本发明公开了一种改善淡水鱼肠道功能的微生物制剂及其用途,微生物制剂的成分及其重量份为:辣蓼13‑17份、金银花19‑23份、板蓝根12‑16份、山楂10‑14份、穿心莲8‑12份、五倍子5‑8份、野菊花5‑8份、麦冬3‑5份、莲雾3‑5份、海藻3‑5份、鼠李糖乳杆菌0.8‑1.1份、戊糖乳杆菌0.7‑1份、蜡样芽孢杆菌1‑2份。有益效果为:本发明的微生物制剂能够维持淡水鱼肠道正常微生物菌群,改善肠道功能和新陈代谢,提高对饲料营养物质的消化和吸收,改善饲料转化率,提高动物免疫力,增强抗病力,可添加到淡水鱼饲料中,使得饲料除具有中草药和益生菌的双重优点。

技术领域
本发明涉及水产品饲料加工技术领域,特别是涉及一种改善淡水鱼肠道功能的微生物制剂及其用途。
背景技术
肠道微生物在鱼类生长发育过程中起着非常重要的作用,良好的肠道微生物菌群可以帮助鱼类更好地消化吸收营养物质,维持鱼类机体的健康。健康的肠道微生物环境可以调节鱼类的免疫功能,帮助分解吞食后不能消化的纤维素、木质素、几丁质和果胶等。并能阻止有害菌在肠道中的定植和扩散。野生鱼类由于处在微生态系稳定性较高的水环境中,故很少发病;而人工饲养的鱼类处于多变微生态系统中,易给条件病原菌创造引起微生物失调的机会。在此背景下,抗生素类饲料添加剂受到热捧,被广泛应用于饲料业中,在一定程度上促进了水产养殖业的发展,但是抗生素带来的高残留、高污染和抗药性等问题愈发的严重。当前社会,食品安全事故频发,人们在满足温饱的同时也更加注重饮食健康,绿色健康的水产品备受关注。微生物制剂是对传统中药加工技术的改革和创新,具有诸多特点和优势。将此技术引入中药饲料添加剂的研究,可使其科技含量大幅提升,效果更加显著,质量更加稳定,效益更加明显,可以显著提高机体免疫力,增强水产动物抵抗疾病的水平,改善肠道功能,促进消化,对于解决当前水产养殖业中水产动物的多种问题有着重要的应用价值,这对于注重养殖成本的水产养殖业显得尤为重要。
现有技术如授权公告号为CN 101983582 B的中国发明专利,公开了一种防治草鱼肠炎的中药微生态制剂及其制备方法,包括以下重量百分比的组分:枯草芽孢杆菌25~35%、醋酸杆菌30~40%、酪酸梭状芽孢杆菌20~35%。该添加剂绿色安全,能预防和治疗草鱼肠炎病,增强机体免疫力、抗病力、并可提高饲料的转化率,也有利于净化水质,保护和改善生态环境。但是该中药微生态制剂只是针对草鱼肠炎,对改善淡水鱼肠道功能的效果不明显。
发明内容
本发明的目的在于提供一种提高动物机体免疫力,可抑制和阻止有害菌的发生,维持健康的肠道菌群,可以高效改善淡水鱼肠道功能的改善淡水鱼肠道功能的微生物制剂及其用途。
本发明针对上述技术中提到的问题,采取的技术方案为:一种改善淡水鱼肠道功能的微生物制剂,成分及其重量份为:辣蓼13-17份、金银花19-23份、板蓝根12-16份、山楂10-14份、穿心莲8-12份、五倍子5-8份、野菊花5-8份、麦冬3-5份、莲雾3-5份、海藻3-5份、鼠李糖乳杆菌0.8-1.1份、戊糖乳杆菌0.7-1份、蜡样芽孢杆菌1-2份。鼠李糖乳杆菌、戊糖乳杆菌和蜡样芽孢杆菌购于中国工业微生物菌种保藏管理中心;上述微生物制剂中各成分配伍合理,能够维持淡水鱼肠道正常微生物菌群,改善肠道功能和新陈代谢,提高对饲料营养物质的消化和吸收,改善饲料转化率,提高淡水鱼的生长率,降低死亡率,同时能提高动物免疫力,增强抗病力,刺激免疫系统细胞产生不同类型细胞因子,在诱导和调节免疫系统起重要作用。
一种改善淡水鱼肠道功能的微生物制剂的制备方法,制备方法具体包括以下步骤:
超声波浸提:按配方量取辣蓼、金银花、板蓝根、山楂、穿心莲、五倍子、野菊花、麦冬、莲雾、海藻,粉碎后以料液比为1:7-10加水,混合均匀,超声波浸提25-35min,离心,即得浸提液,由于超声波具有独特的机械粉碎作用和空化效应,用于浸提混合药粉时,可以增加物料分子运动频率和速度,提高物料分子浸出速度和有效活性成分提取量,大大缩短浸提时间,减少活性成分的氧化破坏;
发酵液配制:按重量份取浸提液95-100份、蛋白胨0.6-0.8份、蔗糖1-2份、葡萄糖1-2份、磷酸二氢钠0.2-0.3份、氯化钠0.1-0.2份、活性多肽0.001-0.002份,混合搅拌均匀,将pH调至7-7.5,灭菌,即得发酵液,上述活性多肽的氨基酸序列为TDHKLCVCVNCFCCGAPFVVSWKYGVAGFYTHCGPNFCCCIS,该活性多肽具有蛋白质和氨基酸不可比拟的生理特性,其吸收机制优于蛋白质和氨基酸,具有很高的营养价值,富含氨基酸,溶解性好,不需要经过蛋白质转化为氨基酸的步骤,可直接被细菌吸收利用,从而加速了微生物的大量繁殖,增加微生物的发酵效率,大大缩短了发酵时间;

