Abstract
本发明提供一种高效催化UDP‑鼠李糖生物合成的基因CsRHMb、其编码蛋白及工程菌,该基因是从茶叶鲜叶中分离并克隆获得的,其编码蛋白在以UDP‑葡萄糖为底物,且在NADPH存在的反应体系中,该蛋白或由工程菌表达的重组蛋白能将UDP‑葡萄糖高效转化生成UDP‑鼠李糖。本发明提供了一种高效安全的UDP‑鼠李糖生物合成技术,利用生物工程方法优化了酶的最适反应条件,为实现UDP‑鼠李糖的商品化生产提供了基础。

技术领域
本发明涉及分子生物学及代谢工程学领域,具体地说,涉及一种高效催化UDP-鼠李糖生物合成的基因CsRHMb及其编码蛋白和应用。
背景技术
茶叶作为一种无酒精饮料广泛被世界各国人民接受和青睐。在茶树中,多酚化合物主要以糖苷化形式存在于茶鲜叶中。而黄酮糖苷是茶树中主要的次生代谢产物,多酚化合物与茶叶品质及人体保健密切相关。其中,黄酮醇及它们的衍生化产物如黄酮醇糖苷是决定茶叶品质和质量的重要组成成分。UDP-鼠李糖作为鼠李糖活化分子的一种重要的化合物,它不仅参与到植物细胞壁合成途径中,而且作为黄酮醇糖苷合成前体物质。例如,芦丁作为一种重要的黄酮醇糖苷,它的含量高低决定了茶汤滋味的好坏。UDP-鼠李糖作为芦丁生物合成所必须的前体物质。UDP-鼠李糖不仅可作为黄酮糖苷生物合成的前体物质,更是多种药用化合物合成必须物质。它在植物细胞壁的形成,化合物合成途径中,科学研究等领域,都有着不可替代的作用。UDP-鼠李糖在植物体内的含量极低,其纯化难度大,效率低等限制该化合物的获得。利用现代化学合成手段也存在诸多问题,例如,合成成本高,过程繁琐,并伴随着污染环境的化学物质产生。因此,采用传统方法很难纯化合成出UDP-鼠李糖。迄今为止,市场上还没有得到商品化的UDP-鼠李糖。而采用生物合成的方式,可以有效避免各种限制因素对UDP-鼠李糖合成的影响。
发明内容
本发明的目的是提供一种高效催化UDP-鼠李糖生物合成的基因CsRHMb及其编码蛋白和应用。
为了实现本发明目的,本发明从茶叶鲜叶中分离并克隆获得一种高效催化UDP-鼠李糖生物合成的基因CsRHMb,基因CsRHMb为:
i)SEQ ID NO:1所示的核苷酸序列;或
ii)SEQ ID NO:1所示的核苷酸序列经取代、缺失和/或增加一个或多个核苷酸且表达相同功能蛋白质的核苷酸序列;或
iii)在严格条件下与SEQ ID NO:1所示序列杂交且表达相同功能蛋白质的核苷酸序列,所述严格条件为在含0.1%SDS的0.1×SSPE或含0.1%SDS的0.1×SSC溶液中,在65℃下杂交,并用该溶液洗膜;或
iv)与i)、ii)或iii)的核苷酸序列具有90%以上同源性且表达相同功能蛋白质的核苷酸序列。
本发明还提供由基因CsRHMb编码的蛋白,其氨基酸序列如SEQ ID NO:2所示。基因CsRHMb编码的蛋白在以UDP-葡萄糖为底物,且在NADPH存在的反应体系中,该蛋白或重组蛋白能将UDP-葡萄糖高效转化生成UDP-鼠李糖。
本发明还提供含有所述基因CsRHMb的表达盒。
本发明还提供含有所述基因CsRHMb或所述表达盒的载体。优选地,出发载体为pMAL-c2X。
本发明还提供含有所述基因CsRHMb、所述表达盒或所述载体的工程菌、转基因细胞系。
在本发明的一个具体实施方式中,将所述基因CsRHMb插入pMAL-c2X载体的多克隆位点中构建得到重组载体pMAL-c2X-CsRHMb,然后将重组载体转化至大肠杆菌Novablue(DE3)中得到携带有基因CsRHMb的重组工程菌。
本发明还提供由所述基因CsRHMb编码的蛋白,或者携带有所述基因CsRHMb的重组工程菌在催化UDP-葡萄糖合成UDP-鼠李糖中的应用。
前述的应用,其是向含有UDP-葡萄糖、NAD和NASPH的反应体系中,加入由所述基因CsRHMb编码的蛋白,或者从所述工程菌中分离纯化的重组蛋白rCsRHMb,通过酶催化反应,生物合成UDP-鼠李糖。
