本发明公开了一种降血糖酵素的生产方法,包括以下步骤:(1)将质量百分比5%的干猫须草和质量百分比10%的玉米须粉碎、混合,加入经过灭菌的质量百分比20%的水倒入发酵罐中,将已经发酵的质量百分比61%的陈皮柑发酵母液倒入发酵罐中,加入质量百分比2%的赤糖或白砂糖,加入质量百分比1%的玉米浆,加入质量百分比1%的益生菌;(2)2天后封闭罐口,并保持发酵罐温度35‑45度,连续不断搅拌5‑7天后,测定PH值并达到2.6‑3.5、乳酸含量:1645.00‑2155.00mg/kg、柠檬酸含量:2450.00‑2950.00mg/kg,结束发酵,转移至储存罐中储存90天,制得降血糖酵素原液。制得的降血糖酵素原液利用猫须草、玉米须、益生菌和新会地区的新鲜柑肉等作为原料制得,具有清热祛湿、排石利尿、降压、利胆、降血糖等作用。

背景技术
广东省江门市新会区生产的陈皮,是国家地理标志保护产品,是新会柑的干果皮,具有很高的药用及经济价值,也是传统的香料及调味品。新会陈皮自宋代以来,就已成为南北贸易的“广货”之一,更被许多华侨带到南洋、中北美洲,由于其经过长时间的储存,可以散发出芳香扑鼻的香味,几百年来被当做名贵的香料在传播、交易。近代研究表明,陈皮具有祛痰、平喘、抗胃溃疡、利胆、降低毛细血管通透性的作用;其挥发油对胃肠道有温和的刺激作用;乙醇提取液可降低血压但对心肌氧耗无影响。
由于陈皮具有独特的药用及食用价值,近年来种植面积逐年扩大,而加工方法也在不断创新。目前市场上有许多产品形态:陈皮茶、陈皮饮料、陈皮果脯、陈皮酒、柑普茶,以及陈皮为原料的各种菜谱和调味品。但往往陈皮是应用和使用了,果肉却没有得到很大的利用价值,而本发明是利用果肉为原料开发的产品,利用新会柑肉所制成的降血糖酵素的生产方法。

本发明的目的是解决现有技术所存在的缺陷,提供一种降血糖酵素的生产方法,该降血糖酵素的生产方法所制得的降血糖酵素原液利用猫须草、玉米须、益生菌和新会地区的新鲜柑肉等作为原料制得,可有效降低大鼠血清尿素氮和肌酐含量,并可改善贫血症状;能够抑制肾小球系膜细胞增殖,对金色葡萄球菌atcc25923、草绿色链球菌556、肺炎链球菌、大肠杆菌等都有一定的抗菌作用。同时具有清热祛湿、排石利尿的功效,有利尿、降压、利胆、降血糖等作用。
本发明所采用的技术方案为:一种降血糖酵素的生产方法,包括以下步骤:
(1)将质量百分比5%的干猫须草和质量百分比10%的玉米须粉碎、混合,加入经过灭菌的质量百分比20%的水倒入发酵罐中,将已经发酵的质量百分比61%的陈皮柑发酵母液倒入发酵罐中,加入质量百分比2%的赤糖或白砂糖,加入质量百分比1%的玉米浆,加入质量百分比1%的益生菌;
(2)2天后封闭罐口,并保持发酵罐温度35-45度,连续不断搅拌5-7天后,测定PH值并达到2.6-3.5、乳酸含量:1645.00-2155.00mg/kg、柠檬酸含量:2450.00-2950.00mg/kg,结束发酵,转移至储存罐中储存90天,制得降血糖酵素原液。
采用了干猫须草、干玉米须为主要原料的发酵物作为制备降血糖药物或者饮品的用途,应用了陈皮柑的发酵液作为发酵原料之一。猫须草为唇形科肾茶属植物,现代研究表明:从猫须草地上部分提取分离出14个黄酮类化合物,2个黄酮醇,8个酚酸类成分;猫须草提取液可有效降低大鼠血清尿素氮和肌酐含量,并可改善贫血症状;用猫须草提取液处理原代培养的大鼠肾小球系细胞2小时,在50ug/ml-250ug/ml浓度范围内,能够抑制肾小球系膜细胞增殖;此外,猫须草提取物抗菌研究发现,对变形杆菌9的抗菌作用最强,最低抑菌溶度小于0.016ug/hl,对金色葡萄球菌atcc25923、草绿色链球菌556、肺炎链球菌、大肠杆菌等都有一定的抗菌作用,同时具有清热祛湿、排石利尿的功效。玉米须最早药用记载见于1476年的《滇南本草》,后《中国药典》1977年收载,为《中华人民共和国卫生部药材标准》1985版收录的常用药材品种,是我国传统的中药材,含糖类、苹果酸、柠檬酸、维生素K、无机盐(钾盐特多),有利尿、降压、利胆、降血糖等作用。陈皮柑味苦、性温,有理气、健脾、化痰功能;柑汁的可溶性固形物为9-12%,有机酸(以柠檬酸计)为0.7-1.2g/100毫升,并含有丰富维生素C。而陈皮柑的囊衣主要成分是纤维素、半纤维素、果胶、葡聚糖,是良好的膳食纤维的原料。上述材料在益生菌(乳酸菌、酵母菌及嗜热链球菌的混合菌株)的作用下,在PH2.6-3.5经过35-45度连续搅拌5-7天,大分子被分解为人体可吸收的小分子,同时产生了许多生物酶(淀粉酶、ß-葡聚糖酶、半纤维素酶、植酸酶、果胶酶、木聚糖酶、肽酶)、维生素、氨基酸、微量元素及小肽。
进一步的, 所述步骤(1)中的益生菌为乳酸菌、酵母菌和嗜热链球菌的混合菌株。
进一步的,所述的步骤(1)中的陈皮柑发酵母液的制备步骤为:将新鲜柑肉放入发酵罐中,并于发酵罐中放质量百分比5%的赤糖或白砂糖,质量百分比2.5%的玉米浆,质量百分比2.5%的益生菌混合菌株,益生菌混合菌株为乳酸菌、酵母菌和嗜热链球菌,质量百分比90%的果肉,调整碳氮比为1:25,进行有氧发酵,得到陈皮柑发酵母液。
进一步的,所述的降血糖酵素原液制备过程中,前期采用好氧发酵,后期采用厌氧发酵。
进一步的,所述的步骤(1)中的各组分装载总容量为发酵罐总容量的80%。
进一步的,所述的发酵罐为带夹套的不锈钢发酵罐,使用夹套的不锈钢发酵罐,是为了保证在低温情况下,维持益生菌发酵的最佳温度。
终止发酵的指标为:PH值2.6-3.5,乳酸含量:1645.00-2155.00mg/kg,柠檬酸含量:2450.00-2950.00mg/kg。
进一步的,所述的步骤(2)中所制得的降血糖酵素原液还需经过过滤和取样,当微生物指标、重金属指标达到饮料国标GB/T31121,其它指标达到行业标准T/CBFIA08003-2016,滤液用管道输入车间,灌装得到降血糖酵素成品。
综上所述,本发明的降血糖酵素的生产方法所制得的降血糖酵素原液利用猫须草、玉米须、益生菌和新会地区的新鲜柑肉等作为原料制得,可有效降低大鼠血清尿素氮和肌酐含量,并可改善贫血症状;能够抑制肾小球系膜细胞增殖,对金色葡萄球菌atcc25923、草绿色链球菌556、肺炎链球菌、大肠杆菌等都有一定的抗菌作用。同时具有清热祛湿、排石利尿的功效,有利尿、降压、利胆、降血糖等作用。
具体实施方式
实施例1
本发明实施例1所描述的降血糖酵素的生产方法,生产前准备,清洗粉碎机,安装水泵并消毒,清洗、消毒发酵罐;具体的制备步骤为:
(1)将质量百分比5%的干猫须草和质量百分比10%的玉米须粉碎、混合,加入经过灭菌的质量百分比20%的水倒入发酵罐中,将已经发酵的质量百分比61%的陈皮柑发酵母液倒入发酵罐中,加入质量百分比2%的赤糖或白砂糖,加入质量百分比1%的玉米浆,加入质量百分比1%的益生菌于发酵罐中部;

