Description

Oral care composition for children and morning and evening benefiting combined toothpaste for children
Technical Field
The invention relates to the technical field of oral care, in particular to oral care for children 9-12 years old, and specifically relates to a children oral care composition and morning and evening combined children toothpaste.
Background
Oral problems in children are a great problem that afflicts children and parents, the most common of which are tooth decay and periodontal disease.
The children 9-12 years old are in the late mixed dentition stage, and at this stage, existing deciduous teeth and permanent teeth exist in the oral cavity, which are the main growth and development periods of the jaw and dental arch of the children and the key period for establishing the permanent teeth. If the deciduous teeth have decayed teeth, the growth and development of permanent teeth can be influenced; the newly erupted permanent teeth are decayed or eroded by bacterial plaque, which can cause tooth defect and even loss, induce gingivitis and the like, and influence the facial development of children. In addition, 9-12 years old is the best period for orthodontic treatment of children, and if the oral cavity is not cleaned in place, bacterial plaque is accumulated to cause periodontal diseases, such as halitosis, which can cause great harm to physical and mental health of the children.
In addition, children have differences in their oral environment in the morning and evening. In the daytime, children frequently eat the food, the sugar content is high, the soil is fertile by bacteria, plaque is easily formed, a large amount of bacteria ingest sugar in saliva, the sugar is decomposed to produce acid, the acidity of the saliva is increased after eating, the saliva is in an acidic state for a long time after eating for many times, and the children are more easily suffered from decayed teeth compared with eating the same amount of food once. At night, children sleep for a long time, the oral cavity is in a static state, the saliva secretion is less, the self-cleaning effect is poor, the bacterial reproduction is facilitated, and the probability of caries is increased. Therefore, the daytime oral environment and the nighttime oral environment of children are greatly different, so that aiming care needs to be carried out aiming at different oral environments in the morning and evening, common oral pathogenic bacteria in the oral cavity of children mainly comprise streptococcus mutans, porphyromonas gingivalis, fusobacterium nucleatum, lactobacillus acidophilus and the like, and the bacteria are the root causes of dental caries, halitosis and gum problems.
There are researchers who use probiotics for oral care, but the use of probiotics in toothpaste is somewhat limited: 1. probiotics generally need to be stored in a low-temperature environment to ensure effective activity, and low-temperature storage of toothpaste in the processes of production, logistics, sale and use is almost impossible. For example, the invention patent application with chinese patent publication No. CN1947698A discloses a probiotic toothpaste using natural mineral salts as a culture medium, wherein sodium chloride with a proper concentration is used to make active probiotics dormant, but the concentration of sodium chloride in the toothpaste system will change with the passage of time, which cannot ensure the stability of the amount of probiotics in the toothpaste; 2. when the probiotics are added into the toothpaste, the stability of the probiotics in the toothpaste is difficult to control, the toothpaste is easy to yellow and has faint yellow aggregates with the diameter of about 1-3 mm which can be seen by naked eyes, and the changes of the toothpaste are mainly caused by metabolic wastes of the probiotics, so that the attractiveness and the use of the toothpaste are seriously influenced.
Most of the children's mothproof oral care products on the market at present contain fluoride. Although fluoride has proved to have a good anticariogenic effect, the body has a high content of fluorine, which has a great influence on teeth and bones, and particularly children have an incomplete swallowing function, and if the fluorine is excessively taken, the children are harmful to their bodies, so that children are not advised to use the fluorine-containing toothpaste.
There are many children's toothpaste on the market, but mostly in the age bracket of 2 ~ 12 years old, a small number is 6 ~ 12 years old children's toothpaste, but all has a problem, and the age bracket scope is too wide, does not aim at the nursing, consequently, research and development neotype nursing product to children's oral cavity problem not only accords with consumers's in general demand, has wide market space moreover.

Compared with the prior art, the invention has the following beneficial effects:
1. the children oral care composition contains two active ingredients, namely oral cavity probiotics and a magnolia bark extract, wherein the magnolia bark extract is obtained by extraction through a supercritical carbon dioxide extraction technology, a reasonable extraction process ensures that the active ingredients in the magnolia bark extract are extracted to the maximum extent, the magnolia bark extract is perfectly matched with the oral cavity probiotics, the synergistic effect is realized, the growth of cariogenic bacteria and other oral cavity pathogenic bacteria is inhibited, the types and the capability of the bacteria inhibition are obviously superior to those of a single active substance, and therefore, the efficacies of preventing decayed teeth, preventing and treating periodontitis, preventing and treating ulcer, refreshing breath, keeping the oral cavity healthy, strengthening gums and firming teeth superior to those of the single active substance can be provided.
