EXAMPLE 5 bacteriostatic experiments
The experimental samples of this example were the morning and evening primrose toothpaste of example 1, the morning and evening primrose toothpaste of example 2, the morning and evening primrose toothpaste of example 3, control 1, control 2, and control 3; the toothpaste formulation of control 1 was identical to the formulation of example 1 except that the children's oral care composition of the present invention was not included; the children's oral care composition in the toothpaste formulation of control 2 consisted of oral probiotics only, and the remaining formulation materials were the same as in example 1; the children's oral care composition in the toothpaste formulation of control 3 consisted of Magnolia bark extract only, with the remainder of the formulation material being in accordance with example 1.
The experimental strains of this example were streptococcus mutans, porphyromonas gingivalis, and fusobacterium nucleatum, and suspensions of each strain were prepared according to the conventional procedures in the art.
Preparation of solid medium (nutrient agar): 5.0g of beef extract, 10g of peptone, 25g of agar powder, 5.0g of sodium chloride and 1000mL of distilled water, heating and dissolving the components except agar, adjusting the pH value to 7.2-7.3 by NaOH, adding the agar powder, heating and boiling to dissolve the agar, subpackaging in a flask, sealing, and sterilizing with 0.1MPa steam for 20 min.
The bacteriostatic test of each experimental sample in this example includes the following specific steps:

In this example, no sample group was added at the same time. Respectively adding 45mL of PBS and 5mL of bacterial suspension into 250mL of triangular flask containing glass beads without adding samples, mixing, respectively mixing 1.0mL of bacterial suspension and PBS before and after 2min of oscillation, and performing gradient dilution to 10-2. Viable bacteria culture counting was carried out according to the method of Disinfection Specification (2002) 2.1.1.2.3.
The bacteriostatic rate was calculated and the results are shown in table 2.

As can be seen from table 2, the control 1 toothpaste without the children's oral care composition of the present invention had the worst antibacterial rate, while the control 2 toothpaste and the control 3 toothpaste, which comprised either only oral probiotics or only magnolia bark extract, had better antibacterial rates than the control 1 toothpaste, but were significantly worse than the examples 1, 2 and 3.
Therefore, the children oral care composition has good inhibition effect on oral harmful bacteria such as streptococcus mutans, porphyromonas gingivalis and fusobacterium nucleatum, and the synergistic inhibition effect of the oral probiotics and the magnolia officinalis extract on the oral harmful bacteria is obviously better than that of the oral probiotics or the magnolia officinalis extract which is singly used.
Example 6 Children use experiment
In this example, 20 children subjects of 9-12 years old who have oral problems such as dental caries and halitosis are selected, and the morning and evening benefit toothpaste prepared in example 2 is used for one month and 2 times a day, and then the test results are tested, and the specific test procedures are as follows:
saliva of 20 subjects without the toothpaste of example 2 for morning and evening and saliva of 20 subjects after 1 month of the toothpaste of example 2 for morning and evening are respectively collected by using sterile test tubes, saliva samples are respectively diluted by sterile PBS appropriately, 0.2mL of the diluted saliva samples are taken and coated on a BHI agar plate, the BHI agar plate is eluted by 2-3 mL of sterile PBS after anaerobic culture at 37 ℃ for 72 hours, and eluents are collected.