一种改善淡水鱼肠道功能的微生物制剂,成分及其重量份为:辣蓼15份、金银花21份、板蓝根14份、山楂12份、穿心莲10份、五倍子6份、野菊花7份、麦冬4份、莲雾4份、海藻4份、鼠李糖乳杆菌1份、戊糖乳杆菌0.8份、蜡样芽孢杆菌1.5份。
一种改善淡水鱼肠道功能的微生物制剂的制备方法,制备方法具体包括以下步骤:
1)超声波浸提:按配方量取辣蓼、金银花、板蓝根、山楂、穿心莲、五倍子、野菊花、麦冬、莲雾、海藻,经机械粉碎至60目,然后以料液比为1:9加水,搅拌混合均匀,在超声功率为315W、温度为26℃的条件下超声波浸提30min,然后置于离心机中,在转速为4800r/min下离心14min,上清液即为浸提液,由于超声波具有独特的机械粉碎作用和空化效应,用于浸提混合药粉时,可以增加物料分子运动频率和速度,提高物料分子浸出速度和有效活性成分提取量,大大缩短浸提时间,减少活性成分的氧化破坏;

4)后处理:将发酵物中以重量比为1:1.3加入无菌脱脂米糠,充分混合,自然风干至水分为8%,即得微生物制剂,该微生物制剂不仅可以高效改善淡水鱼肠道功能,保持肠道微生态平衡和最佳免疫状态,提高营养物质的消化吸收率,提高动物机体免疫力,而且该微生物制剂具有清热解毒、杀菌抗菌的作用,可抑制和阻止病原大肠菌、梭状芽孢杆菌、沙门氏菌等有害菌的发生,增加肠道益生菌,维持健康的肠道菌群,降低恶臭及有害成分形成,使排出的粪便消除恶臭,在保护和改善养殖环境的同时提高鱼肉质品质。