2. In the toothpaste for the early-benefiting children, xylitol and a calcium-containing compound are combined for use, so that the pH value of an oral cavity can be relieved, and the acid resistance of enamel can be improved. The enamel and dentin of the milk tooth are thin, the mineralization degree is low, the acid resistance is weak, and the milk tooth enamel is easy to decalcify under the action of a decalcifying agent and is also easy to remineralize under the action of a remineralizing agent compared with the permanent tooth enamel. In the daytime, children frequently eat the feed, the acidity of saliva is increased after eating the feed, the saliva is in an acidic state for a long time after eating the feed for many times, xylitol enters bacterial cells to form xylitol-5-phosphate and is accumulated in the cells, the xylitol-5-phosphate is dephosphorylated, and then the xylitol is discharged out of the cells, so that an energy-consuming ineffective cyclic process is formed, the growth and acid production of the streptococcus mutans are inhibited, and the quantity of the streptococcus mutans can be reduced by the xylitol. Xylitol is not beneficial to the increment of acid-resistant bacteria, plays roles of promoting remineralization and inhibiting demineralization, and does not form extracellular polysaccharide in the metabolic process of xylitol, thereby reducing the formation of bacterial plaque. Xylitol and calcium-containing compound can inhibit microbial fermentation to produce acid, improve oral cavity pH value, and supplement calcium ion, so as to enhance remineralization rate and improve enamel acid resistance.
3. The toothpaste for children in the evening adopts the oligosaccharide, because the oral cavity of the children is in a static state at night, the saliva secretion is less, the self-cleaning effect is poor, the bacteria reproduction is facilitated, and the oligosaccharide can promote the growth of beneficial bacteria, inhibit the reproduction of harmful bacteria, maintain the microecological balance of the oral cavity, maintain the pH value of the oral cavity at a normal level, and achieve the effect of preventing decayed teeth.
4. In the morning and evening benefiting combined children toothpaste, the amino acid is added in a composition form, so that the stability of the amino acid in the toothpaste quality guarantee period can be improved, and the vitamins are also added in the toothpaste in a compound form, so that the stability of the vitamins in the toothpaste quality guarantee period is also improved.
5. The early-benefiting and late-benefiting combined children toothpaste bacteria adopt the mild surfactant, and the mild surfactant is a mixture consisting of any one of sodium lauroyl sarcosinate, sodium lauroyl glutamate, cocoyl propyl betaine, N-methyl cocoyl sodium taurate or poloxamer and sodium lauryl sulfate according to the weight ratio of 1 (0.1-3), so that the matching can not only clean the oral cavity and remove oil stains and stains adhered to the teeth, but also does not stimulate the viscosity of the oral cavity, and simultaneously improves the explosive force of essence, improves the mouthfeel, can also generate fine foam, is easy to wash clean with clear water and does not remain; for children who do not like to rinse for many times when brushing teeth, the frequency of rinsing can be reduced.
6. Aiming at the condition that children eat frequently and have high sialic acid property and are beneficial to acid-resistant bacterium proliferation, the toothpaste for the early-benefiting children inhibits the bacterial reproduction of bacterial plaque by utilizing xylitol and calcium-containing compounds, promotes remineralization and inhibits demineralization, thereby resisting bacterial erosion and firming teeth; aiming at the condition that saliva of the oral cavity of the children is less and bacteria reproduction is facilitated, the toothpaste for the children is beneficial to promoting the growth of beneficial bacteria, inhibiting the reproduction of harmful bacteria, maintaining the microecological balance of the oral cavity and keeping the pH value of the oral cavity at a normal level, thereby achieving the anti-moth effect; the toothpaste for the early-benefiting children and the toothpaste for the late-benefiting children are used in a matching way, so that the decayed teeth of the deciduous teeth of the children can be effectively prevented, and the teeth of the children are protected to be healthy.

TABLE 1 toothpaste stability test results
Figure BDA0001218714930000111
As can be seen from the results in table 1, the children morning and evening benefit toothpaste of examples 1 to 3 still has stable performance after being placed at a high temperature of 45 ℃ for 3 months, which indicates that the toothpaste of the present invention has good stability and the paste does not yellow.
EXAMPLE 5 bacteriostatic experiments
The experimental samples of this example were the morning and evening primrose toothpaste of example 1, the morning and evening primrose toothpaste of example 2, the morning and evening primrose toothpaste of example 3, control 1, control 2, and control 3; the toothpaste formulation of control 1 was identical to the formulation of example 1 except that the children's oral care composition of the present invention was not included; the children's oral care composition in the toothpaste formulation of control 2 consisted of oral probiotics only, and the remaining formulation materials were the same as in example 1; the children's oral care composition in the toothpaste formulation of control 3 consisted of Magnolia bark extract only, with the remainder of the formulation material being in accordance with example 1.
The experimental strains of this example were streptococcus mutans, porphyromonas gingivalis, and fusobacterium nucleatum, and suspensions of each strain were prepared according to the conventional procedures in the art.
Preparation of solid medium (nutrient agar): 5.0g of beef extract, 10g of peptone, 25g of agar powder, 5.0g of sodium chloride and 1000mL of distilled water, heating and dissolving the components except agar, adjusting the pH value to 7.2-7.3 by NaOH, adding the agar powder, heating and boiling to dissolve the agar, subpackaging in a flask, sealing, and sterilizing with 0.1MPa steam for 20 min.
