背景技术
高血脂症是指身体内的脂肪代谢异常或运转异常导致血浆脂超出正常值,即血脂异常,也就是说血液中的高密度脂蛋白胆固醇(HDL-C)过低或者甘油三酯(TG)和胆固醇(TC)过高而引起的一种致命性的疾病。它是诱发动脉粥样硬化、冠状动脉粥样硬化性心脏病、脑卒中的主要危险因素之一。据统计,全球每年约有1200万人死于心脑血管疾病,而高血脂症是引起心脑血管疾病的主因之一,中国高血脂症患者多达1.6亿人,而且,发病人群正在不断增加,严重危害了我国的人口健康及生活质量。对于高血脂症的防治,目前世界上尚无根治的方法。所以,饮食疗法或保健品对于高血脂症的防治研究显得更加必要和迫切。
高血脂症现代综合治疗的措施包括高血脂症的教育,合理饮食,适当运动,必要的降脂药物和病情监测5个方面。据调查,大部分高血脂症患者需要药物治疗。目前,降低血脂主要以化学药物为主,常用的有:胆固醇生物合成酶抑制剂即HMG-CoA还原酶抑制剂、胆酸螯合剂、烟酸类药物及贝特类药物、胆固醇吸收抑制剂等。这些化学药物均容易引起胃肠反应、肌痛、肝异常等副作用。而中药从整体调节出发,扶正固本,通过抑制脂类吸收和合成,将胆固醇转化为胆汁酸,有效减少人体内胆固醇的含量,达到治疗高血脂症的目的,同时很少引起不良反应,更体现了中医药的特点与长处。对以药食同源物品为原料制备制剂或保健品的研究比较多。

近年来,由于中医药治疗高血脂症的独特优势,并随着在理论与临床研究方面的不断深化,保健食品的研究越发引起了人们的关注与重视。保健食品大多采用中药原料,并根植于中药药膳食疗理论,借助于传统药物学经验和中医药理论指导,因此保健食品的研发具有悠久的历史渊源、丰厚的理论积淀以及美好的发展前景。虽然已有多篇文献报道证实多种中药都具有降低血脂的作用,但将多种中药药材制成复合咀嚼片并作为防治高血脂症的保健食品,目前还未见报道。
咀嚼片为保健食品的主要剂型之一,中国药典2000年版(二部)首次收载了这一剂型。咀嚼片是指于口腔中咀嚼或吮服,使药物溶化后吞服的片剂,是近年来发展起来的一种速效制剂。与普通剂型相比,咀嚼片具有物理、化学稳定性好,生产成本低,崩解时间短,分散状态佳,药物溶出迅速,生物利用度高,服用与携带方便等优点。
本研究基于课题组前期的大量研究工作,将具有降血脂作用且具有宁夏地方特色的三种天然植物:枸杞、葫芦巴及仙人掌,科学配伍研制成长期应用具有良好降低血脂效果的保健食品平脂咀嚼片。
枸杞(Barbary Wolfberry Fruit)是名贵的药材和滋补品,性甘、平,归肝肾经,具有滋补肝肾,益气安神、养肝明目、强身健体的功效。中医很早就有“枸杞养生”的说法。《本草纲目》记载:“枸杞,补肾生精,养肝……明目安神,令人长寿”。枸杞除了具有抗氧化、抗衰老、抗肿瘤、抗脂肪肝、降血糖、增强肌体免疫作用等多种功能外,还具有明显的降低血脂的作用。已有研究证实,枸杞具有明显降低血液中血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)的作用以及降低肝内TC、TG的作用,能够降低血脂。枸杞是宁夏回族自治区的特色药用植物,宁夏有悠久的枸杞种植历史和较为完善的枸杞产品产业化生产开发的技术及设备。
仙人掌(Opuntia dillenii)为仙人掌科仙人掌属植物,在我国分布广,资源丰富,药用历史悠久。其性寒,味苦,具有解毒镇痛、消肿排脓、行气活血之功效。临床上常用于治疗流行性腮腺炎、乳痈、静脉炎、心动过速、胃及十二指肠溃疡、热嗽、手癣、足跟痛、小儿惊风、支气管哮喘、蛇伤以及由于注射引起的硬结、肿块、感染等病症。国外文献报道,其茎有降低血脂和血糖的作用。已有研究表明,仙人掌干粉和浸出液毒副作用小,可降低高血脂症小鼠血清TC,TG含量及AI水平,明显升高HDL-C含量,而发挥降血脂的作用。
葫芦巴为豆科植物胡芦巴(Trigonella foenum-graecum L.)的干燥成熟种子,俗称香豆子、香草子,具有温肾、祛寒、止痛作用,主要用于肾脏虚冷、小腹冷痛、小肠疝气、寒湿脚气等疾病。胡芦巴被作为一种草药,其种子已知具有驱风,滋补和抗糖尿病作用,科学研究指出具有降低血糖之功效,而且在某种剂量上还具有抗溃疡之作用。另有研究亦指出其可溶性膳食纤维部分可抑制碳水化合物的消化与吸收,降低第二型糖尿病之餐后血糖数值之作用。现代科学研究证实胡芦巴还能够减少胆固醇,控制和治疗糖尿病和绝经期综合症。在动物实验中,发现胡芦巴具有降血脂的作用。
发明内容
本发明根据现有技术的不足,提供一种具有降血脂功能的咀嚼片,并提供该咀嚼片的制备方法。所述平脂咀嚼片的制备方法具有极其显著的工艺操作简单、安全、节能并适合大规模生产等优点,能够有效保证枸杞、仙人掌和葫芦巴的营养和功效作用。
本发明通过以下技术方案实现:
一种平脂咀嚼片,由包括如下重量百分比的原料制成:枸杞提取物20%-30%,仙人掌提取物10%-20%,葫芦巴提取物5%-15%,填充剂35%-55%,矫味剂0.5%-4.5%和润滑剂0.5%-1%;
所述枸杞提取物、葫芦巴提取物和仙人掌提取物是将枸杞、葫芦巴和仙人掌分别加入重量为其6-10倍的水或6-10倍的体积浓度为45-80%的乙醇中,回流提取1-3小时,过滤,除去滤渣,减压浓缩滤液,得到浸膏,再干燥、研磨,分别得到枸杞提取物、葫芦巴提取物和仙人掌提取物。
所述填充剂为甘露醇或糊精;所述糊精较佳的为β-环糊精。
所述矫味剂剂为甜菊糖、柠檬酸或薄荷香精中的至少一种。
所述润滑剂为硬脂酸镁或滑石粉。
一种平脂咀嚼片的制备方法,包括如下步骤:
分别将枸杞、仙人掌和葫芦巴进行处理后得到枸杞提取物、仙人掌提取物和葫芦巴提取物。
将枸杞提取物、仙人掌提取物和葫芦巴提取物与矫味剂、填充剂进行均匀混合,得到制软材后,过筛、干燥、整粒后加入润滑剂并搅拌混合后,进行压片,得到平脂咀嚼片。
作为优选方案,所述压片是在单冲压片机中进行的。
有益效果:
对枸杞、葫芦巴、仙人掌等单味药物的性味研究,以及参与复方用于高血脂症的相关研究比较多见,但是仅将这三种中药原料配伍制成防治高血脂症的保健食品,目前国内外尚未见报道。通过中医理论将三味中药按科学比例进行组方,使用其提取物和填充剂、矫味剂共同制粒、压片所得到的平脂片,此平脂片具有缓解高血脂的功效。而压片时使用的单充压片机,性能优良,适应性强,使用方便,易于维修,体积小,重量轻,本机只装一付冲模,物料的充填深度,压片厚度均可调节,能适应制药行业压制各种中、西药制剂的要求和其它行业压制各种类似产品的要求。本研究旨在发扬中医药特色,推动宁夏特色植物的开发和利用,促进宁夏资源优势经济化。特别是应罹患者之需,开发一种疗效确切,安全可靠,服用方便,工艺先进,剂型稳定,质量可控的治疗高血脂症的保健食品,造福于日益增多的广大患者。为促进宁夏特色药食两用、回药植物的进一步开发和利用,并为宁夏将资源优势转化为经济优势提供支持。

