Description

Compound propolis injection and preparation method thereof
Technical Field
The invention belongs to the technical field of veterinary medicines, and particularly relates to a preparation method of a veterinary injection medicine with propolis and acanthopanax as effective components.
Background
With the gradual change of the small-scale workshop-type operation of taking families as a unit to the large-scale centralized operation of the existing livestock breeding, the original breeding mode, the breeding environment and the facilities are difficult to adapt to the new breeding mode. According to the original feeding mode, even a large-scale feeding mode is included, the livestock only pursues the weight and ignores others in the feeding process, so that the livestock lacks necessary nutrition in the growth process, the immunity is low, meanwhile, the adverse stress factors are more, and the pathogenic factor invasion caused by intensive culture can also reduce the immunity of the livestock, so that the disease coping ability of the livestock is greatly reduced, once diseases are generated, the livestock is rapidly overflowed to the whole population, and economic loss is brought to people. However, most farmers do not consider improving the immunity of livestock and treat the livestock until diseases are found, so that the possibility of slaughtering sick pigs is high, and the health of consumers is influenced to a certain extent.
Disclosure of Invention
The invention aims to provide a compound propolis injection and a preparation method thereof, and aims to solve the problems.
In order to achieve the purpose, the technical scheme provided by the invention is as follows:
the effective components of the compound propolis injection comprise propolis and acanthopanax, and the preparation method of the compound propolis injection based on the effective components comprises the following steps:
1) extracting propolis: firstly, selecting unprocessed propolis, storing the propolis in a refrigerator for 24 hours at the temperature of minus 18 ℃, taking out, crushing and sieving the propolis by a 14-mesh sieve, and screening propolis particles to be less than 1.4 mm; adding the crushed particles into 90% ethanol solution, soaking at 20-25 ℃, stirring once every 3 hours, stopping stirring after 12 hours, standing for 3 hours, discharging supernatant for later use, and repeating the step twice; collecting the supernatant liquid of the three times, merging and filtering for 2 times; fourthly, recovering ethanol under reduced pressure at the temperature of 60-80 ℃, wherein the rest is propolis extract, and the weight of the obtained propolis extract is half of that of the unprocessed propolis;
2) extraction of acanthopanax senticosus: firstly, selecting 80g of acanthopanax senticosus powder, adding 800ml of water to soak the acanthopanax senticosus powder for 12 hours, decocting the acanthopanax senticosus powder in water for 2 hours, filtering residues to obtain juice, operating a high-speed centrifuge for 5 minutes, collecting supernate, adding 800ml of water to the filter residues to decoct the filter residues for 2 hours, filtering the residues to obtain juice, and repeating the steps of adding water to decoct the filter residues again and filtering the residues to; ② merging the liquid medicines of the three times, concentrating to 50ml, and standing to room temperature; ③ adding 150ml of ethanol solution with the concentration of 95 percent for three times, preserving for 24 hours at the temperature of 8 ℃, collecting supernatant and filtering; fourthly, recovering the ethanol under reduced pressure, and obtaining acanthopanax extract;
3) preparing a compound propolis injection: mixing 50ml of propylene glycol and 20ml of 95% ethanol solution, homogenizing, heating to 50 ℃, adding 2g of propolis extract, stirring for dissolving, adding 160g of acanthopanax extract equivalent to acanthopanax senticosus powder, stirring for dissolving, adding 1ml of benzyl alcohol, homogenizing, and adjusting the pH value to 6 for later use; ② taking propylene glycol and DMF (dimethyl formamide) to be uniformly mixed according to the volume ratio of 1: 1 as a diluent; thirdly, the solution in the first step is subjected to constant volume by using the diluent in the second step, and the constant volume is 100 ml; and fourthly, filtering the solution obtained in the previous step through a 0.8 mu m filter membrane, filling the solution into 20 ml/ampoule bottles, and sterilizing the solution by using a high-pressure steam sterilization method at 100 ℃ for 30min to obtain the compound propolis injection.
In the invention, in the step of extracting the propolis, the weight of the ethanol added each time is 3-5 times of the weight of the propolis particles.
In the invention, in the acanthopanax root extraction step, an ethanol solution is added by a water extraction and alcohol precipitation method, so that the alcohol content of the solution is 60%, 70% and 80%.
Propolis is a kind of colloidal substance which is obtained by collecting resin from plant tree buds, bark and other parts by bees, mixing with gland secretions of bee tongue glands, wax glands and the like, and transforming through bee processing, is known as purple gold, can be used as a medicine, has the effects of moistening skin, promoting granulation, diminishing inflammation and relieving pain, and can treat diseases such as gastric ulcer, oral ulcer, burn and scald, skin cracking pain and the like. The propolis also can improve organism immunity and disease resistance, and the flavonoids and polysaccharides in the propolis have the effects of regulating organism metabolism and enhancing immunity, and also have antibacterial, antiviral, antifungal, antiprotozoal effects, etc.
The acanthopanax root bark has the functions of expelling wind-damp, strengthening muscles and bones, warm nature, pungent and bitter taste, expelling wind-damp, tonifying liver and kidney, strengthening muscles and bones and inducing diuresis to alleviate edema, is used for treating rheumatism, flaccidity of muscles and bones, retardation of children, asthenia, hypodynamia, edema and beriberi, and has obvious functions of resisting fatigue, radiation, stress and oxygen deficiency, improving the adaptability of a body to temperature change and detoxifying.
Compared with the prior art, the invention has simple process, obvious curative effect and no side effect, improves the disease resistance while improving the immunity by combining the propolis and the acanthopanax senticosus and mutually matching and supplementing the propolis and the acanthopanax senticosus, not only can be independently used as an antiviral disease treatment medicine, but also can be used as an auxiliary medicine in the western medicine treatment process; in the preparation process, the extraction of the effective components of acanthopanax senticosus is realized through three water extractions and one alcohol precipitation in the extraction link of acanthopanax senticosus, and compared with the three water extractions and alcohol precipitation in the prior art, the method reduces twice alcohol precipitation, reduces alcohol waste, accelerates the progress of traditional Chinese medicine extraction, saves time and material consumption, and reduces cost.
Detailed Description
