补充剂的原料还包括下述重量份的:大蒜提取物1.05-1.20。
6.如权利要求5所述的儿童全功能型营养补充剂,所述大蒜提取物通过下述方法制得:取新鲜大蒜去皮后榨汁,所得榨汁液经离心后上清液利用截流分子量为500-600的超滤膜进行超滤,所得滤液经冷冻干燥后即得到大蒜提取物。
7.如权利要求6所述的儿童全功能型营养补充剂,其中各原料的重量份是:无水葡萄糖20;脱脂乳粉 4.4;全脂乳粉 6.5;乳清蛋白 3.3;低聚半乳糖 4.5;低聚果糖 4.95;水苏糖3.15;乳糖 7.3;DHA藻油 1.69;固体玉米糖浆 31;抗坏血酸钠 0.039;酪蛋白酸钠 0.72;柠檬酸钠 1.21;磷脂 2.60;L-抗坏血酸0.18;β-胡萝卜素 1.05;烟酸 0.015;核黄素0.002;醋酸维生素A 0.0022;D-泛酸钙 0.015;叶酸 0.0004;生育酚 0.012;盐酸吡哆醇0.002;盐酸硫胺素0.003;氰钴胺 0.000003;维生素D3 0.000003;牛磺酸 0.12;碳酸钙1.9;碳酸镁 0.46;葡萄糖酸锌 0.082;乙二胺四乙酸铁钠 0.175;酪蛋白磷酸肽 0.24;大蒜提取物 1.15。
8.如权利要求1-7任一项所述的儿童全功能型营养补充剂,其特征在于所述儿童全功能型营养补充剂按本领域常规的制备冲剂或咀嚼片剂的方法,进一步制得本儿童全功能型营养补充剂冲剂或咀嚼片剂。
9.如权利要求1-7任一项所述的儿童全功能型营养补充剂在制备儿童食品中的应用。
10.如权利要求1-7任一项所述的儿童全功能型营养补充剂在制备儿童保健品中的应用。