一种平脂咀嚼片,由如下重量百分比的原料制成:枸杞提取物26%,仙人掌提取物17%,葫芦巴提取物13%,填充剂42%,矫味剂1%和润滑剂1%;所述枸杞提取物、葫芦巴提取物和仙人掌提取物是将枸杞、葫芦巴和仙人掌分别加入重量为其8倍的水中,回流提取2小时,过滤,除去滤渣,减压浓缩滤液,得到浸膏,再干燥、研磨,分别得到枸杞提取物、葫芦巴提取物和仙人掌提取物。
所述润滑剂为硬脂酸镁,其具有流动性好、可压性高等特点。
所述填充剂为甘露醇。
所述矫味剂为甜菊糖本发明中含有葫芦巴,其具有苦味,通过添加矫味剂使其易于咀嚼吞服。

将枸杞提取物420g、仙人掌提取物280g和葫芦巴提取物187g混合均匀后,加入到含有200g的β-环糊精的200mL纯水中,利用超声机超声5分钟,得到混合溶液,加入290g甘露醇、2g甜菊糖、7g柠檬酸进行均匀混合,制软材,过14目筛制粒,50℃干燥120min。
干燥后用14目筛整粒,加入14g滑石粉混匀,放入压片机中压片,包装,放置室温20℃存储。

干燥后用14目筛整粒,加入0.3g硬脂酸镁混匀,放入压片机中压片,包装,放置室温20℃存储。

取宁夏医科大学实验动物中心小鼠120只,随机分成6组,每组20只。6组小鼠在试验环境下饲喂普通饲料7天后,其中5组小鼠饲喂高脂饲料,1组空白对照组从实验开始至结束都饲喂普通饲料,均3次/日,每3天称重。高脂饲养小鼠饮用含0.2%丙基硫氧嘧啶蒸馏水,空白对照组小鼠饮用蒸馏水,4周后,各组小鼠禁食12小时但不禁水,眼后静脉丛取血0.4ml测血脂浓度,从中挑选出高脂血症小鼠。
将高脂血症小鼠50只,随机分成5组:阴性对照组(蒸馏水)、平脂片低剂量组(2g/kg)、平脂片中剂量组(4g/kg)、平脂片高剂量组(8g/kg)、阳性药物对照组(脂必妥)(0.5g/kg)。连续灌胃给药4周后,对各组小鼠进行称重,眼后静脉丛取血0.4ml,测定血清中胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)浓度。实际数据以±s表示,数据用统计SPSS13.0软件处理,组间用t检验。实验数据如下表:

结果分析:高脂饲料喂养4周后,小鼠的体重、TC、LDL均增加并与普通饲料组小鼠差异有显著性(P<0.01),血清中TG也增加,但程度不显著(P<0.05),见表1.实验结果显示。
高血脂小鼠灌胃3个剂量平脂片4周后,小鼠血清TC、TG均降低,高剂量组与脂必妥组差异有显著性(P<0.01),结果见表2;同时,给药组血清中的LDL能显著降低(P<0.01),中剂量和低剂量的HDL不增加,高剂量HDL略增加,但不显著。实验结果表明,本发明平脂咀嚼片具有降血脂药理作用,能够用于预防和治疗高血脂症。
Claims (10)
1.一种平脂咀嚼片,由包括如下重量百分比的原料制成:枸杞提取物20%-30%,仙人掌提取物10%-20%,葫芦巴提取物5%-15%,填充剂35%-55%,矫味剂0.5%-4.5%和润滑剂0.5%-1%。
2.根据权利要求1所述的平脂咀嚼片,其特征在于,所述枸杞提取物、葫芦巴提取物和仙人掌提取物是将枸杞、葫芦巴和仙人掌分别加入重量为其6-10倍的水或6-10倍的体积浓度为45-80%的乙醇中,回流提取1-3小时,过滤,除去滤渣,减压浓缩滤液,得到浸膏,再干燥、研磨,分别得到枸杞提取物、葫芦巴提取物和仙人掌提取物。
3.根据权利要求1所述的平脂咀嚼片,其特征在于,所述填充剂为甘露醇或糊精。
4.根据权利要求3所述的平脂咀嚼片,其特征在于,所述糊精为β-环糊精。
5.根据权利要求1所述的平脂咀嚼片,其特征在于,所述矫味剂剂为甜菊糖、柠檬酸或薄荷香精中的至少一种。
6.根据权利要求1所述的平脂咀嚼片,其特征在于,所述润滑剂为硬脂酸镁或滑石粉。
7.根据权利要求1所述的平脂咀嚼片,其特征在于,所述平脂咀嚼片由包括如下步骤的方法制备:将枸杞提取物、仙人掌提取物和葫芦巴提取物与矫味剂、填充剂进行均匀混合,得到制软材后,过筛、干燥、整粒后加入润滑剂并搅拌混合后,进行压片,得到平脂咀嚼片。
8.根据权利要求7所述的平脂咀嚼片,其特征在于,所述平脂咀嚼片由如下方法制备:
将枸杞提取物420g、仙人掌提取物280g和葫芦巴提取物187g混合均匀后,加入到含有200g的β-环糊精的200mL纯水中,利用超声机超声5分钟,得到混合溶液,加入290g甘露醇、2g甜菊糖、7g柠檬酸进行均匀混合,制软材,过14目筛制粒,50℃干燥120min;
干燥后用14目筛整粒,加入14g滑石粉混匀,放入压片机中压片,包装,放置室温20℃存储;
所述枸杞提取物、葫芦巴提取物和仙人掌提取物是将枸杞、葫芦巴和仙人掌分别加入重量为其10倍的体积浓度为80%的乙醇中,回流提取3小时,过滤,除去滤渣,减压浓缩滤液,得到浸膏,再干燥、研磨,分别得到枸杞提取物、葫芦巴提取物和仙人掌提取物。
9.制备权利要求1-6任一所述的平脂咀嚼片的方法,包括如下步骤:
将枸杞提取物、仙人掌提取物和葫芦巴提取物与矫味剂、填充剂进行均匀混合,得到制软材后,过筛、干燥、整粒后加入润滑剂并搅拌混合后,进行压片,得到平脂咀嚼片。
10.根据权利要求9所述的平脂咀嚼片的制备方法,包括如下步骤:
将枸杞提取物、仙人掌提取物和葫芦巴提取物混合均匀后,加入到含有β-环糊精的纯水中,利用超声机超声5分钟,得到混合溶液,加入甘露醇、甜菊糖、柠檬酸进行均匀混合,制软材,过14目筛制粒,50℃干燥120min;
干燥后用14目筛整粒,加入滑石粉混匀,放入压片机中压片,包装,放置室温20℃存储;
所述枸杞提取物、葫芦巴提取物和仙人掌提取物是将枸杞、葫芦巴和仙人掌分别加入重量为其10倍的体积浓度为80%的乙醇中,回流提取3小时,过滤,除去滤渣,减压浓缩滤液,得到浸膏,再干燥、研磨,分别得到枸杞提取物、葫芦巴提取物和仙人掌提取物。