总反应体系为50μL:含有3mM UDP-葡萄糖、3mM NAD和3mM NASPH,反应缓冲液为100mM的NaH2PO4/Na2HPO4混合液(pH8.0-10.0,优选pH:9.5),以及10-20μg由所述基因CsRHMb编码的蛋白,或者所述重组蛋白rCsRHMb。
在本发明的一个优选实施方案中,所述重组蛋白rCsRHMb的氨基酸序列如SEQ IDNO:3所示。
最适酶反应条件为:pH控制在8.0-10.0的NaH2PO4/Na2HPO4缓冲液,反应温度25-45℃。反应时间为1h左右。
本发明进一步提供由所述高效催化UDP-鼠李糖生物合成的CsRHMb基因编码的蛋白在植物细胞壁形成中的应用。
本发明提供了一种催化UDP-鼠李糖生物合成的基因及其编码蛋白和应用,首次从茶树中克隆并验证了催化UDP-鼠李糖生物合成的CsRHMb基因,本发明还提供了含有CsRHMb基因的重组质粒、转基因工程菌和重组蛋白。本发明提供了一种高效安全的UDP-鼠李糖生物合成技术,利用生物工程方法优化了酶的最适反应条件,为实现UDP-鼠李糖的商品化生产提供了基础。UDP-鼠李糖作为鼠李糖活化分子的一种重要化合物,它不仅参与到植物细胞壁合成途径中,而且作为多种药用物质合成前体物质。例如,可以利用UDP-鼠李糖和糖苷配体为底物,在糖基转移酶酶的催化作用下,生物合成各种糖苷化合物。该类化合物多具有药理活性以及生物活性,已在各个科研领域得到了广泛的认可和利用,为植物的生长发育调控,以及药用化合物的生物合成奠定了坚实基础。

具体实施方式
以下实施例用于说明本发明,但不用来限制本发明的范围。若未特别指明,实施例均按照常规实验条件,如Sambrook等分子克隆实验手册(Sambrook J&Russell DW,Molecular Cloning:a Laboratory Manual,2001),或按照制造厂商说明书建议的条件。
实施例1高效催化UDP-鼠李糖生物合成的基因CsRHMb的克隆及表达
一、材料
1、茶树品种:农抗早(Camellia sinensis(L.)O.Kuntze.var.sinensis cultivarNongkangzao),采集茶树鲜叶,迅速用液氮冷冻,储存于-80℃冰箱中备用;
2、大肠杆菌Novablue(DE3)表达宿主菌:购于上海北诺生物科技有限公司;
3、LB培养基:称取10g NaCl,5g酵母提取物,10g胰蛋白胨,加入950mL超纯水搅拌溶解,用1mol/L NaOH调pH至7.0,加水定容至1000mL,高压蒸汽灭菌15min,即获得LB液体培养基,LB固体培养基是在LB液体培养基中加入15g琼脂粉;
4、质量比为40%的葡萄糖溶液:称取40g葡萄糖,加入超纯水溶解搅拌均匀,定容至100mL,110℃灭菌10min;
5、氨苄青霉素母液(Amp+,100mg/mL):称取1g氨苄青霉素Amp,溶于10mL灭菌水,过滤除菌,分装小管,-20℃保存;
6、1mol/L的IPTG(异丙基硫代-β-D-半乳糖苷):称取2.383g IPTG,溶于灭菌超纯水,定容至10mL,过滤除菌,分装-20℃保存;
7、蛋白纯化缓冲液:上柱缓冲液:称取0.37g EDTA,11.67g NaCl,2.42g Tris,0.15g DTT于足量纯水中,搅拌使其充分混匀。用稀盐酸调其pH至7.4,定容至1L,即得上柱缓冲液。洗脱缓冲液:1L上柱缓冲液中加入3.60g麦芽糖,溶解搅拌均匀;
8、1.5%(v/v)三乙胺乙酸流动相缓冲液:用移液管量取15mL色谱级三乙胺溶液于1L容量瓶中,用超纯水定容至1L,最后用色谱级乙酸调pH至7.0。
二、CsRHMb基因的克隆:
1、设计带有表达载体pMAL-c2X的多克隆酶切位点BamHI和PstI的特异引物,引物序列如SEQ ID NO:4和5所示:
正向引物:
CsRHMb-BamHI-F 5’-GAAGGATTTCAGAATTCGGATCCATGGCTCCATATACTCCGAAGAAC-3’
反向引物:
CsRHMb-PstI-R 5’-CCAGTGCCAAGCTTGCCTGCAGTTATATCGCGGAATTTTTCTTGTTG-3’;
2、按照TaKaRa RNAiso试剂盒和RNAiso Plus试剂盒说明书,从茶树品种农抗早鲜叶中提取出总RNA,并利用TaKaRa公司提供的反转录酶反转录为cDNA;
3、以反转录的产物cDNA为模板,用SEQ ID NO:4和5所示的引物进行扩增,扩增程序为:94℃预变性30s;94℃变性10s,62℃退火20s,72℃延伸50s,30个循环;72℃继续延伸10min,得到PCR产物;
4、利用PCR纯化试剂盒纯化PCR产物,并利用Vazyme biotech公司提供的克隆融合酶,最终连接到表达质粒pMal-c2X上,并获得带目的基因的重组表达质粒pMal-c2X-CsRHMb(图7)。
三、CsRHMb基因的原核表达及功能验证
本实施例中所用到的原核表达和及其功能验证技术手段为本领域的普通技术人员常用或完全可以理解的技术手段。
1、将pMal-c2X-CsRHMb重组质粒转化到大肠杆菌Novablue(DE3)表达宿主菌中,接种到100μL LB液体培养基内,37℃180r/min下培养45-60min;取100μL菌液涂布于含100μg/mL Amp+的LB平板上,37℃倒置培养;
2、经过菌落PCR验证,挑取阳性菌落,接种至含2g/L的100mL灭菌的LB液体培养基中,28℃200r/min下震荡培养,直至OD600≈0.6,获得转基因的工程菌;
3、在上述转基因的工程菌中加入IPTG至终浓度为1mmol/L,37℃过夜培养,收集菌体,加入10mL上柱缓冲溶液,充分悬浮菌体,置于-20℃过夜,将菌体置于冰上解冻,待解冻后置于超声破碎仪中以20%功率超声破碎30min,12000rpm离心收集上清液;利用直链淀粉树脂亲和柱纯化重组蛋白(affinity chromatography on an amylase resin,NewEngland Biolabs,MA,USA),利用本领域常用的SDS-PAGE方法检测蛋白表达和纯化效果,结果如图1所示。
从图1可以看出,pMal-c2X-CsRHMb重组质粒转化表达宿主菌Novablue(DE3),诱导表达后,与诱导前(泳道1)相比,该基因在诱导后(泳道2)有重组蛋白的表达,且重组蛋白条带的大小与预测的一致,加上42.5kDa麦芽糖结合蛋白(MBP)重组标签后,在100~110kDa之间有明显的重组蛋白rCsRHMb(SEQ ID NO:3)条带;诱导后菌体经超声破碎离心后,上清中有可溶性重组蛋白(泳道3),可用于进一步纯化分析;上清蛋白经直链淀粉树脂柱纯化后,得到较纯的重组蛋白(泳道4),纯化的蛋白可用于进一步的酶学分析。
实施例2 rCsRHMb重组蛋白的酶学活性检测分析
1、对UDP-葡萄糖底物的酶活检测
总反应体系为50μL,100mM NaH2PO4/Na2HPO4(pH:9.5)缓冲溶液包含3mM UDP-葡萄糖作为反应底物,3mM的NAD和3mM的NASPH作为辅酶、10-20μg纯化后的重组蛋白rCsRHMb。
上述酶反应体系在35℃水浴1h后,加沸水加热10min终止反应,反应均以死酶蛋白作为对照,获得酶反应产物。
2、HPLC检测
酶反应产物经HPLC进行鉴定,所述HPLC检测条件如下:Agela Technologies色谱柱(Venusil XBPCI8(A),5μL,250mm×4.6mm);流速为1mL/min;进样体积为5μL;流动相为含1.5%(v/v)三乙胺乙酸溶液;光谱检测波长扫描范围为260nm。结果如图2所示,UDP-葡萄糖出峰时间为19min,UDP-鼠李糖出峰时间为21min。