Claims (7)
1.一种降血糖酵素的生产方法,其特征在于, 包括以下步骤:
(1)将质量百分比5%的干猫须草和质量百分比10%的玉米须粉碎、混合,加入经过灭菌的质量百分比20%的水倒入发酵罐中,将已经发酵的质量百分比61%的陈皮柑发酵母液倒入发酵罐中,加入质量百分比2%的赤糖或白砂糖,加入质量百分比1%的玉米浆,加入质量百分比1%的益生菌;
(2)2天后封闭罐口,并保持发酵罐温度35-45度,连续不断搅拌5-7天后,测定PH值并达到2.6-3.5、乳酸含量:1645.00-2155.00mg/kg、柠檬酸含量:2450.00-2950.00mg/kg,结束发酵,转移至储存罐中储存90天,制得降血糖酵素原液。
2. 根据权利要求1所述的一种降血糖酵素的生产方法,其特征在于, 所述步骤(1)中的益生菌为乳酸菌、酵母菌和嗜热链球菌的混合菌株。
3.根据权利要求2所述的一种降血糖酵素的生产方法,其特征在于,所述的步骤(1)中的陈皮柑发酵母液的制备步骤为:将新鲜柑肉放入发酵罐中,并于发酵罐中放质量百分比5%的赤糖或白砂糖,质量百分比2.5%的玉米浆,质量百分比2.5%的益生菌混合菌株,益生菌混合菌株为乳酸菌、酵母菌和嗜热链球菌,质量百分比90%的果肉,调整碳氮比为1:25,进行有氧发酵,得到陈皮柑发酵母液。
4.根据权利要求3所述的一种降血糖酵素的生产方法,其特征在于,所述的降血糖酵素原液制备过程中,前期采用好氧发酵,后期采用厌氧发酵。
5.根据权利要求4所述的一种降血糖酵素的生产方法,其特征在于,所述的步骤(1)中的各组分装载总容量为发酵罐总容量的80%。
6.根据权利要求5所述的一种降血糖酵素的生产方法,其特征在于,所述的发酵罐为带夹套的不锈钢发酵罐。
7.根据权利要求6所述的一种降血糖酵素的生产方法,其特征在于,所述的步骤(2)中所制得的降血糖酵素原液还需经过过滤和取样,当微生物指标、重金属指标达到饮料国标GB/T31121,其它指标达到行业标准T/CBFIA08003-2016,滤液用管道输入车间,灌装得到降血糖酵素成品。