一种改善淡水鱼肠道功能的微生物制剂,成分及其重量份为:辣蓼17份、金银花20份、板蓝根15份、山楂10份、穿心莲12份、五倍子6份、野菊花8份、麦冬3份、莲雾5份、海藻3.8份、鼠李糖乳杆菌1.1份、戊糖乳杆菌0.9份、蜡样芽孢杆菌1.6份。
一种改善淡水鱼肠道功能的微生物制剂的制备方法为:
按配方量取辣蓼、金银花、板蓝根、山楂、穿心莲、五倍子、野菊花、麦冬、莲雾、海藻,粉碎后以料液比为1:10加水,混合均匀,超声波浸提32min,离心,即得浸提液;按重量份取浸提液100份、蛋白胨0.8份、蔗糖1份、葡萄糖2份、磷酸二氢钠0.3份、氯化钠0.2份、活性多肽0.0018份,混合搅拌均匀,调pH至7.5,灭菌,即得发酵液;将发酵液中接种配方量鼠李糖乳杆菌、戊糖乳杆菌和蜡样芽孢杆菌,在10℃下厌氧发酵12h,将所得发酵物中以重量比为1:1.4加入无菌脱脂米糠,充分混合,自然风干即得微生物制剂。
一种改善淡水鱼肠道功能的微生物制剂的用途,将该微生物制剂添加到淡水鱼饲料中,混合均匀后投喂,添加量为鱼饲料重量的0.1%。
本发明的操作步骤中的常规操作为本领域技术人员所熟知,在此不进行赘述。

Claims (10)
1.一种改善淡水鱼肠道功能的微生物制剂,其特征在于:所述微生物制剂的成分及其重量份为:辣蓼13-17份、金银花19-23份、板蓝根12-16份、山楂10-14份、穿心莲8-12份、五倍子5-8份、野菊花5-8份、麦冬3-5份、莲雾3-5份、海藻3-5份、鼠李糖乳杆菌0.8-1.1份、戊糖乳杆菌0.7-1份、蜡样芽孢杆菌1-2份。
2.根据权利要求1所述的一种改善淡水鱼肠道功能的微生物制剂,其特征在于:所述添加剂的制备方法包括以下步骤:
1)超声波浸提:按配方量取辣蓼、金银花、板蓝根、山楂、穿心莲、五倍子、野菊花、麦冬、莲雾、海藻,粉碎,加水混合均匀,超声波浸提,离心,即得浸提液;
2)发酵液配制:按发酵液配方量取各配料,混合搅拌均匀,调pH,灭菌,即得发酵液;
3)发酵:将发酵液中接种配方量鼠李糖乳杆菌、戊糖乳杆菌和蜡样芽孢杆菌,厌氧发酵,即得发酵物;
4)后处理:将发酵物中加入无菌脱脂米糠,充分混合,自然风干,即得微生物制剂。
3.根据权利要求2所述的一种改善淡水鱼肠道功能的微生物制剂的制备方法,其特征在于:所述步骤2中发酵液的成分及其重量份为:浸提液95-100份、蛋白胨0.6-0.8份、蔗糖1-2份、葡萄糖1-2份、磷酸二氢钠0.2-0.3份、氯化钠0.1-0.2份、活性多肽0.001-0.002份。
4.根据权利要求3所述的一种改善淡水鱼肠道功能的微生物制剂的制备方法,其特征在于:所述步骤2中活性多肽的氨基酸序列为TDHKLCVCVNCFCCGAPFVVSWKYGVAGFYTHCGPNFCCCIS。
5.根据权利要求2所述的一种改善淡水鱼肠道功能的微生物制剂的制备方法,其特征在于:所述步骤2中发酵液的pH为7-7.5。
6.根据权利要求2所述的一种改善淡水鱼肠道功能的微生物制剂的制备方法,其特征在于:所述步骤1中混合药粉与水的料液比为1:7-10。
7.根据权利要求2所述的一种改善淡水鱼肠道功能的微生物制剂的制备方法,其特征在于:所述步骤1中超声波浸提25-35min。
8.根据权利要求2所述的一种改善淡水鱼肠道功能的微生物制剂的制备方法,其特征在于:所述步骤3中厌氧发酵的温度为33-40℃,时间为10-12h。
9.根据权利要求2所述的一种改善淡水鱼肠道功能的微生物制剂的制备方法,其特征在于:所述步骤4中发酵物与无菌脱脂米糠的重量比为1:1.2-1.4。
10.一种改善淡水鱼肠道功能的微生物制剂的用途,其特征在于:所述用途为:将微生物制剂添加到淡水鱼饲料中,混合均匀后投喂,添加量为鱼饲料重量的0.08-0.1%。