3、酶的高效转化率反应条件的检测
为了排除因缓冲液不同而对酶活性产生的影响,分别选取4种具有不同缓冲区间的缓冲液,摸索酶的最适缓冲液和最适反应pH范围。4种缓冲液分别为:100mM柠檬酸/柠檬酸钠(pH5.0-8.0),100mM Tris-HCl缓冲溶液(pH7.0-10.0),100mM NaH2PO4/Na2HPO4(pH7.0-9.5),100mM碳酸钠/碳酸氢钠(pH9.0-11.0)。在总反应体系为50μL中,以3mM的UDP-葡萄糖作为反应底物,3mM的NAD和NASPH作为辅酶。在35℃水浴条件下反应1h,分别对酶反应的最适缓冲液和最适pH进行了检测。最后在浓度100mM pH9.5的NaH2PO4/Na2HPO4缓冲液条件下进行酶反应的最适温度分析。
结果显示,rCsRHMb蛋白酶的最适反应缓冲液为NaH2PO4/Na2HPO4混合液,最适的反应pH范围为8.0-10之间(图3)。在最适缓冲液和最适pH条件下,酶的最适反应温度范围为25-45℃(图4)。因此,本发明通过建立rCsRHMb蛋白酶的最适反应条件,获得了高效转化生成UDP-鼠李糖的反应体系。
4、产物的鉴定
1H-NMR,13C-NMR以及二维谱鉴定:通过在最适条件下进行大体系的酶活反应,利用HPLC制备色谱分离纯化反应产物。分离纯化得到的产物用于核磁分析。结果如表1所示,根据1H-NMR和13C-NMR的数据表明,rCsRHMb重组蛋白催化生成的产物为UDP-鼠李糖。同时将纯化得到的化合物进行二维谱TOCSY和NOESY分析。结果如图5A和B所示,在二维谱上明显的看出有UDP-鼠李糖的H1、H2、H3、H5以及H6的信号存在,因此进一步确定产物为UDP-鼠李糖具有如图5C所示的结构。从以上核磁数据分析表明,CsRHMb基因编码的蛋白具有如图6所示的功能,能在体外通过酶反应催化UDP-葡萄糖生成UDP-鼠李糖。

Claims (10)
1.催化UDP-鼠李糖生物合成的基因CsRHMb,其特征在于,基因CsRHMb为:SEQ ID NO:1所示的核苷酸序列。
2.由权利要求1所述基因CsRHMb编码的蛋白。
3.根据权利要求2所述的蛋白,其特征在于,其氨基酸序列如SEQ ID NO:2所示。
4.含有权利要求1所述基因CsRHMb的表达盒。
5.含有权利要求1所述基因CsRHMb或权利要求4所述表达盒的载体。
6.含有权利要求1所述基因CsRHMb、权利要求4所述表达盒或权利要求5所述载体的工程菌。
7.根据权利要求6所述的工程菌,其特征在于,其是将所述基因CsRHMb插入pMAL-c2X载体的多克隆位点中构建得到重组载体pMAL-c2X-CsRHMb,然后将重组载体转化至大肠杆菌Novablue(DE3)中得到的。
8.权利要求2或3所述蛋白,或者权利要求6或7所述工程菌在催化UDP-葡萄糖合成UDP-鼠李糖中的应用。
9.根据权利要求8所述的应用,其特征在于,其是向含有UDP-葡萄糖、NAD和NADPH的反应体系中,加入权利要求2或3所述蛋白,或者从权利要求6或7所述工程菌中分离纯化的重组蛋白rCsRHMb,通过酶催化反应,生物合成UDP-鼠李糖。
10.根据权利要求9所述的应用,其特征在于,总反应体系为50 μL:含有3 mM UDP-葡萄糖、3 mM NAD和3 mM NADPH,反应缓冲液为100 mM pH8.0-10.0的NaH2PO4/Na2HPO4混合液,以及10-20 μg权利要求2或3所述蛋白,或者所述重组蛋白rCsRHMb;
最适酶反应条件为:pH控制在8.0-10.0的NaH2PO4/Na2HPO4缓冲液,反应温度25-45℃。