The bacteriostatic test of each experimental sample in this example includes the following specific steps:
step 1
To a 250mL Erlenmeyer flask containing glass beads, 45mL of PBS (phosphate buffered saline) and 5mL of each of the bacterial suspensions were added to give a concentration of 1X 10 in PBS4CFU/mL~5×104CFU/mL;
Step 2
Weighing 0.50g of experimental toothpaste sample, placing into the Erlenmeyer flask containing PBS and bacterial suspension in step 1, shaking for 2min, sucking 1mL, and diluting to 10% with PBS as gradient-2As a sample solution before oscillation;
weighing 0.50g of experimental toothpaste sample, putting the sample into the Erlenmeyer flask containing PBS and bacterial suspension in the step 1, shaking for 2h at the temperature of 20-25 ℃ at the speed of 300r/min, sucking 1mL of experimental toothpaste, and diluting the sample to 10 degrees by using PBS as a gradient-2As a post-oscillation sample solution;
step 3
Agar pour-on plates (viable cell culture counting according to the protocol specified in Disinfection Specification (2002)): respectively sucking 1mL of sample liquid before and after oscillation, placing the sample liquid in a sterilization plate, inoculating two plates to each sample liquid, transferring the sample liquid into the plate, injecting about 15mL of nutrient agar culture medium cooled to 46 ℃ into the plate in time, and rotating the plate to uniformly mix the nutrient agar culture medium and the plate. After the agar is solidified, the plate is turned over, placed at the constant temperature of 37 ℃ for culturing for 48 hours, taken out and the colony number is recorded.
In this example, no sample group was added at the same time. Respectively adding 45mL of PBS and 5mL of bacterial suspension into 250mL of triangular flask containing glass beads without adding samples, mixing, respectively mixing 1.0mL of bacterial suspension and PBS before and after 2min of oscillation, and performing gradient dilution to 10-2. Viable bacteria culture counting was carried out according to the method of Disinfection Specification (2002) 2.1.1.2.3.
The bacteriostatic rate was calculated and the results are shown in table 2.
TABLE 2 toothpaste bacteriostasis rate results
Figure BDA0001218714930000121
Figure BDA0001218714930000131
As can be seen from table 2, the control 1 toothpaste without the children's oral care composition of the present invention had the worst antibacterial rate, while the control 2 toothpaste and the control 3 toothpaste, which comprised either only oral probiotics or only magnolia bark extract, had better antibacterial rates than the control 1 toothpaste, but were significantly worse than the examples 1, 2 and 3.
Therefore, the children oral care composition has good inhibition effect on oral harmful bacteria such as streptococcus mutans, porphyromonas gingivalis and fusobacterium nucleatum, and the synergistic inhibition effect of the oral probiotics and the magnolia officinalis extract on the oral harmful bacteria is obviously better than that of the oral probiotics or the magnolia officinalis extract which is singly used.
Example 6 Children use experiment
In this example, 20 children subjects of 9-12 years old who have oral problems such as dental caries and halitosis are selected, and the morning and evening benefit toothpaste prepared in example 2 is used for one month and 2 times a day, and then the test results are tested, and the specific test procedures are as follows:
saliva of 20 subjects without the toothpaste of example 2 for morning and evening and saliva of 20 subjects after 1 month of the toothpaste of example 2 for morning and evening are respectively collected by using sterile test tubes, saliva samples are respectively diluted by sterile PBS appropriately, 0.2mL of the diluted saliva samples are taken and coated on a BHI agar plate, the BHI agar plate is eluted by 2-3 mL of sterile PBS after anaerobic culture at 37 ℃ for 72 hours, and eluents are collected.
Blank groups in this example refer to: the eluent obtained by the above operation of the saliva without the morning and evening toothpaste of example 2 was diluted 10 times to 1X 10 with sterile PBS solution6~5×106Concentration ofObtaining bacterial suspension, sucking 1.0mL of bacterial suspension, properly diluting, carrying out anaerobic culture for 72h, and then counting the number of colonies.
The experimental groups of this example refer to: the eluent obtained by the operation of the saliva after 1 month of the toothpaste for morning and evening of example 2 is diluted to 1X 10 times by sterile PBS solution with 10 times6~5×106Obtaining bacterial suspension at the concentration, sucking 1.0mL of bacterial suspension, properly diluting, carrying out anaerobic culture for 72h, and then calculating the number of colonies.
The sterilization rate was calculated according to the following formula:
Figure BDA0001218714930000132
the number of colonies in the blank group of 20 subjects was calculated to have an average value of 6.2X 106CFU/mL, the number of colonies in the experimental group of 20 subjects was calculated as the average<10CFU/mL, and then calculating according to the formula to obtain the bacteriostasis rate of 100%.
The results showed that the amount of anaerobic bacteria in the saliva of children was significantly suppressed after one month using the morning and evening primrose toothpaste containing the children's oral care composition of the present invention, thus demonstrating that the morning and evening primrose toothpaste containing the children's oral care composition of the present invention is indeed effective in preventing dental caries, preventing and treating periodontitis, ulcerating, freshening breath, health of the oral cavity, strengthening gums, and strengthening teeth.
